Dexamethasone is a synthetic glucocorticoid widely used to treat allergic and inflammatory processes. This drug is used in three main situations, are used to contain acute or chronic inflammatory processes, or like immunosuppressive drug's. In these cases the patient will receive high doses for a chronic period and, therefore, has a much greater chance of adverse side effects, such as hypertension, diabetes and dyslipidemia. Dexamethasone promotes deleterious effects on the arachidonic acid pathway, when administered in high doses, because it is a potent anti-inflammatory drug. We recently demonstrated that dexamethasone significantly reduces the protein expression of vascular endothelial growth factor (VEGF) in both skeletal muscle and heart, but the mechanisms involved remain unclear. Meanwhile, exercise has been shown to be effective against high blood pressure, diabetes and dyslipidemia, promoting, among other factors, the increase in VEGF and angiogenesis. One possible explanation for these effects would be the creation of new vessels mediated by inflammation, or by the stimulation of the formation of products of the metabolism of arachidonic acid (AA), such as prostaglandin E2 (PGE2) and VEGF, by increasing the stimulation of the enzymes cyclooxygenase 1 and 2 (COX-1 and COX-2). Little is known about the preventive effects of training on the action of dexamethasone in the arachidonic acid pathway. Therefore, the aim of this study was to determine whether aerobic exercise training, performed before and concomitant treatment with dexamethasone, was able to prevent the effects of the dexamethasone in the protein expression of COX-2 and VEGF. For this, we used young Wistar rats (n = 40) which were randomly divided into 4 groups: sedentary control (SC), sedentary and treated with dexamethasone (SD), trained control (TC) and trained and treated with dexamethasone (TD). The purpose of this work was to evaluate the biological compatibility of the Sealapex, Apexit, Sealer 26 and Ketac Endo endodontic cements. Polyethylene tubes containing these cements were implanted in the subcutaneous tissue of 40 (forty) rats. The animals were sacrificed after 14 and 90 days. A descriptive analysis of the reactions found in the connective tissue by contact with the cements was performed. The magnitude of inflammatory infiltrate, the presence and predominance of cell types and their distribution as to the filling material and reparative phenomena, such as fibroblastic and angioblastic proliferation and formation of fibrous capsules, were subjectively measured. After 90 days, all cements presented statistically significant reduction of the inflammatory reaction, presence of a fibrous tissue capsule in contact with the opening of the tubes containing the filling materials, and reduction of fibroblastic proliferation. Angioblastic proliferation decreased only for the Sealer 26 and Ketac Endo groups. All cements tested were either partially or totally phagocyted, and the mildest inflammatory response was found for the Sealer 26 group at both evaluation periods.

The biocompatibility of two dentin adhesives (Scotchbond MultiPurpose- 3M and Optibond Multi-Use - Kerr) was evaluated in rats subcutaneous connective tissue implants. Polyethylene tubes filled with the adhesives were surgically implanted into the dorsal subcutaneous tissues of rats. The tubes were left implanted for periods of 14, 30, 60 and 84 days, following wich the animais were killed and the implants, together with the surrounding tissue, were excised. The specimens were then fixed in 10 per cent formalin, sectioned serially and stained with hematoxylin and eosin and then, examined by light microscopy. After 14 and 30 days the tubes were surrounded by a mild to moderate inflammatory infiltration. After 60 and 84 days microscopic examination revealed the formation of a connective tissue capsule around each tube. The microscopic examination of the biopsy specimens allowed to conclude that: a) both dentin adhesives are considered biologically acceptable when placed in contact with rats subcutaneous connective tissue; b) in the observation period of 30 days there were moderate tissue reaction to the Optibond Multi-Use -light cure- (Kerr); c) after 60 and 84 days the use of both dentine adhesives did not interfere in the healing of the surrounding connective tissue
