The effect of increased protein intake on the muscle mass gain, nitrogen balance and N-15-glycine kinetics was studied in six young, healthy subjects practitioners of strength training (> 2 years), without use of anabolic steroids and in agreement with the ethical principles of the research. All athletes received adequate diet (0.88g protein/kg/day) during 2 weeks prior the study (D1), and thereafter with diet providing 1.5g of protein/kg/day and 30kcal/g of protein (D2 diet) for the subsequent 2 weeks. Later on, they all received diet with 2.5g of protein/kg/day (D3 diet) and 30 kcal/g protein for the last two weeks. Body composition, food intake, blood biochemistry, nitrogen balance (NB) and 15N-glycine kinetics were determined at the beginning, after D1 (M0) and in the last days of the D2 (M1) and D3 (M2). The results showed at the end of the study (4 weeks) significant increase in muscle mass (1.63 +/- 0.9kg), without difference between D2 and D3. The NB followed the protein/energy consumption (M0 = -7.8g/day; M1 = 5.6g/day and D3 = 16.6g/day), the protein synthesis followed the NB, with M0 < (M1= M2) (M1 = 49.8 +/- 12.2g N/day and M2 = 52.5 +/- 14.0g N/day). Protein catabolism rate was similarly kept among diets. Thus, the results of the NB and N-15-glycine kinetics indicate that the recommended protein intake for these athletes is higher than the one for sedentary adults (0.88g/kg) and lower than 2.5g/kg, around 1.5g of protein/kg/day, with adjustment of the energy consumption to 30 kcal/g of protein.

The objective of this study was to evaluate whether seasonality affects human-assisted reproduction treatment outcomes. For this, 1932 patients undergoing intracytoplasmic sperm injection (ICSI) were assigned to a season group according to the day of oocyte retrieval: winter (n = 435), spring (n = 444), summer (n = 469) or autumn (n = 584). Analysis of variance was used to compare the ICSI outcomes. The fertilization rate was increased during the spring (winter: 67.9%, spring: 73.5%, summer: 68.7% and autumn: 69.0%; p < 0.01). In fact, a nearly 50% increase in the fertilization rate during the spring was observed (odds ratio 1.45, confidence interval 1.20-1.75; p < 0.01). The oestradiol concentration per number of oocytes was significantly higher during the spring (winter: 235.8 pg/mL, spring: 282.1 pg/mL, summer: 226.1 pg/mL and autumn: 228.7 pg/mL; p = 0.030). This study demonstrates a seasonal variability in fertilization after ICSI, where fertilization is higher during the spring than at any other time.

Consistent information on meat products consumed by the public is essential. The technique of stable isotopes is a powerful tool to recover consumers' confidence, as it allows the detection of animal byproduct residues in poultry meat, particularly in quail meat. This study aimed at checking the presence of poultry byproduct mixtures in quail diets by applying the technique of carbon (13C/12C) and nitrogen (15N/14N) stable isotopes in quail breast muscle, keel, and tibia. Sixty four one-day-old male quails were obtained from a commercial farm. Birds were housed in an experimental house from one to 42 days of age, and were randomly distributed into 8 experimental treatments, and fed diets containing poultry offal meal (POM), bovine meat and bone meal (MBM) or poultry feather meal (PFM), or their mixtures. Four birds per treatment were slaughtered at 42 days of age, and breast (Pectoralis major), keel, and tibia were collected for analyses. The inclusion of animal byproducts in quail diets was detected by 13C e 15N analyses in the tissues of the birds; however, it was not possible to specify which byproducts were used. It was concluded that quail meat can be certified by the technique of stable isotopes.

Mammalian oocytes can undergo spontaneous meiotic maturation when they are liberated from their follicles and cultured in vitro; however, the zona pellucida (ZP) becomes resistant to chymotrypsin digestion, or hardens, when spontaneous maturation occurs in serum-free medium. Schroeder et al. [Biol. Reprod. 43 (1990) 891] described that fetuin, a component of fetal calf serum (FCS), inhibits ZP hardening during oocyte maturation. The aim of this experiment was to study the effect of the presence of cumulus cells and addition of hormones to maturation media on bovine zona hardening and embryo development in medium with and without fetuin. In Experiment 1, different concentrations of fetuin were added to the maturation medium. The time necessary for digestion of 50% of the ZP (d50) was not different when oocytes were matured in presence of 10% FCS, 1 mg/ml polyvinyl alcohol (PVA), or 4, 1 and 0.25 mg/ml of fetuin; cleavage rates were also similar. However, significantly more blastocysts (P < 0.05) were formed when FCS was used compared to PVA and 0.25 mg/ml of fetuin. In Experiment 11, we examined the influence of the presence of cumulus cells and hormones during the maturation of oocytes in media with PVA, BSA, FCS and fetuin. The d50 was significantly higher (P < 0.05) when oocytes were matured in presence of cumulus cells. The cleavage rate of cumulus-intact oocytes was similar for all groups. However, when oocytes were partially stripped before maturation, the cleavage rate was significantly higher (P < 0.05) when FCS or fetuin was used. In both stripped and non-stripped groups, significantly more blastocysts (P < 0.05) were formed when oocytes were matured with FCS compared to BSA and PVA. These results indicate that zona hardening, as described for mouse and human oocytes, does not have a large effect on bovine cumulus-intact oocytes. Apparently fetuin can be used as a substitute for FCS during bovine oocyte maturation, since it leads to similar developmental rates as FCS in intact and partially stripped oocytes. Effect of fertilization in water quality and in zooplankton community in open plankton-culture ponds
Resumo:
The effect of swine manure fertilization on the water quality of zooplankton artificial culture in two ponds was measured in diel cycles on different months (October 1999, January and March 2000). Fertilization affected directly water quality; values for total phosphorus were above 1.4 mgL(-1) and maximum rates for ammonia reached 108 mu gL(-1). Zooplankton community comprised four species, namely, Brachionus calyciflorus, B. falcatus, Moina sp. and Thermocyclops sp. B. falcatus (Rotifera) and Moina sp (Cladocera) were dominant respectively in January and in March. There was no difference in abundance of zooplankton between the two ponds (p>0.05), although adult cyclopoid Thermocyclops sp was different between diel cycles (p<0.05). There were significant differences (p<0.05) in dissolved oxygen, ammonia, total phosphorus, orthophosphate, nitrite and nitrate between ponds. In fact, only dissolved oxygen and pH were significantly different (p<0.05) between the ponds' surface and bottom. Organic fertilization also provided greater amount of nutrients and higher pH and conductivity, coupled to greater oscillations in the concentration of dissolved oxygen, directly affecting the zooplanktonic composition.

The objective of this study was to evaluate the effect of fertilization with zinc or boron on the growth and dry matter production, nutritional value and accumulation of nutrients in white oats. The study comprised two experiments conducted in glasshouses, the first consisting of the application of four doses of zinc (0, 0.2, 0.4 and 0.6 mg/dm³) in the form of zinc sulphate (20% Zn), and the second consisting of the application of four doses of boron (0, 0.2, 0.4 and 0.6 mg/dm³) in the form of Borax (11% B). The experimental design in each case was a randomized block design, with five replicates. Fertilization with zinc and boron increased the growth of white oats, but had no significant effect on the nutritional value of the forage. Higher levels of absorption and accumulation of nutrients in plant tissues were observed following the application of boron and zinc at rates of up to 0.60 mg/dm³ of soil.
