The PKC1 gene in the yeast Saccharomyces cerevisiae encodes protein kinase C that is known to control a mitogen-activated protein (MAP) kinase cascade consisting of Bck1, Mkk1 and Mkk2, and Mpk1. This cascade affects the cell wall integrity but the phenotype of Pkc1 mutants suggests additional targets which have not yet been identified. We show that a pkc1Δ mutant, as opposed to mutants in the MAP kinase cascade, displays two major defects in the control of carbon metabolism. It shows a delay in the initiation of fermentation upon addition of glucose and a defect in derepression of SUC2 gene after exhaustion of glucose from the medium. After addition of glucose the production of both ethanol and glycerol started very slowly. The V max of glucose transport dropped considerably and Northern blot analysis showed that induction of the HXT1, HXT2 and HXT4 genes was strongly reduced. Growth of the pkc1Δ mutant was absent on glycerol and poor on galactose and raffinose. Oxygen uptake was barely present. Derepression of invertase activity and SUC2 transcription upon transfer of cells from glucose to raffinose was deficient in the pkc1Δ mutant as opposed to the wild-type. Our results suggest an involvement of Pkc1p in the control of carbon metabolism which is not shared by the downstream MAP kinase cascade. © 2002 Federation of European Microbiological Societies. In addition to the bio-guided investigation of the antifungal activity of Plinia cauliflora leaves against different Candida species, the major aim of the present study was the search for targets on the fungal cell. The most active antifungal fraction was purified by chromatography and characterized by NMR and mass spectrometry. The antifungal activity was evaluated against five Candida strains according to referenced guidelines. Cytotoxicity against fibroblast cells was determined. The likely targets of Candida albicans cells were assessed through interactions with ergosterol and cell wall composition, porosity and architecture. The chemical major component within the most active antifungal fraction of P. cauliflora leaves identified was the hydrolysable tannin casuarinin. The cytotoxic concentration was higher than the antifungal one. The first indication of plant target on cellular integrity was suggested by the antifungal activity ameliorated when using an osmotic support. The most important target for the tannin fraction studied was suggested by ultrastructural analysis of yeast cell walls revealing a denser mannan outer layer and wall porosity reduced. It is possible to imply that P. cauliflora targeted the C. albicans cell wall inducing some changes in the architecture, notably the outer glycoprotein layer, affecting the cell wall porosity without alteration of the polysaccharide or protein level. © 2013 by the authors.

The external morphology of seeds and post-germination developmental stages of Angelonia salicariifolia Bonpl. (Scrophulariaceae) were investigated using scanning electron microscopy. Some structural features of the seed exotesta and seedling in Angelonia are presented for the first time and are of potential taxonomic value for this neotropical genus. The seeds are very small (0.9-1.7 mm long and 0.5-0.9 mm wide), ovate, with a reticulate-crested exotesta, reticules arranged uniformly in longitudinal rows, with a high density of microcilia-like projections on the cell wall of the reticule base and on the edge of the crests. The hilum is located beside the micropyle at the narrow end of the seed. Germination is epigeal. During germination the radicle develops, followed by elongation of the hypocotyl and primary root. At this stage dense root hairs develop on the lower part of the hypocotyl. The apical bud-located between the cotyledons-begins to develop after the cotyledons have unfolded. The cotyledons are equal in size, sessile and ovate. The seedlings have two types of trichomes, one characteristic of the cotyledons and first pair of leaves (glandular, sessile, four-celled head with quadrangular shape) and the other characteristic of the hypocotyl and epicotyl (stalked, erect, elongate and three-celled with dome-shaped unicellular head). The effects of age on microbiota composition, gut fermentation end-product formation and peripheral lymphocyte numbers were compared between old and young adult Beagle dogs fed four kibble diets differing in yeast cell wall contents. The experiment had a double 4 x 4 Latin square design, one with four mature dogs (4 years old) and the other with four old dogs (10 years old), with four replicates (diets) per dog. In each period a 15d adaptation period preceded a 5d total collection of faeces for the digestibility trial. on day 21, fresh faecal samples were collected for the determination of bacterial enumeration, pH, biogenic amine and short-chain fatty acid. Flow cytometry was used for immunophenotypic evaluation. Dogs were fed four kibble diets with similar composition with 0, 0.15, 0.30 and 0.45% of yeast cell wall (as-fed), respectively. Data were evaluated using general linear models of Statistical Analysis Systems statistical software (P<0.05). No evidence of a difference in faecal bacteria counts between ages was found (total aerobes, total anaerobes, Bifidobacterium, Lactobacillus, Clostridium and Escherichia coli: P. 0.15). Faecal concentrations of butyrate, histamine, agmatine and spermine were lower (P <= 0.05) and faecal pH was higher (P=0.03) in older dogs than in mature adult dogs, suggesting an alteration in bacterial metabolic activity, or in the rate of intestinal absorption of these compounds. Concentrations of T-lymphocytes, T-cytotoxic lymphocytes and B-lymphocytes were also lower (P <= 0.01) in older dogs than in mature adult dogs. The study confirmed alterations in peripheral lymphocytes and revealed a reduced concentration of some fermentation end products in the colon of old dogs.

Foi investigado o efeito da substituição de antibióticos por prebiótico, probiótico e simbiótico em dietas para frangos de corte de 1 a 45 dias de idade. Foram utilizados 750 pintos de 1 dia de idade, distribuídos em cinco tratamentos, sendo: T1-sem aditivos, T2-antibiótico (Olaquindoxâ e Nitrovinâ ), T3-prebiótico (0,2% de parede celular de S. cerevisiae), T4-probiótico (300 ppm de B. subtilis) e T5-simbiótico (T3 + T4). O desempenho dos frangos de 1-45 dias de idade foi influenciado pelos diferentes tratamentos, sendo o melhor ganho de peso observado em aves que receberam o simbiótico, seguido daquelas com antibiótico, prebiótico e probiótico. O pior ganho de peso foi observado nas aves que não receberam qualquer tipo de aditivo na dieta. A conversão alimentar, no período de 1 a 45 dias de idade, também foi influenciada pelo tipo de aditivo. As aves que não receberam suplementação apresentaram pior conversão alimentar quando comparadas com as aves dos demais tratamentos. Os resultados deste experimento permitem concluir que a substituição de antibióticos por simbióticos na ração de frangos é uma alternativa viável, pois não compromete o desempenho das aves, contudo a ausência de aditivos na dieta piora o desempenho das mesmas.

This study evaluated the effects of dietary supplementation with 0.3% Saccharomyces cerevisiae yeast cell wall and of vaccination against Streptococcus agalactiae on the cellular component of acute inflammation induced in the coelomic cavity of Nile tilapia Oreochromis niloticus and on survival of the fish after challenge. A total of 84 tilapia of mean (+/- SD) weight 125.0 +/- 1.5 g were distributed among twelve 310 l fiberglass tanks according to a 2 x 2 x 3 factorial design in the following manner: with and without supplementation; 2 stimulations (oily solution without S. agalactiae vaccine and vaccination); 15 d later all fish were intracoelomically challenged with 10(8) CFU ml(-1) of a homologous strain of S. agalactiae, and evaluated after 6, 24 and 48 h, with 7 replicates. The fish received the non-supplemented or supplemented diet for a total of 77 d. The vaccination was performed on the 60th day, intracoelomically, as a single injection of 0.5 ml of the vaccine containing 10(8) CFU ml(-1). Fifteen days later, all the fish were challenged with S. agalactiae by means of an intracoelomic inoculation of 10(8) CFU ml(-1). No mortality was observed among the supplemented fish. The fish that were fed the non-supplemented diet and immunized with the bacterium presented a mortality rate of 28.5%. Among the non-supplemented and non-immunized fish, the mortality rate was 38.09%. Supplementation, in both vaccinated and non-vaccinated fish, induced larger accumulations of thrombocytes, lymphocytes and macrophages at the inflammatory focus. The results suggest that supplementation with 0.3% yeast cell wall, in both vaccinated and non-vaccinated fish, improved the inflammatory response of the fish and protected against the challenge. Vaccination increased the defense response, but the effect was stronger when associated with supplementation with S. cerevisiae.

The inflammatory response and hernatological parameters among Nile tilapia (Oreochromis niloticus) supplemented with Saccharomyces cerevisiae were evaluated six and 24 h after inoculation with inactivated Aeromonas hydrophila into the swim bladder. Six groups were formed (n = 10 each): G1 was treated with non-supplemented feed+injection with 0.65% saline solution; G2 with non-supplemented feed+ inoculation with A. hydrophila: G3 with feed containing 2% yeast+ injection with saline; G4 with feed containing 2% yeast + inoculation with A. hydrophila: G5 with feed containing 0.3% cell wall + injection with saline: and G6 with feed containing 0.3% cell wall + inoculation with A. hydrophila. In the groups inoculated with bacteria, the responses were more intense (P<0.05) than in those injected with saline. The groups receiving supplement that were inoculated with A. hydrophila accumulated a greater total number of cells at the lesion site (P<0.05) than did the non-supplemented groups, after six and 24 h. The groups receiving cell wall presented greater total accumulation of cells (P<0.005) that did those receiving yeast. The differential count showed that there were significantly greater number of thrombocytes (P< 0.05) and lower number of neutrophils, macrophages and lymphocytes (P<0.05) in the groups that received supplement, after 6 and 24 h, in relation to the non-supplemented groups. The values in the erythrocyte count, hemoglobin concentration and blood measurement indices did not differ statistically. The variation in circulating thrombocyte and leukocyte counts suggests that the inflammatory stimulus caused recruitment from reserve compartments to the blood. The groups that received yeast or yeast cell wall supplements presented increased nonspecific acute inflammatory response, thus suggesting that this has a beneficial effect on the immunological defense system. Com objetivo de determinar o consumo e a digestibilidade in vivo, bem como a composição química de dietas com níveis crescentes (0, 50 e 100%) de feno de catingueira (FC), utilizaram-se 15 ovinos Morada Nova, durante um período de 22 dias. Foi usado delineamento inteiramente casualizado com três tratamentos e cinco repetições. O FC apresentou 91,5% de matéria seca (MS), 93,89% de matéria orgânica (MO), 11,25% de proteína (PB), 4,31% de extrato etéreo (EE), 45,47% de fibra em detergente neutro (FDN), 37,08% de fibra em detergente ácido (FDA), 8,39% de hemicelulose (HE), 24,37% de celulose (CE), 12,52% de lignina e 6,30% de tanino condensado. Os consumos de MO e MS, em g/dia, g/kg0,75 e %PV; energia bruta (EB), em kcal/kg/dia e kcal/kg0,75; e dos componentes da parede celular foram reduzidos linearmente pelo aumento de FC na dieta. Os consumos de PB, proteína digestível (PD) e EE aumentaram linearmente com o aumento do FC na dieta. Os coeficientes de digestibilidade da MS, MO, PB e EB das dietas não foram afetados pela inclusão do FC. No entanto, observou-se redução linear para os coeficientes de digestibilidade da FDN, FDA, CE e EE e efeito quadrático para o coeficiente de digestibilidade da HE. Os consumos médios de MS e PB verificados nos níveis intermediários de FC atenderam as exigências de mantença dos animais.

O experimento foi conduzido com o objetivo de avaliar a composição química e a digestibilidade in vitro da matéria seca (MS) do bagaço de cana-de-açúcar contendo 60% de MS submetido a doses crescentes de hidróxido de sódio (NaOH) (0; 2,5; 5 e 7,5% de uma solução 2:1 de água:NaOH na MS) em diferentes períodos de tratamento (1, 3, 5 e 7 dias). Foram utilizados baldes plásticos com capacidade de 10 L, mantidos em uma câmara climática à temperatura constante de 25ºC. Não foi verificado efeito dos tratamentos (dose de NaOH e dias de tratamento) sobre os teores de PB, que apresentaram valor médio de 1,6%. A MS aumentou com os dias de tratamento, não sendo observadas alterações para essa variável em relação às doses crescentes de NaOH. Foi observada redução das frações de FDN, FDA, celulose (CEL), hemicelulose (HEM) e lignina (LIG). A digestibilidade in vitro da matéria seca (DIVMS) e o teor de sódio aumentaram quando o bagaço de cana foi submetido a doses crescentes de NaOH, mas não foi observado efeito do período de tratamento sobre essas variáveis. O valor nutritivo do bagaço de cana é melhorado com a adição de NaOH, comprovado pela redução nos constituintes da parede celular e pelo aumento na DIVMS.
