1. The synthesis of heat shock protein 70 (Hsp70) mRNA and the expression of Hsp70 in the liver of broiler chickens submitted to acute heat stress (35 degrees C for 5 h) was investigated.2. Hsp70 expression was detected by SDS-PAGE and Western blot analysis using a polyclonal antiserum against Hsp70 of Blastocladiella emersonii. The specific signal of Hsp70 mRNA was analysed by Northern blot using as probe a Hsp70 cDNA of B. emersonii.3. An increase in the amount of Hsp70 was detected from the first up to the fifth hour of acute heat exposure. This increase in the amount of Hsp70 was accompanied by an increase in Hsp70 mRNA which peaked at 3 h.4. This study shows that the heat induced increase in Hsp70 mRNA and protein in broiler liver, in vivo, are time dependent, similar to that in mammals.

The first diagnosis of botulism in cattle in Brazil and it's epizootiology are reviewed. The high prevalence of the disease raised on phosphorus deficient pastures in Savanna regions has caused severe economic losses in the past.The temperature induced microcomplement fixation test (TIMCF) confirmed the clinical-pathological diagnosis in all of the 24 cases studied by this method.The most important reason why botulism has not been controlled satisfactorily in Brazil is the lack of an available effective vaccine (type C and D). Additional prophylactic measures are phosphorus supplementation and removal of carcasses from the pasture.

The purpose of the present study was to investigate the effect of thermal conditioning, (through exposure to heat stress), during pre-hatch development on some physiological responses of post-hatch broilers to a post-natal heat stress challenge. Exposure to heat stress at this stage, we hope, may possibly induce epigenetic heat adaptation. Incubating eggs were exposed to temperature of 39.0degreesC for 2 h from Day 13 to 17 of incubation. At 33, 35, 37, 39, 41 and 43 d of age, the broilers hatched from these eggs were housed individually in open-circuit respiration cells. The climatic chambers were set to 22degreesC and increased to 30degreesC for 4 h. O-2 consumption and CO2 production of each chicken was monitored continuously in order to calculate the heat production. Blood samples were obtained before and during the 4 h heat stress. Thermal conditioning during incubation did not affect the plasma T-4, corticosterone, glucose, uric acid and CK concentrations. Temperature challenge, decreased plasma T-3 of broilers of both groups but the decrease was greater in pre-conditioned broilers compared with controls. A similar trend was observed for triglycerides. These changes did not affect total heat production. Since decreased T3 and triglyceride levels are part of the mechanisms for thermoregulation, these suggest that thermal conditioning during incubation can improve the broiler chicken capability for thermotolerance at later post-hatch age. This study investigated the effect of non-ventilation of the incubator during the first 10 days of incubation and its combination with dexamethasone administration at day 16 or 18 of incubation on hatching parameters and embryo and post-hatch chick juvenile physiology. A total of 2400 hatching eggs produced by Cobb broiler breeders were used for the study. Blood samples were collected at day 18 of incubation, at internal pipping stage (IP), at the end of hatch (day-old chick) and at 7-daypost-hatch for T-3, T-4 and corticosterone levels determination. From 448 to 506 h of incubation, the eggs were checked individually in the hatcher every 2 h for pipping and hatching. The results indicate that non-ventilation during the first 10-day shortened incubation duration up to IP, external pipping (EP) and hatch, had no effect on hatchability and led to higher T-3 levels at IP but lower corticosterone levels at 7-day-post-hatch. The injection of dexamethasone at days 16 and 18 of incubation affected hatching and blood parameters in both the ventilated and non-ventilated embryos differentially and the effect was dependent on the age of the embryo. Dexamethasone increased T-3 levels and T-3/T-4 ratios but the effect was greater with early non-ventilation of eggs. Dexamethasone decreased hatchability but the effect was greater when injected at day 16 and especially in ventilated embryos. The effects of incubation protocols and dexamethasone treatments during incubation were still apparent in the hatched chicks until 7 days of age. The changes in T-3, T-4 and corticosterone levels observed in response to the early incubation conditions and late dexamethasone treatments in this study suggest that incubator ventilation or non-ventilation may influence the hypothalamic-pituitary-adrenal axis (HPA) regulation of stress levels (in terms of plasma corticosterone levels) and thyroid function in the embryo with impact on incubation duration, hatching events and early post-hatch life of the chick. Our results also suggest that some stages of development are more sensitive to dexamethasone administration as effects can be influenced by early incubation protocols. (c) 2006 Elsevier B.V. The serological response to Salmonella pullorum and S. gallinarum infection in chickens was studied with an indirect enzyme-linked immunosorbent assay (ELISA). In broiler chickens, a more virulent strain of S. pullorum produced a significantly lower serum IgG titer than did a less virulent strain. In laying hens, the serum and egg-yolk IgG titers were very similar. In chickens infected with S. gallinarum, high IgG titers persisted for 30 weeks. In chickens reinfected with this strain, each reinfection was followed by transitory increases in IgG lasting no longer than 2 weeks. Serum samples from Brazil taken from a laying flock with evidence of fowl typhoid showed much higher antibody levels than did those from three uninfected flocks. Using lipopolysaccharide as the detecting antigen, infections caused by these salmonellae could be differentiated from those caused by other groups. Incorporation of the appropriate flagella antigen in the ELISA allowed differentiation between infections caused by S. pullorum and S. enteritidis.

The objective was to evaluate the effects of genetic group and age on growth, carcass, and meat traits of rabbits. A total of 144 straightbred Botucatu and White German Giant x Botucatu crossbred rabbits were involved. Rabbits were weaned at 35 d and sequentially, slaughtered, four per genetic group x sex combination, at: 42, 49, 56, 63, 70, 77, 84 and 91 d. A 2x2 factorial arrangement was employed in a completely randomized design with repeated measures for growth traits, and a split-plot for carcass and meat traits. Crossbred rabbits were heavier (2032 vs. 1962 g; P < 0.01), consumed more feed (143.5 vs. 131.0 g/d; P < 0.01), and presented higher slaughter weight (2169 vs. 2093 g, P=0.02) and dressing percentage (59.0 vs. 58.2%; P=0.07) than straightbreds throughout the experiment. No difference between genetic groups was detected for feed conversion and empty gastrointestinal weight corrected for slaughter weight (SW). Crossbreds showed higher skin weight (308.2 vs. 299.7 g, P = 0.06) and distal parts of leg weight (75.7 vs. 71.4 g; P < 0.01), both corrected for SW. No genetic group effect was detected on dissectible fat and hind part weights. Chilled commercial carcass (1284 vs. 1229 g: P=0.02), chilled reference carcass (1036 vs. 1000 g, P=0.06), fore part (297.9 vs. 283.3 g; P=0.01) and loin (308.7 vs. 295.5 g; P=0.05) were heavier in crossbreds than in straightbreds, but these differences were attributed to differences in SW. Uncorrected weights of head, kidneys, liver and thoracic viscera were higher in the crossbred group, but only head (116.6 vs. 113.6 g; P=0.06) and thoracic viscera (30.4 vs. 28.6 g; P=0.01) were, in fact, proportionately heavier in crossbreds than in straightbreds. No effect of genetic group was detected on meat to bone ratio, muscle ultimate pH and chemical composition of the Longissimus dorsi muscle. All traits, except for ash and fat contents of the Longissimus muscle, showed age effects (P < 0.01). Crossbreeding may be recommended for the production of whole commercial carcasses, but it is not clearly advantageous for the production of retail cuts. Slaughter should take place between 63 and 70 d of age for both genetic groups.

This experiment was carried out to evaluate the effect of the addition of fumaric acid to broiler diet on birds performance. One thousand and eighty Hubbard day-old broiler chicks were alloted in a randomized block design, six treatments with two replicates of males, and four replicates of females. Three basal diets were formulated to meet the nutritional requirements in each growing phase: from 1 to 21 days, 21 to 37 days and 37 to 45 days. Treatments consisted first in the addition of growth promoter to 0.75 and 1.0%, of fumaric acid. The group treated without of both growth promoter and fumaric acid presented a higher intake and a smaller feed:gain ratio in relation to the fumaric acid treated groups. However, there were no differences among groups treated with growth promoter and fumaric acid. The contrasts did not show any difference among weight gain, carcass yield and abdominal fat. The addition of fumaric acid levels to the diets promoted reduction of feed intake, without any effect on weight gain, improving, therefore, the feed/gain ratio. A digestibility experiment was carried out, using 30 Hy-Line roosters, to determine apparent metabolyzed energy (AME), corrected by nitrogen of the diets containing 0.0, 0.5 and 1.0% of the acid. An increase on the AME of the diets was observed with fumaric acid addition.

Salmonella enterica serovar Enteritidis-lysing bacteriophages isolated from poultry or human sewage sources were used to reduce Salmonella Enteritidis in vitro and in experimentally infected chicks. Cocktails of 4 different bacteriophages obtained from commercial broiler houses (CB4O) and 45 bacteriophages from a municipal wastewater treatment plant (WT45O) were evaluated. In experiment 1, an in vitro crop assay was conducted with selected bacteriophage concentrations (105 to 101 pfu/mL) to determine ability to reduce Salmonella Enteritidis in the simulated crop environment. Following 2 h at 37 degrees C, CB40 or WT45O reduced Salmonella Enteritidis recovery by 1.5 or 5 log, respectively, as compared with control. However, CB40 did not affect total SE recovery after 6 h, whereas WT45O resulted in up to a 6-log reduction of Salmonella Enteritidis. In experiment 2, day-of-hatch chicks were challenged orally with 3 x 103 cfu /chick Salmonella Enteritidis and treated cloacally with 1 X 109 WT45O pfu/chick I h postchallenge. One hour later, chicks were treated or not with a commercially available probiotic (Floramax-B11). Both treatments significantly reduced Salmonella Enteritidis recovery from cecal tonsils at 24 h following vent lip application as compared with controls, but no additive effect was observed with the combination of bacteriophages and probiotic. In experiment 3, day-of-hatch chicks were challenged orally with 9 x 103 cfu/chick Salmonella Enteritidis and treated via oral gavage with I X 108 CB40 pfu/chick, 1.2 x 108 WT45O pfu/chick, or a combination of both, I h postchallenge. All treatments significantly reduced Salmonella Enteritidis recovered from cecal tonsils at 24 h as compared with untreated controls, but no significant differences were observed at 48 h following treatment. These data suggest that some bacteriophages can be efficacious in reducing SE colonization in poultry during a short period, but with the bacteriophages and methods presently tested, persistent reductions were not observed.

This research was designed to study the effect of addition of 5, 10 e 15% of textured soy protein (TSP) on sensorial, chemical, and physical-chemical parameters of chicken loaf with ground meat. The sensorial properties were not affected by textured soy protein. The addition of textured soy protein in preparing chicken loaf increased the water absorption and holding capacity, and decreased the moisture and ether extract content, but did not increase the final product yield.

The effect of feed restriction and enzymatic supplementation on intestinal and pancreatic enzyme activities and weight gain was studied in broiler chickens. Quantitative feed restriction was applied to chickens from 7 to 14 d of age. An enzyme complex mainly consisting of protease and amylase was added to the chicken ration from hatching to the end of the experiment. Birds subjected to feed restriction whose diet was not supplemented showed an increase in sucrase, amylase, and lipase activities immediately after the restriction period. Amylase, lipase, and chymotrypsin activities were higher in chickens subjected to feed restriction and fed a supplemented diet than in those only subjected to feed restriction. Trypsin activity increased after feed restriction and after supplementation, but there was no interaction between these effects. Early feed restriction had no effect on enzyme activity in 42-d-old chickens. Chickens subjected to early restriction and fed the supplemented diet presented higher sucrase, maltase, and lipase activities than nonsupplemented ones (P < 0.05). There was no effect of early feed restriction or diet supplementation on weight gain to 42 d. Percentage weight gain from 14 to 42 d of age was equivalent in feed-restricted and ad libitum fed birds. Feed-restricted broilers fed a supplemented diet showed a higher percentage weight gain than nonsupplemented birds. We conclude that enzymatic supplementation potentiates the effect of feed restriction on digestive enzyme activity and on weight gain.

It was studied the effect of energy levels in the diet, obtained from vegetal oil inclusion, on the performance, diet cost per kilogram of broiler and fat content of the carcass, There were used 4,800 broiler chicks, 2,400 males and 2,400 females, and they were distributed in six treatment (energy levels in kcal ME/kg of diet) in three different phases: start diet from one to 20 days of age: A - 2930, B - 2980, C - 3030, D - 3080, E - 3130, F - 3180, growth diet from 21 to 40 days: A - 2980, B - 3030, C - 3050, D - 3130, E - 3150, F - 3230, finishing diet from 41 to 45 days of age: A - 3030, B - 3080, C - 3130,, D - 3180, E - 3230, F - 3280. The protein content was 22, 20 and 18% in each phase, respectively, There were evaluated the feed intake, energy intake, protein intake, weight gain, feed conversion mortality, diet cost per kg/bw. Sixty broilers were slaughtered, five males and five females per treatment, randomly chosen to determine the carcass composition considering the dry matter content, lipids content and protein content, total fat in carcass and fat per kilogram of broiler. The data were submitted to variance analysis and the means were compared using SNK. The broilers fed with medium and low levels of dietary energy showed the highest weight gain. At energy levels of the treatment D in the three phases, the lowest cost per kilogram of broiler was obtained. The females presented the highest fat content per kilogram of broiler produced.

One-day-old broiler chicks received cecal microflora (CM) cultured under aerobic, anaerobic conditions, or both (mixed) and were then infected with Salmonella Enteritidis, in order to compare the efficacy of these different types of culture in terms of the number of chicks infected, cecal colonization and faecal excretion of the challenging bacteria. Regardless of culture type, CM always led to a smaller number of S. Enteritidis for any of the parameters studied compared to untreated chicks. Aerobic CM demonstrated better efficacy in reducing the number of infected chicks and cecal colonization by S. Enteritidis, followed by mixed CM. No difference was observed in faecal excretion of S. Enteritidis between the chicks that received different types of CM culture. (C) 2002 Elsevier B.V. B.V. All rights reserved.

This research was designed to study the elaboration of chicken loaf with ground meat. The products were elaborated in laboratory scale utilizing broiler meat bought at the market. The products were conditioned in vacuum-packages and stored for 133 days at 5 and 10 degrees C. Chemical, sensorial and microbiological analyses of the elaborated products were performed. The storage temperature affected the total bacterial counts, pH, characteristic aroma and rancid aroma. In both treatments (storage temperature: 5 and 10 degrees C), depletion of nitrites, reaching 24 ppm in 133 days of storage, and exudate formation after 77 days were observed. The storage of chicken leaves, vacuum-packaged, at 5 degrees C for a maximum of 77 days is recommended.

This study was undertaken to investigate the toxic effect of Crotalaria spectabilis seeds added to the ration for commercial broilers during the final phase of growth. Ground seeds were added at different concentrations to the ration: 0.0% (control), 0.01%, 0.1% and 0.4%. Rations containing 0.4% caused symptoms of intoxication, beginning in the second week of the study. During the third week birds showed bristling, apathy, general weakness, distended abdomen and agglomeration. In the fourth week, four animals died. Necropsy revealed prominent ascites and severe lesions of liver, kidney and lung. Microscopic examination revealed necrosis of hepatocytes, inflammatory cell infiltration, hypertrophy and hyperplasia of biliary duct cells leading to atresia and cholestasis. Prominent cartilaginous and osseous nodules in the lungs were also present as well as degenerative, changes in the kidney tubules and necrosis of cells of the bursa. All the broilers receiving a ration with 0.1% of Crotalaria seeds showed ascites and slight lesions of the liver. The remaining groups showed no reduction in weight gains, lesions or clinical symptoms. It is concluded that broilers during the final phase of growth are sensitive to administrations of seeds of C. spectabilis in their ration. The presence of ascites and cartilaginous and osseous nodules in the lungs of the affected birds was also considered important.

Economic losses caused by enzootic bovine leukosis (EBL) have been of interest since World War II, when the neoplastic form of EEL increased dramatically in Europe. Olson (1974) and House et al. (1975) ed that animals with lymphosarcoma caused by the bovine leukosis virus (BLV) had reduced milk yields. a less efficient reproductive performance and high veterinary costs and mortality rates, while many carcasses were rejected at slaughter. However, the actual impact of BLV infection in cattle without lymphosarcoma is not; clear. The purpose of the study reported here was to compare some productive and reproductive responses of cattle that were antibody-positive (BLV+) or negative (BLV-) for BLV.Holstein dairy cows in commercial dairy farms were used in this study. Blood samples were collected and subjected to BLV serological examination by the agar gel immunodiffusion test of Miller & van der Maaten (1976). Animals were then grouped as BLV+ or BLV- according to their serological response to the BLV antigen. Productive and reproductive histories were obtained from individual animal records and the following factors were considered: milk production, calving interval and birth rate. For milk production, we had the daily milk yields of 547 animals, and for calving interval the time between two successive parturitions for 444 cows. These values were examined by ANOVA and when this was significant a Student's t test was carried out for each age group. Birth rates, the percentage of animals that calved in 1 gear, were available for 557 animals and were examined with the Z-two proportion test. For all analyses, P < 0.05 was considered significant.
