A comparative study is reported between C-18 bonded silica gel and powdered polyethylene (PE) as sorbent for Cd, Cu, and Pb determination using ammonium diethyldithiophosphate (ADTP) as the complexing agent in a flow injection system. The complexes were formed in 0.14 mol L-1 HNO3 and processed in a simple flow system comprising a peristaltic pump, a manual injector-commutator, and a sorbent-packed minicolumn. Ethanol was selected as the eluent and analytes in the eluate were determined by flame atomic absorption spectrometry. The optimum concentration of the complexing agent was 0.1% (m/v) ADTP for Cu and Pb determination using either C-18 or PE, and 0.25% (m/v) ADTP for Cd determination using PE. The sample loading flow rates were 5.0, 3.6, and 3.0 mL min(-1) for Cu, Pb, and Cd, respectively. The best elution flow rate was 6.5 mL min(-1). For a 60-sec preconcentration time, the sampling rate was 40 h(-1) and the enrichment factors of 33, 36, and 11 times (C-18) or 18, 22, and 23 times (PE) were obtained for Cu, Pb, and Cd, respectively. The limits of detection (LOD) were 1.6 mug L-1 Cu, 11 mug L-1 Pb, and 2.0 mug L-1 Cd using C-18 or 2.9 mug L-1 Cu, 19 mug L-1 Pb, and 1.0 mug L-1 Cd using PE, respectively. The relative standard deviations (n = 12) were typically <2%, <2%, and <6% for Cd, Cu, and Pb, respectively. The recoveries of Cd, Cu, and Pb added to wine samples varied from 96-99%, 97-102%, and 90-99%, respectively, using C-18 or PE. Accuracy was checked for Cd, Cu, and Pb determination in six wine samples digested by block digestor and open-vessel microwave-assisted digestion systems. The results revealed that C-18 was more efficient for Cu and Pb determination, while PE was the best sorbent for Cd.

A procedure for the standardization of ensembles of gold nanodisk electrodes (NEE) of 30 nm diameter is presented, which is based on the analytical comparison between experimental cyclic voltammograms (CV) obtained at the NEEs in diluted solutions of redox probes and CV patterns obtained by digital simulation. Possible origins of defects sometimes found in NEEs are discussed. Selected NEEs are then employed for the study of the electrochemical oxidation of iodide in acidic solutions. CV patterns display typical quasi-reversible behavior which involves associated chemical reactions between adsorbed and solution species. The main CV characteristics at the NEE compare with those observed at millimeter sized gold disk electrodes (Au-macro), apart a slight shift in E1/2 values and slightly higher peak to peak separation at the NEE. The detection limit (DC) at NEEs is 0.3 mu M, which is more than one order of magnitude lower than DL at the Au-macro (4 RM). The mechanism of the electrochemical oxidation of iodide at NEEs is discussed. Finally, NEEs are applied to the direct determination of iodide at micromolar concentration levels in real samples, namely in some ophthalmic drugs and iodized table salt. (c) 2006 Elsevier B.V. An on-line dialysis flow system coupled to inductively coupled plasma mass spectrometry to determine trace elements in serum samples by isotope dilution is presented. Isotope dilution was performed on samples incubated with enriched Cu-65, Zn-66, Se-77 and Pb-206 for 24 h at 36degreesC prior to dialysis to quantified total element concentrations. The sample and acceptor solutions flowed through the dialysis unit with cellophane membrane placed in between the compartments. The serum sample (1 mL) was left to recycle in a closed path while the acceptor solution was continuously pumped along the dialyzer channel and through a cationic AG50W X-8 resin column. After 10 min, around 70% of Na, K and Cl migrate from the sample. Three replicate injections of 0.1 mL were performed for the clean sample after each separation step. The on-line coupling of the dialyzer to ICP-MS allowed isotope dilution for total element determination either in the cleaned sample or by eluting the cations retained in the resin to be carried out. Results demonstrated no matrix effects from alkaline elements or spectral interference from ArNa+ on Cu-63, ArCl+ on Se-77 and (SO2+)-S-34 on Zn-66. The precision of isotope ratio measurements for Cu and Zn was around 1% and for Se and Pb was around 2.5%. The values found for the reference serum sample IMEP-17 were in good agreement with the certified values for Cu, Zn and Se.
Effects of breeder feeding and trace mineral source on leg health and bone traits of broiler progeny

The purpose of this paper is to characterize the lability/inertness metal fractions complexed by aquatic humic substances (HS) in relation to pH, complexation time, and HS concentration. HS were preconcentrated by ultrafiltration and complexed with bivalent metal ions. These fractions were characterized by ion exchange with the chelating collector cellulose Hyphan by applying batch procedure. The metals were determined by atomic absorption spectrometry. The results show that the distribution coefficients, Kd, decreased with HS presence, and that the relative lability of metal fractions complexed by HS is dependent on variables such as pH, complexation time, and HS concentration. Until c.a. 15 min, the metal change between aquatic HS and ion exchanger occurs following a 2 order reaction. Afterwards, the remaining metal fraction in the HS reacts following a 1st order reaction. For traces of metal ions bound to dissolved HS, the lability orderPb > Mn > Cd, Ni > Cu is revealed. ©1997 Soc. Bras. Química.

Within the next decade, the improved version 2 of Global Ozone Monitoring Experiment (GOME-2), a ultraviolet-visible spectrometer dedicated to the observation of key atmospheric trace species from space, will be launched successively on board three EUMETSAT Polar System (EPS) MetOp satellites. Starting with the launch of MetOp-1 scheduled for summer 2006, the GOME-2 series will extend till 2020 the global monitoring of atmospheric composition pioneered with ERS-2 GOME-1 since 1995 and enhanced with Envisat SCIAMACHY since 2002 and EOS-Aura OMI since 2004. For more than a decade, an international pool of scientific teams active in ground-and space-based ultraviolet-visible remote sensing have contributed to the successful post-launch validation of trace gas data products and the associated maturation of retrieval algorithms for the latter satellites, ensuring that geophysical data products are/become reliable and accurate enough for intended research and applications. Building on this experience, this consortium plans now to develop and carry out appropriate validation of a list of GOME-2 trace gas column data of both tropospheric and stratospheric relevance: nitrogen dioxide (NO 2), ozone (O 3), bromine monoxide (BrO), chlorine dioxide (OClO), formaldehyde (HCHO), and sulphur dioxide (SO 2). The proposed investigation will combine four complementary approaches resulting in an end-to-end validation of expected column data products.

Saliva plays important roles in facilitation of a bloodmeal, lubrication of mouthparts, and parasite transmission for some vector insects. Salivary composition changes during the lifetime of an insect, and differences in the salivary profile may influence its functions. In this report, the amount and profile of salivary gland protein of the American visceral leishmaniasis vector Lutzomyia longipalpis (Lutz & Neiva, 1912) were analyzed at different times of insect development and diet. Protein content from unfed female sand flies increased significantly with age, and a significant difference was observed in sugar-fed females during the first 10 d of adult life. Salivary protein content sharply decreased 1 d after blood feeding, with gradual increase in concentration the following days. SDS-polyacrylamide gel electrophoresis analysis revealed that most polypeptides present in the saliva of sugar-fed also were present in the saliva of blood-fed females. Understanding changes in sand fly's saliva contents at distinct days after emergence and the influence of a bloodmeal in this aspect may reveal the role played by saliva during leishmaniasis transmission. © 2008 Entomological Society of America.

Were analyzed 29 samples, 16 wet and 13 dry cat food of different flavors. The iodine levels ranged from 2.7 to 3.4 (average 2.95 mg/kg/MS) in dry food and 2.9 to 4.0 (average 3.4 mg/kg/MS) in the wet food. Eight samples (27.6%) specified on the package the assurance level (maximum) of iodine, with a dry (2.0 mg/kg/diet) and seven wet (0.04 mg/kg/diet). All non-compliant, because the values were higher than declared. Cats fed commercial diets consume proportionally more iodine in wet food than cats fed dry diets, but both with high levels of iodine. All samples analyzed were above the amounts stated on the labels. Accordingly, the wet and dry commercial food for cats show no reliable values with those reported, resulting in a possible excessive intake of this trace element for animals in question.

Breeder feed restriction may negatively affect broiler progeny immunity. Sources of trace minerals (TM) with higher bioavailability in breeder diets have been reported to enhance humoral and cellular immunity in broiler progeny. An experiment was conducted to examine the effects of breeder feeding programs and TM dietary sources on maternal antibody transfer and humoral immune response of progeny to a live vaccine against Newcastle disease virus (NDV). Cobb 500 breeders were fed according to 2 feed allocation programs, sigmoid late fast and sigmoid late slow, from 14 to 29 wk of age. From 56 to 62 wk of age, breeders were fed with either inorganic TM or an organic source (OTM) to replace 30% of Cu, Zn, and Mn. Progeny broilers were vaccinated intraocularly with La Sota NDV vaccine at 7 d of age. Blood samples were collected at hatching, 4, and 14 d postvaccination. Serum antibody levels against NDV were assessed by ELISA and cytokine expression by real time PCR. At hatching, late slow breeder progeny fed diets with 30% OTM had higher antibody titers as compared with progeny of breeders fed 100% inorganic TM. Similar results were observed 2 wk postvaccination. Breeder feeding programs and TM sources affected the expression level of IL-4 in NDV vaccinated broiler progeny. It was concluded that breeder feeding programs influenced humoral immune response to NDV vaccine in the broiler progeny, and 30% OTM may increase these responses. © 2013 Poultry Science Association, Inc.
