Excessive rearfoot eversion is thought to be a risk factor for patellofemoral pain development, due to the kinesiological relationship with ascendant adaptations. Individuals with patellofemoral pain are often diagnosed through static clinical tests, in scientific studies and clinical practice. However, the adaptations seem to appear in dynamic conditions. Performing static vs. dynamic evaluations of widely used measures would add to the knowledge in this area. Thus, the aim of this study was to determine the reliability and differentiation capability of three rearfoot eversion measures: rearfoot range of motion, static clinical test and static measurement using a three-dimensional system. A total of 29 individuals with patellofemoral pain and 25 control individuals (18-30 years) participated in this study. Each subject underwent three-dimensional motion analysis during stair climbing and static clinical tests. Intraclass correlation coefficient and standard error measurements were performed to verify the reliability of the variables and receiver operating characteristic curves to show the diagnostic accuracy of each variable. In addition, analyses of variance were performed to identify differences between groups. Rearfoot range of motion demonstrated higher diagnostic accuracy (an area under the curve score of 0.72) than static measures and was able to differentiate the groups. Only the static clinical test presented poor and moderate reliability. Other variables presented high to very high values. Rearfoot range of motion was the variable that presented the best results in terms of reliability and differentiation capability. Static variables do not seem to be related to patellofemoral pain and have low accuracy values.

Stair ascent is an activity that exacerbates symptoms of individuals with patellofemoral pain. The discomfort associated with this activity usually results in gait modification such as reduced knee flexion in an attempt to reduce pain. Although such compensatory strategy is a logical approach to decrease pain, it also reduces the normal active shock absorption increasing loading rates and may lead to deleterious and degenerative changes of the knee joint. Thus, the aims of this study were (i) to investigate whether there is reduced knee flexion in adults with PFP compared to healthy controls; and (ii) to analyze loading rates in these subjects, during stair climbing. Twenty-nine individuals with patellofemoral pain and twenty-five control individuals (18-30years) participated in this study. Each subject underwent three-dimensional kinematic and kinetic analyses during stair climbing on two separate days. Between-groups analyses of variance were performed to identify differences in peak knee flexion and loading rates. Intraclass correlation coefficient was performed to verify the reliability of the variables. On both days, the patellofemoral pain group demonstrated significantly reduced peak knee flexion and increased loading rates. In addition, the two variables obtained high to very high reliability. Reduced knee flexion during stair climbing as a strategy to avoid anterior knee pain does not seem to be healthy for lower limb mechanical distributions. Repeated loading at higher loading rates may be damaging to lower limb joints.

Analysis of genomic data is increasingly becoming part of the livestock industry. Therefore, the routine collection of genomic information would be an invaluable resource for effective management of breeding programs in small, endangered populations. The objective of the paper was to demonstrate how genomic data could be used to analyse (1) linkage disequlibrium (LD), LD decay and the effective population size (NeLD); (2) Inbreeding level and effective population size (NeROH) based on runs of homozygosity (ROH); (3) Prediction of genomic breeding values (GEBV) using small within-breed and genomic information from other breeds. The Tyrol Grey population was used as an example, with the goal to highlight the potential of genomic analyses for small breeds. In addition to our own results we discuss additional use of genomics to assess relatedness, admixture proportions, and inheritance of harmful variants. The example data set consisted of 218 Tyrol Grey bull genotypes, which were all available AI bulls in the population. After standard quality control restrictions 34,581 SNPs remained for the analysis. A separate quality control was applied to determine ROH levels based on Illumina GenCall and Illumina GenTrain scores, resulting into 211 bulls and 33,604 SNPs. LD was computed as the squared correlation coefficient between SNPs within a 10 mega base pair (Mb) region. ROHs were derived based on regions covering at least 4, 8, and 16 Mb, suggesting that animals had common ancestors approximately 12, 6, and 3 generations ago, respectively. The corresponding mean inbreeding coefficients (F ROH) were 4.0% for 4 Mb, 2.9% for 8 Mb and 1.6% for 16 Mb runs. With an average generation interval of 5.66 years, estimated NeROH was 125 (NeROH>16 Mb), 186 (NeROH>8 Mb) and 370 (NeROH>4 Mb) indicating strict avoidance of close inbreeding in the population. The LD was used as an alternative method to infer the population history and the Ne. The results show a continuous decrease in NeLD, to 780, 120, and 80 for 100, 10, and 5 generations ago, respectively. Genomic selection was developed for and is working well in large breeds. The same methodology was applied in Tyrol Grey cattle, using different reference populations. Contrary to the expectations, the accuracy of GEBVs with very small within breed reference populations were very high, between 0.13-0.91 and 0.12-0.63, when estimated breeding values and deregressed breeding values were used as pseudo-phenotypes, respectively. Subsequent analyses confirmed the high accuracies being a consequence of low reliabilities of pseudo-phenotypes in the validation set, thus being heavily influenced by parent averages. Multi-breed and across breed reference sets gave inconsistent and lower accuracies. Genomic information may have a crucial role in management of small breeds, even if its primary usage differs from that of large breeds. It allows to assess relatedness between individuals, trends in inbreeding and to take decisions accordingly. These decisions would be based on the real genome architecture, rather than conventional pedigree information, which can be missing or incomplete. We strongly suggest the routine genotyping of all individuals that belong to a small breed in order to facilitate the effective management of endangered livestock populations.

High-frequency ultrasound is a non-invasive tool used in skin ageing research to assess dermis thickness and echogenicity. This study evaluated the reliability of a range of high-frequency ultrasound parameters and tested their correlation with age and a validated clinical scale for the assessment of forearm skin photoageing; the difference between two body sites according to environmental exposition patterns was also investigated. Twenty-three volunteers aged 28-82 years were divided into three groups according to forearm photoageing degree. A 20 MHz ultrasound unit was used to obtain cross-sectional images of the skin by two trained investigators on two different sites: the dorsal forearm (chronically photoexposed skin) and the proximal medial arm (non-photoexposed skin). Several echogenicity parameters were studied for each skin compartment: total dermis, upper dermis and lower dermis, and the ratio between upper and lower dermis. The intraclass correlation coefficient (for complete agreement) between investigators was higher for upper and total dermis echogenicity measures compared with the lower dermis. At the non-photoexposed site, the upper and lower dermis parameter ratio was better correlated with age. At the photoexposed area, total dermis parameters demonstrated higher correlations with clinical score. The authors discuss the choice of parameters for forearm photoageing assessment using high-frequency ultrasound.

This work describes instrumental strategies for the determination of Mn in a wide range concentration by high-resolution continuum source flame atomic absorption spectrometry technique (HR-CS F AAS) by means of different atomic lines (primary at 279.482 nm, secondary at 403.075 nm and alternative at 209.250 nm). These lines provided complementary concentration intervals, and large sample dilutions became unnecessary. The proposed method was applied to tap water, metal alloy certified material and foliar fertilizer. Accuracy for secondary line were evaluated by tests of significance (t Student test) with reference materials from the Institute of Technological Research of São Paulo, and the results were in agreement at the 95% confidence level. For primary and alternative lines, recovery is were in the 84-116% range and the RSD were 6.1% for all wavelengths. Analytical curves in the 0.1 - 2.0 mg L-1 (279.482 nm), 2.0 - 25 mg L-1 (403.075 nm), 25 - 500 mg L-1 (209.250 nm) intervals were obtained with linear correlation coefficient better than 0.9991. The detection limits were 3.3x10-3 mg L-1 (279.482 nm), 7.4 x 10-3 mg L-1 (403.075 nm), 3.9 mg L- 1 (209.250 nm). The found Mn concentrations were < 3.3x10-3 mg L-1 (tap water), 1.00 ± 0.04 (% m/m) (alloy IPT 25), 7235 ± 175 mg L-1 (foliar fertilizer 1), 4990 ± 132 mg L-1 (foliar fertilizer 2). A method was developed to detect interference of Fe in the Mn primary line (279.482 nm) using the ratio of absorbances of other lines of the triplet (279.827 nm and 280,108 nm).

The following research project deals with the development of new analytical procedures aimed at the identification and quantification of the element zinc in different fractions of vegetable drugs used for weight loss, using the high-resolution continuum source atomic absorption spectrometry in flame. The determination of the levels of zinc was made in an AnalytikJena ContrAA 300 spectrometer in the principal line of Zn at 213.857 nm and wavelength integrated absorbance equal to 3 pixels. Calibration curves were obtained with linear dynamic range from 0.10 to 1.00 mg L-1, correlation coefficient equal to 0.9991 and limit of detection equal to 0.020 mg L-1. The total Zn content in the samples obtained after microwave-assisted acid digestion system were: 28.06 mg kg-1 (Artichoke), 31.49 mg kg-1 (Chamomile), 77.16 mg kg-1 (Gorse), 105.38 mg kg-1 (Horsetail), 32.37 mg kg-1 (Fennel) and 5.00 mg kg-1 (Senna). The extraction in aqueous environment assisted by microwave radiation produced extracts containing: 5.40 mg kg-1 (Artichoke), 10.65 mg kg-1 (Chamomile), 14.83 mg kg-1 (Gorse), 18.90 mg kg-1 (Horsetail), 7.80 mg kg-1 (Fennel), 3.24 mg kg-1 (Senna), corresponding to 19%, 34%, 19%, 18%, 24% and 64% of their total content of Zn, respectively. The amounts of Zn in the extracted samples from some plants indicate that this metal was strongly bounded to the plant's matrix. The sequential extraction using solvents and mixtures with different polarities (n-hexane, ethyl acetate, propanol, methanol, ethanol-water 1:1 (v/v), water) aimed the separation of different Zn containing species and water was considered the most efficient extractor. The Zn contents extracted were: 14.82 mg kg-1 (Artichoke), 16.82 mg kg-1 (Chamomile), 34.09 mg kg-1 (Gorse), 4.75 mg kg-1 (Senna), corresponding to 53% in the first two samples, 44% and 95% for the last two, respectively. Darunavir (DRV) is a protease inhibitor used in the treatment of HIV infection, which constitutes a keystone in the therapy of patients infected with this virus. There is no monograph described in official compendia. The literature provides few methods of analysis for the determination of DRV in pharmaceuticals which include TLC, IR, UPLC, HPLC, HPLC-MS, HPLC-MS/MS, but there are no reports of the use of capillary electrophoresis (CE) for the determination of this drug. Thus, this research proposed the development and validation of a CE method for the determination of DRV in tablets. The method was completely validated according to the International Conference on Harmonization guidelines, showing linearity, selectivity, precision, accuracy and robustness. The migration was achieved in less than 1 minute using fused-silica uncoated capillary with an id of 50 μm and total length of 21 cm and voltage of +20 kV. The sample injection was performed in the hydrodynamic mode. The method was linear over the concentration range of 50-200 μg mL-1 with correlation coefficient 0.9998 and limits of detection and quantification of 7.29 and 22.09 μg mL-1, respectively. The drug was subjected to acid, base, oxidation and photolysis degradation. Degradation products were found interfering with the assay of DRV, therefore the method can be regarded as stability indicating. The validated method is useful and appropriate for the routine quality control of DRV in tablets.
