The presence of Mycobacterium bovis in bovine carcasses with lesions suggestive of tuberculosis was evaluated. Seventy-two carcass samples were selected during slaughter inspection procedures in abattoirs in the state of Mato Grosso do Sul, Brazil. Seventeen (23.6%) of samples showed colonies suggestive of mycobacteria that were confirmed to be acid-fast bacilli by Ziehl-Neelsen staining. Polymerase chain reaction (PCR) using primers specific for M. bovis identified M. bovis in 13 (76.5%) isolates. The PCR-restriction enzyme pattern analysis using gene encoding for the 65-kDa protein and two restriction enzymes identified the remaining four isolates that were represented by two M. tuberculosis complex and two nontuberculous mycobacteria. The results are indicative of infection of slaughter cattle by M. bovis and other mycobacteria in the state of Mato Grosso do Sul.

Atualmente, no cenário nacional, a cultura da soja em plantio direto é muito utilizada na integração agricultura-pecuária. No ano agrícola de 2006/2007, no município de Selvíria (MS), entre as latitudes de 20 ° 18 ' 05S e 20 ° 18 ' 28S e as longitudes de 52 ° 39 ' 02W e 52 ° 40 ' 28W, foi analisada a produtividade da soja, em plantio direto, em função de alguns atributos físicos de um Latossolo Vermelho distroférrico. O objetivo foi selecionar, entre os atributos do solo pesquisados, aquele que melhor se apresentasse para explicar a variabilidade espacial da produtividade agrícola. Para isso, foi instalada a malha geoestatística para a coleta dos dados do solo e da planta, contendo 120 pontos amostrais, numa área de 4.068 m² e declive homogêneo de 0,025 m m-1. do ponto de vista linear e espacial, a elevada produtividade de grãos de soja pôde ser explicada em função da densidade do solo e da umidade volumétrica. A baixa variabilidade da maioria dos atributos do solo indicou que o plantio direto é um sistema que proporciona a homogeneização do ambiente físico do solo.

No ano agrícola de 2006/07, no município de Selvíria (MS), foi analisada a produtividade do feijão em função de alguns atributos físicos de um Latossolo Vermelho distroférrico, cultivado nas condições de elevado nível tecnológico de manejo pelo sistema plantio direto irrigado com pivô central. O objetivo foi selecionar, entre os atributos pesquisados do solo, aquele com a melhor representatividade para explicar a variabilidade da produtividade agrícola. Foi instalada a malha geoestatística, para a coleta de dados do solo e da planta, com 117 pontos amostrais, numa área de 2.025 m² e declive homogêneo de 0,055 m m-1. do ponto de vista linear e espacial, a produtividade de feijão foi explicada em função da densidade de partículas e da umidade gravimétrica do solo, respectivamente. Assim, os valores de densidade de partículas entre 1,95 e 2,20 kg dm-3 e de umidade gravimétrica entre 0,205 e 0,239 kg kg-1; ambas coletadas na camada intermediária, resultaram em sítios com as mais elevadas produtividades (2.200-2.600 kg ha-1). Entretanto, para as condições do elevado nível tecnológico de manejo empregado, a produtividade média do feijão não atingiu seu máximo potencial, uma vez que o agravado estado de compactação no qual o solo se encontrava proporcionou à produtividade redução de 26,6 %. Dessa forma, os valores médios da densidade do solo e da resistência do solo à penetração, respectivamente nas faixas de 1,36-1,41 kg dm-3 e de 1,56-3,24 MPa, típicos de solos argilosos compactados, atestaram a substancial queda da produtividade de grãos de feijão.

A resistência mecânica à penetração do solo pode influenciar, negativamente, o crescimento radicular da soja, afetando, sobremaneira, sua produtividade. No ano agrícola 2008/2009, no município de Selvíria (MS), no Cerrado brasileiro, foi analisada a produtividade da soja, em função da resistência mecânica à penetração de um Latossolo Vermelho distroférrico, no sistema de semeadura direta. O objetivo foi selecionar, entre os atributos pesquisados do solo, aquele com a melhor correlação, linear e espacial, para explicar a variabilidade da produtividade da soja. Foi instalada uma malha geoestatística, para a coleta de dados do solo e da planta, com 120 pontos amostrais, em área de 8,34 ha e declive homogêneo de 0,055 m m-1. Dessa forma, visando-se a auxiliar futuros trabalhos voltados à agricultura de precisão, os quais venham a utilizar os mesmos atributos do presente estudo, os valores dos alcances da dependência espacial a serem empregados deverão estar compreendidos entre 38,1 m e 114,7 m. do ponto de vista linear, a resistência mecânica à penetração do solo, quando avaliada na camada 0,10-0,20 m, apresentou boa correlação exponencial inversa com a produtividade de grãos de soja. Entretanto, do ponto de vista espacial, o número de vagens por planta apresentou-se como bom indicador da produtividade de grãos de soja.

Soil porosity influences plant development since root growth and crop yield are determined by the root depth. The objective of this study was to investigate the linear and spatial variability and correlations between common bean yield and soil porosity. The bean grain yield of the irrigated cultivar Carioca IAC was analyzed in the growing season 2004/2005, in Selviria-MS, as well as macroporosity (MA), microporosity (MI) and total porosity (TP), in a Dystroferric Red Latosol, at four depths: 1 (0.0-0.10 m), 2 (0.10-0.20 M), 3 (0.20-0.30 m) and 4 (0.30-0.40 m). Soil and plant data were collected in a geostatistical grid with 135 points spaced 10 m apart, covering an area of 50 x 150 m. The data of the studied attributes did not vary randomly and the values were intermediate to low. They followed well-defined spatial standards, reaching between 11.70-104.40 m. on the other hand, the linear correlation between the plant and soil attributes was low, due to the high number of observations. Grain yield had the best linear correlations with MA1b, MI1 and TP3. From the spatial point of view, the inverse correlation between PG and #TP2 was outstanding. At the sites where #TP2 diminished (0.030-0.045 m(3) m(-3)) the yield varied from 2,173 to 3,529 kg ha(-1) and where it increased (0.045-0.076 m(3) m(-3)), the yield was between 1,630 and 2,173 kg ha(-1). Therefore, the total soil porosity, evaluated in the 0.10-0.20 m layer (#TP2), indicated the importance of the contact root/soil and was in turn a satisfactory indicator of soil physical quality, with a view to the grain yield of irrigated common bean.

The soybean is the crop most cultivated in Brazil, with great socioeconomic importance. In the agriculture year 2008/09 in Selviria County, Mato Grosso do Sul State, in the Brazilian Savannah, was analyzed the production components and the soybean yield cultivated in a Typic Acrustox on no-tillage. The main purpose objective was select among the production components number of pods per plant, number of grains per pod, number of grains per plant, mass of a thousand grains, mass of grains per plant and population of plants, which of the best linear and spatial correlation aiming explain the soybean yield variability. The irregular geostatistical grid was installed to collect of data, with 120 sampling points, in an area of 8.34 ha. The values of spatial dependence range to be utilized should be among 38.1 and 114.7 meters. The model of the adjusted semivariograma was predominantly the spherical. of the lineal and spatial point of view, the number of pods per plant and the mass of grains per plant they were correlated in a direct way with the soybean yield, demonstrating be the best components to esteem her.

As pastagens cultivadas do Cerrado brasileiro são responsáveis pela metade da produtividade da carne bovina destinada à alimentação da população, enquanto as culturas agrícolas o são pela terça parte de sua produtividade de grãos, desempenhando importante papel econômico-financeiro para o país. Dessa forma, no ano agrícola 2005/2006, na Fazenda de Ensino e Pesquisa da Faculdade de Engenharia de Ilha Solteira - FEIS/UNESP, foram estudadas a variabilidade e a dependência espacial entre atributos físicos do solo e a produtividade da soja, quando rotacionados após a pastagem degradada com Brachiaria, sobre um Latossolo Vermelho distroférrico. Também foram estudadas correlações lineares e espaciais entre esses atributos, apurando condições que proporcionassem aumento da produtividade agrícola. Para isso, na área com a referida pastagem instalou-se uma malha contendo 124 pontos amostrais, com espaçamentos de 10,0 x 10,0 e de 5,0 x 5,0 m entre eles, numa área total de 7.500 m². Dos pontos de vista linear e espacial, a elevada produtividade de grãos de soja pode ser explicada em razão do número de grãos por planta e da macroporosidade do solo. A alta variabilidade obtida na maioria dos atributos do solo denotou que a integração lavoura-pecuária é um sistema que proporciona heterogeneidade do ambiente físico do solo.

This study evaluated the microtensile bond strength test (mu T), micromorphology of resin-enamel interface (RET) and etching patterns (EP) promoted by the etch-and-rinse adhesive, Prime&Bond NT (PB), and two self-etching adhesives, Clearfil SE Bond (SE) and Adper Prompt L-Pop (APR), to ground bovine enamel surfaces' when applied at temperatures of 5 degrees C (C), 40 degrees C (H) and 20 degrees C (R). Materials and Methods. Sixty-three bovine incisors were randomly divided into nine experimental groups (n=7) according to adhesive systems and temperatures. The buccal enamel surfaces were flattened with 600-grit SiC paper and abraded with a diamond bur under water-cooling. The adhesive systems were applied according to the manufacturer's instructions. After the restorative procedures, the specimens were sectioned into five slabs. Four slabs were prepared for mu T and one for interface analysis. For etching pattern analysis, the remaining 16 bovine enamel fragments were used (n=2). The adhesives were applied and the surfaces were rinsed with organic solvents after application. The specimens for RET and EP analysis were prepared for SEM analysis. Results. No significant differences among the adhesives were found at R temperature. However, at 5 degrees C, PB and APR presented lower bond strength than SE. At H temperature, higher bond strength was observed for PB than for APR and SE. At C and H temperature, formation of the interdiffusion zone was impaired and the treated enamel surfaces presented an undefined EP.

A porosidade do solo é de extrema importância à produtividade agrícola, uma vez que, em condições adversas, pode dificultar a penetração das raízes e, consequentemente, limitar o adequado aproveitamento dos nutrientes e da água disponível. Na Fazenda Bonança, em Pereira Barreto - SP, em 2005, foi analisada a produtividade de forragem do milho (MSF), a macroporosidade (MA), a microporosidade (MI) e a porosidade total (PT) de um Latossolo Vermelho distrófico (Acrustox Háplico), sob plantio direto, em três profundidades. O objetivo foi estudar as correlações lineares e espaciais entre os atributos da planta e do solo, na tentativa de selecionar, entre os do solo, um indicador da sua qualidade física de boa representatividade para a produtividade da forragem. Foi instalada a malha geoestatística para a coleta dos dados do solo e da planta, contendo 125 pontos amostrais numa área de 2.500 m². Os atributos estudados apresentaram variabilidade entre baixa e muito alta. Também, seguiram padrões espaciais claramente definidos, com alcances da dependência espacial entre 6,6 e 31,1 m. Apesar da correlação linear simples entre a MSF e a MI na profundidade de 0,10-0,20 m ter sido baixa, foi extremamente significativa. Contudo, do ponto de vista espacial, houve elevada correlação inversa entre tais variáveis. Assim, a MI2 apresentou-se como satisfatório indicador da qualidade física do solo de Pereira Barreto - SP, quando destinado à produtividade de forragem do milho.

To identify genes specifically or predominantly expressed in the stigmas/styles and to establish their possible function in the reproductive process of plants, a tobacco stigma/style cDNA library was constructed and differentially screened, resulting in the isolation of several cDNA clones. The molecular characterization of one of these clones is described here. After sequencing the cDNA and the isolated genomic clone, it was determined that the corresponding gene encodes a protein containing an ATP-binding cassette, characteristic of ABC transporters. This gene, designated as NtWBC1 (Nicotiana tabacum ABC transporter of the White-Brown Complex subfamily), encodes a protein that contains the typical structure of the 'half-transporters' of the White subfamily. To establish the spatial expression pattern of the NtWBC1 gene, northern blot and real-time RT-PCR analyses with total RNA from roots, stems, leaves, sepals, petals, stamens, stigmas/styles, ovaries, and seeds were performed. The result revealed a transcript of 2.5 kb present at high levels in stigmas and styles and a smaller transcript (2.3 kb) present at a lower level in stamens. NtWBC1 expression is developmentally regulated in stigmas/styles, with mRNA accumulation increasing toward anthesis. In situ hybridization experiments demonstrated that NtWBC1 is expressed in the stigmatic secretory zone and in anthers, at the stomium region and at the vascular bundle. NtWBC1 is the first ABC transporter gene with specific expression in plant reproductive organs to be identified and its expression pattern suggests important role(s) in the reproductive process.

The aim of this study was to know the yeast biodiversity from fresh olive (Olea europaea L.) fruits, olive paste (crush olives) and olive pomace (solid waste) from Arbequina and Cornicabra varieties. Yeasts were isolated from fruits randomly harvested at various olive groves in the region of Castilla La Mancha (Spain). Olive paste and pomace, a byproduct of the processing of this raw material, were also collected in sterile flasks from different oil mills. Molecular identification methodology used included comparison of polymerase chain reaction (PCR) amplicons of their 5.8S rRNA gene and internal transcribed spacers ITS1 and ITS2 followed by restriction pattern analysis (RFLP). For some species, sequence analysis of the 5.8S rDNA gene was necessary. The results were compared to sequences held in public databases (BLAST). These techniques allowed to identify fourteen different species of yeasts, belonging to seven different genera (Zygosaccharomyces, Pichia, Lachancea, Kluyveromyces, Saccharomyces, Candida, Torulaspora) from the 108 yeast isolates. Species diversity was thus considerable: Pichia caribbica, Zygosaccharomyces fermentati (Lachancea fermentati) and Pichia holstii (Nakazawaea holstii) were the most commonly isolated species, followed by Pichia mississippiensis, Lachancea sp., Kluyveromyces thermotolerans and Saccharomyces rosinii. The biotechnological properties of these isolates, was also studied. For this purpose, the activity of various enzymes (beta-glucosidase, beta-glucanase, carboxymethylcellulase, polygalacturonase, peroxidase and lipase) was evaluated. It was important that none of species showed lipase activity, a few had cellulase and polygalacturonase activities and the majority of them presented beta-glucanase, beta-glucosidase and peroxidase activities. An uncommon disseminated Mycobacterium tuberculosis infection is described in a 12-year-old female dog presenting with fever, dyspnea, cough, weight loss, lymphadenopathy, melena, epistaxis, and emesis. The dog had a history of close contact with its owner, who died of pulmonary tuberculosis. Radiographic examination revealed diffuse radio-opaque images in both lung lobes, diffuse visible masses in abdominal organs, and hilar and mesenteric lymphadenopathy. Bronchial washing samples and feces were negative for acid-fast organisms. Polymerase chain reaction (PCR)-based species identification of bronchial washing samples, feces, and urine revealed M. tuberculosis using PCR-restriction enzyme pattern analysis-PRA. Because of public health concerns, which were worsened by the physical condition of the dog, euthanasia of the animal was recommended. Rough and tough colonies suggestive of M. tuberculosis were observed after microbiological culture of lung, liver, spleen, heart, and lymph node fragments in Löwenstein-Jensen and Stonebrink media. Background: Mycobacterium spp. is one of the most important species of zoonotic pathogens that can be transmitted from cattle to humans. The presence of these opportunistic, pathogenic bacteria in bovine milk has emerged as a public-health concern, especially among individuals who consume raw milk and related dairy products. To address this concern, the Brazilian control and eradication program focusing on bovine tuberculosis, was established in 2001. However, bovine tuberculosis continues to afflict approximately 1,3 percent of the cattle in Brazil. In the present study, 300 samples of milk from bovine herds, obtained from both individual and collective bulk tanks and informal points of sale, were cultured on Löwenstein-Jensen and Stonebrink media. Polymerase chain reaction (PCR)-based tests and restriction-enzyme pattern analysis were then performed on the colonies exhibiting phenotypes suggestive of Mycobacterium spp., which were characterized as acid-fast bacilli.Results: Of the 300 bovine milk samples that were processed, 24 were positively identified as Mycobacterium spp.Molecular identification detected 15 unique mycobacterial species: Mycobacterium bovis, M. gordonae, M. fortuitum, M. intracellulare, M. flavescens, M. duvalii, M. haemophilum, M. immunogenum, M. lentiflavum, M. mucogenicum, M. novocastrense, M. parafortuitum, M. smegmatis, M. terrae and M. vaccae. The isolation of bacteria from the various locations occurred in the following proportions: 9 percent of the individual bulk-tank samples, 7 percent of the collective bulk-tank samples and 8 percent of the informal-trade samples. No statistically significant difference was observed between the presence of Mycobacterium spp. in the three types of samples collected, the milk production profiles, the presence of veterinary assistance and the reported concerns about bovine tuberculosis prevention in the herds.Conclusion: The microbiological cultures associated with PCR-based identification tests are possible tools for the investigation of the presence of Mycobacterium spp. in milk samples. Using these methods, we found that the Brazilian population may be regularly exposed to mycobacteria by consuming raw bovine milk and related dairy products. These evidences reinforces the need to optimize quality programs of dairy products, to intensify the sanitary inspection of these products and the necessity of further studies on the presence of Mycobacterium spp. in milk and milk-based products. © 2013 Franco et al.; licensee BioMed Central Ltd.
