Photodynamic Therapy (PDT) is a therapeutic method which employs a photosensitizer and light to cause cellular death. The chemical compounds have low or none toxicity for hosts cells. Under the incidence of light, in an appropriated wavelength, these chemical compounds produce reactive oxygen which affects the biomolecules of the target-cells. The specific illumination of the affected area increases the selectivity of the therapy, since the photodynamic process occurs only in the irradiated area. Pythiosis, for instance, is a life-threatening emerging disease caused by a fungus-like organism called Pythium insidiosum. The disease occurs in man and other animals, being mostly observed in horses. Human pythiosis may present as ophthalmic, cutaneous-subcutaneous and systemic forms of lesions. Due to the fact that P. insidiosum is not a true fungus, it is refractory to most antifungal drugs and the treatment of the disease is difficult. Extensive surgery procedures, such as limb amputation, are the treatment of choice, however relapses may occur frequently. Although not totally effective, the use of immunotherapy associated to surgery have shown some results. Considering that pythiosis is an emerging disease few explored in its etiological and therapeutic aspects, which are limited and few effective, it is of great importance to encourage the development of researches for new strategies of treatment. In this sense, it was evaluated the effect of PDT on in vitro growth of the pathogen employing two chemical compounds as photosensitizer, porphyrin and chlorine, at different concentrations in combination with several energetic dosages. Porphyrin showed inhibition of growth at 25mg/mL with 100J/cm2 of energetic dosage and chlorine showed similar results employing low concentrations (0,7, 1,0 and 1,3mg/mL) with 70J/cm2 of energetic dosage... The Canine Visceral Leishmaniasis is a chronic disease of endemic character, caused by Leishmania Chagasi in Americas. The inoculation of the promastigote form in the individual triggers a local and widespread immune reaction with formation of inflammatory infiltrates and deposition of immune complexes in tissues. Initial clinical symptoms of the disease are: weight reduction, hepatomegaly, splenomegaly and, according to the disease chronicity, signs such as alopecia, erythema, onychogryphosis, arthropathies, renal diseases, pyoderma, seborrheic dermatitis, muscle atrophy and Ocular diseases. Ocular diseases are often reported and are result of the direct parasitism or immune-mediated mechanisms caused by the disease. The Leishmania spp have greater affinity for the anterior segment, so that anterior uveitis is one of the most frequently diagnosed injuries. Blepharitis diffuse and Keratoconjunctivitis also appear as important ocular changes. In histological section, inflammatory infiltrates and macrophages with amastigote form are observed in all ocular tissues, with the exception of the retina and optical nerve. In the clinical analysis and disease diagnosis, should be considered the differential diagnosis, such as Ehrlichiosis and systemic hypertension, because these may cause some ocular manifestations similar to those observed in leishmaniasis

Sugarcane (Saccharum sp.) is an important grass cultivated in tropical and subtropical regions of the world, such as in Br, Ind, and Ch, and has its biomass being raw material for production of sugar, fuel ethanol, and some other derivatives. Fungal diseases infect sugarcane fields worldwide, damaging crops and thus, causing great economic losses. Fungal specialized structures act during all Pathogen-Host Relationship Cycle (PHRC) (survival, dissemination, infection, colonization, and reproduction of pathogen), maintaining fungal populations in cultivation soil, infecting plants in following crops and vegetative propagation of sugarcane by infected seeds may allow fungal transportation into regions where diseases haven’t occured before. Biotechnological methods and approaches have significantly contributed to understanding of the relationship among parasite and host, as to diseases management (control, detection, and prevention). Some techniques have daily applications in Agriculture, while others are only used in research and to breeding of host resistant varieties. Among notable diseases, smut (Sporisorium scitamineum) and pineapple disease (Ceratocystis paradoxa) are important because they cause damage and losses in sugarcane regions, although there are different periods for each one to occur. This work aims to review the PHRC for each patosystem, the biotechnological methods and approaches and its perspectives in the study and management of these diseases. As environment is an important factor to the effectiveness of PHRC, one chapter is dedicated to Global Climate Change (GCC) and its possible influences over these diseases in a longterm period

A fagocitose de células apoptóticas é um processo dinâmico e de fundamental importância para homeostase dos tecidos após uma injúria. A fagocitose de células apoptóticas promove a síntese de mediadores anti-inflamatórios como PGE2, TGF-β e IL-10, podendo resultar na supressão da resposta imune do hospedeiro contra agentes infecciosos. Entretanto, um elegante estudo utilizando células apoptóticas infectadas demonstrou que a fagocitose destas células promove a geração não apenas de citocinas anti-inflamatórias como TGF-β mas também de IL-6 e IL-23, promovendo um efeito imunoestimulador, a diferenciação de células Th17. A atuação da PGE2 na imunidade adaptativa vem sendo investigada quanto à diferenciação e ativação de linfócitos Th1, Treg e Th17. Os resultados aqui apresentados demonstram que o protocolo de diferenciação de células dendríticas utilizado foi capaz de gerar em torno de 85% de CD imaturas evidenciado pela expressão de um perfil fenotípico CD11c+CD11b+MHCIIlowCD80lowCD86low. Quanto à produção de PGE2, a fagocitose de AC+PAMP por células dendríticas foi capaz de induzir níveis elevados deste mediador lipídico nas diferentes proporções de células apoptóticas utilizadas. Os níveis de PGE2 encontrados no sobrenadante de cultura foi proporção dependente evidenciando uma relação direta entre fagocitose de AC+PAMP e a produção de PGE2. A fim de mimetizar a cinética da carga bacteriana durante uma infecção, ou seja, inicialmente uma menor carga bacteriana que tende a aumentar conforme ocorre a colonização, os animais foram inoculados com alta (high - 106 UFC de E. coli) e baixa (low - 105 UFC de E. coli) cargas bacterianas, gerando desta forma o que chamamos de AC+PAMPhigh e AC+PAMPlow, respectivamente. Os resultados aqui apresentados demonstram que diante de uma alta carga bacteriana há uma maior produção dos mediadores

This work provides a systematic and critical review of part of the scientific production of its author. It includes three research topics, namely salmonellosis, brucellosis, and leishmaniasis. Each one of these topics is discussed in individual chapters. Most of the data discussed here refers to pathology, pathogenesis, hostpathogen interactions, innate immunity, and diagnosis of these diseases. Instead of being organized as a traditional literature reviews, the chapters actually emphasize the scientific contribution of the author’s publications in each of these fields.
