Avaliou-se a progressão dos sinais radiográficos de cães com displasia coxofemoral ou potencialmente displásicos tratados pela técnica da sinfisiodese púbica. Foram utilizados 14 cães, seis machos e oito fêmeas, com idades entre quatro e seis meses. Após exame físico, procedeu-se ao exame radiográfico para avaliação da articulação coxofemoral, segundo padrão estabelecido pelo Colégio Brasileiro de Radiologia Veterinária. Três deles foram também avaliados pelo método PennHIP. Pelo histórico clínico, nove (64%) animais foram trazidos por manifestarem sinais de dificuldade locomotora com os membros pélvicos, manifestada especialmente por claudicação unilateral ou bilateral e dificuldade de se levantar. Cinco (36%) cães não tinham qualquer sinal clínico, e em quatro (29%) realizou-se o exame radiográfico como prevenção. O sinal físico mais evidente foi dor à palpação da articulação coxofemoral, e dois (14%) cães apresentaram sinal de Ortolani positivo. Com exceção de um cão que apresentou infecção nos pontos cutâneos, todos os demais evoluíram sem complicações no período pós-operatório. Radiograficamente houve progressão das lesões articulares em seis (43%) animais e oito (57%) mantiveram a classificação inicial. Concluiu-se que cães tratados pela sinfisiodese púbica não apresentam melhora dos sinais radiográficos iniciais.

Foi realizado um estudo retrospectivo dos resultados dos exames citológicos provenientes dos arquivos do Serviço de Citopatologia do Laboratório de Patologia Animal, Faculdade de Medicina Veterinária e Zootecnia, Universidade Estadual Paulista, Botucatu, São Paulo, entre janeiro de 1994 e dezembro de 2008. do total de 139.986 animais atendidos no Hospital Veterinário da instituição, 11.468 (8,2%) foram encaminhados para exame citológico. Desses diagnósticos, 57,28% corresponderam a lesões neoplásicas, 19,28% a lesões inflamatórias e 14,79% a processos não neoplásicos (imunológicos, degenerativos, hiperplásicos ou displásicos). Não foi possível concluir o diagnóstico por meio do exame citológico em 7,28% dos casos e não haviam informações diagnósticas registradas em 1,37% dos casos. Dentre os animais que mais receberam indicação para exame citológico destacaram-se os caninos (92,06%), seguidos pelos felinos (4,08%), bovinos (1,77%) e equinos (1,30%). Lesoes neoplásicas foram as mais comumente observadas e foram mais prevalentes nos cães (59,18%), afetando principalmente animais com idade média de 119,60 meses e principalmente fêmeas (61,61%). O incremento anual observado ao longo dos 15 anos de aplicação da citologia demonstra que o exame citológico é um método de suma importância para confirmar, sugerir ou afastar o diagnóstico de diversas afecções, inclusive neoplasia, em todas as espécies animais.

Foram realizados três ensaios com o objetivo de se determinar o nível de toxicidade da contaminação de sementes de Cassia occidentalis na alimentação de frangos de corte. Nos dois primeiros ensaios foram utilizadas 640 aves de 1 dia de idade. Os níveis de adição da semente no primeiro ensaio foram, 0; 2; 4 e 6%, obtendo-se 5,77; 84,62; 100 e 100% de mortalidade, respectivamente. No segundo ensaio, os níveis utilizados foram 0; 0,5; 1 e 2%, obtendo-se 0; 3,29; 15,73 e 89,47% de mortalidade, respectivamente. No terceiro ensaio, utilizou-se 960 aves de 3 dias de idade. Adicionou-se a ração inicial (4-31 dias de idade) e final (32-52 dias) sementes moídas de fedegoso aos níveis de 0; 0,1; 0,2; 0,3; 0,4 e 0,5%. O peso médio final das aves foi 2,01; 1,95; 1,95; 1,90; 1,77 e 1,58 kg, respectivamente, observando-se diferença significativa (P < 0,05%) em relação ao controle para 0,3; 0,4 e 0,5% de adição. Os valores médios de consumo (4,33; 4,32; 4,32; 4,28; 4,08 e 3,80 kg, respectivamente) e da conversão alimentar (2,15; 2,21; 2,22; 2,25; 2,31 e 2,41, respectivamente) foram significativamente diferentes (P < 0,05%) para 0,4 e 0,5% de adição das sementes em relação ao grupo controle. O aspecto histológico de órgãos e tecidos das aves que receberam C.occidentalis, sacrificadas no término das fases inicial e final, foi característico de um processo degenerativo observado no coração, fígado, pâncreas, rins, músculo estriado esquelético e intestinos. Os percentuais de mortalidade foram 2,77; 2,08; 2,08; 0,69; 0,69 e 0%, respectivamente.

The RT of domestic quail from Italian variety showed mainly an albuginic pattern being represented by tortuous channels lined predominately by a simple cubic epithelium. RT channels extended along the testicular albuginea and penetrates into the epididymal region (ER) through its myoconnective matrix. Passageways were continuous to proximal efferent ducts of the ER. Epithelium lining ultrastructure of RT passageways showed some differences between the spring and the inactive phase at middle fall, concerning the quail testicular reproductive cycle. The features observed in RT epitheliocytes in fall were the low cytoplasmic electrodensity, paucity of supranuclear vesicles, which were abundant and variable in form and shape in spring, and some degenerative aspects of cell organelles mainly in ER lamellae. Moreover, presence of apical cytoplasmic extrusions were verified in the fall. No marked features were seen in the RT ultrastructure during the winter and summer comparatively to the active phase of spring.

The present study describes the morphology and ultrastructure of the salivary glands of semi-engorged females of the southern cattle-tick Rhipicephalus (Boophilus) microplus. The acini that compose these glands, at that specific feeding stage, show cells featuring degenerative process of the salivary glands, such as: vacuolated cytoplasm, condensed chromatin, fragmented nuclei, and presence of apoptotic bodies. In addition, the presence of microorganisms was detected, with morphology typical of protozoa, inside these organs. (c) 2005 Elsevier Ltd. All rights reserved.

This study describes the morphology of salivary glands of Rhipicephalus (Boophilus) microplus female ticks at beginning of feeding (24-48 h of attachment) and semi-engorged (4-5 days of attachment) to verify the degenerative characteristics of these organs and the secretory phase in which the process begins. At the beginning of feeding, secretion granules had been observed only in the cytoplasm of cells b, c(1), c(2), c(4) (type II acinus) and d (type III acinus), as well as large nuclei with regular and preserved morphology. In the semi-engorged females the acini presented few normal cells, few partially preserved ones, and the remaining ones in several stages of degeneration, that is, with retraction and cytoplasmic vacuolization, and nuclei with chromatin in several stages of condensation, picnotic and/or in fragmentation. In type I acinus and in the excretory ducts of the studied glands, at both feeding stages, no degenerative characteristic was observed. In females of R. (B.) microplus, the salivary glands degenerate asynchronically and precociously when compared with those of others tick's species. (c) 2006 Elsevier B.V. All rights reserved.

This study presents the morphology of the ovary, as well as the dynamics of the vitellogenesis process in oocytes of the cattle-tick Boophilus microplus. The ovary of these individuals is of the panoistic type; therefore, it lacks nurse cells. This organ consists of a single tubular structure, continuous, and composed of a lumen delimitated by a wall of small epithelial cells with rounded nuclei. In this tick species, the oocytes were classified into six stages varying from I to VI and according to: cytoplasm appearance and presence of the germ vesicle, yolk granules, and chorion. Oocytes of various sizes and at different developmental stages remain attached to the ovary through a cellular pedicel until completing stage V. Afterwards, they are liberated into the lumen and from there to the exterior. Some oocytes (classified as type VI) showed an atypical appearance indicating that some of the cellular components would be undergoing a degenerative process and/or reabsorption. The secretory cycle of hypopharyngeal glands (HPGs) in Scaptotrigona postica resembles that of Apis mellifera: in newly emerged workers the HPGs are in prefunctional state, their maximum development happens in the nurse workers and in forager workers they show signs of reabsorption. In S. postica these glands are also present in queens and males where they are more developed in newly emerged individuals. The ultrastructural features of the HPG secretory cycle in workers of S. postica and A. mellifera are alike: granular endoplasmic reticulum well developed, large secretion masses around the intracellular canaliculus in nurse workers and extensive degenerative structures in forager workers. Then it is suggested that the HPG secrete similar substances in both species. A second secretory cycle seems to occur in early foragers, may be with production of enzymes. The role of the HPGs in queens and males remains unknown but one possibility is enzyme production.

In a previous study, we showed that the Polybia paulista wasp venom causes strong myonecrosis. This study was undertaken to characterize the myotoxic potency of mastoparan (Polybia-MPII) isolated from venom (0.25 mu g/mu l) and injected in the tibial anterior (TA) muscle (i.m.) of Balb/c mice. The time course of the changes was followed at muscle degenerative (3 and 24 h) and regenerative (3, 7, and 21 days) periods (n = 6) after injection and compared to matched controls by calculation of the percentage of cross-sectional area affected and determination of creatine kinase (CK) activity (n = 10). The results showed that although NIP was strongly myotoxic, its capacity for regeneration was maintained high. Since the extent of tissue damage was not correlated with the CK serum levels, which remained very low, we raised the hypothesis that the enzyme underwent denaturation by the peptide. Evidence suggested that MP induced the death of TA fibers by necrosis and apoptosis and had the sarcolemma as its primordial target. Given its amphiphilic polycationic nature and based on the vast spectrum of functions attributed to the peptide, we suggest that MP interaction with cell membrane impaired the phosphorylation of dystrophin essential for sarcolemma mechanical stability, and disturbed Ca2+ mobilization with obvious implications on sarcoplasmic reticulum and mitochondrial functioning. The morphological and histochemical features of degeneration in honeybee (Apis mellifera) salivary glands were investigated in 5th instar larvae and in the pre-pupal period. The distribution and activity patterns of acid phosphatase enzyme were also analysed. As a routine, the larval salivary glands were fixed and processed for light microscopy and transmission electron microscopy. Tissue sections were subsequently stained with haematoxylin-eosin, bromophenol blue, silver, or a variant of the critical electrolyte concentration (CEC) method. Ultrathin sections were contrasted with uranyl acetate and lead citrate. Glands were processed for the histochemical and cytochemical localization of acid phosphatase, as well as biochemical assay to detect its activity pattern. Acid phosphatase activity was histochemically detected in all the salivary glands analysed. The cytochemical results showed acid phosphatase in vesicles, Golgi apparatus and lysosomes during the secretory phase and, additionally, in autophagic structures and luminal secretion during the degenerative phase. These findings were in agreement with the biochemical assay. At the end of the 5th instar, the glandular cells had a vacuolated cytoplasm and pyknotic nuclei, and epithelial cells were shed into the glandular lumen. The transition phase from the 5th instar to the pre-pupal period was characterized by intense vacuolation of the basal cytoplasm and release of parts of the cytoplasm into the lumen by apical blebbing; these blebs contained cytoplasmic RNA, rough endoplasmic reticule and, occasionally, nuclear material. In the pre-pupal phase, the glandular epithelium showed progressive degeneration so that at the end of this phase only nuclei and remnants of the cytoplasm were observed. The nuclei were pyknotic, with peripheral chromatin and blebs. The gland remained in the haemolymph and was recycled during metamorphosis. The programmed cell death in this gland represented a morphological form intermediate between apoptosis and autophagy.

The modern approach to the development of new chemical entities against complex diseases, especially the neglected endemic diseases such as tuberculosis and malaria, is based on the use of defined molecular targets. Among the advantages, this approach allows (i) the search and identification of lead compounds with defined molecular mechanisms against a defined target (e.g. enzymes from defined pathways), (ii) the analysis of a great number of compounds with a favorable cost/benefit ratio, (iii) the development even in the initial stages of compounds with selective toxicity (the fundamental principle of chemotherapy), (iv) the evaluation of plant extracts as well as of pure substances. The current use of such technology, unfortunately, is concentrated in developed countries, especially in the big pharma. This fact contributes in a significant way to hamper the development of innovative new compounds to treat neglected diseases. The large biodiversity within the territory of Brazil puts the country in a strategic position to develop the rational and sustained exploration of new metabolites of therapeutic value. The extension of the country covers a wide range of climates, soil types, and altitudes, providing a unique set of selective pressures for the adaptation of plant life in these scenarios. Chemical diversity is also driven by these forces, in an attempt to best fit the plant communities to the particular abiotic stresses, fauna, and microbes that co-exist with them. Certain areas of vegetation (Amazonian Forest, Atlantic Forest, Araucaria Forest, Cerrado-Brazilian Savanna, and Caatinga) are rich in species and types of environments to be used to search for natural compounds active against tuberculosis, malaria, and chronic-degenerative diseases. The present review describes some strategies to search for natural compounds, whose choice can be based on ethnobotanical and chemotaxonomical studies, and screen for their ability to bind to immobilized drug targets and to inhibit their activities. Molecular cloning, gene knockout, protein expression and purification, N-terminal sequencing, and mass spectrometry are the methods of choice to provide homogeneous drug targets for immobilization by optimized chemical reactions. Plant extract preparations, fractionation of promising plant extracts, propagation protocols and definition of in planta studies to maximize product yield of plant species producing active compounds have to be performed to provide a continuing supply of bioactive materials. Chemical characterization of natural compounds, determination of mode of action by kinetics and other spectroscopic methods (MS, X-ray, NMR), as well as in vitro and in vivo biological assays, chemical derivatization, and structure-activity relationships have to be carried out to provide a thorough knowledge on which to base the search for natural compounds or their derivatives with biological activity.
