Photodynamic therapy, term introduced by von Tapeiner in 1900, can be defined as the administration of a non toxic drug, i.e., a dye, known as photosensitizer (FS), which subsequently will be illuminated with light of specific wavelength. PDT is based on the interaction among FS, oxygen and light, which through photochemical reactions cause cell death. The FS molecules must have a high probability to form the singlet state after the excitation, which can induce chemical changes in the neighborhood in two ways, called reactions type I and type II. The type II reaction is based on the exchange of energy to molecular oxygen, exciting it to its state of higher energy (singlet), which is highly reactive. The proposed mechanisms for cell death are linked to damage to the DNA, mitochondria and to the cytoplasmic membrane. Several pre‐clinical and clinical trials have been carried out and the PDT is already used in many countries for treatment mainly against certain types of cancer. The therapy also has been gaining strength in antimicrobial control, since the microorganisms have appeared increasingly resistant to current antibiotics. Another attempt to use the PDT is for the inactivation of macro‐organisms, such as micro‐crustaceans and mosquitoes. To this end I tested whether the photosensitizers methylene blue, rose Bengal and the chlorophyll a has insecticidal activity against the yellow fever and dengue vector mosquito, Aedes aegypti. Since these diseases have no effective treatments, its control is linked to the vector control, which has shown resistance to chemical pesticides used. Based on this, this work shows its importance, because it is a new type of mosquito control since all the photosensitizers used are low cost, do not generate toxic products at the concentrations used and showed good results in mortality. The aim of present study was to evaluate frozen canine semen with ACP-106 (R) (Powder Coconut Water) using an in vitro sperm-oocyte interaction assay (SOIA). Ten ejaculates from five stud dogs were diluted in ACP-106 (R) containing 20% egg yolk, submitted to cooling in a thermal box for 40 min and in a refrigerator for 30 min. After this period, a second dilution was performed using ACP-106 (R) containing 20% egg yolk and 12% glycerol. Samples were thawed at 38 degrees C for 1 min. Post-thaw motility was evaluated by light microscopy and by using a computer aided semen analysis (CASA). Plasma membrane integrity and sperm morphology/acrosomal status were evaluated by fluorescent probes (C-FDA/PI) and Bengal Rose respectively. Moreover, frozen-thawed semen was analysed by a SOIA. Subjective post-thaw motility was 52.0 +/- 14.8% and it was significant higher than the total motility estimated by CASA (23.0 +/- 14.8%) because this system considered the egg yolk debris as immotile spermatozoa. Although normal sperm rate and acrosomal integrity evaluated by Bengal Rose stain was 89.6 +/- 3.1 % and 94.3 +/- 3.1 %, respectively, post-thaw percentage of intact plasma membrane was only 35.1 +/- 14.3%. Regarding SOIA, the percentage of interacted oocytes (bound, penetrated and bound and/or penetrated) was 75.3%. Using regression analysis, it was found significant relations between some CASA patterns and data for SOIA. In conclusion, the freezing-thawing procedure using ACP-106 (R) was efficient for maintain the in vitro fertility potential of dog spermatozoa.

Combining the data from conventional semen analysis with oocyte penetration assays should improve the assessment of the fertilizing ability of a semen sample. Thus, the objective of the present study was to evaluate the prognostic value of various semen parameters on the in vitro interactions between frozen-thawed canine sperm and homologous oocytes. Ten ejaculates from five stud dogs (two ejaculates/dog) were collected by digital manipulation. Semen samples were evaluated, extended in Tris-egg yolk-glycerol, frozen and stored in liquid nitrogen, and thawed several weeks later. Samples were evaluated for motility and sperm populations by computer-aided semen analysis (CASA), plasma membrane integrity (carboxy-fluorescein diacetate and propidium iodide), and sperm morphology (Bengal Rose). Thawed spermatozoa were also incubated with homologous oocytes for 18 h in an atmosphere of 5% CO2 and 95% air at 38 degrees C and sperm-oocyte interactions were evaluated. Simple linear regression models were calculated, with sperm parameters as independent variables and sperm-oocyte interactions as the dependent variable. There were significant associations between: percentage of oocytes bound to spermatozoa and beat cross frequency (BCF; R-2 = 63%); percentage of oocytes that interacted with spermatozoa and BCF (R-2 = 73%); and number of penetrated spermatozoa and velocity average pathway (VAP; R-2 = 64%) and velocity straight line (VSL; R-2 = 64%). Although plasma membrane integrity and sperm morphology had little prognostic value for in vitro interactions between canine frozen-thawed sperm and homologous oocytes, some motility patterns (evaluated by CASA) were predictive of in vitro sperm-oocyte interactions. The objectives of this study were to evaluate the effect of type of cuttings (apical, intermediate and basal) and different concentrations of IBA (indole-3-butyric acid) on rooting of Red success rose (Rosa sp.) leafy cuttings, during two season of cuttings collection (summer and winter). The investigation was carried out in the farm Irmaos Van Schaik in Holambra-SP, Brazil, from February to April and August to October/1993. The experimental design was a randomized block in a factorial arrangement. It consisted of 12 treatments (3 types of cuttings combined with IBA powder in 4 different concentrations - 0, 1000, 2000 and 4000 ppm) with 3 replicates during 2 seasons. The investigation permitted the following conclusions: the apical and intermediate cuttings showed better results in general when compared to the basal ones in both seasons; the average rooting at the transplanting time were 76%, 70% and 47% (summer) and 80%, 69% and 33% (winter) for apical, intermediate and basal cuttings, respectively; the utilization of IBA did not stimulate rooting. The average rooting for the control cuttings reached 85% (summer) and 78% (winter), regardless the type of cuttings.

The objectives of this study were to evaluate the effects of rooting media (vermiculite, carbonized rice bark, fenolic spume and sand), presence of IBA (indole-3-butyric acid) and presence of fertilization during the rooting of rose (Rosa sp.) leafy cuttings, Dalas cultivar. This investigation was carried out in Jaboticabal (Sao Paulo State - Brazil) during the period of March to April 1997. The experimental design was a randomized block in a factorial arrangement. It consisted of 16 treatments (the 4 rooting media combined with IBA concentrated solutions - 0 and 1.000 ppm and fertilizer - 0 and commercial product with macro and micronutrients, applied weekly) with 3 replicates. The evaluations were 30 days after cuttings showed that the best rooting percentage occurs in sand (98%), followed by vermiculite (90%) and fenolic spume (87%) and cuttings treated with IBA (95%), while the fertilization showed no promoting effects. The number of cuttings roots wasn't affected with rooting media or DBA treatment, while the fertilization showed effective.
