It is known that there is a wide variety of bioavailable trace metals in aquatic systems, and its determination is fundamental to predict impacts to organisms in these environments. However, the determination of the total concentration of chemicals in aquatic systems, despite its importance, does not provide necessary information for decision making or even may lead to misinterpretation of the procedures and potential risks to the system in question. This study aimed to use the technique of diffusion in thin films by concentration gradients (DGT) to evaluate the anion exchange membrane Whatman DE 81 as binder alternative in determining Cr (VI). Experiments were carried out to determine the diffusion coefficients for the material used diffusion (agarose gel). The behaviors of the binders were analyzed before the main variables of the systems, immersion time, pH and ionic strength. Then was made the assessment of potential interferences, to finally proceed with testing using actual samples in the laboratory and in situ

The hardness has an important role in quality control, in research studies and metallurgical and mechanical specification, selection and comparison of various materials. This property is of extreme importance in the oil industry because it is a determining factor to ascertain the safety of the material used in pressure vessels and pipelines. Due to the inability to stop the equipment while checking the hardness, the hardness testers are widely used portable method UCI, its great advantage is the fact that an essay fast, simple realization and not be considered a non-destructive testing with a good relationship money. The objective is to determine if there is significant difference in hardness measurements between 80 and 1200 sandpaper using a portable hardness tester UCI method, the material applied in gas storage spheres composition ASTM 516 Gr 70. After determining the number of homogeneity, we performed the hardness profile to isolate the major factors influencing the hardness part: cold rolling and segregation of impurities. Factors Cooling and sanding were analyzed using the method of design of experiments (DOE), in which it was demonstrated that neither variables nor their interactions, has significant influence on the hardness measurements by portable MIC 10. This fact will lead to reduction in time and cost for surface preparation

O estudo de novas alternativas terapêuticas no tratamento de infecções microbianas tornou-se crescente no cenário científico devido à grande variação genética desses micro-organismos, que resultou na resistência aos antimicrobianos existentes. A grande diversidade na flora brasileira e a ampla utilização das plantas como medicamentos pela população justificam os estudos e o crescente interesse pela descoberta de novos compostos bioativos isolados dos vegetais. Plantas usualmente utilizadas na agricultura, apenas como adubação verde, por exemplo, são atualmente alvo de estudos científicos com potenciais promissores de atuação como de produtos terapêuticos. Plantas da família Leguminosae, amplamente conhecidas e utilizadas como fornecedoras de nitrogênio ao solo, vem sendo estudadas por diversas áreas da saúde para comprovarem a ação de compostos isolados entre estes os alcalóides pirrolidizínicos como antiinflamatórios, antibióticos e até como veneno para pragas. O objetivo do presente estudo foi determinar, a partir de extrativos de Crotalaria pallida, a atividade antimicrobiana in vitro utilizando cepas padrões de: Staphylococccus aureus, Escherichia coli, Salmonella sp e da levedura Candida albicans. Para determinação dessa atividade foi utilizada a técnica de diluição em microplaca que possibilitou o estudo da atividade do extrativo vegetal e da concentração inibitória mínima, isto é, concentração bactericida e/ou bacteriostática mínima e concentração fungiostática e/ou fungicida mínima. A Crotalaria pallida, não apresentou atividade frente aos micro-organismos testados nas condições padronizadas neste estudo

The objective of this study was to compare the power corresponding to maximal lactate steady state determined through continuous (MLSSC) and intermittent protocol with active recovery (MLSSI). Ten trained male cyclists (25 ± 4 yr, 72.5 ± 10.6 kg, 178.5 ± 4.0 cm), performed the following tests on different days on a cycle ergometer: (1) incremental test in order to determine the anaerobic threshold (AT) and maximal power (Pmax); (2) two to five constant workload tests to determine MLSSC, and; 3) two to three constant workload tests to determine MLSSI, consisting on 8 x 4 minutes bouts interspersed by two minutes of active recovery at 50% Pmax (i.e., 46 min of exercise protocol). MLSS intensity was defined as the highest workload at which blood lactate concentration did not increase by more than 1 mM between minutes 10 and 30 of the constant workload. The workload corresponding to MLSSC (273.2 ± 21.4 W) was significantly lower than that corresponding to MLSSI (300.5 ± 23.9 W). With base on these data, it can be verified that the intermittent exercise mode utilized in this study, allows an increase of 10% approximately, in the exercise intensity corresponding to MLSS.

s not degraded in the rumen but destroy ureasis. Soybean meal is one of the protein ingredients commonly used in formulations of animal feeds. In the diets of monogastric bran has high protein content provided by a greater separation of soybean hulls. For the ruminant protein value of the meal is lower with the inclusion of soybean hulls to reduce the level of protein. With the increased consumption of meal high protein, due to increases in production of pigs and poultry in recent years, increasing the availability of soybean hulls on the market, this is because soybean hulls is little understood in the feeding of monogastric . In this context, the main objective of this study was to determine the times and temperatures needed to ensure the effective inactivation of ureasis present in the bran and soybean hulls. According to the assessments, to make the determination of temperatures and times required for the inactivation of ureasis present in the bran and soybean hulls are 170 º C and 25 minutes for soybean meal and 140 ° C 10 minutes for soybean hulls, respectively

O uso de plantas medicinais especialmente na América do Sul contribui significativamente para os cuidados básicos com a saúde. Para o tratamento de infecções comuns, muitas plantas são utilizadas no Brasil na forma de extrato bruto, infusões ou emplastros, sem nenhuma evidência científica de sua eficácia (PESSINI et al., 2003). AYRES et al. (2008) afirma que a busca de substâncias com atividades antimicrobianas tem direcionado a atenção sobre os produtos naturais e, entre estes, os derivados das plantas superiores têm, nos últimos anos, despertado a investigação para o potencial da flora brasileira. Byrsonima pachyphylla Griseb é uma árvore típica do cerrado. Na medicina popular, a casca é utilizada como antifebril, contra tosses e doenças pulmonares, os ramos com folhas são diuréticos e os frutos são laxantes brandos (SILVA JÚNIOR et al., 2005). Levantamento no NAPRALERT indicou que espécies deste gênero são comumente empregadas como antiasmáticas, contra a febre e infecção de pele (MENDES et al., 1999). Este trabalho teve como objetivo determinar qual o melhor método de extração, sendo eles a maceração, a maceração dinâmica, a digestão e a digestão + maceração dinâmica, visando à obtenção de extratos brutos de folhas de Byrsonima pachyphylla Griseb., considerando sua atividade biológica frente a bactéria Gram-positiva: Staphylococcus aureus; as bactérias Gram-negativas: Escherichia coli e Pseudomonas aeruginosa, e a levedura Cândida albicans, testada pelo método de diluição seriada de extratos em microplacas; além de comparar perfis cromatográficos dos extratos obtidos em cromatografia de camada delgada (CCD), bem como pesquisar as principais classes de metabólitos secundários nos extratos. Os resultados obtidos na cromatografia em camada delgada e na triagem fitoquímica preliminar possibilitaram sugerir a presença de taninos, flavonoides, terpenos e saponinas na espécie ...

Changes in circulating angiogenic factors have been implicated in the pathogenesis of preeclampsia. Thus, evaluation of angiogenesis agonist and antagonist factors is of greater importance to understand the mechanisms responsible for this disorder. The objective of the present study was to evaluate whether circulating angiogenic and anti-angiogenic factors may differentiate early-onset from late-onset preeclampsia. The study was conducted in 86 women with preeclampsia diagnosed in the third trimester of pregnancy. Preeclampsia was classified according to the onset of clinical manifestation in early-onset (before 34 weeks of gestation; n=31) or in late-onset (from 34 weeks of gestation on; n=55) preeclampsia. Serum was obtained from the patients in the moment of the diagnosis and assayed for placental growth factor (PlGF), vascular endothelial growth factor (VEGF), soluble Endoglin (sEng) and soluble form of vascular endothelial growth factor receptor (sVEGFR-1) determination by enzyme-linked immunosorbent assay. The results showed that early-onset preeclampsia was characterized by significant lower levels of PlGF (median 38.3 vs 123.5 pg/mL) and VEGF (median 23.1 vs 35.3 pg/mL) in serum as well as by higher serum levels of sEng (median 54.7 vs 42.1 pg/mL) and sVEGFR-1 (median 5211.0 vs 4657.6 pg/mL) compared with late-onset preeclampsia. In this study serum levels of angiogenic and anti-angiogenic factors prove useful in differentiating early-onset from late-onset preeclampsia in the third trimester of pregnancy. Therefore, these findings suggest that angiogenic factors determination may indicate that early- and late-onset preeclampsia have different pathophysiology

Worldwide environmental degradation is an undesirable byproduct resulting from the increasing demand for natural resources. Water sources are suffering intense contamination since they usually receive a huge amount of domestic and industrial effluents - which are mostly wasted without proper treatment - inserting a large number of pollutants in the environment, heavy metals included. Mercury holds great toxicological importance because, under some physicochemical conditions in a water environment, Hg (II) ion turns into methylated compounds stemming from this element, such as methylmercury CH3Hg, which is highly toxic for the aquatic community in which bioaccumulation occurs. Nowadays passive sampling techniques are being developed to enable the analytical procedures which are applied in environmental monitoring. Diffusive gradients in thin-films technique (DGT) has been proven an interesting tool for the determination of labile metal species due to its in situ application. The DGT technique consists of a piston-like device on which the following series of agents is disposed: a binding agent (conventionally Chelex 100 resin), a diffusive agent, usually a polyacrylamide gel, and a membrane filter. Nevertheless, the agents conventinally used for this technique don't usually show satisfactory results in mercury sampling. The main goal of this study was to evaluate the phosphate-treated cellulose membrane (Whatman P 81), an alternative material, as binding agent in the DGT to determine labile mercury fractions in aquatic systems. In this context, we conducted a study of the behavior of this material in relation with system variables, pH and ionic strength. A predição da resposta do tumor a radioterapia e a questão mais importante durante o tratamento de pacientes com câncer. Como consequência, a predição de genes que sejam responsivos a radiação ionizante e uma possibilidade para a melhoria dos resultados clínicos e a otimização das doses as quais os pacientes são submetidos ao longo do tratamento. Juntamente com esses dados, é possível obter respostas sobre os mecanismos de resistência a radiação dos tumores e até mesmo a identificação de biomarcadores responsáveis pela resistência a radiação ionizante que podem ser potenciais para o desenvolvimento de novas drogas visando a proteção de tecidos saudáveis. A determinação experimental dos genes que sejam responsivos à radiação ionizante é algo caro e que demanda muito tempo e trabalho; porém, se utilizarmos uma forma computacional de direcionar os estudos experimentais diretamente aos genes que têm mais potencial para serem responsivos à radiação ionizante, as pesquisas podem ser mais direcionadas e específicas. Para determinar essa característica, construímos, analisamos e determinamos os dados da topologia da rede integrada de interações moleculares entre genes humanos, contendo interações físicas entre proteínas, interações metabólicas e interações de regulação transcricional. Os dados topológicos foram utilizados como atributos de treinamento para o aprendizado de máquina, no qual os genes conhecidamente responsivos à radiação ionizante foram apresentados a um algoritmo de árvore de decisão que gerou modelos de predição com índices de sensibilidade e precisão de 5% e 72%, respectivamente. Os índices de acerto obtidos para os conjuntos de teste foram satisfatórios, retornando 91% dos genes conhecidos como responsiveis à radiação ionizante utilizados para o treinamento da árvore de decisão. There are cassava varieties that present compounds as carotenoids, beta-carotene, lycopene and minerals important for human and animal health. The present study evaluated the antioxidant activity of the white, yellow and pinkish varieties of Manihot esculenta, by mean of the DPPH test and by the ferrous ion-chelating activity. Furthermore, the total phenols, carotenoids, beta-carotene, lycopene and zinc contents were also determined. Utilizing the DPPH test it was possible to find that extracts of boiled samples presented higher antioxidant activity (89.53% - pinkish) in comparison to the fresh samples (1.97% - white). For the ferrous ion-chelating test, the highest activity was found for the boiled pinkish variety extract (63.43%) and the lowest was for fresh yellow extract (17.34%) the white sample did not present activity. The highest concentration of total phenols and zinc content was obtained for the boiled pinkish variety extract 136.12 mg EAG/g of extract and 0,811ppm, respectively, in the concentration of 1000 µg/mL. The pinkish variety presented also higher quantity of pigments, including carotenoid (29.40 µg/g), beta-carotene (9.14 µg/100g) and lycopene (68.92%). According to the results obtained in this study it was possible to conclude that the yellow and pinkish varieties of M. esculenta present quantity of phenolic compounds and minerals sufficient to attribute the antioxidant activity and may thus contribute to reduce oxidative damage and be used as nutraceuticals or directly ingested in the diet to maintain good health.
