A leishmaniose é um conjunto de doenças infecciosas causadas por parasitas do gênero Leishmania, que afligem milhões de pessoas no mundo, tendo a cada ano dois milhões de novos casos e 70 mil mortes. As drogas utilizadas no tratamento das diferentes formas clínicas da doença apresentam alta toxicidade e baixa eficácia, além de apresentarem muitos efeitos colaterais e colaborarem com o aparecimento de parasitas e vetores resistentes. Devido a estas adversidades, o desenvolvimento de novas terapias para o tratamento desta parasitose é incentivada pela Organização Mundial da Saúde (OMS) e um maior conhecimento sobre a biologia molecular destes protozoários poderá facilitar estas descobertas. Ultimamente, os telômeros, estruturas nucleoproteícas nos terminais dos cromossomos de eucariotos, têm sido alvo intenso de estudos, que visam utilizá-los como alvo terapêutico contra tumores malignos e microrganismos patogênicos. Os telômeros geralmente se apresentam como estruturas dinâmicas onde ocorrem interações entre o DNA e proteínas, resultando na formação do complexo telomérico, que é responsável pela proteção e manutenção dos cromossomos e estabilidade do genoma, caracterizando uma função imprescindível para viabilidade celular. Componentes do complexo telomérico de Leishmania amazonensis foram identificados por ensaios bioquímicos em extratos positivos para a atividade de telomerase. Entre estes componentes identificou-se as proteínas LaRPA-1 (L. amazonensis Replication Protein A-1) e a LaRbp38 (L. amazonensis RNA binding protein) as quais mostraram habilidade de interagir com o DNA telomérico in vitro e in vivo. Mais recentemente, utilizando-se ensaios de imunoprecipitação e de captura por “pull-down” foi demonstrado que estas proteínas fazem parte de um mesmo complexo nos telômeros do parasita. A glicerol quinase é uma proteína tetramérica que cataliza a fosforilação do glicerol a glicerol-3-fosfato, composta por quatro subunidades - sua análise eletroforética fornecerá apenas uma banda se for constatada a sua pureza num gel não desnaturante - e sua reação com o glicerol é dependente de magnésio e de ATP. A eletroforese de proteínas utiliza como suporte um gel de poliacrilamida, que é formado pela polimerização de monômeros de acrilamida ao longo da sua cadeia e ligações cruzadas de cadeias pela inclusão de um co-monômero bifuncional apropriado, usualmente a N,N’ – metileno-bis-acrilamida ou apenas Bis . A eletroforese de proteínas mais comum é a que utiliza SDS para um gel de poliacrilamida desnaturante. O SDS é um sal chamado Dodecil Sulfato de Sódio que tem por característica se ligar a cadeia proteína nos resíduos de aminoácidos apolares, deixando com a carga negativa, sendo assim, toda proteína fica com carga total negativa, migrando para o anodo na eletroforese. As técnicas de purificação utilizadas foram ultrafiltração e precipitação com ácido tricloro acético, etanol gelado e sulfato de amônio. A dupla precipitação com etanol resultou na recuperação de maior quantidade de proteínas. A coloração do gel com prata foi mais sensível do que Comassie blue. O gel de eletroforese mostrou quatro bandas, correspondentes às quatro subunidades da glicerol quinase quando revelados com prata em gel de SDS-PAGE.

The metropolitan region of São Paulo is the most populous of the country, this happens because of its great importance in the national economy and the job opportunities that are offered to the population. These factors result in intense population growth and urban expansion, reaching some non-habitable places of the metropolis, as areas of pipelines, which are very important for the transportation of natural gas, oil and its derivatives. Before the population growth of the region, these sites were unoccupied, do not presenting problems for the population. However, with the disorderly occupation is generated great anthropogenic pressure on the pipeline stitches, causing risks to people who are around them. Therefore it is extremely important to monitor the strip of pipelines through products and techniques of remote sensing and geoprocessing, enabling, through high spatial resolution images, identification of objects or phenomena that occur on Earth's surface that can alter the functioning and safety of pipelines. Therefore, this study aims to monitor a stretch of the area of the pipeline mesh GASPAL/OSVAT and Capuava Refinery (RECAP), located on the outskirts of the metropolitan area of São Paulo in the city of Mauá, who suffer great human pressure, proving thus the techniques of remote sensing and geographic information system (GIS) as effective tools for monitoring phenomena occurred in urban areas of great complexity. The monitoring was done by object-based classification applied in orbital images Ikonos II and RapidEye, of high spatial resolution and, image processing, detection of objects, segmentation, classification and editing were developed through the eCognition and ArcGis softwares. The biometric characteristics have been used increasingly as a way to identify an individual, mainly for security reasons. Among them, the fingerprint is the most used biometric characteristic around the world, because it is relatively simple and very efficient. In this scene, there was a significant increase in the size of databases containing information on fingerprints, necessary to perform the recognition of a person. The task of classifying them beforehand has become extremely important as it reduces dramatically the size of the problem during a search, because it is not necessary to go through the whole database. Considering its importance, in the last thirty years, many techniques have been developed to try to increase the efficiency of the classification process. This project followed the rules-based approach and the Software Development Kit (SDK) VeriFinger 6.1 was used to assist in the detection of cores and deltas. Additionally, the classification was also implemented by means of directional map and the Poincar´e index. We know that play is directly linked to the development and growth of the child. Thinking in this context, were created recreational spaces, more known as playrooms or toy libraries. This research seeks to understand the playful objects used in more visits with children, realized CPA, FC UNESP - Bauru. Therefore, it was necessary to identify the objects necessary to the demands of users of CPA, they were psychology trainees, fellows extension projects, graduate students, teachers and the subjects treated population served. Composed over 1000 objects, the collection must be appropriate to the needs of the CPA. The research in question is characterized as a case study, is related to our shares a scholarship project Playing in the Center for Applied Psychology - CPA, held in the collection playful and Toy CPA, FC UNESP - Bauru, used, in this case, as a field for this research. Watching the playful collection, daily, some questions have arisen about this space became in this study. The data collection period was from September 2011 to September 2012. As an instrument for data collection was mounted a notebook control in order to check the movement of the objects of the collection, also applied a questionnaire to teachers and trainees working in the CPA. At the end of the study we can say that the symbolic games and rules are the objects that are related to care provided in the CPA because of its capabilities to assist in various aspects of the development of children, with the most frequently used symbolic games with children aged 2 to 7 years and the games rules with children aged 7-12 years or more

Proteínas que apresentam atividades no núcleo e possuem sequência de localização nuclear (NLS) tem seu deslocamento dependente do heterodímero importina-α/β. A importina- (ImpA) é responsável pelo reconhecimento inicial do substrato a ser importado através da interação com os NLS. Os sinais são caracterizados por apresentar um ou mais grupos de aminoácidos básicos, denominados como sequências monopartidas e bipartidas. O fungo Neurospora crassa vem sendo utilizado há mais de 70 anos como organismo modelo em estudos de expressão gênica, desenvolvimento e diferenciação celular, ritmo circadiano, defesa do genoma, bem como outros aspectos da biologia de eucariotos. A presença de um grande número de genes no genoma de N. crassa ainda com funções desconhecidas aponta este organismo como um promissor modelo para o estudo de novos mecanismos genéticos e bioquímicos ainda não identificados. Considerando a importância do metabolismo do glicogênio para os organismos, o presente trabalho teve como objetivo o estudo estrutural de complexos de ImpA com peptídeos NLSs (NCM e NCB) de proteínas envolvidas no metabolismo de glicogênio do fungo N. crassa, utilizando técnicas de cristalografia de proteínas. Monocristais dos complexos ImpA-NCM e ImpA-NCB foram obtidos para a coleta dos dados de difração de raios-X, resultando em dois conjuntos de dados à 2,1Å e 2,45Å de resolução, respectivamente. Após elucidação da estrutura da ImpA, mapas de densidade eletrônica gerados revelaram uma densidade eletrônica no sítio principal de reconhecimento de NLS da ImpA de ambas estruturas, possibilitando modelagem dos peptídeos. Em uma comparação do mapa de densidade eletrônica obtido de ambos complexos com um mapa de uma estrutura nativa de ImpA (70-529) coletada à 2,0Å de resolução, a qual usualmente apresenta um peptídeo “contaminante” no sitio de ligação... During the orthodontic correction planning in addition to dental-jaw correction, facial aesthetics is the orthodontist's and patient's major concern. To prevent facial aesthetics damage is really important to take into account the type of craniofacial growth: mesofacial (balanced), dolichofacial (vertical) or brachyfacial (horizontal). We evaluated 152 documentation files from the Department of Orthodontics of Dental University of Sao Jose dos Campos- UNESP in order to analyze what kind of growth occurs in most Class I malocclusions, Class II and Class III Angle of treated individuals from 6 to 12 years old. From the randomly collected samples, 15 of them belonged to Class I Angle malocclusions; 123 belonged to Angle Class II and 14 to Class III malocclusion. The results showed that in Class I, 66.67% were classified as dolicocephalic; in Class II, 64.23% were classified as dolicocephalic and in Class III, 50% were brachycephalic. We conclude that the dolichofacial was the type which ocurred the most, both in females and males and both in malocclusion Class I and Class II. The brachyfacial type most occurred in Class III malocclusion and the mesofacial type occurred in smaller numbers in the three malocclusions studied

Over the last decades changes have occurred in communication within and between enterprises, made easier by technologies suchas E-commerce, Internet, ERP systems and remote meetings and there was a rapid progress in network technology, which has changed the way business is done. A standardized way to offer services over the internet is using web services. Web services are a kind of remote procedure call and are generally used to integrate systems, independent of language, both client and server. It is common to use several web services run in sequence to perform a business process. To this type of process, gives the name of workflow. Thus, Web services are the primary components of workflows. A tool that provides a way of visualizing the behavior of a workflow can assist the administrator and is required. The present work presents the development of a tool that allows the administrator to classify visually services components and evaluate their importance in the final performance of a workflow. As proof of concept we used several virtual servers and computers where each computer has received a set of web services. A proxy was added between each call of workflows collecting relevant information and storing them in a database for later analysis. The analysis was based on Quality of Service parameters

In this project the Pattern Recognition Problem is approached with the Support Vector Machines (SVM) technique, a binary method of classification that provides the best solution separating the data in the better way with a hiperplan and an extension of the input space dimension, as a Machine Learning solution. The system aims to classify two classes of pixels chosen by the user in the interface in the interest selection phase and in the background selection phase, generating all the data to be used in the LibSVM library, a library that implements the SVM, illustrating the library operation in a casual way. The data provided by the interface is organized in three types, RGB (Red, Green and Blue color system), texture (calculated) or RGB + texture. At last the project showed successful results, where the classification of the image pixels was showed as been from one of the two classes, from the interest selection area or from the background selection area. Paediatric diagnostic radiology can be considered as a separate specialty and with distinct characteristics of the radiology applied in adult patients. This in reason of the variability in the anatomical structures size and bigger sensitivity of tissues. The literature present in its majority methodologies for segmentation and tissue classification in adult patients, and works on tissue quantification are rare. This work had for objective the development of a biological tissue classifier and quantifier algorithm, from histograms, and that converts the quantified average thickness of these tissues for its respective simulator materials. The results will be used in the optimization process of paediatrics images, in future works, since these patients are frequently over exposed to the radiation in the repeated attempts of if getting considered good quality radiographic images. The developed algorithm was capable to read and store the name of all the archives, in the operational system, to filter artifacs, to count and quantify each biological tissues from the histogram of the examination, to obtain the biological tissues average thicknesses and to convert this value into its respective simulator material. The results show that it is possible to distinguish bone, soft, fat and pulmonary tissues from histograms of tomographic examinations of thorax. The quantification of the constituent materials of anthropomorphic phantom made by the algorithm, compared with the data of literature shows that the biggest difference was of 21,6% for bone. However, the literature shows that variations of up to 30% in bone thickness do not influence of significant form in the radiographic image quality. The average thicknesses of biological tissues, quantified for paediatrics patients, show that one phantom can simulate patients with distinct DAP ranges, since variations... As plantas apresentam moléculas que detectam a luz, entre elas estão os fitocromos que detectam mudanças na qualidade da luz nas regiões de vermelho (V) e vermelho-extremo (VE). O fitocromo age no crescimento das plantas possibilitando-as perceber o sombreamento por outras plantas, o que pode ocasionar no processo de estiolamento, ou seja, alongamento do caule. Assim, conforme aumenta o sombreamento, a razão V:VE diminui. Nas plantas de sol, quanto maior a proporção de VE, maior a taxa de estiolamento, enquanto que as plantas de sombra mostram pouca ou nenhuma redução na taxa de alongamento dos caules. Na sucessão florestal as plantas podem ser classificadas como pioneiras, sendo aquelas dependentes de luz e intolerantes à sombra, e as de transição ou clímax, que são tolerantes à sombra. Assim, este trabalho teve o intuito de analisar o processo de estiolamento de plântulas de espécies arbóreas nativas selecionadas, mantidas em condições de luz diferentes e correlacionar com a classe seral correspondente citada na literatura. As seguintes espécies foram analisadas: Bixa orellana, Caesalpinia peltophoroides, Dalbergia nigra, Erythrina speciosa, Eugenia uniflora, Hevea brasiliensis, Hymenaea courbaril, Licania tomentosa, Pachira aquatica, Piptadenia gonoacantha, Psidium guajava, Schizolobium parahyba. Todos os experimentos foram desenvolvidos no jardim experimental da UNESP de Rio Claro. As espécies foram expostas a seis condições luminosas diferentes, com razão V:VE de: 0,1; 0,19; 0,21; 0,35; 1,15 e 1,17. E radiação solar incidente de: 3,16UM; 44,78UM; 27,7UM; 73,54UM; 1987,00UM; 1453,00UM. Foram tiradas medidas do comprimento dos hipocótilos e epicótilos. Posteriormente foram realizadas análises estatísticas com nível de significância de 5%. As espécies consideradas pioneiras foram D. nigra e S. parahyba.
