AimTo evaluate the antibiofilm activity of sodium hypochlorite (NaOCl) and chlorhexidine (CHX) solutions associated with cetrimide (CTR), and QMiX using confocal laser scanning microscopy.MethodologyEnterococcus faecalis (ATCC- 29212) biofilms were induced on bovine dentine blocks for 14days. The dentine blocks containing biofilm were immersed for 1min in the following solutions: 2.5% NaOCl; 2.5% NaOCl+0.2% CTR; 2% CHX; 2% CHX+0.2% CTR; 0.2% CTR; QMiX. After contact with the solutions, the dentine blocks were stained with Live/Dead((R)) BacLight for analysis of the remaining biofilm using confocal laser scanning microscope. Images were evaluated using the BioImage_L software to determine the total biovolume (m(3)), the green biovolume (live cells) (m(3)) and the percentage of substrate coverage (%). The data were subjected to nonparametric statistical test using Kruskal-Wallis and Dunn's tests at 5% significance level.ResultsAfter exposure to irrigants, the total biovolume observed for CHX, CHX+CTR, CTR, QMiX was similar to distilled water (P>0.05). NaOCl and NaOCl+CTR had the lowest total and green biovolume. The CTR and QMiX had intermediate green biovolume, with greater antibacterial activity than CHX and CHX+CTR (P<0.05). The NaOCl and NaOCl+CTR solutions were associated with microorganism removal and substrate cleaning ability.ConclusionsNaOCl and NaOCl+CTR solutions were effective on microorganism viability and were able to eliminate biofilm. The addition of cetrimide did not influence antibacterial activity.

Aim To assess (i) heat generated by pluggers during warm vertical compaction of gutta-percha and investigation of temperature changes on the external root surface during canal filling, and (ii) the chemical changes of root canal sealers induced by heat.Methodology Four sealers, namely AH Plus, MTA Plus and two other experimental sealers based on tricalcium silicate, were characterised. External temperatures generated on the root surface during warm vertical compaction of gutta-percha with different sealers inside the root canal were monitored using an infrared thermography camera. Chemical changes induced by heating the sealers were assessed by Fourier transform infrared (FT-IR) spectroscopy.Results MTA Plus and the experimental sealers were composed of a cement and radiopacifier, with epoxy resin or a water-soluble polymer as dispersant, whilst AH Plus was epoxy resin-based. The heat generated at the tips of the continuous wave pluggers was found to be lower than the temperature set and indicated on the device LCD display. The sealers reduced the heat generated on the external root surfaces during the heating phase. AH Plus sustained changes to its chemical structure after exposure to heat, whilst the other sealers were unaffected.Conclusions The temperatures recorded at the tips of continuous wave pluggers varied with their taper and were lower than the temperature set on the System B LCD display. Root canal sealers reduced the dissipation of heat generated during warm vertical compaction, with the temperature at the external root surface maintained at 37-41 degrees C, a temperature below that is necessary to cause irreversible damage to bone and periodontium. The use of AH Plus sealer during warm vertical compaction techniques results in chemical changes in the sealer. The effect on sealer properties needs to be further investigated.

The aim of this study it was to evaluate the use of irrigating solution used during root canal preparation on the adhesive cementation of prefabricated fiberglass pins. The bond strength between different regions of the root and the fiberglass pins cemented into the root canal were evaluated by push-out assay. For this study, 36 human teeth were used, all equally prepared. The irrigating solutions used for biomechanical preparation were: saline solution, sodium hypochlorite 2,5% and glycolic extract of salvia 20%. The dual cure resin cement was used for cementation of pins. After cementation, specimens were sectioned to give 3 slices of approximately 2 mm each (cervical, middle and apical), which were submitted to mechanical push-out test. Were performed ANOVA and Tukey's test with a significance level of 5%, for the analysis of mechanical test. Comparing the thirds we observed that the only solution that presented statistical difference in the bond strength was sodium hypochlorite, which showed higher values for the middle and apical third compared with the cervical third, but when compared without taking into consideration the dental thirds, solutions studied showed no differences in bond strength. Therefore, it was concluded that the solutions studied can be used during endodontic treatment without causing harmful effects on the final adhesive restoration

With the emergence of new filling materials with different properties and behaviors, the approach of endodontic treatment must be readjusted so that the appropriate result can be achieved. New endodontic sealers include methacrylate resin-based, plant resin-based and the evolution of epoxy-based sealers. This study verified the behavior of new materials that presents controversial results in the literature, about coronal bacterial leakage. That for, 56 single-rooted human teeth were prepared in the direction crown-apex and filled with gutta-percha points with taper of 4% using the single cone technique. Roots were divided randomly into 4 groups according to the sealer (Apexit Plus, AH Plus, EndoREZ and Polifil). After filling, the roots were incorporated in a leakage model, which upper chamber contained a suspension of Streptococcus mutans, and lower chamber a broth, leaving 3 mm of root apical portion immersed. Leakage was assessed for turbidity in lower chamber every day for 60 days. Survival analysis was performed using the nonparametric Kaplan- Meier method (p<0,05). All experimental groups presented leakage during the study’s period, however the maximum time achieve was 22 days. The medium time of leakage was: Apexit Plus 6,3 days, AH Plus 6,3 days and Polifil 5,1 days, but in EndoREZ all specimens infiltrated in the first day, presenting shorter capacity of impermeabilization compared to the other groups. Concluding that none of the sealers tested was able to prevent coronal bacterial leakage

The biological principles of osseointegration caused the rehabilitation treatment with osseointegrated implant become a safe, well alternative accepted by the dental community for the high success rate, allowing the preparation of functional and aesthetic prostheses in edentulous and partial patients. We passed the initial phase of functional vision for an aesthetic approach, too, depending on the demands of the patient and the quest for excellence by professionals. Over these last years, implant treatment has undergone many changes in surgical and prosthetic protocols. The less invasive surgical techniques and the development of restorative materials, especially ceramics allow the prosthetic rehabilitation of high functional quality and aesthetics. The installation of implants in sockets immediately after extraction of teeth involved by fracture, periodontal disease, endodontic lesions, is part of the arsenal of techniques for rehabilitating a number of advantages that we get to the outcome of multidisciplinary treatment. For this, we must consider several criteria in its planning as a fundamental unit, adjacent teeth, multiple losses, system, form and number of implants, type of retention of the prosthesis components, the patient expectation of the outcome perspective, gingival biotype, standard of ridge resorption, the smile line, etc. occlusal analysis. With the three-dimensional placement of the implant can immediately join the biological processes of repair of the socket, implant osseointegration, speeding treatment time. Much has been made by companies in the surface treatment of endosseous implants to be osseointegration shortened and restorative procedures started earlier, bringing benefits to the patient and professional. Among the main advantages we can emphasize the preservation of the structures adjacent to the teeth replaced, minor resorption of bone tissue involved... The maxillary lateral incisor has a very peculiar and internal morphology that can be influenced by the its external anatomy such as supernumerary roots and ruts deep root, which often hinders an effective endodontic treatment. Generally, this tooth have a single root with one root canal, but teeth with two to four channels can be found. The main canal can present ramifications such as accessory canals, laterals, collaterals, recorrents and apical deltas. All this factors interfere directally on endodonthics treatment. The objective of this work is a study of the internal anatomy of the maxillary lateral incisors by digital and conventional radiographics methods, comparing the results obteined by each method Six hundred upper lateral incisor were used in this study teeth of the collection of the Department of Anatomy ICT – UNESP. All the teeth were radiographed by vestibular face, using a digital radiographic unit(Dabi Atlante). After this step, all the teeth underwent by conventional radiographic technique(Kodak 2200) so that the results obtained were compared. To classify the channel system, it was adopted the classification of Pucci and Reig. All data obtained by digital technique were compared with those obtained in the conventional technique and were subjected to statistical analysis

The objective of this study is to evaluate the measurement accuracy of endodontic files obtained by digital and conventional radiographies in primary teeth. Kerr and Hedströen files (# 20), with the reference as the apparent length of tooth, were inserted in the root canal of 18 extracted primary teeth, which were x-rayed by digital and conventional techniques. Measurements from a reference point to the apical end were carried out by an experienced operator twice in a week. An electronic ruler was used for the digital method and a caliper was used for the conventional method. The data were subjected to Pearson correlation test and Student´s t test (p = 0.05). The correlation between the first and the second measurements was r = 0.99, regardless the type of file and method. Comparing the measurements within the methods, the agreement was r = 0.96 for Kerr and r = 0.95 for Hedströen files. The values of length files in the digital radiographies were statistically lower than that obtained in the conventional radiographies (p = 0.02). However, the values obtained by the two methods were statistically similar to real length of teeth for Kerr files (p = 0.29) and for Hedströen files (p = 0.18). The digital radiography was a more trustful method to obtain the lengths of endodontic files.

Aim To assess the initial cytotoxicity and the late phenotype marker expression of odontoblast-like cells (MDPC-23) subjected to less aggressive in-office bleaching therapies. Methodology A 17.5% hydrogen peroxide (H2O2) gel was applied for 45, 15 or 5 min to enamel/dentine discs adapted to trans-wells positioned over cultured MDPC-23 cells. No treatment was performed on the negative control. Immediately after bleaching, the cell viability, gene expression of inflammatory mediators and quantification of H2O2 diffusion were evaluated. The ALP activity, DSPP and DMP-1 gene expression and mineralized nodule deposition (MND) were assessed at 7, 14 or 21 days post-bleaching and analysed statistically with Mann–Whitney U-tests (α = 5%). Results H2O2 diffusion, proportional to treatment time, was observed in all bleached groups. Reductions of approximately 31%, 21% and 13% in cell viability were observed for the 45-, 15- and 5-min groups, respectively. This reduction was significant (P < 0.05) for the 45- and 15-min groups, which also presented significant (P < 0.05) over-expression of inflammatory mediators. The 45-min group was associated with significant (P < 0.05) reductions in DMP-1/DSPP expression at all periods, relative to control. The ALP activity and MND were reduced only in initial periods. The 15-min group had less intense reduction of all markers, with no difference to control at 21 days. Conclusions The 17.5% H2O2 applied to tooth specimens for 5 min caused no alteration in the odontoblast-like cells. When this gel was applied for 45 or 15 min, a slight cytotoxicity, associated with alterations in phenotypic markers, was observed. However, cells were able to recover their functions up to 21 days post-bleaching.
