O objetivo deste trabalho foi desenvolver um oligonucleotídeo iniciador para reação em cadeia da polimerase (PCR) específico para as estirpes de Xylella fastidiosa que causam o mal de Pierce (PD) em videira (Vitis vinifera). Amplificações de DNA de 23 diferentes hospedeiros, usando o conjunto de oligonucleotídeos REP1-R (5'-IIIICGICGIATCCIGGC-3') e REP 2 (5'-ICGICTTATCI GGCCTAC-3') utilizando o programa: 94 ºC/2 min; 35 X (94 ºC/1 min, 45 ºC/1 min; 72 ºC/1 min and 30 s) 72 ºC/5 min, produziu um fragmento de 630 pb que diferenciou as estirpes de videiras dos demais. Entretanto, padrões de bandeamento REP não são considerados confiáveis para detecção devido ao par de oligonucleotídeos REP 1 e REP 2 corresponderem a seqüências repetitivas encontradas por todo o genoma bacteriano. Desse modo, o produto amplificado de 630 pb foi eluído do gel de agarose, purificado e seqüenciado. A informação da seqüência nucleotídica foi usada para identificar e sintetizar um oligonucleotídeo específico para o isolado de X. fastidiosa causadora do mal de Pierce denominado Xf-1 (5'-CGGGGGTGTAGGAGGGGTTGT-3'), que foi utilizado juntamente com o oligonucleotídeo REP-2 nas condições 94 ºC/2 min; 35 X (94 ºC/1 min, 62 ºC/1 min; 72 ºC/1 min and 30 s) 72 ºC/10 min. Os DNAs das estirpes de X. fastidiosa de outros hospedeiros [amêndoa (Prumus amygdalus), citros (Citrus spp.), café (Coffea arabica), olmo (Ulmus americana), amora (Morus rubra), carvalho (Quercus rubra), vinca (Catharantus roseus), ameixa (Prunus salicina) e ragweed (Ambrosia artemisiifolia)] e de bactérias Gram negativas e positivas foram submetidos a amplificação com o conjunto de oligonucleotídeos Xf-1/REP 2. Um fragmento, de aproximadamente 350 pb, foi amplificado apenas com o DNA de X. fastidiosa isolada de videira.

Em janeiro de 2003 foram constatados sintomas típicos de ferrugem em folhas maduras de videira (Vitis vinifera), nas áreas experimentais da Universidade Estadual Paulista, Campus de Ilha Solteira, em Selvíria - MS e Ilha Solteira - SP. Este é o primeiro relato desta doença da videira no Centro-Oeste do Brasil. Considerando a intensidade da doença, a importância da cultura da videira na região e a falta de informações sobre a ferrugem, os objetivos deste trabalho foram: reconfirmar a patogenicidade de Phakopsora euvitis; estudar a influência da luz e da temperatura na germinação de urediniósporos; estudar o efeito do tipo de superfície na germinação de urediniósporos; determinar a longevidade in vivo de urediniósporos e avaliar a eficiência agronômica de fungicidas para o controle da ferrugem. Sintomas da ferrugem foram verificados somente em folhas maduras de videira. A temperatura de 25 ºC, no escuro, e a superfície inferior da folha de videira foram as melhores condições para a germinação de urediniósporos de P. euvitis. A germinação dos urediniósporos foi reduzida em 94%, em folhas de videira que foram retiradas das plantas e mantidas durante sete dias na superfície do solo. em dois experimentos realizados em campo constatou-se que os fungicidas tebuconazole, propiconazole e azoxystrobin proporcionaram as menores intensidades da doença. Tebuconazole apresentou-se como o fungicida mais eficiente no controle da ferrugem. No geral, maior intensidade da ferrugem foi observada em videira conduzida no sistema de espaldeira.

O objetivo deste trabalho foi selecionar o pré-tratamento químico mais apropriado para a secagem de uvas cv. Rubi para a produção de passas. Foram obtidas curvas de secagem convectiva com ar a 50ºC, em um secador de bandejas, para uvas submetidas a pré-tratamentos químicos com diferentes concentrações de carbonato de potássio e azeite de oliva, e diferentes tempos de imersão, de acordo com planejamentos fatoriais. Também foram obtidas curvas de secagem convectiva, para uvas pré-tratadas em suspensões aquosas de lecitina de soja, em várias concentrações de lecitina e diferentes tempos de imersão. O modelo de Page foi ajustado às curvas de secagem experimental, e os tempos de secagem calculados mostraram que o melhor pré-tratamento consistiu na imersão das uvas por 2 minutos, em uma emulsão de 5% de azeite de oliva e 6% de K2CO3, a 50ºC, o que resultou em tempos de secagem próximos aos do pré-tratamento com 2,5% de azeite de oliva, mas com um menor consumo dessa substância. A©m disso, a imersão das uvas em uma suspensão aquosa de 2% de lecitina de soja, a 50ºC, por 5 minutos, resultou em um tempo de secagem total apenas levemente superior ao do pré-tratamento mais efetivo.

Xylella fastidiosa causes citrus variegated chlorosis (CVC) disease in Brazil and Pierce's disease of grapevines in the United States. Both of these diseases cause significant production problems in the respective industries. The recent establishment of the glassy-winged sharpshooter in California has radically increased the threat posed by Pierces disease to California viticulture. Populations of this insect reach very high levels in citrus groves in California and move from the orchards into the vineyards, where they acquire inoculum and spread Pierce's disease in the vineyards. Here we show that strains of X. fastidiosa isolated from diseased citrus and coffee in Brazil can incite symptoms of Pierce's disease after mechanical inoculation into seven commercial Vitis vinifera varieties grown in Brazil and California. Thus, any future introduction of the CVC strains of X. fastidiosa into the United States would pose a threat to both the sweet orange and grapevine industries. Previous work has clearly shown that the strains of X. fastidiosa isolated from Pierce's disease- and CVC-affected plants are the most distantly related of all strains in the diverse taxon X. fastidiosa. The ability of citrus strains of X. fastidiosa to incite disease in grapevine is therefore surprising and creates an experimental system with which to dissect mechanisms used by X.,fastidiosa in plant colonization and disease development using the full genome sequence data that has recently become available for both the citrus and grapevine strains of this pathogen.

This work aimed to study aspects related to vegetative and productive development of plants and characteristics and chemical components of bunches of six seedless grape varieties at environmental conditions of São Francisco river's valley, Northeastern Brazil. The experiment was carried out during 1997 and 1998. The varieties tested were grafted on IAC 572 ('Jales') rootstock. The experimental model utilized was totally randomized, and with parcels subdivided into different production cycles. The results showed significative differences among varieties at different pruning times. The average weight of bunches varied from 164.8 g for 'Marroo Seedless' and 203.5 g for 'Beauty Seedless'. The average diameter of berries was superior to 15.7 mm for all of the varieties. The total soluble solids varied from 14.05°Brix in 'Canner' to 19.6°Brix in 'Venus'. The total titratable acidity (ATT) was lower than 0.91 g tartaric acid/100 mL of juice. The ratio SST/ATT varied from 19.05 in 'Beauty Seedless' to 28.57 in 'Vênus'. 'Vênus' and 'Marroo Seedless' were the most productive varieties, with the average annual production of 24 ton/ha and 20 ton/ha, respectively.

Purpose: This paper aims to evaluate the consumer acceptance of Bordô and Isabel wines from innovative winemaking in order to increase red wine consumption by consumers to obtain nutritional benefits. Design/methodology/approach: All wines were produced by a standard procedure of vinification. Pre-drying treatment aimed at drying the grapes up to 22°Brix and static pomace wines presented the constant contact between the must and pomace. Sensory acceptance was carried out by 80 consumers who evaluated eight samples (six experimental wines and two commercial wines) concerning the attributes: appearance, aroma, body, flavor and overall acceptance. Findings: Experimental wines presented higher acceptance when compared to commercial wines and cluster analysis shows the splitting of consumer preferences, highlighting the higher acceptance of traditional Bordô wine and static pomace samples in all sensory attributes. Pre-drying process enhances the concentration of coloured compounds, to highlight the appearance acceptance of these samples. Research limitations/implications: Although this paper has limited the consumer acceptance of red table wines, the innovative treatments will be applied in Vitis vinifera red wines. Practical implications: The wide acceptance of static pomace and pre-drying wines has promoted further information about innovative winemaking that can be applied in Brazilian and worldwide wineries. Social implications: The innovative treatments can change sensorial features of wines and therefore influence the choice of consumers as well as enhancing the nutritional benefits of red wines. Originality/value: The pre-drying and static pomace winemaking are the novelty of this study and the wide acceptance of the sensory attributes concerning these treatments indicates the potential for application in wineries. © Emerald Group Publishing Limited.

The almost complete absence or misdistribution of water as a natural resource frequently constitute a limiting factor for plant growth and development in the semi-arid northeastern Brazil. In this context, the use of appropriate irrigation techniques is an essential and indispensable factor for proper functioning of the primary and secondary metabolisms in plants. This study aimed to assess the metabolism of the Syrah grapevine in the semi-arid northeastern Brazil, by using three irrigation strategies (controlled deficit irrigation [CDI], deficit irrigation [DI], and full irrigation [FI]). The research was conducted at Embrapa Semiarid (Embrapa-Brazilian Agricultural Research Corporation), Experimental Field of Bebedouro, municipality of Petrolina, state of Pernambuco, Brazil. The statistical design was randomized blocks, with a 3 x 6 factorial scheme. Six samplings were performed throughout the experiment cycle at 54, 60, 73, 87, 101, and 115 days after pruning (DAP). We analyzed the total soluble sugars, reducing sugars, total soluble protein, and invertase activity. Analysis of variance and the F test were performed for all analyzed variables. The means were compared using the Tukey test at 5% significance. At the end of the experiment cycle, DI was found to increase the average acid invertase activity in the plant cell wall. Further, by the end of the experiment cycle, the total soluble sugars and reducing sugars increased in all plants in the three irrigation strategies. Thus, we conclude that it is possible to reduce water consumption in vineyards of the semi-arid northeastern Brazil, without significantly affecting the protein and sugar metabolisms in the plants.

Brazilian wines are appreciated by consumers for having fruity odor and taste which are typical from Vitis labrusca cultivars. The rusticity of these cultivars, the winemaking and the physicochemical properties are some of the key factors to obtain a quality beverage. In this context, the aim of this article was to analyze the quality of three white wines from wineries located in northwest region of São Paulo state by determining physicochemical properties and sensory profile. Data were analyzed by statistical tests and by Principal Component Analysis (PCA). It was possible to observe statistical differences (P<0,05) in most physicochemical properties. Furthermore, the PCA showed that the attributes body and color significantly explained the variance of the first and second component, respectively, higlighting the Vitis vinifera wine. Thus the connection between the physicochemical and sensory properties of white wines was important for characterization of these beverages produced in the northwest of São Paulo.
