The retrovirus HTLV-1 is the etiological agent of the adult T-cell leukemia and HTLV-1 associated myelopathy/tropical spastic paraparesis. The proviral genome has 9,032 base pairs, showing regulatory and structural genes. The env gene encodes for the transmembrane glycoprotein gp 21. The development of methodologies for heterologous protein expression, as well as the acquisition of a cellular line that constituently expresses the recombinant, were the main goals of this work. The DNA fragment that encodes for gp 21 was amplified by nested-PCR and cloned into a pCR2.1-TOPO vector. After which, a sub-cloning was realized using the expressing vector pcDNA3.1+. The transfection of mammalian cells HEK 293 was performed transitorily and permanently. Production of the recombinant gp 21 was confirmed by flux cytometry experiments and the cell line producing protein will be used in immunogenicity assays.

HTLV-1 is the virus that causes T cell lymphoma/leukemia in adults and a neurological disorder known as HTLV-associated myelopathy or tropical spastic paraparesis. One of the transmission means is through contaminated blood and its byproducts. Because of the risk of HTLV-associated infections, screening for HTLV was introduced for Brazilian blood donors in 1993. Most of the diagnostic kits used in the national blood banks are bought from foreign companies. Brazil does not have the technology to produce this material and there is a need to produce diagnostic systems with national technology. In this study, we show the expression of gp21/HTLV-1 in Escherichia coli and its reactivity towards monoclonal antibodies and the antibodies of infected patients. Expressing these proteins is the first step towards obtaining diagnostic kits with Brazilian biotechnology.

Mammary tumors are among the most common neoplasia of canine females. The occurrence of metastasis may be detected by proteic markers. Among them, exist the E-cadherin, a member of cadherin family known for its important role in the regulation of intercellular adhesion in epithelial tissues. Studies suggest that E-cadherin may function as a tumor and invasion suppressor molecule. Cadherin activity is regulated by multiple mechanisms, including interaction with other proteins such as catenins. In this review, the authors approach the cadherin family and other related adhesion proteins including its function, physiopathology and potential use as marker for diagnosis and prognosis of canine mammary tumors.

In bovine females the release of prostaglandin F 2α (PGF 2α) is induced in vivo by estradiol (E 2). It is believed that E 2 stimulates the synthesis of essential proteins for the production of PGF 2α. This study aimed to evaluate the effect of E 2 in increasing the concentration of total protein and modifying the protein composition of endometrial explants from bovine females treated with E 2 at the 17 th day of estrous cycle. Crossbred heifers were treated at 17 th day of estrous cycle intravenously with 0 mg (Control Group; n = 6) or 3 mg of E 2 (E 2 Group; n = 6) and killed two hours after. Endometrial explants were isolated, subjected to extraction of total protein, quantified and were analyzed by onedimensional electrophoresis on polyacrylamide gel 10% SDS-PAGE. The concentration of total protein did not differ between groups, 6296.10 + 439.90 μg/mL for the Control Group and 8426.56 + 1156.00 μg/mL for E 2 Group (p = 0.1158). There was no significant difference (p > 0.05) in the protein profile of endometrial explants in gels stained with Coomasie Blue. In gels stained with Silver Nitrate it was verified in E 2 Group greater relative percentage of the bands referring to the molecular weight of 75 to 76 kDa (8.40% vs. 4.89% in E 2 Group and Control respectively; p < 0.05) and 108 to 110 Kda (6.85% vs. 3.84% in E 2 Group and Control respectively, p < 0.05). It was observed in E 2 Group lower relative percentage of the band referring to the molecular weight of 90 kDa (5.78% vs. 9.83% in E 2 Group and control respectively; p < 0.05). We concluded that the E 2 does not increase the protein concentration in the endometrium, however, it modifies the proteinic composition in the endometrial explants, indicating that E 2 alters the expression of specific proteins.

Introduction: The autologous serum skin test (ASST) may suggest an autoimmune etiology in chronic urticaria (CU). A new laboratory technique called basophil activation test (BAT) has been currently employed for its diagnosis. Objective: To analyze ASST in relation to BAT as well as to evaluate interleukin 3 (IL3) receptors (CD123) and non-specific immunoglobulin G (IgG) autoantibodies bound to basophils in patients with chronic urticaria. Methods: We studied 33 adults with CU and mean age of 42.5 + 14 years. After stimulation by serum from patients with CU, CD63 expression on basophils from one atopic donor was analyzed by flow cytometry. Furthermore, we investigated CD123 and IgG autoantibody expressions. Results: The odds ratio (OR) between ASST and BAT was 1.00 (95% confidence interval [CI]: 0.22 to 4.5). The ASST for autoimmune CU diagnosis showed an accuracy of 54.5%, sensitivity of 66%, specificity of 33%, positive predictive value of 63%, and negative predictive value of 36%. There was no statistical difference between the studied groups as to mean non-specific IgG and CD123 expressions (for a p < 0.05). Discussion: This study demonstrated that ASST has low accuracy in the diagnosis of autoimmune CU. Concerning other analyzed aspects, there was no statistical difference between positive ASST and negative ASST. Conclusions: Due to insufficient studies in this area and the relevance of this issue, further investigation is required.
