To investigate the genetic characteristics of phosphoprotein (P) and matrix protein (M) genes of variable rabies virus (RV) prevalent in Brazil, the authors genetically characterized the P and M genes from 30 Brazilian RV field isolates. Phylogenetic analysis based on the P and M genes revealed the presence of six RV variants that consisted primarily of three insectivorous bats, the vampire bat, dog and fox in Brazil. Specific amino acid substitutions corresponding to these phylogenetic lineages were observed, with ASP(42) and GlU(62) in the P protein found to be characteristic of Brazilian chiroptera- and carnivora-related RVs, respectively. Amino acid sequence motifs predicted to associate with a viral function in the P and M proteins were conserved among Brazilian RV variants.

Hybrid organic - inorganic nanocomposites doped with Fe-II and Fe-III ions and exhibiting interesting magnetic properties have been obtained by the sol - gel process. The hybrid matrix of these ormosils ( organically modified silicates), classed as di-ureasils and termed U( 2000), is composed of poly( oxyethylene) chains of variable length grafted to siloxane groups by means of urea crosslinkages. Iron perchlorate and iron nitrate were incorporated in the diureasil matrices, leading to compositions within the range 80 greater than or equal to n greater than or equal to 10, n being the molar ratio of ether-type O atoms per cation. The structure of the doped diureasils was investigated by small-angle X-ray scattering (SAXS). For Fe-II-doped samples, SAXS results suggest the existence of a two-level hierarchical structure. The primary level is composed of spatially correlated siloxane clusters embedded in the polymeric matrix and the secondary, coarser level consists of domains where the siloxane clusters are segregated. The structure of Fe-III-doped hybrids is different, revealing the existence of iron oxide based nanoclusters, identified as ferrihydrite by wide-angle X-ray diffraction, dispersed in the hybrid matrix. The magnetic susceptibility of these materials was determined by zero-field-cooling and field-cooling procedures as functions of both temperature and field. The different magnetic features between Fe-II- and Fe-III-doped samples are consistent with the structural differences revealed by SAXS. While Fe-II-doped composites exhibit a paramagnetic Curie-type behaviour, hybrids containing Fe-III ions show thermal and field irreversibilities.

The effects of prenatal exposure of rat pups to 0.08 mg/kg deltamethrin (DTM) on physical, reflex and behavioral developmental parameters, on forced swimming and open-field behaviors, and on striatal monoamine levels at 60 days of age were observed. Maternal and offspring body weight, physical and reflex development were unaffected by the exposure to the pesticide. At 21 days of age, open-field locomotion frequency and immobility duration of male and female offspring were not different between control and exposed animals. However, male rearing frequency was increased in experimental animals. A decreased immobility latency to float and in general activity after the swimming test in male offspring was observed at adult age; no interference was detected in the float duration during the swimming test. In addition, these animals presented higher striatal 3,4-dihydroxyphenylacetic acid (DOPAC) levels without modification in dopamine (DA) levels and an increased DOPAC/DA ratio. These data indicate a higher activity of the dopaminergic system in these animals. Noradrenaline (NA) levels were increased, while MHPG levels were not detectable in the system studied. Serotonin (5-HT) and 5-hydroxyindolacetic acid (5-HIAA) levels, as well as the homovanillic acid (HVA)/DA ratio, were not modified by the exposure to the pesticide. No changes were observed in swimming and open-field behaviors nor were there any changes in striatal monoamines or their metabolites in the female experimental group. In relation to the pesticide formula, the present data showing that prenatal exposure to DTM alters latency to float and the activity of striatal dopaminergic system might reflect a persistent effect of the pesticide on animal motor activity, mainly in males. on the other hand, the decrease in general activity observed in experimental male rats suggests higher levels of emotionality induced by previous exposure to the swimming behavior test in relation to control animals. The magnetic-field and confinement effects on the Land, factor in AlxGa1-xAs parabolic quantum wells under magnetic fields applied parallel or perpendicular to the growth direction are theoretically studied. Calculations are performed in the limit of low temperatures and low electron density in the heterostructure. The g factor is obtained by taking into account the effects of non-parabolicity and anisotropy of the conduction band through the 2 x 2 Ogg-McCombe Hamiltonian, and by including the cubic Dresselhaus spin-orbit term. A simple formula describing the magnetic-field dependence of the effective Land, factor is analytically derived by using the Rayleigh-Schrodinger perturbation theory, and it is found in good agreement with previous experimental studies devoted to understand the behavior of the g factor, as a function of an applied magnetic field, in semiconductor heterostructures. Present numerical results for the effective Land, factor are shown as functions of the quantum-well parameters and magnetic-field strength, and compared with available experimental measurements.

Many studies have assessed the process of forest degradation in the Brazilian Amazon using remote sensing approaches to estimate the extent and impact by selective logging and forest fires on tropical rain forest. However, only a few have estimated the combined impacts of those anthropogenic activities. We conducted a detailed analysis of selective logging and forest fire impacts on natural forests in the southern Brazilian Amazon state of Mato Grosso, one of the key logging centers in the country. To achieve this goal a 13-year series of annual Landsat images (1992-2004) was used to test different remote sensing techniques for measuring the extent of selective logging and forest fires, and to estimate their impact and interaction with other land use types occurring in the study region. Forest canopy regeneration following these disturbances was also assessed. Field measurements and visual observations were conducted to validate remote sensing techniques. Our results indicated that the Modified Soil Adjusted Vegetation Index aerosol free (MSAVI(af)) is a reliable estimator of fractional coverage under both clear sky and under smoky conditions in this study region. During the period of analysis, selective logging was responsible for disturbing the largest proportion (31%) of natural forest in the study area, immediately followed by deforestation (29%). Altogether, forest disturbances by selective logging and forest fires affected approximately 40% of the study site area. Once disturbed by selective logging activities, forests became more susceptible to fire in the study site. However, our results showed that fires may also occur in undisturbed forests. This indicates that there are further factors that may increase forest fire susceptibility in the study area. Those factors need to be better understood. Although selective logging affected the largest amount of natural forest in the study period, 35% and 28% of the observed losses of forest canopy cover were due to forest fire and selective logging combined and to forest fire only, respectively. Moreover, forest areas degraded by selective logging and forest fire is an addition to outright deforestation estimates and has yet to be accounted for by land use and land cover change assessments in tropical regions. Assuming that this observed trend of land use and land cover conversion continues, we predict that there will be no undisturbed forests remaining by 2011 in this study site. Finally, we estimated that 70% of the total forest area disturbed by logging and fire had sufficiently recovered to become undetectable using satellite data in 2004. (C) 2010 Elsevier B.V. An experiment was conducted to study the effects of liming and drying method on Ca nutrition, fungus infection and aflatoxin production potential on peanut (Arachis hypogea) grains. Peanut cv. Botutatu was grown in the absence or presence of liming to raise the base saturation of the soil from 20 to 56%. Calcium contents of the soil were increased from 5.5 to 14.6 mmol((c))kg-1 and pH from 4.2 to 4.9. After harvest, plants and pods were dried in (1) shade, (2) field down to 100 g water kg-1 (3) field down to 250 g water kg-1 and transferred to a forced-air oven at 30°C, (4) field down to 360 g water kg-1 and transferred to a forced-air oven at 30°C. Calcium contents were analyzed in the grains, pericarps and seed coats. The incidence of Aspergillus spp., Penicillium spp., Rhizopus spp. and potential aflatoxin production in vitro were evaluated, as well as the seed coat thickness. The seed coat was thicker when peanut was grown in the presence of lime, leading to a decrease in seed infection by Aspergillus spp. and Penicillium spp. When plants were dried in shade, the growth of aflatoxinogenic fungi was independent of liming. However, in plants dried in the field or field + oven, the development of these fungi was decreased and even suppressed when the Ca content of the seed coat was increased from 2.2 to 5.5 g kg-1.

In order to evaluate the bean yield under different water table levels as well as the moisture and nitrate distribution in the soil profile, a field experiment was carried out at the experimental area from the College of Agronomic Sciences - UNESP, Botucatu, SP, Brazil. Beans were grown in field lysimeters and subjected to five water table depths:30; 40; 50; 60 and 70 cm. The moisture in the soil profile was gravimetrically determined through samples obtained at 10; 20; 30; 40; 50; 60 and 70cm of depth. The water table depths of 30cm and 40cm showed the highest productivities (3,228.4 kg.ha-1 and 3,422.1 kg.ha-1, respectively), showing no statistical differences between each other. The highest productivity was related to the two most elevated water table levels (30 and 40cm), which provided the highest moisture average values on basis of volume in the soil profile (33.3 e 31%) as well as the consumptive use of water (416 and 396 mm). The nitrate content during the bean cycle at the extraction depth of 60cm has been under the safe drinking limit of 10 mg.1-1 for water table depths of 30; 40; 50 and 60cm, showing the denitrification effectiveness as a way of controlling water table from nitrate pollution. The water table handling allowed the attainment of high bean productivity levels, as well as the reduction of the nitrate level.

A high surface area silica gel (737 ± m2 g-1) was synthesized modified through a two-step reaction with a 4-amino-2-mercaptopyrimidine ligand and applied to Cu(II) and Cd(II) adsorption from an aqueous medium. The modified material was characterized by FTIR, which showed that attachment of the molecule occurred via thiol groups at 2547 and 2600 cm-1, and by elemental analysis that indicated the presence of 0.0102 mmol of ligand. The data from adsorption experiments were adjusted to a modified Langmuir equation and the maximum adsorption capacity was 6.6 and 3.8 μmol g-1 for Cu(II) and Cd(II), respectively. After adjusting several parameters, the material was applied in the preconcentration of natural river water using a continuous flow system before and after sample mineralization, and the results showed a 10-fold enrichment factor. The proposed method was validated through preconcentration and analysis of certified standard reference material (1643e), whose results were in agreement with the values provided by the manufacturer.

The research on multiple classifiers systems includes the creation of an ensemble of classifiers and the proper combination of the decisions. In order to combine the decisions given by classifiers, methods related to fixed rules and decision templates are often used. Therefore, the influence and relationship between classifier decisions are often not considered in the combination schemes. In this paper we propose a framework to combine classifiers using a decision graph under a random field model and a game strategy approach to obtain the final decision. The results of combining Optimum-Path Forest (OPF) classifiers using the proposed model are reported, obtaining good performance in experiments using simulated and real data sets. The results encourage the combination of OPF ensembles and the framework to design multiple classifier systems. © 2011 Springer-Verlag.

The study aimed to evaluate the occurrence of pest and management of cotton genetically modified with the introduction of the bacterium Bacillus thunringiensis compared to conventional cotton in the 'Cerrado' region, installing the field experiment in 2007/2008 in Chapadão do Sul-MS. The experimental design was randomized blocks in a 2 × 3 factorial, with two managements of insecticides (with and without insecticides to control lepidopteran) and 3 modes of use of cultivars: 100% transgenic (NUOPAL), 100% non transgenic (Deltaopal) and interior area with transgenic (80% NUOPAL) with non-transgenic border (20% Deltaopal) with five replications. Evaluations were performed weekly of pest infestation, observing in 15 plants per plot the number of Alabama argillacea, Heliothis virescens and Spodoptera frugiperda. Based on these results it was concluded that: the occurrence of A. argillacea and H. virescens was lower in treated transgenic cultivar. There were no differences between transgenic and conventional farming on the occurrence of S. frugiperda.

The polyphagous pests belonging to the genus Spodoptera are considered to be among the most important causes of damage and are widely distributed throughout the Americas'. Due to the extensive use of genetically modified plants containing Bacillus thuringiensis genes that code for insecticidal proteins, resistant insects may arise. To prevent the development of resistance, pyramided plants, which express multiple insecticidal proteins that act through distinct mode of actions, can be used. This study analyzed the mechanisms of action for the proteins Cry1Ia10 and Vip3Aa on neonatal Spodoptera frugiperda, Spodoptera albula, Spodoptera eridania and Spodoptera cosmioides larvae. The interactions of these toxins with receptors on the intestinal epithelial membrane were also analyzed by binding biotinylated toxins to brush border membrane vesicles (BBMVs) from the intestines of these insects. A putative receptor of approximately 65. kDa was found by ligand blotting in all of these species. In vitro competition assays using biotinylated proteins have indicated that Vip3Aa and Cry1Ia10 do not compete for the same receptor for S. frugiperda, S. albula and S. cosmioides and that Vip3Aa was more efficient than Cry1Ia10 when tested individually, by bioassays. A synergistic effect of the toxins in S. frugiperda, S. albula and S. cosmioides was observed when they were combined. However, in S. eridania, Cry1Ia10 and Vip3Aa might compete for the same receptor and through bioassays Cry1Ia10 was more efficient than Vip3Aa and showed an antagonistic effect when the proteins were combined. These results suggest that using these genes to develop pyramided plants may not prove effective in preventing the development of resistance in S. eridiana. © 2012 Elsevier Inc.

A novel composite formed by interaction of a octa(3-chloropropyl)octasilsesquioxane modified with Purpald®, and its subsequent reaction with silver and hexacyanoferrate (III) (AgHSP), was synthesized and initially characterized by Fourier transform infrared spectra (FTIR) and cyclic voltammetry. The cyclic voltammogram of the modified graphite paste electrode with AgHSP, showed one redox couple with formal potential Eθ'=0.64V (vs Ag/AgCl, KNO3, 1.0 mol L-1; v = 20 mV s-1), attributed to the Fe2+(CN)6/ Fe3+(CN)6 process. The redox couple presents an electrocatalytic response for determination of sulfite. The modified electrode showed a linear response from 7.0×10-5 to 1.0×10-3 mol L-1 with the corresponding equation Y(μA) = 18.05 + 29.983×103 [sulfite], and a correlation coefficient of r=0.999. The method showed a detection limit of 0.115×10-4 mol L-1 with a relative standard deviation of ± 4% (n = 3) and amperometric sensitivity of 29.983×10-3A mol L-1. The modified electrode showed a excellent stability and good reproducibility during experiments. © 2013 by ESG.

In this work, thermoplastic starch (TPS)-clay bionanocomposites were obtained by an innovative methodology using a combination of methodologies commonly used in the composites and nanocomposites preparations. The main objectives or novelties were to confirm efficiency of the processing methodology by field emission gun scanning electron microscopy and investigate the effect of clay content on the spectroscopic, bulk and surface hydrophilic/hydrophobic properties of these bionanocomposites. Raman and FTIR spectroscopies confirmed the changes in the spectroscopic properties of the TPS bionanocomposites with the addition of the clay materials. Water absorption and contact angle measurements were also used to analyze the effect of the clay content on the hydrophilic properties of the TPS bionanocomposites. The results also showed that the addition of the cloisite-Na+ clay increased the bulk and surface hydrophobicities of the TPS matrix, which may increase its industrial application, particularly in manufacturing of food containers. © 2013 Elsevier B.V.
