Impermeabilidade do tegumento da semente à água ocorre em muitas espécies, inclusive em Schizolobium amazonicum Huber ex Ducke. Para promover a germinação de sementes com tegumento impermeável um dos métodos recomendados é o uso de ácido sulfúrico (H2SO4). O objetivo desse estudo foi identificar o melhor tempo de escarificação com ácido sulfúrico na superação da dormência em sementes de S. amazonicum, escarificadas durante 20, 40 e 60 minutos. Parte das sementes foi semeada imediatamente após a escarificação e parte após 24 horas de imersão em água, em uma mistura de areia e serragem (1:1), onde foram quantificadas a porcentagem e a velocidade de germinação. O delineamento foi inteiramente casualizado com quatro repetições de 50 sementes. A análise estatística da germinação foi efetuada aos seis, nove, 12, 15, 18, 21 e 24 dias após a semeadura em esquema fatorial. Para a velocidade de germinação as médias foram comparadas pelo teste de Tukey. Foi observada interação entre tratamentos para superação da dormência e o tempo de imersão em água após a escarificação na maioria das avaliações. A imersão em água acelerou o início da germinação. Os tratamentos para superação da dormência promoveram a germinação das sementes, entretanto, as sementes escarificadas durante 60 minutos apresentaram melhor germinação, 92 e 86,5% quando semeadas imediatamente e após 24 horas, respectivamente. O índice de velocidade de germinação foi superior nas sementes escarificadas durante 60 minutos e imersas em água. A escarificação durante 60 minutos constitui uma alternativa para redução da dormência das sementes.

As rodas compactadoras das semadoras-adubadoras têm como principal função, propiciar o bom contato entre o solo e a semente para garantir a germinação das sementes e a emergência das plântulas. Objetivou-se, neste trabalho, estudar a influência de três modelos de rodas compactadoras, três profundidades de semeadura e três níveis de carga sobre a roda compactadora sob a temperatura e o teor de água do solo. O trabalho foi desenvolvido na pista de ensaios de semeadura, localiza em Uberaba, MG, em Latossolo Vermelho distrófico, utilizando-se o delineamento experimental de parcelas sub-subdivididas, com 27 tratamentos e quatro repetições. As rodas com maior área de contato com o solo proporcionaram maiores valores do teor médio de água e da temperatura do solo. O nível de carga sobre a roda compactadora não afetou a temperatura e o teor de água do solo.

Seeds of Mimosa bimucronata are heavily infested (pre-dispersal predation) by the bruchid beetle Acanthoscelides schrankiae in Brazil. In this study, firstly we set up experiments to assess seed germination under seven and six different light and temperature regimes, respectively, and then we evaluated the ability of seeds to germinate after predation. We tested the hypothesis that the non-predated seeds from infested fruits may respond differently when set for germination than those seeds of non-infested fruits. We also hypothesized that predation may increase the production of unviable seeds. Seeds under 18 hours of light presented the highest percentage of germination, and the alternating temperature 20-30 degrees C was considered as optimum for germination (abnormal seedlings were not considered as a successful germination). Germination of seeds from non-infested fruits was significantly higher than germination of non-predated seeds from infested fruits, and predation also caused a significant increase in the proportion of dead seeds. Our results also show a positive correlation between proportions of unviable seeds and predated seeds. These results demonstrated that seeds of M. bimucronata are strongly affected by predation because predated seeds did not germinate and non-predated seeds had their viability reduced when located in infested fruits, supporting our hypothesis.

The objective of the present research was to determine the allelopathic potential of Ipomoea fistulosa plants on germination and growth of seedlings. In laboratory, lettuce and tomato seeds were placed to germinate in the presence of water or Ipomoea fistulosa leaf extract, in the concentrations of 5, 10 e 15% (p/v). In greenhouse, seeds were placed to germinate in trays containing Plantmax (R) substrate and Plantmax (R) substrate + Ipomoea fistulosa dry leaf residue, at 3% (w/w). The aqueous extract inhibited germination, raised the mean germination time of the seeds and reduced the radicle and hypocotyl growth in all used concentrations. In greenhouse, the plant height, leaf number, and dry mass accumulation also were reduced in the presence of the leaf residue in the substrate. The results suggest the presence of allelochemicals in Ipomoea fistulosa and demonstrate the highest sensitivity of lettuce that tomato to these compounds.

With the advance of agriculture, there was consequent rising in environment degradation. Therefore, It is interesting the use of plants which have in their chemical composition substances capable of helping in the control of weeds to minimize the use of pesticides. Hence, the objective of this paper was to evaluate solutions of leucena extract on weeds that are frequent on soybean crop. The treatments were leucena extracts at 0, 20, 40, 60, 80 and 100% at room temperature water and water heated to 80 degrees C. The extracts were obtained by crushing 200g of leucena leaves and mixing them with one liter of distilled water with the help of a blender. Seeds were kept in B.O.D. at a temperature of 25 degrees C, with a photoperiod of 12 hours of light. Evaluations were done oil a daily basis, from the first to the last day of the experiment, varying according to the species. Results show a negative interference of the extracts, both hot and cold, in the root length of Ipomoea grandifolia, in the percentage of germination and in the root length of Arrowleaf sida and Hair beggarticks. There was no negative interference in the analyzed parameters for soybean seeds. Thus, the results indicate allelopatic potential of Leucaena leuucocephala as an alternative for the management of weeds without interfering on the development of the soybean crop.

The aim of this work was to evaluate the allelopathic effect of water extract from sunflower fresh leaves on the germination and initial development of conventional and transgenic soybean cultivars, and of the invasive hairy beggartick (Bidens pilosa L.). Experiments were carried out at Laboratory of Plant Physiology, Universidade Estadual do Oeste do Parana, in 2007. Fresh leaves from sunflower were ground in a blender at the proportion 200g/1L distilled water, resulting in crude extract (100%), in which pH was measured. Dilutions of 80%, 60%, 40% and 20% were done from the crude extract, and only distilled water was used as control. Statistical analysis (Tukey 5% probability) indicated that only the highest extract concentrations (60%, 80% and 100%) influenced the analyzed parameters for conventional and transgenic soybean. Beggartick seeds, however, had germination percentage completely inhibited when 40% water extract was applied, which indicates that sunflower straw could be used as a natural herbicide. Nevertheless, field studies must be carried out to prove such effect.

The family Meliaceae has been identified as one of the most promising for biological control, as most species have biologically active compounds. Among the species, the Neem (Azadirachta indica A. Juss) is that stands out most for its efficiency and low toxicity, and it is utilized to control insects, fungi and nematodes. The aim of this work was to verify the allelopathic potential of aqueous extract of fresh leaves of Neem on germination of lettuce, soybeans, maize, beans and Bidens pilosa, as a way to control this weed. The extract was obtained by grinding 200g/L of fresh leaves of the Neem (considered extract 100%) and diluting in distilled water at concentrations of 80, 60, 40, and 20%, and a control with only water. The seeds were kept in the germination chamber at a temperature of 25 degrees C, with photoperiod of 12 hours of light. The aqueous extract affected the germination percentage and average length root of lettuce, with values of 1% and 0.1, cm respectively. For the soybeans, the extract influenced negatively the average time of germination and average speed of germination. There was a drastic reduction of the average root length at higher concentration for soybean, corn and beans. For Bidens pilosa all the parameters analyzed were negatively affected, which means sensitivity to the extract. The bioassay performed in the laboratory showed that the aqueous extract of fresh leaves of Azadirachta indica has allelopathic effect on all species.

Bioassays were carried out to identify and characterize the allelopathic potential effects of forage legumes leucena (Leucaena leucocephala), mineirão (Stylosanthes guianensis cv. Mineirão) and calopogônio (Calopogonium mucunoides) on germination and radicle elongation of the pasture weeds desmódio (Desmodium adscendens), guanxuma (Sida rhombifolia) and assa-peixe (Vernonia polyanthes). Aqueous seeds and shoot extracts were prepared in a concentration of 10% (w/v). The pH and osmotic potential were measured in each extract. The effects of the osmotic potential on the results were calculated considering the regression equations adjusted to the variations of osmotic potential in each parameter and the osmotic potential of the extracts. The results showed that the pH did not constitute in a source of variation of the results. The donor species indicated allelopathic potential that varied in function of donor and receiver species and part of the donor plant. The aqueous shoot extract of mineirão and calopogônio showed inhibition potential higher than the extract from seeds, while for leucena the effects more evident were obtained with the extract from seeds. Comparatively, the radicle elongation was a more sensitive indicator than germination to the effects of the extracts.The receiver species assa-peixe was the less sensitive to the effects of the extract.

The isolation of Clostridium botulinum from honey samples is described. Botulism is characterized as an intoxication provoked by ingestion of contaminated foods with this toxin. Infant botulism happens by the ingestion of spores of C. botulinum together with food that in special conditions of the intestinal tract, such as those present in babies of less than 1 year old, will allow the germination and colonization of the intestine with production and absorption of botulinic toxin. The samples were subjected to dilution and to a thermal shock and cultivated in modified CMM (Difco). Cultures were subjected to Gram smears and toxicity tests in mice. The toxic cultures were purified in RFCA (Oxoid) plates and incubated in anaerobic jars. Positive samples were typed using the mouse assay neutralization test. From the 85 honey samples analyzed, six were positive for C. botulinum (7.06%), and identified as producers of type A, B, and D toxins.

The species is a very popularly used plant as woundwort. Leaves, top flowers and rhizomes are used. The objective of this work is a contribution to the germination study, photoblastism and more adequate temperature for reproduction of the species. The best temperature for the germination was found to be 20°C and no seeds germinated under continuous darkness, within 14 days; therefore, the seeds are positive photoblastics. The seed germination percentage was low, in relation to the mass of the achene produced, varying from 8,2 to 20,0%. The alternated temperatures of 20° and 30°C did not influence the seed germination and the average of the weight of 100 seeds was 0,026 1 g.

The aim of the present work was to evaluate plasma membrane integrity, motility, vigor and morphology of fresh and frozen goat spermatozoa with or without seminal plasma. Semen samples were diluted in Tris solution, before and after thawing, with a combination of carboxifluorescein diacetate and propidium iodide. The results showed differences (P < 0.01) for motility and minor defects in the presence or absence of seminal plasma, for both fresh and frozen samples. Periods of collection had a significant effect on motility, probably due to changes in the photoperiod. Plasma membrane integrity was significantly reduced by the freezing process, whether seminal plasma was present or absent. In conclusion, removal of seminal plasma decreased motility and vigor rates in frozen samples. The photoperiod probably decreased the testosterone level, contributing negatively to the high percentage of sperm abnormalities, mainly damaged membranes. The use of fluorescent probes allowed a better estimation of the percentage of functional cells, instead of only estimating the percentage of motile cells or morphology defects. © 2001 Elsevier Science B.V.
