Idiosyncratic hepatotoxicity is a well-known complication associated with aromatic antiepileptic drugs (AAED), and it has been suggested to occur due to the accumulation of toxic arene oxide metabolites. Although there is clear evidence of the participation of an immune process, a direct toxic effect involving mitochondria dysfunction is also possible. The effects of AAED on mitochondrial function have not been studied yet. Therefore, we investigated, in vitro, the cytotoxic mechanism of carbamazepine (CB), phenytoin (PT) and phenobarbital (PB), unaltered and bioactivated, in the hepatic mitochondrial function. The murine hepatic microsomal system was used to produce the anticonvulsant metabolites. All the bioactivated drugs (CB-B, PB-B, PT-B) affected mitochondrial function causing decrease in state three respiration, RCR, ATP synthesis and membrane potential, increase in state four respiration as well as impairment of Ca(2+) uptake/release and inhibition of calcium-induced swelling. As an unaltered drug, only PB, was able to affect mitochondrial respiration (except state four respiration) ATP synthesis and membrane potential; however, Ca(2+) uptake/release as well as swelling induction were not affected. The potential to induce mitochondrial dysfunction was PT-B > PB-B > CB-B > PB. Results suggest the involvement of mitochondrial toxicity in the pathogenesis of AAED-induced hepatotoxicity. In social insects, cuticular hydrocarbons are involved in species, kin, caste and nestmate recognition. Gas chromatography and mass spectrometry were used to compare the cuticular hydrocarbon composition of workers, males and queens of Melipona bicolor. The cuticular hydrocarbon composition of this species was found to consist mainly of C23, C25:1, C25, C27:1, C27, C29:1 and C29, which are already present in imagoes that have not yet abandoned the brood cell. This composition varied quantitatively and qualitatively between and within the castes and sexes. The newly emerged workers and young queens (virgins) had similar cuticular hydrocarbon profiles, which were different from those of the males. When the females start executing their tasks in the colony, the cuticular hydrocarbon profile differences appear. The workers have less variety, while the queens conserve or increase the number of cuticular hydrocarbon compounds. The queens have more abdominal tegumentary glands than the workers, which apparently are the source of the new cuticular compounds.

Gastric ulcer is an excoriated area of the gastric mucosa. It is among the predominant gastrointestinal chronic diseases. The essential oils represent an important part of the traditional pharmacopoeia in many countries and have been successfully used for gastroprotection and ulcer healing. Thus, this review presents the experimental activity of essential oils traditionally used in the gastric ulcer prevention and treatment, considering their families, part of the plant studied, bioassays, and their mechanisms of gastroprotection and ulcer healing, with the aim of stimulating novel studies in the search for a new phytomedicine to treat gastric diseases. © 2012 The Authors Fundamental and Clinical Pharmacology © 2012 Société Française de Pharmacologie et de Thérapeutique.

Aromatic amines are environmental pollutants and represent one of the most important classes of industrial and natural chemicals. Some types of complex effluents containing these chemical species, mainly those originated from chemicals plants are not fully efficiently treated by conventional processes. In this work, the use of electrochemical technology through an electrolytic pilot scale flow reactor is considered for treatment of wastewater of a chemical industry manufacturer of antioxidant and anti-ozonant substances used in rubber. Experimental results showed that was possible to remove between 65% and 95% of apparent colour and chemical oxygen demand removal between 30 and 90% in 60 min of treatment, with energy consumption rate from 26 kWh m-3 to 31 kWh m-3. Absorbance, total organic carbon and toxicity analyses resulted in no formation of toxic by-products. The results suggest that the presented electrochemical process is a suitable method for treating this type of wastewater, mainly when pre-treated by aeration. This work describes the mutagenic response of Sudan III, an adulterant food dye, using Salmonella typhimurium assay and the generation of hazardous aromatic amines after different oxidation methods of this azo dye. For that, we used metabolic activation by S9, catalytic oxidation by ironporphyrin and electrochemistry oxidation in order to simulate endogenous oxidation conditions. The oxidation reactions promoted discoloration from 65% to 95% of Sudan III at 1×10-4molL-1 and generation of 7.6×10-7molL-1 to 0.31×10-4molL-1 of aniline, o-anisidine, 2-methoxi-5-methylaniline, 4-aminobiphenyl, 4,4'-oxydianiline; 4,4'-diaminodiphenylmethane and 2,6-dimethylaniline. The results were confirmed by LC-MS-MS experiments. We also correlate the mutagenic effects of Sudan III using S. typhimurium with the strain TA1535 in the presence of exogenous metabolic activation (S9) with the metabolization products of this compound. Our findings clearly indicate that aromatic amines are formed due to oxidative reactions that can be promoted by hepatic cells, after the ingestion of Sudan III. Considering that, the use of azo compounds as food dyestuffs should be carefully controlled. © 2013 Elsevier Ltd.

Oil is a hydrocarbon mixture of various sizes, including saturated and aromatic compounds. Natural gas is a mixture of gaseous hydrocarbons and its main component is methane. In our society, the great demand for these fuels requires fast extraction, transportation and refining, increasing the number of accidents that compromise the environment. Oil is a finite resource and it is necessary to reduce the problems related to the question concerning environmental pollution which has encouraged the search for alternative fuel sources in our country. So today we have two major biofuels: ethanol and biodiesel. Concurrently, many studies have been done directed toward the isolation of microorganisms capable of degrading petrochemical industrial wastes, most of them using as a source of isolation soil and water collected in a contaminated environment. Isolation from alternative substrates has emerged as a new strategy that has provided satisfactory results. In this work, we present the leaf-cutter ants of the Attini tribe as a source for the isolation of micro-fungi with the potential for hydrocarbon degradation. These insects have a social way of life and a highly specialized system of intra and interspecific communication, which is based on the recognition of individuals through volatile chemical compounds, the majority hydrocarbons, stored in their exoskeleton. The micro-environment exoskeleton of Attini ants (genus Atta) used in this work proved to be a rich source of microbial biodiversity, as other studies have found. The flotation isolation technique applied here allowed the achievement of 214 micro-fungi, 118 representatives of the dematiaceous fungi group and 96 hyaline filamentous fungi. They were submitted to toluene degradation tests and at least one strain of each genus presented good results, namely Teratosphaeria, Exophiala, Cladosporium, Penicillium, Aspergillus... Spills can ocurr during oil productive chain and contaminate various environments due to the toxicity of monoaromatics hidrocarbons. Toluene stands out for being agressive to the nervous sistem and teratogenic, with high mobility and solubility in water, which facilitates environmental impact. Studies show that fungi are potential aromatic compounds assimilators, encouraging new researches about its use on the recovery of contaminated sites. This study aimed to select and characterize fungus with potential for biorremediation of toluene. 50 fungi were selected of the Collection of Microorganisms of Interest for Oil Gas and Biofuels, of UNESP Rio Claro, all of which were isolated from sites contaminated with monoaromatic hydrocarbons. Two trials were realized to select the microorganism with greater potential. The first test evaluated fungal growth under toluene saturated atmosphere. 24 fungi were chosen because its greater biomass production to participate in the next trial, the degradation in plates test, where the blue redox agente, DCPIP, indicates the degradation reaction, turning colorless. From this teste was possible to select one isolate which showed higher growth and stronger medium discoloration as the microorganism with the greatest potential to assimilate toluene. The Trichoderma cf. koningii had its potential evaluated through gas cromatography. The experiment proved the efficiency of the methodology, with positives results from the method validation and the effectiveness demonstrated of the LA-PHA-PACK bottles to prevent the volatilization of toluene during the 21 days of experiment. Being reliable its use for monitoring toluene decay associating it with degradation. This results are important because there aren't many methodologies and vials efficient to the purpose of this work. A multicomponent reaction of β-naphthol, dimethyl malonate, and aromatic aldehydes in the presence of NbCl5 as promoter is described. Under similar conditions, aromatic aldehydes with different substituents exhibited different behaviors than β-naphthol and dimethyl malonate. In these MCRs, 4-aryl-3,4-dihydrobenzo[f]coumarins are obtained as the major products (41–93%) and 14-aryl-14H-dibenzo[a,j]xanthenes as the minor products (1–38%).
