O objetivo do presente trabalho foi avaliar uma equação de predição das exigências de proteína bruta (PB) para reprodutoras pesadas na fase de produção. O experimento foi realizado com 600 aves reprodutoras pesadas, Hubbard HI-Y, durante o período de 31 a 46 semanas de idade, alojadas em boxes num delineamento experimental inteiramente casualizado com três tratamentos e cinco repetições de 40 aves. Os tratamentos consistiram de: T1- Fornecimento de PB de acordo com o manual da linhagem (controle), T2- Fornecimento de PB de acordo com a equação de predição determinada, utilizando os dados de desempenho médio das aves do tratamento controle para predizer as exigências e T3- Fornecimento de PB de acordo com a equação de predição determinada, utilizando os dados de desempenho de cada parcela experimental para predizer as exigências, onde a equação de predição avaliada foi: PB=2,282.P0,75+0,356.G+0,262.MO, sendo PB a exigência de proteína bruta (g/ave/dia), P o peso corporal (kg), G o ganho de peso (g) e MO a massa de ovos (g). As rações foram formuladas para atender as exigências nutricionais e quando necessário eram incluídos os aminoácidos sintéticos, metionina, lisina, triptofano, treonina e arginina. As aves alimentadas de acordo com a equação ingeriram menores quantidades de proteína (20,8g/dia) quando comparadas às alimentadas de acordo com as recomendações (23,80g), entretanto isto levou a menores pesos dos ovos refletindo no peso dos pintos. A equação de predição proporcionou melhores resultados quanto à eficiência protéica. Assim, concluiu-se que a equação de predição não forneceu a quantidade mínima de proteína bruta para atender as exigências dos aminoácidos não suplementados na dieta.

Laboratory tests were carried out on larvae and adults of the cattle tick Boophilus microplus to determine the toxicity of macrocyclic lactone acaricides (MLs). Technical and commercial MLs were used in larval packet test (LPT), larval immersion test (LIT) and adult immersion test (AIT). Tn LIT and AIT the toxicity of MLs was much higher than for LPT. In the AIT, diluting the injectable formulation of MLs in water was as effective as dilution in ethanol + Triton X-100. LC50, LC99.9 and 95% confidence limits were determined so that a discriminating dose (DD) could be set for larval and adult tests in order to diagnose potential resistance to MLs in field samples of the tick. These DDs are for Australian strains of B. microplus and may not be suitable for other strains until further work is carried out.The value of these diagnostic tests can only be verified if or when resistance to MLs emerges in ticks. In the present investigation the locomotor and the metabolic activity of Gymnostreptus olivaceus were studied, using 24-hr cycles at different photoperiods and constant temperature and RH. Locomotor activity was studied by the actography method and was reported as coefficients of nocturnalism [CN (N/N + D). 100]. The results showed a nocturnalism coefficient of 98,71% under normal photoperiod conditions and of 29,58% under inverted photoperiod conditions. In constant illumination, the CN of G. olivaceus was 88,22%, and in constant darkness, its rhythm was close to that of the normal photoperiod (CN = 94,92%). The metabolic activity was studied by manometric Warburg respirometry and lit was reported as mu l O-2 . g(-1). h(-1). The respiratory rate of G. olivaceus under normal photoperiod condition was higher at night than during the day (52,52 x 28,76), coinciding with the pattern of nocturnal locomotor activity of the animal. However, under conditions of inverted photoperiod, the millipede maintained its tendency toward a more intense nocturnal respiratory rate (50,35 x 39,14). Similar behaviours were observed under constant illumination and constant darkness, in which G. olivaceus again presented higher nocturnal respiratory rates than diurnal ones(85,84 x 53,48 and 73,18 x 57,0, respectively). The present experimental data suggests the persistence of an endogenous rhythm where the light may not be an important exogenous synchronizer of the activity of G. olivaceus, because it was insufficient to block the start of the biological clock and the natural tendency of higher nocturnal activities of millipedes, principally when the tests were performed in constant illumination or darkness (free-running tests).

The aim of this investigation was to study the effect of organic acids and/or anaerobic cecal microflora (ACM) on systemic and digestive infection of broilers by Salmonella typhimurium and S. enteritidis. ACM was used without previous bacterial identification. The treatment with ACM increased the resistance to Salmonella spp infection. Infection was more evident in caeca, followed by rectum and crop and did not interfere on body weight of broilers. Treated and control groups showed the same degree of infection at the end of the experiment. The use of ACM isolated or combined with acetic acid, reduced the colonization of the chick's digestive system by S. typhimurium and S. enteritidis. Acetic acid added to ACM did not potentiate the reduction of digestive system colonization. Except for the crop, the isolated use of acetic, propionic or formic acids did not reduce S. typhimurium and S. enteritidis, in caeca and rectum. The use of organic acids and ACM had little effect on reduction of caecum pH. The treatment with ACM reduced the quantity of S. enteritidis in the faeces. The reduction of caecum pH did not reduce the quantity of S. enteritidis in faeces. S. enteritidis was much more invasive than S. typhimurium and use of organic acids and ACM had little effect on reduction of systemic infection.

This study investigated the effect of non-ventilation of the incubator during the first 10 days of incubation and its combination with dexamethasone administration at day 16 or 18 of incubation on hatching parameters and embryo and post-hatch chick juvenile physiology. A total of 2400 hatching eggs produced by Cobb broiler breeders were used for the study. Blood samples were collected at day 18 of incubation, at internal pipping stage (IP), at the end of hatch (day-old chick) and at 7-daypost-hatch for T-3, T-4 and corticosterone levels determination. From 448 to 506 h of incubation, the eggs were checked individually in the hatcher every 2 h for pipping and hatching. The results indicate that non-ventilation during the first 10-day shortened incubation duration up to IP, external pipping (EP) and hatch, had no effect on hatchability and led to higher T-3 levels at IP but lower corticosterone levels at 7-day-post-hatch. The injection of dexamethasone at days 16 and 18 of incubation affected hatching and blood parameters in both the ventilated and non-ventilated embryos differentially and the effect was dependent on the age of the embryo. Dexamethasone increased T-3 levels and T-3/T-4 ratios but the effect was greater with early non-ventilation of eggs. Dexamethasone decreased hatchability but the effect was greater when injected at day 16 and especially in ventilated embryos. The effects of incubation protocols and dexamethasone treatments during incubation were still apparent in the hatched chicks until 7 days of age. The changes in T-3, T-4 and corticosterone levels observed in response to the early incubation conditions and late dexamethasone treatments in this study suggest that incubator ventilation or non-ventilation may influence the hypothalamic-pituitary-adrenal axis (HPA) regulation of stress levels (in terms of plasma corticosterone levels) and thyroid function in the embryo with impact on incubation duration, hatching events and early post-hatch life of the chick. Our results also suggest that some stages of development are more sensitive to dexamethasone administration as effects can be influenced by early incubation protocols. (c) 2006 Elsevier B.V. Salmonella enterica serovar Enteritidis-lysing bacteriophages isolated from poultry or human sewage sources were used to reduce Salmonella Enteritidis in vitro and in experimentally infected chicks. Cocktails of 4 different bacteriophages obtained from commercial broiler houses (CB4O) and 45 bacteriophages from a municipal wastewater treatment plant (WT45O) were evaluated. In experiment 1, an in vitro crop assay was conducted with selected bacteriophage concentrations (105 to 101 pfu/mL) to determine ability to reduce Salmonella Enteritidis in the simulated crop environment. Following 2 h at 37 degrees C, CB40 or WT45O reduced Salmonella Enteritidis recovery by 1.5 or 5 log, respectively, as compared with control. However, CB40 did not affect total SE recovery after 6 h, whereas WT45O resulted in up to a 6-log reduction of Salmonella Enteritidis. In experiment 2, day-of-hatch chicks were challenged orally with 3 x 103 cfu /chick Salmonella Enteritidis and treated cloacally with 1 X 109 WT45O pfu/chick I h postchallenge. One hour later, chicks were treated or not with a commercially available probiotic (Floramax-B11). Both treatments significantly reduced Salmonella Enteritidis recovery from cecal tonsils at 24 h following vent lip application as compared with controls, but no additive effect was observed with the combination of bacteriophages and probiotic. In experiment 3, day-of-hatch chicks were challenged orally with 9 x 103 cfu/chick Salmonella Enteritidis and treated via oral gavage with I X 108 CB40 pfu/chick, 1.2 x 108 WT45O pfu/chick, or a combination of both, I h postchallenge. All treatments significantly reduced Salmonella Enteritidis recovered from cecal tonsils at 24 h as compared with untreated controls, but no significant differences were observed at 48 h following treatment. These data suggest that some bacteriophages can be efficacious in reducing SE colonization in poultry during a short period, but with the bacteriophages and methods presently tested, persistent reductions were not observed.

Crystallization of piratoxin I, a myotoxic Lys49-phospholipase A(2) homologue isolated from the venom of Bothrops pirajai. Toxicon 36, 547-551, 1998.-Large single crystals of piratoxin I, a Lys49-PLA(2) homologue with low enzymatic activity, have been obtained. The crystals belong to the orthorhombic system space group p2(1)2(1)2(1) and diffract X-raps to a resolution of 2.8 Angstrom. Previously, we reported that thermal conditioning at 39degreesC on days 13-17 of incubation of broiler eggs enabled thermotolerance during post-hatch growth (J. Therm. Biol. 28 (2003) 133). Tolerance to a temperature of 30degreesC was accompanied by changes in thyroid hormones and metabolic parameters. In the current study, we determined the mechanism of epigenetic heat adaptation during embryonic age by measuring blood physiological parameters that may be associated with the ultimate effects of thermal conditioning. Hatching eggs from Ross breeders were subjected to heat treatment of 39degreesC at days 13, 14, 15, 16 and 17 of incubation for 2 h per day. Control eggs were incubated at 37.6degreesC. Samples of eggs were withdrawn on each day of thermal conditioning and at internal pipping (IP) to obtain blood samples from embryos. The remaining eggs were weighed at day 18 and transferred to hatchers. The timing of IP, external pipping (EP) and hatching were monitored every 2 h. At hatch, chicks were weighed and hatchability was determined. Blood samples were obtained from samples of day-old chicks. T3, T4, corticosterone, pCO(2), pO(2) levels were determined in the blood. Blood pH was measured and T3/T4 ratios were calculated. Heat conditioning significantly increased corticosterone and pO(2) levels and blood pH but depressed pCO(2) at day 14. These were followed by a significant depression of T4 level on day 15. Remarkably, at day 16, all these parameters were back to normal as in the control embryos. Hatching was delayed by thermal conditioning probably as a result of the depressed corticosterone levels at IP. Hatchability was also lower in the heat-treated group but 1-day old chick weights were comparable to those of the controls. The result suggests that epigenetic thermal conditioning involves changes in these physiological parameters and probably serve as a method for epigenetic temperature adaptation since the same mechanisms are employed for coping with heat during post-embryonic growth. It also suggests that days 14-15 may be the optimal and most sensitive timing for evoking this mechanism during embryonic development. The adverse effects of heat treatment observed in this study may have been due to the continued exposure to heat until day 17. Fine-tuning thermal conditioning to days 14-15 only may improve these production parameters. (C) 2003 Elsevier Ltd. All rights reserved.

We studied the effect of feed and water deprivation on gastrointestinal tract and intestinal mucosa development of chicks at 24, 48, and 72 h posthatching. The treatments were water and feed ad libitum, water ad libitum and no feed, no water but feed ad libitum, and no water and no feed. The relative weight of the yolk sac was not influenced by the treatments. However, at 48 and 72 h posthatching, the relative weight of the liver increased, and the gizzard + proventriculus weight decreased in birds receiving feed ad libitum. An increase in jejunum and ileum relative weights and lengths was observed when the birds were supplied with feed and water. The lack of water produced the same effect as the lack of feed, both causing a higher number of villi per area with reduction in villus size, when compared with feed and water ad libitum treatments. The results of this study revealed that feed and water are able to affect intestinal villus development after hatching, indicating that both feed and water must be supplied to the chicks immediately after hatching.

In Neoponera villosa ants, we found ovaries of the polytrophic meroistic type which is characterized by the presence of nurse cells forming together with the oocyte, the so-called follicles. The nurse cells have the primary function of supplying the oocyte with RNA, but they contribute to the supply of other elements such as glycogen. With the objetive of detecting the presence of this substance in the ovarioles of workers and queens of N.villosa ante the ovaries were removed and processed according to electron microscopy technic for glycogen detection. Glycogen is a common element in insect oocytes and is abundantly distributed in the cytoplasm of N.villosa workers and queens. However, in ovarian follicles it can only be detected at stages ET and lit of development. Glycogen synthesis probably occurs predominantly in nurse cells which transfer it into the oocyte through the nourish pore. This process requires high energy expenditure that justify the large numbers of mitochondria associated with glycogen in the nurse cell cytoplasm. The amount of glycogen in the nurse cells of queens is slightly greater than workers.

This study was conducted to assess the effects of incubation temperature (34 C, 36[degree]C and 38[degree]C) and relative humidity (RH, 50% and 60%) on egg weight loss, embryo mortality, hatchability, incubation time and chick weight in eggs from red-winged tinamou. The eggs were placed in incubators that were operated at 34[degree]C, 36[degree]C, or 38[degree]C and 50% or 60% RH (mean wet bulb temperatures of 28[degree]C and 30[degree]C, respectively) from day 1 to hatching. Each treatment had two replicate groups of 30 eggs each. Hatchability varied with incubation temperature and RH and was highest for eggs incubated at 36[degree]C and 60% RH and lowest for eggs incubated at 38[degree]C. Early, intermediate and late embryo mortality were highest at 38[degree]C, 38[degree]C/50% RH, and 50% RH, respectively. Incubation period was longest at 34[degree]C and shortest at 38[degree]C/50% RH. Present results show the highest hatchability of red-winged tinamou eggs after incubation at 36[degree]C and 60% RH; highest embryo sensitivity to high temperature in the early period of incubation (1 to 7 days), to high temperature and low RH in the second period of incubation (8-14 days) and to low RH in the late period of incubation (after 15 days) and shortest incubation period with increasing temperature and RH.

A performance trial was conducted with broiler chicks to Study the effect of phytase (PHY) supplementation in diets formulated With reduced AME, Ca, and P. The nutrient digestibility was determined during the 14- to 21-d and 28- to 35-d periods. The treatments consisted of 3 diets (NC1, NC2. NC3) differing ill nutrient content and each diet with Or without supplemental PHY (NC1, 0 oi 500; NC2, 0 or 750; NC3, 0 or 1,000 U of PHY/kg feed) and I positive control diet (PC). Compared with the PC diet. negative control diets (NC) resulted in lower AME and apparent ileal amino acid digestibility for some amino acids. Phytase Supplementation of the NC diets increased AME. apparent ileal amino acid digestibility, and apparent ileal crude protein digestibility. Phytase addition also increased mineral absorption in 2 1 - and 35-d-old broilers fed NC diets. Reduced nutrient digestibility appears to be I factor in the weight gain and feed intake results. Reducing Ca and P content reduced feed intake in a stepwise fashion in the NC diets. Phytase increased feed intake and generally improved nutrient digestibility, which resulted in an increase in digestible nutrient intake. Averaged across NC diets. PHY improved body weight. Bone-breaking strength was the most consistent predictor of Ca and P reduction. All NC diets had significantly lower bone-breaking strength than the PC. Phytase supplementation of the NC diets gave bone-breaking strengths that were comparable to the PC. Diets with PHY had the highest bioeconomic index.

Mast cells are present in the eye of chick embryos from the 14th day onward, displaying metachromatic granules, mainly in the iris anterior surface and pectinate ligament. Ultrastructurally these cells show electron-dense granules and a few thin and short cytoplasmic projections in close contact with fibroblasts. Sometimes these contacts are extensive, with long fibroblast projections partially involving the mast cells. Gap junctions between mast cells and fibroblasts are observed only in the eyes of 16- and 20-day-old embryos. These intercellular specializations are represented by a close apposition of cytoplasmic membranes with an extension up to 300 nm. Gap junctions between mast cells and fibroblasts were not observed previously in vivo or in vitro, although in vitro studies have shown that a number of functionally critical interactions may occur between these cells. Our morphological findings suggest that, in vivo, fibroblasts interact with mast cells and may influence their maturation.
