Within the genus Prochilodus, the species Prochilodus lineatus is certainly the most studied one from a cytogenetic point of view. In this study, a cytogenetic characterization of specimens of P. lineatus from Mogi-Guacu River was performed in the period from 2003 to 2007, through utilization of cytogenetic markers, such as Giemsa, Ag-NOR, C-banding and cytogenetical-molecular markers (FISH) to detect both 5S and 18S ribosomal genes. All analyzed individuals presented 2n=54 meta/submetacentric chromosomes, besides bearing up to 7 supernumerary Microchromosomes. Polymorphic NORs were detected on a single chromosomal pair. The constitutive heterochromatin was distributed at centromeric region of all chromosomes in the A complement, while the microchromosomes were totally heterochromatic. A syntenic distribution of 5S and 18S ribosomal genes was detected, comprising the NOR-bearing chromosomal pair. No additional ribosomal clusters in other chromosomes were observed. Although the karyotype features are commonly conserved within the genus Prochilodus, the few differences on the distribution of both ribosomal genes and heterochromatin when compared to specimens of P lineatus from other localities can be associated to the evolutionary changes that these repetitive sequences have undergone through the years.

A large number of functional foods, including those that contain P-glucan, have been shown to prevent the development of cancer and other chronic diseases. The aim of the present study was to elucidate its mechanism of action, as well as to understand its effects as an antigenotoxic, anticlastogenic agent, and to determine its capacity to preserve cell viability. The investigation was carried out in the CHO-k1 and CHO-xrs5 cell lines. The cytokinesis-blocked micronucleus assay indicated that the different doses of beta-glucan examined (5, 10, 20 and 40 mu g/ml) did not show clastogenic effects. In the CHO-k1 cell line, a chemopreventive effect could be observed in all the protocols tested: pre-treatment (% reduction of 35.0-57.3), simultaneous treatment (simple - 5 reduction of 19.7-55.6 and with pre-incubation - of 42.7-56.4) and post-treatment (% reduction of 17.9-37.6). This finding indicates mechanisms of action involving desmutagenesis and bio-antimutagenesis, albeit the latter having a lesser role. However, in the repair-deficient CHO-xrs5 cells, beta-glucan did not show a protective effect with post-treatment (% reduction of 2.96), thus supporting the involvement of bioantimutagenesis. The comet assay in CHO-k1 cells demonstrated that beta-glucan has neither a genotoxic nor an antigenotoxic effect. Cell viability tests indicated that beta-glucan preserves cell viability in both cell lines, preventing apoptotic events. These findings suggest that beta-glucan, when present in foods, could provide them with nutraceutical characteristics and act as a dietary supplement, or that P-glucan could be used in new drug development. The Spot-billed Toucanet (Selenidera maculirostris) is an endemic member of the Ramphastidae occurring in the Atlantic Forest in Brazil. There is anecdotal literature about this species breeding in the wild, but no data are available about parental behavior and nest morphometry. We describe observations of parental behavior of the Spot-billed Toucanet including measurements of one nest in Ilha do Cardoso State Park, São Paulo, Brazil. The nest was inside a hollow of a Lauraceae tree with the entrance hidden by leaves of Aechmea sp. (Bromeliaceae). The Spot-billed Toucanet, based on our observations and review of the literature, nests in tree cavities between 2 and 7 to above ground and both parents provision the nestlings. Este trabalho descreve a investigação do óleo essencial, obtido das partes aéreas de Turnera diffusa (Turneraceae) submetida a descontaminação usando irradiação com 60Co (raios gama). As analises dos óleos irradiados nos permite verificar diferenças quali e quantitativa nas amostras de óleos.

O presente trabalho visou estabelecer uma comparação entre composição de cachaças produzidas por Saccharomyces cerevisiae (Sc) e estirpes de leveduras selvagens [Pichia silvicola (Ps), Pichia anomala 1 (Pa1), Pichia anomala 2 (Pa2) e Dekkera bruxelensis (Db)], isoladas em destilarias da região de Jaboticabal-SP. Os componentes secundários da fração denominada coração foram determinados por cromatografia gasosa. Os níveis dos componentes secundários foram influenciados pelo pH dos respectivos vinhos, os quais dependem da estirpe de levedura empregada no processo fermentativo. A Saccharomyces cerevisiae apresentou valores ligeiramente superiores de componentes secundários, enquanto as estirpes selvagens produziram maiores teores de álcoois superiores. As estirpes selvagens de leveduras mostraram-se adequadas para obtenção de uma cachaça de boa qualidade.

For the first time, a survey on Giardia in the live-trapped small domestic and wild mammals was perfomed in four regions of state of the São Paulo, Brazil, with special attention to the parasitism of Rattus rattus rattus by Giardia. This species was found infected in all studied sites: Botucatu (15.4%), Conchas (28.5%), Itaporanga (38.7%) and São Roque (100 %). Two new hosts and their frequency of infection were described for Giardia in Nectomys squamipes, an aquatic rodent and in Bolomys lasiurus, a forest rodent (100 % and 14.3 %, respectively). Both G. muris and G. duodenalis groups were found in scrapings of intestinal mucosa of those rodents. Mixed infection was observed in some animals. It is important to emphasize the infection by G. duodenalis in the black rat as this species lives as a comensal with man and in N. squamipes as it may contaminate small streams used for domestic consumption. Therefore, further investigation will be necessary to elucidate the potential of these rodents to act as reservoirs of Giardia for man.

Eight Panthera onca (Po), 13 Felis concolor (Fc), 7 Felis yagouaroundi (Fy), 7 Felis tigrina (Ft) and 5 Felis pardalis (Fp) specimens from São Paulo State zoos were used. All animals were restrained with darts containing 10 mg/kg ketamine and 1 mg/kg xylazine. Venous blood samples were collected as soon as possible (within 15-20 min) and serum was frozen until the time for cortisol quantification. Cortisol was determined using a solid phase radioimmunoassay with an intra-assay coefficient of 8.51%. Data were analyzed statistically by the Kruskal-Wallis test, followed by Dunn's multiple comparisons test, and the one-sample t-test, with the level of significance set at P<0.05. Data are reported as means ± SEM. Cortisol levels differed among the captive felines: Po = 166 ± 33a, Fc = 670 ± 118b, Fy = 480 ± 83b, Ft = 237 ± 42ab, Fp = 97 ± 12a nmol/l (values followed by different superscript letters were significantly different (P<0.001)). Since most of the veterinary procedures on these species involve chemical restraint, these results show the necessity of preventive measures in order to minimize the effect of restraint stress on more susceptible species

A biologia de Bemisia tabaci biótipo B (Genn.) foi avaliada em genótipos de feijoeiro (Phaseolus vulgaris L.) que contêm arcelina em suas sementes. Foi também realizada análise bioquímica de proteínas, em sementes e em folhas dos genótipos de feijoeiro, a fim de verificar se haveria traços de arcelina nas folhas dos materiais a serem avaliados. Os testes foram conduzidos em condições de casa de vegetação, nas épocas das águas e da seca, em dois anos consecutivos, com os seguintes genótipos: ARC 3s, ARC 5s (genótipos selvagens portadores de arcelina); ARC 1, ARC 2, ARC 3, ARC 4 (linhagens quase-isogênicas portadoras de arcelina - Empresa Brasileira de Pesquisa Agropecuária (EMBRAPA)), Porrillo 70, Bolinha e IAPAR MD 808 (genótipos sem arcelina). Os genótipos selvagens, ARC 3s e ARC 5s, apresentaram altos níveis de antibiose, com ênfase para o ARC 5s (as ninfas tiveram alta mortalidade, em torno de 90%). O prolongamento do ciclo de desenvolvimento dos insetos provenientes do genótipo ARC 5s podem sugerir uma resistência do tipo antibiose e/ou não-preferência para alimentação. A resistência dos genótipos selvagens não está relacionada com a presença de arcelina nas sementes, já que nenhum traço dessa proteína foi encontrado nas folhas destes.

The phycoerythrin-deficient strain (green phenotype) of Hypnea musciformis (Rhodophyta) originated from a green branch, which had arisen as a spontaneous mutation in a wild plant (brown phenotype) collected from the Brazilian coast. The present study describes the growth responses to irradiance, photoperiod and temperature variations, pigment contents, and photosynthetic characteristics of the brown and green strains of H. musciformis. The results showed that growth rates increased as a function of irradiance (up to 40 mu mol photons m(-2) s(-1)) but, with further increase in irradiance (from 40 to 120 mu mol photons m(-2) s(-1)), became light-saturated and remained almost unchanged. The highest growth rates of the brown and green strains were observed in temperatures of 20-25 degrees C under long (14:10 h LD) and short (10:14 h LD) photoperiods. The brown strain had higher growth rates than the green strain in the short photoperiod, which could be related to the high concentrations of phycobiliproteins. Phycoerythrin was not detected in the green strain. The brown strain had higher concentrations of allophycocyanin and phycoerythrin in the short photoperiod while the green strain had higher concentrations of phycocyanin. The brown strain presented higher photosynthetic efficiency (alpha), and lower saturation parameter (I-k) and compensation irradiance (I-c) than the green strain. The brown strain exhibited the characteristics of shade-adapted plants, and its higher value of photosynthetic efficiency could be attributed to the higher phycoerythrin concentrations. Results of the present study indicate that both colour strains of H. musciformis could be selected for aquaculture, since growth rates were similar (although in different optimal light conditions), as the green strain seems to be adapted to higher light levels than the brown strain. Furthermore, these colour strains could be a useful experimental system to understand the regulation of biochemical processes of photosynthesis and metabolism of light-harvesting pigments in red algae.

An understanding of isoniazid (INH) drug resistance mechanism in Mycobacterium tuberculosis should provide significant insight for the development of newer anti-tubercular agents able to control INH-resistant tuberculosis (TB). The inhA-encoded 2-trans enoyl-acyl carrier protein reductase enzyme (InhA) has been shown through biochemical and genetic studies to be the primary target for INH. In agreement with these results, mutations in the inhA structural gene have been found in INH-resistant clinical isolates of M. tuberculosis, the causative agent of TB. In addition, the InhA mutants were shown to have higher dissociation constant values for NADH and lower values for the apparent first-order rate constant for INH inactivation as compared to wild-type InhA. Here, in trying to identify structural changes between wild-type and INH-resistant InhA enzymes, we have solved the crystal structures of wild-type and of S94A, I47T and I21V InhA proteins in complex with NADH to resolutions of, respectively, 2.3 angstrom, 2.2 angstrom, 2.0 angstrom, and 1.9 angstrom. The more prominent structural differences are located in, and appear to indirectly affect, the dinucleotide binding loop structure. Moreover, studies on pre-steady-state kinetics of NADH binding have been carried out. The results showed that the limiting rate constant values for NADH dissociation from the InhA-NADH binary complexes (k(off)) were eleven, five, and tenfold higher for, respectively, I21V, I47T and S94A INH-resistant mutants of InhA as compared to INH-sensitive wildtype InhA. Accordingly, these results are proposed to be able to account for the reduction in affinity for NADH for the INH-resistant InhA enzymes. (c) 2006 Elsevier Ltd. All rights reserved.

The selected yeast strains were examined for their ability lo grow, to retain cell viability and to ferment diluted sugar cane juice (15% total sugar, w/v) to ethanol at 40-degrees-C. The degree of agitation (aeration) affects the thermotolerance while the method used for isolation of the strains appears to have no significant effect. The yeast isolated are aerobically fermentative with increased levels of fermentation and growth resulting from agitation (aeration), the exact level of these increases being dependent on the strain used.
