The present study reports on an alternative methodology for the extraction of Humic Substances (HS) from the soil. The extractions were carried out with 0.5 M KOH for 3 h, at a ratio of 1:20 (m/v) under a nitrogen atmosphere. The HS were separated by centrifugation based on their solubility in alcaline solution. This methodology was compared with the usual procedure in three different soil samples and in one sample from vermicompost. The yield, E4/E6 ratio, ash content, and the elemental composition (C, H, O, N) of the extracted HS have been determined. The functional groups were identified by Fourier-transform infrared spectroscopy. This novel procedure adds a new perspective to the extraction of humic substances due to the short time and high performance of the extraction in relation to the usual procedure.

In the laboratory of cytogenetics of the DBAA-UNESP we are studying the karyotipe of some Brazilian Palms. To determine the best protocol, methods of seed germination, inhibition of mitosis, time to pick up the roots and staining were analyzed. The results shown that the seed germination in sphagnum is effective to achieve good roots. The best time to collect the root tips is between 11 to 12 AM., when there are more cell metaphases. The inhibition of mitosis cycle at metaphases may be effective both with 8-hydroxiquinoleine (0,03% -5 hours) or with cold water (0°C - 18-20 hours). The staining with Giemsa 2% showed the best chromosome figures in the metaphases. Now, to get good metaphases slides the method in use in the lab is: 1) seed germination in sphagnum at room temperature and high humidity; 2) The major roots are cut maintaining at least 5 cm, because this technic allows new emergence of roots, increasing the number of roots collected per germinated seed, that is very important in some species with poor germination rates; 3) To get the mitosis inhibition we are using cold water (0°C) treatment for 18-20 hours, following the standard protocols for conservation and hydrolysis; or enzyme digestion with pectinasecellulase 4) the staining procedures are made using Giemsa 2%. The Brazilian palms species studied and their respective chromosome number were: Aiphanes acanthophylla (2n=30), A. caryotaefolia (2n=30), Syagrus quinquifaria (2n=32), S. coronata (2n=32), S. romanzoffiana (2n=32), Euterpe edulis (2n=36), E. oleracea (2n=36), Copernicia prunifera (2n=36), Scheelea lauromuelleriana (2n=32) and Bactris gasipaes (2n=30).

Differences of venom peptide composition as function of two collection methodologies, electrical stimulation (ES) and reservoir disruption (RD), were analyzed by reverse-phase HPLC in Apis mellifera races - A. m. adansonii, A. m. ligustica and Africanized honeybee. The analyses were performed through determination of the relative number and percentage of each molecular form associated to the peaks eluted by chromatography. Comparison of these profiles revealed qualitative and quantitative differences related to the venom collection methodology as well to the three races analyzed. In contrast to data usually found for venom proteins, the three races presented a major number of peaks or molecular forms when venom was collected by ES. Besides, in general, the relative concentration of each peak was higher for ES in relation to RD. That indicates the presence of molecular precursors in the venom obtained by RD. The presence/absence pattern of the peaks, such as their relative concentrations showed a closer similarity between A. m. adansonii and the Africanized honeybees than that observed between these and A. m. ligustica. The obtained data allowed a discussion about the differences in the relative concentration of each venom component according to the collection methodology, and finally the biological action of the venom in different races. So, these results, apart from being useful to establish a peptide profile for each bee race as a function of the venom collection methodology, pointed out once more that the chromatographic techniques are a great tool for the identification of A. mellifera subspecies.

Among the positioning systems that compose GNSS (Global Navigation Satellite System), GPS has the capability of providing low, medium and high precision positioning data. However, GPS observables may be subject to many different types of errors. These systematic errors can degrade the accuracy of the positioning provided by GPS. These errors are mainly related to GPS satellite orbits, multipath, and atmospheric effects. In order to mitigate these errors, a semiparametric model and the penalized least squares technique were employed in this study. This is similar to changing the stochastical model, in which error functions are incorporated and the results are similar to those in which the functional model is changed instead. Using this method, it was shown that ambiguities and the estimation of station coordinates were more reliable and accurate than when employing a conventional least squares methodology.

The main purpose of this work is the development of computational tools in order to assist the on-line automatic detection of burn in the surface grinding process. Most of the parameters currently employed in the burning recognition (DPO, FKS, DPKS, DIFP, among others) do not incorporate routines for automatic selection of the grinding passes, therefore, requiring the user's interference for the choice of the active region. Several methods were employed in the passes extraction; however, those with the best results are presented in this article. Tests carried out in a surface-grinding machine have shown the success of the algorithms developed for pass extraction. Copyright © 2007 by ABCM.

This paper presents a tool box developed to read files describing a SIMULINK® model and translates it into a structural VHDL-AMS description. In translation process, all files and directory structures to simulate the translated model on SystemVision™ environment is generate. The tool box named MS2SV was tested by three models of commercially available digital-to-analogue converters. All models use the R2R ladder network to conversion, but the functionality of these three components is different. The methodology of conversion of the model is presents together with sort theory about R-2R ladder network. In the evaluation of the translated models, we used a sine waveform input signal and the waveform generated by D/A conversion process was compared by FFT analysis. The results show the viability of this type of approach. This work considers some of challenges set by the electronic industry for the further development of simulation methodologies and tools in the field of mixed-signal technology. © 2007 IEEE.

The present study sought biotensoactive production from soybean oil fry waste using Pseudomonas aeruginosa ATCC 10145 and Pseudomonas aeruginosa isolated from the soil of a petroleum station having undergone gasoline and diesel oil spills. The results of the experiments were analyzed using a complete factorial experimental design, investigating the concentration of soybean oil waste, ammonia sulfate and residual brewery yeast. Assays were performed in 250-mL Erlenmeyer beakers containing 50 mL of production medium, maintained on a rotary shaker at 200 rpm and a temperature of 30±1 °C for a 48-hour fermentation period. Biosurfactant production was monitored through the determination of rhamnose, surface tension and emulsification activity. The Pseudomonas aeruginosa ATCC 10145 strain and isolated Pseudomonas aeruginosa were able to reduce the surface tension of the initial mexlium from 61 mN/m to 32.5 mN/m and 30.0 mN/m as well as produce rhamnose at concentrations of 1.96 and 2.89 g/L with emulsification indices of 96% and 100%, respectively.

To carry out the dating by the Fission Track Method (FTM) the international community that works with this method employs methodologies in which the mineral to be dated must be irradiated with neutrons. Such irradiation, performed in a nuclear reactor, demand a relatively long waiting time so that the activity of the sample attain a proper level for handling. The present work aims to establish a methodology that makes possible the dating by FTM using a mass spectrometer instead of a nuclear reactor. This methodology was applied to apatite samples from Durango, Mexico. © 2009 Elsevier Ltd. All rights reserved.

A synbiotic yoghurt based on a combination of soymilk and yacon water extract (from yacon root tubers) was developed as a novel food product fermented with a probiotic culture of Enterococcus faecium CRL 183 and Lactobacillus helveticus ssp jugurti 4l6. Response surface methodology (RSM) was used to optimize the independent variables soymilk protein concentration and percentage of yacon extract in the formulation through a Central Composite Rotatable Design (CCRD), consisting of a 22 factorial design with two levels (-1, +1), two central points (0) and four axial points (± a, 0) (0, ± α). The responses were assessed by consumer acceptance tests. The optimization indicated that a formulation with a soymilk protein concentration of 1.74g/L and 25.86% of yacon extract gave the best average values, 5.91 for the taste and 6.00 for the overall impression responses. The formulation with 40% of yacon extract and the same concentration of soymilk protein achieved similar acceptance values: taste (5.94) and overall impression (5.87), however, with the extra yacon, it probably had a greater content of prebiotic fructooligosaccharides. Consequently, both formulations may give useful functional foods, with sensory properties comparable with those of soy yoghurt (control formulation). This work presents a methodological proposal for acquisition of biometric data through telemetry basing its development on a research-action and a case study. Nowadays, the qualified professionals of physical evaluation have to use specific devices to obtain biometric signals and data. These devices in the most of the time are high cost and difficult to use and handling. Therefore, the methodological proposal was elaborate in order to develop, conceptually, a bio telemetric device which could acquire the desirable biometric signals: oxymetry, biometrics, corporal temperature and pedometry which are essential for the area of physical evaluation. It was researched the existent biometrics sensors, the possible ways for the remote transmission of signals and the computer systems available so that the acquisition of data could be possible. This methodological proposal of remote acquisition of biometrical signals is structured in four modules: Acquisitor of biometrics data; Converser and transmitter of biometric signals; Receiver and Processor of biometrics signals and Generator of Interpretative Graphs. The modules aim the obtention of interpretative graphics of human biometric signals. In order to validate this proposal a functional prototype was developed and it is presented in the development of this work.

Studies carried out with glass electrode in anhydrous ethanol and ethanol-water solutions for measuring pH values have shown that this parameter depends on the solution composition, the contact time with the solution, the utilized temperature, and the type of electrolyte used. It was also observed that the glass electrode behavior in an acid medium differs from an alkaline medium. These studies provided correction factors for pH values from 2 to 12, allowing the realization of proper measurements of the hydrogen ionic activity in the ethanol-water and anhydrous ethanol solutions. However, these correction factors could not be applied to the fuel ethanol. Alternatively, a new method was developed for the correction of the pH values, which can be applied in hydrous and anhydrous fuel ethanol samples. Copyright © 2011 by ASTM International.

This article aims to present proposals for improvement of key standards and resolutions concerned about the methodology for calculating the indicator of total harmonic voltage distortion, and should contribute to the process of examining the compatibility of potentially disturbing loads in electric power quality in distribution systems. These proposals were drawn from the analysis of results from measurement campaigns conducted in a case study including analysis of the connection of a new induction furnace in a foundry served by a distributor of São Paulo state. A general historical situating the quality of electric energy in the electricity sector is presented, and methodological guidelines and procedures used in experimental trials are shown. The analysis and discussion of results are prepared to answer the main questions that arise during the implementation of standards, resolutions and procedures. © 2011 IEEE.

Palatal rugoscopy, or palatoscopy, is the process by which human identification can be obtained by inspecting the transverse palatal rugae inside the mouth. Aim: This study evaluated a digital method for human identification using palatoscopy, by comparing photographs of the palate against the images of cast models of the maxilla photographed with and without highlighting of the palatal rugae. Methods: Condensation silicone impressions were made from the upper arches of 30 adult subjects of both genders and their palates were then photographed. The first impression was made with heavy silicone, the second impression with light silicone, and then the models were cast in improved type IV dental stone. The casts were photographed, the palatal rugae of each one were highlighted with a pencil, and then the models were photographed again. Using a free image-editing software, the digital photographs were overlapped over the images of the palatal rugae of the models with and without highlighting of the palatal rugae, in order to identify the pairs. Results: The result of overlapping the digital photographs with the images of the models without highlighted palatal rugae resulted in 90% positive identification. For the overlapping of the digital photographs with the images of models with highlighted palatal rugae, there was 100% positive identification. Conclusions: The digital method evaluated in this study was proven effective for human identification.

Despite the large use of differential scanning calorimetry (DSC) technique in advanced polymer materials characterization, the new methodology called DSC in high heating rates was developed. The heating rate during conventional DSC experiments varying from 10 to 20°C.min-1, sample mass from 10 to 15mg and standard aluminum sample pan weighting, approximately, 27mg. In order to contribute to a better comprehension of DSC behavior in different heating rates, this work correlates as high heating rate influences to the thermal events in DSC experiments. Samples of metallic standard (In, Pb, Sn and Zn) with masses varying from 0.570mg to 20.9mg were analyzed in multiples sample heating rate from 4 to 324°C. min-1. In order to make properly all those experiments, a precise and careful temperature and enthalpy calibrations were performed and deeply discussed. Thus, this work shows a DSC methodology able to generate good and reliable results on experiments under any researcher choice heating rates to characterize the advanced materials used, for example, for aerospace industry. Also it helps the DSC users to find in their available instruments, already installed, a better and more accurate DSC test results, improving in just one shot the analysis sensitivity and resolution. Polypropylene melting and enthalpy thermal events are also studied using both the conventional DSC method and high heating rate method.
