Diatraea saccharalis F. is one of the greatest pests of the sugar cane culture. This report aimed to characterize the germanium region of the sugarcane borer by light and transmission electron microscopy, emphasizing the morphological steps of the ovarian cluster formation. In the germanium of this insect, four zones could be morphologically identified during the cluster formation. In the most apical end of each ovariole - Zone I - the germ line stem cells undergo complete mitotic division, originating the cystoblasts. In the Zone II, each cystoblast produces a group of eight cells, the cystocytes, which are interconnected by the ring canals. Clusters containing all the cystocytes in the meiosis, characterizes the Zone III. Germ cells with ultrastructural features of apoptosis are also detected in this Zone. In the Zone IV the cystocytes differentiate, morphologically, into one oocyte and seven nurse cells. Interstitial somatic cells and pre-follicle cells exhibit, in their cytoplasm, heterogeneous vacuoles containing degenerated cellular fragments, characterized as apoptotic bodies. Our results pointed out to the morphological evidences related with important control mechanisms for new clusters/follicles production and for the cellular arrangement into the germanium, resulting from the programmed cell death. We believe that the morphological characterization of ovarian cluster formation in D. saccharalis provided valuable information for the understanding of the initial steps of oogenesis and contributed for the knowledge of the cellular mechanisms related with the oocyte production and with reproduction in insects.

The ultrastructure and distribution of gonial and somatic cells in the ovary of Dermatobia hominis was studied during the 3rd larval instar. In larvae weighing between 400 and 500 mg, the ovary is partially divided into basal and apical regions by oblong somatic cells that penetrate from the periphery; these cells show ovoid nucleus and cytoplasm full of microtubules. In both regions, gonial cells with regular outlines, large nucleus and low electron-density cytoplasm are scattered among the interstitial somatic cells. These later cells have small nucleus and electrodense cytoplasm. Clear somatic cells with small nucleus and cytoplasm of very low electron-density are restrict to the apical region of the gonad. Degenerating interstitial somatic cells are seen in the basal portion close to the ovary peduncle. During all this larval period the morphological features of the ovary remain almost the same. At the end of the period there is a gradual deposition of glycogen in the cytoplasm of the somatic cells, increase in the number and density of their mitochondria plus nuclear modification as membrane wrinkling and chromatin condensation in masses.

Ovaries up to the 8th day pupae of Dermatobia hominis were studied by transmission electron microscopy. Ovarioles were recognized in ovaries of 4-day old pre-pupae, surrounded by a thin tunica propria of acellular fibrilar material similar in structure to the internal portion of the external tunica of the ovary. There is continuity of the tunica propria and the ovarian tunica, indicating that the former structure originates from the tunica externa. In 5 to 7-day pupae the interstitial somatic cells from the apical region of the ovary, close to the ovarioles, show delicate filamentous material inside of their rough endoplasmic reticulum cisternae; similar material is seem among these cells. Our observations suggest that interstitial somatic cells do not originate the tunica propria but contribute to its final composition.

The aim of the present study is to characterize the way worker and queen ovaries differentiate in, Apis mellifera, a species with trophic determination of female castes. A morphological study carried out with light and transmission electron microscopy showed that the differences in ovary development between the two castes begin as soon as the differential nursing of larvae is initiated. The decrease in ovariole number in worker ovaries is due to a process of cell death occurring in germinative cells and autophagic regression of somatic cells in the ovarioles that commence in the third instar larvae and proceed until the fifth instar where the process is more intense. Germinative cell death leads to ovariole disintegration and incorporation of the remaining somatic cells of the latter into the stromatic cells in such a way that the total volume of the ovary is little affected during larval development, although the ovariole number decreases. By the end of the larval stage, loss of cells is observed among the stromatic cells of the ovary. As a result, the ovary starts to decrease in volume and takes on the adult form.

Cell death that occurs during ovary differentiation in the honeybee worker's larval development accounts for ovariole reabsorption. From a morphological standpoint, three modes of death were detected. Germinative cells in the ovarioles die by an apoptotic-like process, whereas the somatic cells die by an autophagic process, type 11 cell death; and during pupation, stromatic and ovarian capsular cells die through cytoplasmic disintegration, releasing their components into the hemolymph. These modes of cell death are in part determined by the pattern of tissue organization within which the cell occurs. (C) 2002 Elsevier B.V. Ltd. All rights reserved.

O objetivo deste trabalho foi avaliar o efeito do fotoperíodo sobre o consumo alimentar, produção e composição do leite (gordura, proteína, lactose, sólidos totais) e número de células somáticas de sete ovelhas da raça Bergamácia, submetidas a um fotoperíodo considerado curto (12 horas luz:12 horas escuro), e oito ovelhas da mesma raça, a um fotoperíodo longo (18 horas luz:6 horas escuro), durante as últimas quatro semanas de gestação e três meses de lactação. Para comparação, foi utilizado o teste t para duas amostras independentes. O consumo alimentar avaliado, diariamente, para cada baia com duas ovelhas, não diferiu entre os tratamentos. No fotoperíodo curto, o consumo médio diário foi de 4,48 kg e, no fotoperíodo longo, de 4,40 kg. Não foi observada diferença significativa entre os fotoperíodos, para a produção de leite e teores de proteína, lactose e sólidos totais, quando considerados os 84 dias de lactação. No fotoperíodo curto, a porcentagem média de gordura foi 5,57%, índice superior ao obtido no fotoperíodo longo, de 5,21%. Na análise dos resultados, a cada semana, notou-se que na segunda e na quarta semanas de lactação, a produção de leite foi maior nas ovelhas do fotoperíodo longo. O número de células somáticas não diferiu entre os fotoperíodos e manteve-se baixo por toda a lactação. O maior número de horas de luz estimula a produção de leite no primeiro mês de lactação, principalmente na quarta semana, e as porcentagens de gordura e sólidos totais são menores nos períodos de maior produção de leite.

Este trabalho teve como objetivo o estudo de fatores de risco associados à mastite bovina causada por Prototheca zopfii. Foram analisadas 13 propriedades leiteiras dos Estados do Paraná e de São Paulo, segundo os seguintes critérios de seleção: confirmação prévia de casos de mastite por Prototheca spp., triagem pela pesquisa de Prototheca spp. em tanques de expansão e latões e rebanhos com contagem de células somáticas acima de 5x105cel mL-1. As amostras coletadas consistiram de: leite, água, solo, fezes e swab de teteiras. Prototheca spp. foi isolada de amostras de leite dos quartos mamários com mastite clínica ou subclínica em uma propriedade e de amostras de leite e do ambiente em quatro propriedades, nas quais foi isolada em amostras de: água de bebedouro, abastecimento, esgoto, empoçada no piso de estábulo e sala de ordenha, solo de piquete e pasto, teteiras, fezes de bezerros e suínos. do total de 383 vacas examinadas, Prototheca spp. foi isolada em 20 (5,2%) vacas, sendo caracterizada como P. zopfii em 18. Os fatores de risco associados à mastite causada por P. zopfii foram: criação das vacas a pasto, alimentação dos animais com pasto e silagem, realização de ordenha mecânica em estábulo, permanência das vacas após ordenha em piquete sem alimento, criação de suínos próxima às instalações dos bovinos, existência de cães, gatos e roedores, falta de higienização dos tetos com água, pré-imersão dos tetos em aplicador com retorno e sem a troca do anti-séptico, alimentação dos bezerros com leite de vacas com mastite clínica e serem as vacas da raça holandesa.

Objetivou-se avaliar as alterações fisiológicas, produtivas e comportamentais em cabras da raça Alpina submetidas ao transporte. Foram utilizadas 12 cabras pluríparas em final de lactação. A produção, composição e contagem de células somáticas do leite e as respostas comportamentais das cabras durante a ordenha foram mensuradas três dias antes, no dia e três dias após o transporte. Também foram colhidas amostras de sangue antes (-120 e -10 minutos) e após (10 e 120 minutos) o transporte para determinação dos níveis de cortisol e glicose. Entre os comportamentos estudados durante a ordenha, apenas sobrepasso e coçar foram significativamente influenciados pelo transporte. Também foram observados aumentos significativos nos níveis de glicose e cortisol 10 minutos após o transporte. Os níveis de glicose permaneceram elevados até 120 minutos após o transporte, enquanto os de cortisol aos 120 minutos foram semelhantes aos observados antes do transporte. O transporte com retorno às instalações conhecidas e ao manejo rotineiro de ordenha causa pequenas alterações fisiológicas e comportamentais que não alteram significativamente a produção, a composição e a contagem de células somáticas do leite.

Mastitis is an inflammatory process in the udder that can affect the quality and quantity of milk produced causing economic losses and risks for health. Considering the somatic cell count (SCCs) as indicator of udder health and the milk yield (MY) of buffaloes from São Paulo State, this study aimed to quantify the related losses in milk due to somatic cells count (SCC). 9404 sources of information from 2198 lactations that occurred between 1997 and 2004 were analysed. There was no relation between MY and the SCCs in the buffaloes at first parity. For the second parity in the months 1, 2, 5, 6 and 7 of lactation, there was a negative and significant relationship between SCCs and MY. For parities of three or more there was a significant and negative regression coefficient during every month of lactation betrween MY and SCC. The average losses varied from 0,18 to 2,2 milk liters per unit of SCCs. The results indicated large losses observed in the miltiparous buffaloes and that this category needs received special attention in terms of udder health. The effect of farm, parity and year, must be considered comparison between animals.

The aim of the present study was to evaluate through endoscopy the tracheal aspiration cytology in twenty seven Quarter Horses from Curitiba and surroundings, following the Three Barrel Competittion. Upper respiratory tract secretion was obtained by tracheal aspiration using a polyethylene catheter introduced through the endoscopic fiberoptic working channel, at the level of tracheal bifurcation. Cytologic slides were prepared by smear and stained by diff-quick technique and the differential was performed in 500 cells counting by 1,000X optic microscopy. None of the horses presented abnormality, including epixtasis, at the clinical examination. However, hemosiderophages were detected at cytology in three animals, suggesting that some may be suffering of subclinical pulmonary hemorrhage. Differential cell counting of tracheal aspiration results were, in average: 44.09 ± 35.68% of epithelial cells; 1.10 ± 2.18% of Globet cells; 23.10 ± 35.93% of neutrophils; 0.13 ± 0.37% of lymphocytes; 0.91 ± 2.81% of eosinophils; 30.57 ± 23.62% of macrophages and 0.13 + 0.93% of hemosiderophages. In conclusion, based in the present study, the evaluation of cellular populations with the tracheal aspiration may offer important additional information to the clinician, particularly about the inflammatory processes of lower respiratory tract and pulmonary bleeding.

Mozzarella cheese is traditionally prepared from bubaline (Bubalus bubalis) milk, but product adulteration occurs mainly by addition of or full substitution by bovine milk. The aim of this study was to show the usefulnes of molecular markers to identify the admixture of bovine milk to bubaline milk during the manufacturing process of mozzarella cheese. Samples of mozzarella cheese were produced by adding seven different concentrations of bovine milk: 0%, 1%, 2%, 5%, 8%, 12% and 100%. DNA extracted from somatic cells found in cheese were submitted to PCR-RFLP analysis of casein genes: α-s1-CN - CSN1S1 that encompasses 954 bp from exon VII to intron IX (AluI and HinfI), β-CN - CSN2 including 495 bp of exon VII (Hae III and HinfI), and κ-CN - CSN3, encompassing 373 bp of exon IV (AluI and HindIII). Our results indicate that Hae III-RFLP of CSN2exon VII can be used as a molecular marker to detect the presence of bovine milk in mozzarella cheese. Tick control has been accomplished through the use of synthetic acaricides, which has created resistant individuals, as well as contaminating the environment and nontarget organisms. Substances of plant origin, such as oils and extracts of eucalyptus and neem leaves, have been researched as an alternative to replace the synthetic acaricides. Ricinoleic acid esters from castor oil have recently been shown as a promising alternative in eliminating bacterial contamination during ethanol fermentation, by acting as an effective biocide. The same positive results have been observed when these esters are added to the food given to tick-infested rabbits. This study tested the effect of these substance on the reproductive system of Rhipicephalus sanguineus females, added to rabbit food, more specifically on oogenesis. For this, four groups were established: four control groups (CG1, CG2, CG3, and CG4) and four treatment groups (TG1, TG2, TG3, and TG4) with one rabbit in each (New Zealand White), used as hosts. After full 4 days feeding (semi-engorgement), the females were collected and had their ovaries extracted. In this study, it was observed that R. sanguineus females exposed to esters had their ovaries modified, which was demonstrated through transmission electron microscopy techniques. The addition of ricinoleic esters to the diet of tick-infested rabbits revealed how toxic such substances are for the cytoplasmic organelles of oocytes and pedicel cells. These compounds can change the morphophysiology of germ and somatic cells, consequently influencing their viability and, therefore, confirming that the ricinoleic acid esters from castor oil are a promising substance in the control of R. sanguineus. © 2012 Springer-Verlag Berlin Heidelberg.
