With the increasing development of the brazilian sheep production, the producer was forced to achieve higher production rates. The use of artificial insemination has been shown as an important biotechnological tool in animal breeding. Among the various existing techniques, the superficial cervical insemination with fresh semen is demonstrating fertility rates between 70 and 80% (AISEN, 2008), in addition to its applicability, it does not require sophisticated equipment or manpower to highly specialized implementing these plans greater possibility in your job to maximize reproduction and greater dissemination of superior genetic material on the property. This study aims to address aspects of artificial insemination with fresh semen in sheep and its applicability in commercial herds biotechnology as a tool in assisted reproduction

The Doppler ultrasound allows the assessment of blood flow and the differentiate vessels according to each spectral pattern, location and organ that irrigate, helping the examinations and disease diagnosis. The main methods used are Pulsed Doppler, Color Doppler and Power Doppler. In Brazil veterinary medicine, it shows progress since 1990, but its use is limited by the equipment high cost, the need for operator experience and patient cooperation. There are studies in obstetrics and reproduction of dogs and cats, such as evaluation of the testicular vasculature, including the use of contrast material, and attempts to evaluate prostate in healthy dogs or pathological conditions (cancer and testicular torsion , orchitis and benign prostatic hyperplasia; estrous cycle monitoring, evaluation of irrigation in follicles and corpus luteum, uterus and breasts (mammary tumors) in bitches; monitoring pregnancy from the assessment of maternal, fetal and placental hemodynamics, both in dogs as in cats, and aspirations for early detection of pregnancy in dogs. The Brazilian livestock stands out for having the world largest commercial herd of cattle and leads meat exportation and production of bovine embryos. The in vitro production (IVP) of embryos is considered an effective option to overcome problems such as infertility in cows with high economic value and also for genetic improvement of cattle. The in vitro oocyte maturation is an essential step to the success of IVP, but is still considered poor when compared to in vivo maturation. Recent studies have suggeested an important role of Fibroblast Growth Factor 10 (FGF10) on the in vitro maturation of oocytes, which favored the expression of genes related to oocyte maturation and cumulus cell expansion. Aware that maturity stage influences the final production of blastocysts, we aimed study to verify if the addition of FGF10 into the maturation medium is able to affect positively the IVP of bovine embryos. Hence, FGF10 was added to maturation in five different concentrations: 0.5 ng/mL (group 0.5), 2.5 ng/mL (group 2.5), 5 ng/mL (group 5), 10 ng/mL (group 10) and 50 ng/mL (group 50). Additionally, two other maturation groups were used, group BSA (Bovine Serum Albumin, 4 mg/mL) and group FCS (Fetal Calf Serum, 10%). The rates of cleavage, morula and blastocyst were analyzed by Analysis of Variance (ANOVA), differences of P<0.05 were considered significant. Cleavage rates did not differ between the seven groups. On the other hand, morula rate on FCS group was higher than groups BSA, 0.5, 10 and 50 (P<0.05), but did not differ among groups treated with intermediate doses of FGF10 (2.5 and 5). FCS group presented higher blastocyst rate compared to all other groups that were well below the FCS group (P<0.0001). Therefore, the use of FGF10 during oocyte maturation did not affect positively embryo development on the IVP of bovine embryos

Haddadus binotatus é uma espécie endêmica de Mata Atlântica e ocorre desde o sul de Pernambuco até o Rio Grande do Sul. Apesar da sua ampla distribuição pouco se conhece da história natural dessa espécie. Haddadus binotatus apresenta desenvolvimento direto e ovos são depositados sob a serapilheira ou sob troncos caídos. Há dimorfismo sexual, sendo as fêmeas maiores do que os machos. O objetivo desse estudo é conhecer os padrões reprodutivos de populações de H. binotatus no continente e em ilhas do litoral do estado de São Paulo. Os objetivos específicos são: 1) avaliar as diferenças morfológicas entre populações de H. binotatus entre ilhas e entre ilhas-continente, 2) avaliar se há dimorfismo sexual em cada população, e 3) comparar os padrões reprodutivos entre populações insulares e continentais. Neste estudo, foram utilizados exemplares de H. binotatus, depositados na coleção de Anfíbios (CFBH), do departamento de Zoologia, Instituto de Biociências, UNESP, Campus de Rio Claro. Os exemplares são provenientes de uma localidade no continente (São Sebastião) e três ilhas (Couves, Queimada Grande e Ilhabela). Os exemplares provenientes das ilhas, com exceção daqueles da Ilha das Couves, foram maiores do que os da Juréia. Houve dimorfismo sexual em todas as populações, sendo as fêmeas maiores. As populações insulares apresentam número de ovócitos menor do que a população do continente (Juréia). O número de ovócitos semelhante aos da população da Juréia foi registrado para uma população em Minas Gerais, reforçando o padrão de que populações insulares têm desovas menores. No entanto, não houve diferença no tamanho dos ovos. As diferenças encontradas para as populações insulares podem ser resultado de recursos mais limitados como espaço e alimento, pelo tamanho das presas e/ou ausência de predadores. The dairy sector has undergone considerable economic losses due to low fertility rates due to adverse effects of heat stress on reproduction of cows. Genetic selection for increased production, coupled with the expanding dairy to tropical areas of the planet, and global warming has further aggravated the problem of heat stress. The effects of heat stress are multifactorial and act directly or indirectly at various levels of reproductive tissues, resulting in low fertility of cows, which in practice, results in reduced reproductive efficiency in the property, reducing the producers’ profit. Some strategies related to breeding biotechnology such as fixed-time artificial insemination, embryo transfer and use of BST, can minimize these effects and improve the reproductive efficiency of the herd

Nos peixes, durante a vida reprodutiva o desenvolvimento gonadal é cíclico e anual. As alterações estruturais das gônadas de machos e fêmeas provêm parâmetros úteis para a sua classificação macroscópica e microscópica. Porém, o desenvolvimento das células germinativas, nem sempre é consoante com os diferentes tipos de classificação e, machos e fêmeas nem sempre parecem concatenados. Neste estudo, aplicando-se propostas recentes, utilizadas primariamente para peixes derivados, notadamente os Perciformes marinhos, e tomando como modelo experimental representantes da série Otophysi, Hoplias malabaricus (Characiformes), Tanichthys albonubes (Cypriniformes), Sorubim lima (Siluriformes) e Gymnotus sp. (Gymnotiformes), e dos Perciformes Neotropicais, Cichla temensis, foram descritos, comparativamente, os eventos iniciais da oogênese e da espermatogênese, assim como o desenvolvimento oocitário a fim de traçar um paralelo entre grupos basais e derivados. A partir dos resultados, verificou-se que a maioria dos eventos morfológicos durante o desenvolvimento oocitário é similar entre grupos basais e derivados. Os Perciformes de água doce divergem dos grupos basais por duas características: a presença de inclusões lipídicas, que surgem no citoplasma perinuclear após o início da formação dos alvéolos corticais e deposição de vitelo e por uma tênue proteólise do vitelo durante a maturação final. A foliculogênese e espermatogênese não apresentam divergências e parecem ser processos conservados entre os Teleostei. Durante a fase de Regeneração, os eventos iniciais desses processos em fêmeas e machos, respectivamente, são responsáveis por altos índices de proliferação das células germinativas, quando comparado às demais fases. Of the four lacustrine deltaic models, which were found in the Pendência formation, two are represented in the Serraria field. Respectively the deltaic models 1 and 3 shows the reservoir zones A and B. The Zone A is divided into six sub-areas. Each is representing a smaller cycle of development of sigmoidal lobes of deltaic front. Zone B produces in reservoirs of Model 3, or so called Full delta. The Zone B is formed by overlapping the deltaic plain system over the deltaic/prodeltaic front (model 1). This work uses the method of zooming with the aim to contextualize the geometric aspects of the sand bodies, highlighting the analysis of facies and diagenesis with help of pictures and testimonies of thin sections. The sigmoidal lobes of Zone A are fine to very fine sandstones, well sorted, with a arcosian composition.;practically with a weak compaction and cementation of a kind of film of clay (if very fine) and overgrowth feldspar (fine texture). This silicate phases are succeeded by cementation of poiquilotópica calcite, and after this a stage of dissolution, containing only regular permoporosity for this reservoir. Zone B has a combination of two types of deltaic plain reservoir. One is the rarest of distributary channel and the other the most common of lobes of crevasse. In the channel coarse to medium-grained and poor to moderate sorted sandstones are formed (tuning up), and with a lytic arcosiana nature. Rarely there are cements, including growth of feldspar and rhombohedral dolomite, which prevent a high permoporosity of the reservoir. In the crevasse lobes, the sandstones are laminated, fine and well sorted, arkosic, rarely with overgrowth feldspar and calcite poiquilotópica, and with a good intergranular permoporosity

Climate change in tropical countries, like Brazil, causes major problems in dairy production due to an increase of heat stress effects. In recent years, milk production in Brazil increased 36.07%. The Southeast region remains a leader in production with herds of high producing Holstein cattle (mostly), which is more susceptible to heat stress. Thermal stress decreases fertility in direct and indirect ways. Conception rates are reduced of 40-60% during cooler months of the year and 10-20% in the warmer months. Negative effects of heat stress involve changes in reproductive hormones, follicular development, oocytes, and embryos, and decreased dry matter intake. Several studies discuss change in reproductive hormones, such as reduction in plasma concentration of GnRH, LH, and oestradiol, which lead to decreased detection of estrus and ovulation. Various methods are being studied to bypass these negative effects and increase the fertility of dairy cows under heat stress. Nos peixes, durante a vida reprodutiva o desenvolvimento gonadal é cíclico e anual. As gônadas passam por alterações de forma, de tamanho, de cor e de textura que são utilizadas como parâmetros para a sua classificação macroscópica. Porém, o desenvolvimento das células germinativas, nem sempre é consoante com essa classificação e, machos e fêmeas nem sempre parecem concatenados. Novas propostas de classificação vêm surgindo na tentativa de compatibilizar as informações macroscópicas e histológicas, considerando a atividade do epitélio germinativo de machos e fêmeas. Ocorre, no entanto, que essas propostas vêm sendo desenvolvidas em estudos com Perciformes marinhos. Aqui, tomando-se como modelo o Characiformes, Schizodon nasutus e o Perciformes, Plagioscion squamosissimus analisou-se comparativamente o ciclo reprodutivo de ambas as espécies, frente às classificações tradicionais e às novas propostas de classificação adicionadas das características histológicas das gônadas, e pode-se perceber que a análise macroscópica das gônadas, muitas vezes, não corresponde a sua verdadeira fase do ciclo reprodutivo, analisado microscopicamente. Isso se dá porque muitas das características cruciais para distinção das fases não são visíveis a olho nu, como folículos atrésicos, maturação do oócito ou anastomose de túbulos ou lóbulos testiculares. Assim, para determinação das fases reprodutivas, deve-se sempre contar com a análise microscópica e, no presente estudo, foi considerado que a melhor classificação para tal é a proposta de Brown-Peterson e colaboradores (2009), que divide o ciclo reprodutivo em seis fases reprodutivas, bastante detalhadas. A partir da descrição do ciclo reprodutivo feminino e masculino em S. nasutus e P. squamosissimus, os dados foram correlacionados com o índice gonadossomático, determinando, assim, que o período... In the last years Brazil has achieved a leading position in worldwide cattle breeding. This country is consolidating the practice of an intensive and technical level cattle breeding, due to greater emphasis on reproductive efficiency and genetic improvement. The attention given to reproductive efficiency became to enhance the genetic improvement process, by exploiting the full reproductive potential of the animals. The biotechniques applied to reproduction arised to expand the reproductive potential for greater economic return. Are the most important reproductive biotechniques the FTAI and in vivo and in vitro embryo production for embryo transfer. Different hormonal treatments are consolidating to interfere with normal physiology in order to achieve the desired goals. The FTAI works to increase the reproductive rates of cattle through pre-booked artificial insemination in all the females in reproduction, without the need for estrous detection. The in vivo and in vitro embryo production aims to distribute the genetics of males and females genetically improved by transferring their embryos to genetically inferior females. Superovulation treatments designed to prevent the mechanism of dominance, which results in many follicles ovulating simultaneously for in vivo embryo production. To the in vitro embryo production, the technique of ovarian puncture guided by transvaginal ultrasound allows to aspirate a number of quality oocytes. The application of biotech is based on knowledge of the female and ovarian reproductive physiology and knowledge of their acting in different breeds and environments.

Experimental models are necessary to elucidate pathophysiological mechanisms not yet understood in humans. To evaluate the repercussions of the diabetes, considering two methodologies, on the pregnancy of Wistar rats and on the development of their offspring. In the 1st induction, female offspring were distributed into two experimental groups: Group streptozotocin (STZ, n=67): received the β-cytotoxic agent (100mg STZ/kg body weight - sc) on the 1st day of the life; and Non-diabetic Group (ND, n=14): received the vehicle in a similar time period. In the adult life, the animals were mated. After a positive diagnosis of pregnancy (0), female rats from group STZ presenting with lower glycemia than 120 mg/dL received more 20 mg STZ/kg (ip) at day 7 of pregnancy (2nd induction). The female rats with glycemia higher than 120mg/dL were discarded because they reproduced results already found in the literature. In the mornings of days 0, 7, 14 and 21 of the pregnancy glycemia was determined. At day 21 of pregnancy (at term), the female rats were anesthetized and killed for maternal reproductive performance and fetal development analysis. The data were analyzed using Student-Newman-Keuls, Chi-square and Zero-inflated Poisson (ZIP) Tests (p<0.05). STZ rats presented with increased rates of pre (STZ=22.0%; ND=5.1%) and post-implantation losses (STZ=26.1%; ND=5.7%), reduced rates of fetuses with appropriate weight for gestational age (STZ=66%; ND=93%) and reduced degree of development (ossification sites). Conclusion: Mild diabetes led a negative impact on maternal reproductive performance and caused intrauterine growth restriction and impaired fetal development
