Com o objetivo de verificar o efeito da adição de níveis crescentes de óleo no concentrado sobre a atividade plasmática das enzimas creatina quinase (CK), aspartato aminotransferase (AST) e lactato desidrogenase (LDH) como indicativo de metabolismo energético, foram fornecidas dietas experimentais compostas de cinco níveis de óleo (controle, 6, 12, 18 e 24% do concentrado). Utilizaram-se 20 equinos da raça Árabe, peso médio de 400 kg, submetidos a prova de enduro de 80 km em esteira rolante. O enduro foi dividido em quatro anéis de 20 km, com duração média de 1 hora e dez minutos. A adição de óleo e a distância percorrida tiveram efeitos sobre as variáveis AST, CK e LDH, que apresentaram as respectivas expressões: AST (7,045-0,2292x+0,007991x2+0,008517z- 0,0003282xz), CK (8,06-,07020x+0,05546x2-0,001262x3+0,01204z+0,0006207xz) e LDH (6,624-0,3522x+0,03448x2-0,0008382x3+0,02401z-0,0007489xz) . O óleo é uma importante e bem aproveitada fonte de energia para equinos em exercício, pois sua adição na dieta de animais submetidos a prova de enduro promoveu alteração metabólica que favorece a produção de energia. O metabolismo animal poupou suas reservas energéticas oriundas da glicose, favorecendo a utilização do óleo. A menor atividade plasmática das enzimas AST, CK e LDH com a adição de óleo nas dietas indica direcionamento do metabolismo energético para a β-oxidação. Como apresentam várias isoenzimas, as enzimas estudadas atuam amplamente no metabolismo energético, favorecendo a constante reposição de ATP ao longo do exercício.

Quatro búfalos com cânulas ruminais foram distribuídos aleatoriamente em quadrado latino 4 x 4, compreendendo quatro rações com feno de capim-coastcross (Cynodon dactylon) (50% MS) e níveis crescentes de polpa cítrica peletizada em substituição ao milho em grão moído no concentrado (0, 33, 66 e 100% MS). Foram avaliadas a degradabilidade dos nutrientes e a população de protozoários no rúmen. Houve efeito quadrático da fração solúvel dos nutrientes entre os tratamentos, com valores maiores nos dois extremos. A taxa de degradação da PB do feno e da MS da polpa cítrica apresentou-se linear com aumento da polpa cítrica na ração. As rações contendo polpa cítrica promoveram menores concentrações de Entodinium e número total de ciliados/mL que a dieta só com milho. Houve predominância dos Entodinium nas dietas contendo somente milho ou somente polpa cítrica, enquanto em mistura predominaram os ciliados da subfamília Diplodiniinae. Observou-se efeito de interação da polpa cítrica com o milho, promovendo modificações no metabolismo ruminal, especialmente na solubilidade dos nutrientes, na curva do pH e na composição da fauna.

The putative translation initiation factor 5A (eIF5A) is a highly abundant and conserved protein in all eukaryotes and archaebacteria. This factor is essential for cell viability and is the only cellular protein known to contain the unusual amino acid residue hypusine. In Saccharomyces cerevisiae eIF5A is expressed in aerobic conditions by the gene TIF51A. Although eIF5A has been known for almost 30 years, the biological role of this protein is still obscure. This article reviews the research on the function of eIF5A, discussing the evidence for its involvement in various steps of mRNA metabolism, including translation initiation, nucleocytoplasmic transport and mRNA decay. Moreover, it indicates other studies that have associated eIF5A with cell proliferation and cell cycle progression. Finally, this review presents recent results obtained in our laboratory that reemphasize the role of eIF5A in the translation scenario. Further experiments will be necessary to define the role played by eIF5A in the translational machinery.

Type-1 diabetes patients suffer from frequent episodes of acidosis caused by an increased fatty acid metabolism and consequently increased plasma level of acetoacetate (AcAc) and β-hydroxybutyrate (β-HOB). This article describes a study of the effects of pathological concentrations of AcAc and β-HOB on lipoperoxidation, cell viability and the release of the CXCL8 (IL-8) cytokine by activated neutrophils. Neutrophils from healthy donors were isolated by density gradient (Histopaque® 1077/1119) and incubated with the ketone bodies. Lipoperoxidation was determined as thiobarbituric acid reactive substances (TBARS). The cell viability was evaluated by the release of intracellular lactate dehydrogenase. The release of CXCL8 was measured by ELISA in a 24-h culture of opsonized zymosan-stimulated neutrophils. AcAc, but not β-HOB, provoked a dose-dependent increase in the neutrophil membrane lipoperoxidation (p<0.05; r =0.9915). In the cytotoxicity assay, a dose-dependent release of LDH was observed when the neutrophils were incubated with AcAc in concentrations up to 40 mM (p<0.05). β-HOB was devoid of effect. The release of CXCL8 was inhibited by AcAc and β-HOB in a dose-dependent manner. In conclusion, these results suggest that the accumulation of ketone bodies in diabetic patients could be involved in their usually increased susceptibility to infection.

The maximal oxygen uptake (VO2max) is the maximal quantity of energy that can be produced by the aerobic metabolism in certain time unity. It can be determined direct or indirectly by predictive equations. The objective of this study was to make a specific predictive equation to determine the VO 2max from boys aged 10-16 years-old. Forty-two boys underwent a treadmill running ergospirometric test, with the initial velocity set at 9 km/h, until voluntary exhaustion. By the multiple linear regression was possible to develop the following equation for the indirect determination of the VO 2max: VO2max (ml/min) = -1574.06 + (141.38 x Vpeak) + (48.34 * Body mass), with standard error of estimate = 191.5 ml/min (4.10 ml/kg/min) and coefficient of determination = 0.934. We suggest that this formula is appropriate to predict VO2max for this population.

The aim of this study was to investigate the possible influence of different levels of aerobic fitness (VO2MAX) on the parameters of the running anaerobic sprint test (RAST). Thirty-eight subjects (Age = 18.1 ± 2.5 years, Height = 173 ± 1 cm and Body mass = 65.1 ± 6.5 kg) were classified into two groups, low and high aerobic fitness (LAF: n = 22 and HAF: n = 16). The VO2MAX was determined by an incremental exercise performed until exhaustion. The RAST was composed of six maximal efforts of 35m separated by 10s passive recovery. The VO2MAX was significantly different between groups (LAF = 51.7 ± 1.9 mL.kg -1.min-1; HAF = 58.6 ± 3.1 mL.kg -1.min-1). The mean power (MP) was significantly higher in the LAF (552.7 ± 132.1 W) in relation to the HAF group (463.6 ± 132.8 W). The impulse (ImP) was significantly correlated with the VO 2MAX in HAF. It can be concluded that there is an indication that the aerobic metabolism exerts an influence on the completion of RAST.
