Intensive deforestation and forest fragmentation of the Brazilian forest biomes has contributed to increase the number of trees species under the risk of extinction. Peltophorum dubium (Sprengel) Taubert is one of these species. Some of its populations are being conserved ex situ by the Instituto Florestal de São Paulo through provenance and progeny tests. The aim of this study was to investigate the genetic variation and to estimate genetic parameters at age 24 years in a progeny test of P. dubium established in Luiz Antônio, São Paulo State, Brazil. The trial was established in a random block design, with six replications, five plants per plot and 18 open-pollinated progenies. Measured were: diameter at breast height (DBH), height and stem form. The genetic parameters heritability, genetic variation and effective population size were estimated. Significant genetic differences were observed only for DBH. This trait also presented a high coefficient of genetic variation (CV g=4.8%) and heritability, especially among progeny means (h m 2=0.6607). This indicates that DBH is the most indicated trait for selection in the population. The effective population size conserved ex situ in the test was estimated to be 38.9. Concerning genetic conservation, although the effective population size in the test is small, the values of the genetic variation and of the heritability indicate that the ex situ population has sufficient genetic variation and potential to respond to changes promoted by natural and artificial selection.

Monitoring the survival of nematophagous fungi is needed to establish periods of reapplication of formulations of nematophagous fungi to control the citrus nematode in the field. We monitored the survival of fungi: Arthrobotrys robusta, A. oligospora, A. musiformis Dactylella leptospora and Monacrosporium eudermatum in plots treated with 1, 2, 4, 6 liters of the formulation of fungi/plant or witness without the application, during the period of nine months with the first assessment six months after application and the other with intervals of three months after the first evaluation. The fungus D. leptospora was found only in the evaluation of 6 months after treatment application, indicated a short survival time in the soil. However, the isolated A. robusta, A. musiformis and A. oligospora were recovered in all evaluations and especially in plots treated with higher doses of the formulation and witness. Monacrosporium eudermatum was recovered in all experimental periods and even in assessing the witness portion of nine months after application. The fact of the presence of species of Arthrobotrys and M. eudermatum in control plots indicates that native species that were already orchard.

The software industry has become more and more concerned with the appropriate application of activities that composes requirement engineering as a way to improve the quality of its products. In order to support these activities, several computational tools have been available in the market, although it is still possible to find a lack of resources related to some activities. In this context, this paper proposes the inclusion of a module to aid in the requirements specification to a tool called Requirements Elicitation Support Tool. This module allows to specify requirements in accordance with IEEE 830 standard, thus contributing to the documentation of the requirements established for a software system, besides supporting the learning of concepts related to the requirements specification, which improves the skills of users of the tool. © 2012 IEEE.

Non-conventional database management systems are used to achieve a better performance when dealing with complex data. One fundamental concept of these systems is object identity (OID). Two techniques can be used for the implementation of OIDs: physical or logical. A logical implementation of OIDs, based on an Indirection Table, is used by NuGeM, a multimedia data manager kernel which is described in this paper. NuGeM Indirection Table allows the relocation of all pages in a database. The proposed strategy modifies the workings of this table so that it is possible to reduce considerably the number of I/O operations during the request and release of pages containing objects and their OIDs. Tests show a reduction of 84% in reading operations and a 67% reduction in writing operations when pages are requested. Although no changes were observed in writing operations during the release of pages, a 100% of reduction in reading operations was obtained. © 2012 IEEE.

Bovine mastitis is considered the main disease causing great economic losses in dairy herds. It is usually treated by antimicrobial chemicals that promote drug resistance, residues in food and environmental contamination. However, consumers in different countries requires more natural foods and with higher quality. Thus, the objective was to check the activities of propolis in controlling bovine mastitis. Seventy-two Holstein cows were used. The mastitis was identified by the California Mastitis Test, somatic cell counts and microbiological examination of milk. Four treatments were held: in group EAP1 10ml of a 30% alcoholic propolis extract (EAP) were given orally for seven consecutive days; in group EAP2 the same procedure described for the first group was used, in addition EAP was used for immersion of the teats before and after milking; in group CA alcohol was used for immersion of the teats before and after milking; and in group CT animals were subjected to soaking and disinfection, procedures routinely used by the property. The results were analyzed by the non-parametric variance model for repeated measures complemented by independent groups in multiple comparisons. There was a decrease of the somatic cell count in all groups. The biological activities of propolis provide great prospects; however, under the conditions evaluated, it was not possible to observe differences between treatments. The great diversity in its chemical composition and the complexity of multiple synergistic mechanisms involved in its biological activity require additional clinical trials.

The climates of the central and southern regions of São Paulo state in Brazil favor pathogens such as Puccinia psidii Winter, which causes a common and severe disease in Eucalyptus plantations under 2 years old. We studied genetic parameters including genotype by environment interaction (G × E) of resistance to P. psidii rust in Eucalyptus grandis at nine sites in São Paulo State. Open-pollinated progeny from ten 'provenances' were established in a randomized complete block design; at individual sites there were from 134 to 160 progenies, from four to eight blocks, and five to six trees per plot. Significant provenance and progeny(provenance) differences were detected, as was G × E involving progeny(provenance). However, the G × E involved little if any rank changes, indicating that selection can be done efficiently at a single site, if the disease level is sufficient. The estimated coefficient of genetic variation among the progeny within provenances CVg was high and variable among the sites (ranging from 11 % to 36. 7 %), demonstrating different expression of genetic variability among the sites. The estimated heritability at the individual-tree level h2 and within a plot hw 2 ranged from low to intermediate (ranging from 0. 04 to 0. 46) and was high at the progeny-mean level hf 2 (ranging from 0. 30 to 0. 86). Our study shows good prospects of controlling this disease by selection among and within progenies in a single site. © 2012 Springer-Verlag Berlin Heidelberg.

The relationship between the queens' lipid content and nest growth (population size, biomass and nest architecture) was studied from founding up to 1 year. Nests aged 3, 4, 5, 6, 9 and 12 months were dug in the field, and their dimensions were measured. The ant nest population and fungus garden was also collected. The sample was taken to the laboratory where we counted the worker population and weighed the biomass (fungus plus offspring) and queens. Queens were separated for the determination of lipids. The lipid content in the bodies of queens decreased in the first months, then stabilized (at 4-6 months) before increasing in months 9 and 12. Nest biomass (symbiotic fungus and offspring) and worker population increased over time. The structural growth of the nests was observed by excavating around them. Initially nests (3 months old) had one chamber at an average depth of 15 cm. By 1 year, the nests had three or four deep chambers, and were about 3-4 m deep. Our study contributes to knowledge of the dynamics of the energy-reserve expenditure by queens during colony founding and colony development for up to 1 year. © 2013 Copyright Taylor and Francis Group, LLC.

Ethnopharmacological relevance: In Brazilian traditional medicine, Arctium lappa (Asteraceae), has been reported to relieve gastrointestinal symptoms. Aim of the study: In the present study, we investigated the effects of the lactone sesquiterpene onopordopicrin enriched fraction (ONP fraction) from Arctium lappa in an experimental colitis model induced by 2,4,6 trinitrobenzene sulfonic acid and performed experiments to elucidate the underlying action mechanisms involved in that effect. Materials and methods: ONP fraction (25 and 50 mg/kg/day) was orally administered 48, 24 and 1 h prior to the induction of colitis and 24 h after. The inflammatory response was assessed by gross appearance, myeloperoxidase (MPO) activity, tumor necrosis factor alpha (TNF-α) levels and a histological study of the lesions. We determined cyclooxygenase (COX)-1 and -2 protein expressions by western blotting and immunohistochemistry assays. Results: TNBS group was characterized by increased colonic wall thickness, edema, diffuse inflammatory cell infiltration, increased MPO activity and TNF-α levels. On the contrary, ONP fraction (25 and 50 mg/kg) treatment significantly reduced the macroscopic inflammation scores (p<0.05 and p<0.01, respectively) and morphological alterations associated with an increase in the mucus secretion. Similarly, the degree of neutrophil infiltration and the cytokine levels were significantly ameliorated. Moreover, COX-2 expression was up regulated in TNBS-treated rats. In contrast, ONP fraction (50 mg/kg) administration reduced COX-2 overexpression. Conclusions: We have shown that the ONP fraction obtained from Arctium lappa exert marked protective effects in acute experimental colitis, confirming and justifying, at least in part, the popular use of this plant to treat gastrointestinal diseases. © 2013 Elsevier B.V. This study examined the effects of long-term creatine supplementation combined with resistance training (RT) on the one-repetition maximum (1RM) strength, motor functional performance (e.g., 30-s chair stand, arm curl, and getting up from lying on the floor tests) and body composition (e.g., fat-free mass, muscle mass, and % body fat using DEXA scans) in older women. Eighteen healthy women (64.9 ± 5.0 years) were randomly assigned in a double-blind fashion to either a creatine (CR, N = 9) or placebo (PL, N = 9) group. Both groups underwent a 12-week RT program (3 days week-1), consuming an equivalent amount of either creatine (5.0 g day-1) or placebo (maltodextrin). After 12 week, the CR group experienced a greater (P < 0.05) increase (Δ%) in training volume (+164.2), and 1RM bench press (+5.1), knee extension (+3.9) and biceps curl (+8.8) performance than the PL group. Furthermore, CR group gained significantly more fat-free mass (+3.2) and muscle mass (+2.8) and were more efficient in performing submaximal-strength functional tests than the PL group. No changes (P > 0.05) in body mass or % body fat were observed from pre- to post-test in either group. These results indicate that long-term creatine supplementation combined with RT improves the ability to perform submaximal-strength functional tasks and promotes a greater increase in maximal strength, fat-free mass and muscle mass in older women. © 2012 Springer-Verlag Berlin Heidelberg.

The aim of this study was to investigate the possible influence of different levels of aerobic fitness (VO2MAX) on the parameters of the running anaerobic sprint test (RAST). Thirty-eight subjects (Age = 18.1 ± 2.5 years, Height = 173 ± 1 cm and Body mass = 65.1 ± 6.5 kg) were classified into two groups, low and high aerobic fitness (LAF: n = 22 and HAF: n = 16). The VO2MAX was determined by an incremental exercise performed until exhaustion. The RAST was composed of six maximal efforts of 35m separated by 10s passive recovery. The VO2MAX was significantly different between groups (LAF = 51.7 ± 1.9 mL.kg -1.min-1; HAF = 58.6 ± 3.1 mL.kg -1.min-1). The mean power (MP) was significantly higher in the LAF (552.7 ± 132.1 W) in relation to the HAF group (463.6 ± 132.8 W). The impulse (ImP) was significantly correlated with the VO 2MAX in HAF. It can be concluded that there is an indication that the aerobic metabolism exerts an influence on the completion of RAST.

Despite the improvements in neoadjuvant chemotherapy, the outcome of patients with advanced bladder cancer has changed very little over the past 30 years. In the present study we tested and compared the in vitro antitumor activities of four different inhibitors of Polo-like kinase 1 (PLK1) (BI 2536, BI 6727, GW843682X, and GSK461364), against 3 bladder carcinoma cell lines RT4, 5637 and T24. The impact on radiosensitivity and drug interactions in simultaneous treatments with cisplatin, methotrexate, and doxorubicin were also investigated. Our results showed that PLK1 inhibition prevented cell proliferation and clonogenicity, causing significant inhibition of invasion of tumor cells, though modest differences were observed between drugs. Moreover, all PLK1 inhibitors induced G2/M arrest, with the subsequent induction of death in all 3 cell lines. Drug interactions studies showed auspicious results for all PLK1 inhibitors when combined with the commonly used cisplatin and methotrexate, though combinations with doxorubicin showed mostly antagonistic effects. Comparably, the four PLK1 inhibitors efficiently sensitized cells to ionizing radiation. Our findings demonstrate that irrespective of the inhibitor used, the pharmacological inhibition of PLK1 constrains bladder cancer growth and dissemination, providing new opportunities for future therapeutic intervention. However, further laboratorial and preclinical tests are still needed to corroborate the usefulness of using them in combination with other commonly used chemotherapeutic drugs. © 2013 Landes Bioscience.

Currently, many museums, botanic gardens and herbariums keep data of biological collections and using computational tools researchers digitalize and provide access to their data using data portals. The replication of databases in portals can be accomplished through the use of protocols and data schema. However, the implementation of this solution demands a large amount of time, concerning both the transfer of fragments of data and processing data within the portal. With the growth of data digitalization in institutions, this scenario tends to be increasingly exacerbated, making it hard to maintain the records updated on the portals. As an original contribution, this research proposes analysing the data replication process to evaluate the performance of portals. The Inter-American Biodiversity Information Network (IABIN) biodiversity data portal of pollinators was used as a study case, which supports both situations: conventional data replication of records of specimen occurrences and interactions between them. With the results of this research, it is possible to simulate a situation before its implementation, thus predicting the performance of replication operations. Additionally, these results may contribute to future improvements to this process, in order to decrease the time required to make the data available in portals. © Rinton Press.

Anopheles darlingi is the principal neotropical malaria vector, responsible for more than a million cases of malaria per year on the American continent. Anopheles darlingi diverged from the African and Asian malaria vectors ∼100 million years ago (mya) and successfully adapted to the New World environment. Here we present an annotated reference A. darlingi genome, sequenced from a wild population of males and females collected in the Brazilian Amazon. A total of 10 481 predicted protein-coding genes were annotated, 72% of which have their closest counterpart in Anopheles gambiae and 21% have highest similarity with other mosquito species. In spite of a long period of divergent evolution, conserved gene synteny was observed between A. darlingi and A. gambiae. More than 10 million single nucleotide polymorphisms and short indels with potential use as genetic markers were identified. Transposable elements correspond to 2.3% of the A. darlingi genome. Genes associated with hematophagy, immunity and insecticide resistance, directly involved in vectorhuman and vectorparasite interactions, were identified and discussed. This study represents the first effort to sequence the genome of a neotropical malaria vector, and opens a new window through which we can contemplate the evolutionary history of anopheline mosquitoes. It also provides valuable information that may lead to novel strategies to reduce malaria transmission on the South American continent.
