The muscles can perform the same function in a specific segment (muscles of fast and slow contraction), and at the same time be antagonistic in relation to muscular action (flexors or extensors). The present research aimed to study the morphology, frequency and metabolism of fiber types and the contractile characteristics of extensor and flexors muscles of rabbit. We studied muscles anterior tibialis (AT), flexor digitorum supeficialis (FDS), extensor digitorum longus (EDL) and posterior tibialis (PT). The muscles were submitted to the techniques HE, NADH-TR and myofibrillar ATPase. In EDL and PT extensor muscles, the frequencies of red (SO + FOG) and white fibers (FG) were 68.77% and 31.23% versus 58.87% and 41.13%, respectively. In the AT and FDS flexor muscles, these frequencies were 75.14% and 24.86% versus 73.89% and 26.11%, respectively. In extensor muscles, the percentage of slow contraction fibers was 8.05% in EDL and 9.74% in PT, and in fast contraction, 91.95% in EDL and 90.26% in PT. In flexors, the slow contraction frequencies were 12.35% in AT and 8.17% in FDS, and in fast contraction, 87.65% and 91.83%, respectively. Skeletal muscles with antagonistic muscular actions (flexors and extensors) the morphological, contractile and metabolic characteristics are identical.

The salivary glands of Rhipicephalus sanguineus males at stages: unfed (control), at day seven post-attachment, and at days three and seven post-detachment from the host were examined using methods of enzymatic analysis and cell viability. At these stages of feeding, different staining patterns were observed in the cells of type IV, III, II and I acini, which were affected by degeneration in this sequence. Acid phosphatase reaction was inversely proportional to that of ATPase, while ATPase reaction was proportional to membrane integrity.Salivary gland cells of unfed males exhibited intact nucleus and plasma membrane, suggesting that the acid phosphatase detected may participate in the normal physiology of acini. In males at day seven post-attachment, intact membranes were observed in almost all types of acini, as well as stronger reaction for acid phosphatase, nuclear changes, and decrease in ATPase reaction, changes associated with the degenerative process. At days three and seven post-detachment degeneration progress, being observed loss of membrane integrity, nuclear changes, prominent decrease in ATPase reaction, and an increase in acid phosphatase reaction in the first case and a decreased of it at day seven post-detachment from the host. During cell death, alterations occurred in the following sequence: a) nuclear changes, b) loss of ATPase reaction, c) loss of integrity of the plasma membrane, and d) increase of acid phosphatase. The latter might be associated with the late degradation of cytoplasmic remnants, characterizing the process of cell death in glands of R. sanguineus males as atypical or non-classic apoptosis. Ants frequently harvest seeds from faeces of frugivorous vertebrates. By transporting these seeds to nests, ants may influence seed dispersal success of plants. Seed removal by ants from vertebrate faeces is influenced by the seed species involved. Faeces from different vertebrates differ in chemical composition and physical characteristics. It remains unclear, however, whether the faeces itself affects the ant-seed interaction. In this study experimental seed-containing faecal portions were prepared using defecations of birds, marsupials, and monkeys and seeds of two aroids (Philodendron corcovadense and P. ppendiculatum, Araceae) and one liana species (Schlegelia parviflora, Bignoniaceae). Faecal portions were arranged along a transect established in the understory of a lowland rainforest in southeast Brazil. For P. appendiculatum the probability of detection and the proportion of seeds removed were identical between marsupial and monkey faeces. For P. corcovadense and S. parviflora, the probability of detection was affected by seed species and, apparently, also by the interaction between seed species and type of faeces (P = 0.097), but not by the type of faeces itself (bird or monkey). Both factors (i.e., seed species and type of faeces) affected the proportion of seeds removed (faeces type was marginally significant; P = 0.08), whereas the interaction between them was not significant. The results indicate that seed species affects seed removal by ants, while the type of faeces probably interacts with seed traits to influence faeces detection.

This study evaluated anteroposterior trunk movements and the time spent on activities of load-carrying to surfaces at different heights, among experienced and inexperienced individuals. Thirty-six healthy males (16 experienced and 20 inexperienced in load-carrying) had their trunk movements recorded by an electrogoniometer while transporting boxes (7 and 15 kg) to surfaces of variable heights (low, low intermediate, high intermediate and high). Longer time was spent on trunk flexion when carrying loads to low surfaces and on trunk extension when carrying to high surfaces (p<0.05). Differences in time spent on trunk flexion/extension were identified between loads, and between groups for the heavier load. There were no differences in flexion/extension amplitudes between groups or loads. Although unnecessary flexion/extension occurring prior to a task may increase the exposure of the trunk to risky movements, the amplitudes reported here were relatively small and seemed to have occurred to facilitate the final tasks.Relevance to industryLifting devices and technological assistance are increasingly available in industrial settings. However, distribution centers and delivery services represent new demands on workers and involve an expressive amount of manual handling and carrying activities. omeprazole is a substituted benzimidazole which suppresses gastric-acid secretion by means of H+, K+-ATPase inhibition. It is an optically active drug with the sulfur of the sulfoxide being the chiral center. This pro-drug can be easily converted into its respective sulfenamide at low pH. In this work, omeprazole has been studied in relation to racemization barrier and decomposition reaction. Quantum chemistry coupled to PCA chemometric method were used to find all minimum energy structures. Conformational analysis and calculation of racemization barriers were carried out by PM3 semiempirical method (Gaussian 98). The average racemization energy barrier for all minimum energy structures (43.56 kcal mol(-1)) can be related to the velocity constant in Eyring's equation. The enormous half-life time at 100 degrees C (9.04 x 10(4) years) indicates that the process cannot be observed in human time scale. on the other hand, the difference of free energy change (Delta(Delta G) = -266.78 kcal mol(-1)) for the decomposition reaction shows that the process is favorable to the sulfenamide formation. The highly negative Delta(Delta G) obtained for the decomposition reaction shows that this process is extremely exothermic. This result explains why omeprazole decomposes and does not racemize. We model the electrical behavior of organic light-emitting diodes whose emissive multilayer is formed by blends of an electron transporting material, tris-(8-hydroxyquinoline) aluminum (Alq(3)) and a hole transporting material, N,N-'-diphenyl-N,N-'-bis(1,1(')-biphenyl)-4,4-diamine. The multilayer is composed of layers of different concentration. The Alq(3) concentration gradually decreases from the cathode to the anode. We demonstrate that these graded devices have higher efficiency and operate at lower applied voltages than devices whose emissive layer is made of nominally homogeneous blends. Our results show an important advantage of graded devices, namely, the low values of the recombination rate distribution near the cathode and the anode, so that electrode quenching is expected to be significantly suppressed in these devices. FUNDAMENTO: Vários autores mostraram que a deterioração da função cardíaca associa-se com o grau e a duração da obesidade. Os padrões de expressão gênica após longos períodos de obesidade precisam ser estabelecidos. OBJETIVO: Este estudo testou a hipótese de que a exposição prolongada à obesidade leva à redução nos níveis de RNAm de proteínas envolvidas na homeostase do Ca2+ miocárdico. Além disso, este estudo avaliou se uma diminuição no hormônio tireoidiano causava redução na expressão de RNAm. MÉTODOS: Ratos Wistar machos de 30 dias de idade foram distribuídos em dois grupos: controle (C) e obeso (Ob). O grupo C recebeu uma dieta padrão e o grupo Ob recebeu dietas hiperlipídicas por 15, 30 e 45 semanas. A obesidade foi definida pelo índice de adiposidade. A expressão gênica foi avaliada por PCR em tempo real quantitativa. RESULTADOS: O índice de adiposidade foi maior no grupo Ob do que no C em todas as etapas. Enquanto a obesidade nas semanas 15 e 45 determinou uma redução no RNAm de Ca2+-ATPase do retículo sarcoplasmático (SERCA2a), trocador Na+/Ca2+ (NCX) e calsequestrina (CSQ), observou-se aumento da expressão do RNAm de canal de Ca2+ do tipo L, receptor de rianodina, SERCA2a, fosfolamban (PLB), NCX e CSQ após a semana 30, em comparação ao grupo C. Não houve associação significativa entre os níveis de T3 e a expressão de RNAm. CONCLUSÕES: Nossos dados indicam que a obesidade por curtos ou longos períodos de tempo pode promover alteração na expressão gênica de proteínas reguladoras da homeostase do Ca2+ sem influência do hormônio tireoidiano

FUNDAMENTOS: A leishmaniose tegumentar americana permanece doença endêmica em diversas regiões brasileiras. A sobrevivência do parasita no interior dos macrófagos se deve, em parte, pela atividade de uma K+/H+-ATPase de membrana que pode ser inibida pelo omeprazol. OBJETIVOS: Avaliar a eficácia do omeprazol na prevenção do desenvolvimento de lesões de leishmaniose em hamsters. MÉTODOS: Empregaram-se 18 hamsters, divididos em três grupos: o grupo L recebeu apenas a inoculação de L. brasiliensis na pata anterior direita, o grupo O recebeu apenas doses diárias de 0,4mg de omeprazol subcutâneo, e o grupo L+O recebeu o inóculo de leishmanias e o tratamento com omeprazol desde o dia da inoculação. O estudo foi conduzido por 42 dias, realizaram-se medidas dos diâmetros das patas semanalmente, e, ao final do estudo, foram realizados esfregaços das lesões para verificação dos parasitas. RESULTADOS: Os hamsters dos grupos L e L+O desenvolveram lesões de leishmaniose tegumentar havendo ulceração em duas patas do grupo L e uma do grupo L+O. Ao final do estudo, a mobilidade e vitalidade no grupo L foram menores que em L+O, e estas menores que no grupo O. Os diâmetros das patas inoculadas nos grupos L e L+O foram significativamente maiores que no início do estudo (p<0.05). Não houve diferença significativa entre os diâmetros das patas dos grupos L e L+O ao final do estudo (p0,05), sendo detectados parasitas no esfregaço das lesões dos dois grupos. CONCLUSÕES: Omeprazol, no protocolo utilizado, não evitou o desenvolvimento de lesões de leishmaniose tegumentar em hamsters.

Solenopsis saevissima has a midgut composed of columnar, regenerative, and goblet cells. The midgut epithelium was covered by a basal lamina. Outside the basal lamina, layers of inner oblique, circular, and outer longitudinal muscles were present. Columnar cells showed a basal plasma membrane containing numerous folds, mitochondria, and the nucleus. Rough endoplasmic reticulum, Golgi bodies, membrane bounded vacuoles, and spherocrystals were found in this region. The apical plasma membrane was constituted by microvilli, which were above a region rich in mitochondria. Regenerative cells were found in groups lying by the basal lamina. Goblet cells were associated with an ion-transporting mechanism between the haemolymph and the midgut epithelium. These cells were lying by the midgut lumen and large microvilli were evident, but the cytoplasmic features were similar to the columnar cells.

Skeletal muscle fibers areas, the weight of body, carcass and muscle, the gastric content and the length of intestines and caecum of Norfolk rabbit from neonate to 26th week were measured. Types of fibers and areas were studied in -70-degrees-C frozen blocks of flexor carpo radialis muscle. Some sections were stained with Haematoxylin and Eosin and others were submitted to SDH, NADH-TR and m-ATPase, followed by acid and alkali preincubations. All parameters revealed a high degree of correlation. Carcass weight/body weight ratio reached 55,1% at 10th week. From neonate to 26th week, the areas of SO, FOG and FG muscle fibers increased of x9.1, x12.4 and x22.2. While SO and FOG fibers tend to stabilize their growth at 10th week, FG fibers were active at this age. Due to early stabilization of SO and FOG fibers growth, the additional increase in muscle tissue in attributed mainly to hypertrophy of FG fibers.

Alkaline phosphatase from rat osseous plate is allosterically modulated by ATP, calcium and magnesium at pH 7.5. At pH 9.4, the hydrolysis of ATP and PNPP follows Michaelis-Menten kinetics with K0.5 values of 154 muM and 42 muM, respectively. However, at pH 7.5 both substrates exhibit more complex saturation curves, while only ATP exhibited site-site interactions. Ca2+-ATP and Mg2+-ATP were effective substrates for the enzyme, while the specific activity of the enzyme for the hydrolysis of ATP at pH 7.5 was 800-900 U/mg and was independent of the ion species. ATP, but not PNPP, was hydrolyzed slowly in the absence of metal ions with a specific activity of 140 U/mg. These data demonstrate that in vitro and at pH 7.5 rat osseous plate alkaline phosphatase is an active calcium or magnesium-activated ATPase.

The effect of uroguanylin (UGN) oil K(+) and H(+) secretion in the renal tubules of the rat kidney was studied using in vivo stationary microperfusion. For the study of K(+) secretion, a tubule was Punctured to inject a column of FDC-green-colored Ringer's solution with 0.5 mmol KCI/L 10(-6)(mol UGN/L, and oil was Used to block fluid flow. K(+) activity and transepithelial potential differences (PD) were measured with double microelectrodes (K(+) ion-selective resin vs. reference) in the distal tubules of the same nephron. During perfusion, K(+) activity rose exponentially, from 0.5 mmol/L to stationary concentration, allowing for the calculation of K(+) secretion J(K)). JK increased from 0.63 +/- 0.06 nmol.cm(-2).s(-1) in the control croup to 0.85 +/- 0.06 in the UGN group (p < 0.01). PD was -51.0 +/- 5.3 mV in the control group and -50.3 +/- 4.98 mV in the UGN group. In the presence of 10(-7) mol iberiotoxin/L, the UGN effect was abolished: JK was 0.37 +/- 0.038 nmol-cm(-2).s(-1) in the absence of, and 0.38 +/- 0.025 in the presence of, UGN. indicating its action oil rnaxi-K channels. In another series of experiments, renal tubule acidification was studied, using similar method: proximal and distal tubules were perfused with solutions containing 25 mmol NaHCO(3)/L. Acidification half-time was increased both in proximal and distal segments and, as a consequence, bicarbonate reabsorption decreased in the presence of UGN (in proximal tubules, from 2.40 +/- 0.26 to 1.56 +/- 0.21 nmol-cm(-2).s(-1)). When the Na(+)/H(+) exchanger was inhibited by 10(-4) mol hexamethylene amiloride (HMA)/L, the control and UGN groups were not significantly different. In the late distal tubule, after HMA, UGN significantly reduced J(HCO3)(-). indicating all effect of UGN oil H(+)-ATPase. These data show that UGN stimulated J(K)(+) by actin, oil maxi-K channels. and decreased J(HCO3)(-) by acting on NHE3 in proximal and H(+)-ATPase in distal tubules.
