The mandibular gland in Melipona bicolor workers and queens was studied by scanning and transmission electron microscopy. There is no difference in the gland anatomy between the castes, but the transmission electron microscopy showed variation of the cellular ultrastructure according to the secretory phase of the gland in both castes. Smooth endoplasmic reticulum was abundant in the secretory cells of physogastric queens, indicating that these cells produce lipid secretion that is stored in granules with multi-lamellar bodies. Mitochondrial variations during the cell secretory cycle indicates their participation in the lipid synthesis. After secretion, release in the reservoir lumen through the collecting canals, the secretory cells contain many myelinic bodies, indicative of cellular regression. (C) 2004 Elsevier Ltd. The ultrastructure of digestive cells of newly emerged, nurse and forager worker bees is described. Newly emerged bees have the endoplasmic reticulum characteristically in whorls and stacks as well as in parallel arrays of smooth endoplasmic reticulum. Nurse bees have spherits, autophagic vacuoles, lysosomes and multivesicular bodies. Forager bees have many cells with disorganized cytoplasm containing vacuoles, lamellated bodies, lipid inclusions, microbodies and intranuclear crystalloid inclusions. The basal cell region stays essentially unchanged. The changes observed are discussed in relation to previous observations on other insect species.

The Malpighian tubules of workers of the fire ant Solenopsis saevissima (Myrmicinae) were analyzed by scanning and transmission electron microscopy in order to determine their functional organization and association with the hindgut epithelium. The ants showed six Malpighian tubules with three segments morphologically and structurally different. The proximal segment was long and its cells showed abundant smooth endoplasmic reticulum and lipid droplets, which suggest their role in lipid secretion. The mid segment was long and undulated and it was composed by the cells that showed the typical features of ion transporting epithelia. The distal segment, short and flattened, adheres to the rectum wall. The cells of this segment showed the basal lamina fused to that of the rectum, it is probable that this part of the tubule may play a role in ion and water uptake from the feces. The ultrastructure of the glands responsible for synthesizing the aggregation pheromone rhynchophorol in the beetle Rhynchophorus palmarum (L.) was studied. Glands from live beetles were removed and examined under a light microscope using semithin sections (∼1μm) and under a transmission electron microscope (TEM) using ultrathin sections (60 nm). The glands show an alveolar structure without a reservoir and the cells making up the gland are arranged in a typical acinus pattern. The existence of two types of cells with different electron densities, secretion canaliculi, numerous mitochondria and smooth endoplasmic reticulum in abundance were observed. These latter organelles are characteristic of cells which synthetize non-proteinaceous substances and are thus, likely candidates for the producion of rhynchophorol.

The effects of diets with variable zinc levels on the midgut epithelial cells were studied in Oreochromis niloticus L. One hundred and twenty fry of tilapia were apportioned into 4 experimental groups (I, II, III and IV groups), with 30 fish in each treatment, 5 replicate aquaria per treatment containing 6 fish each. The animals of the 4 groups were fed with isonitrogenous (30% crude protein) and isoenergetic (3000 Kcal/Kg of digestible energy) diets with increasing quantities of zinc (44.59; 149.17; 309.93; 599.67 mg Zn/kg of diet), twice a day, for 93 days. Three fish from each group were sacrificed at 36, 66 and 93 days and samples of midgut were removed for ultrastructural analysis. After 93 days of treatment, 3 animals of each experimental group were used for the analysis of zinc concentration by atomic absorption spectrophotometry. The comparative relative index (CRI) revealed that the animals in groups II, III and IV contained, respectively, 1.99%, 34.67% and 22.78% more zinc than the mean concentration in animals from group I. The ultrastructural analysis showed enterocytes with swelling of smooth surfaced endoplasmic reticulum and dilated mitochondria with variable matrix rarefaction and cristae number reduction in the fish exposed to 599.67 mg Zn/Kg of diet at 66 and 93 days of treatment.

The effects of ingested neem oil, a botanical insecticide obtained from the seeds of the neem tree, Azadirachta indica, on the midgut cells of predatory larvae Ceraeochrysa claveri were analyzed. C. claveri were fed on eggs of Diatraea saccharalis treated with neem oil at a concentration of 0.5%, 1% and 2% during throughout the larval period. Light and electron microscopy showed severe damages in columnar cells, which had many cytoplasmic protrusions, clustering and ruptured of the microvilli, swollen cells, ruptured cells, dilatation and vesiculation of rough endoplasmic reticulum, development of smooth endoplasmic reticulum, enlargement of extracellular spaces of the basal labyrinth, intercellular spaces and necrosis. The indirect ingestion of neem oil with prey can result in severe alterations showing direct cytotoxic effects of neem oil on midgut cells of C. claveri larvae. Therefore, the safety of neem oil to non-target species as larvae of C. claveri was refuted, thus the notion that plants derived are safer to non-target species must be questioned in future ecotoxicological studies. © 2012 Elsevier Ltd.

Melanins are dark, insoluble pigments that are resistant to concentrated acids and bleaching by oxidising agents. Phytomelanin (or phytomelan) is present in the seed coat of some Asparagales and in the fruits of some Compositae. In Compositae fruits, melanin is deposited in the schizogenous spaces between the hypodermis and underlying fibrous layer. Phytomelanin in Compositae is poorly understood, and there are only speculations regarding the cells that produce the pigment and the cellular processes involved in the secretion and polymerisation of phytomelanin. This report describes the cellular processes involved in the secretion of phytomelanin in the pericarp of Praxelis diffusa, a species with a structure typical of the family. The ovaries and fruits at different stages were fixed and processed according to the standard methods of studies of light microscopy and transmission electron microscopy. Hypodermal cells have abundant rough endoplasmic reticulum and mitochondria, and the nuclei have chromatin that is less dense than other cells. These characteristics are typical of cells that synthesise protein/amino acids and suggest no carbohydrate secretion. The fibres, however, have a dense cytoplasm rich in the Golgi bodies that are associated with vesicles and smooth endoplasmic reticulum, common characteristics of carbohydrate secretory cells. Our results indicate that the hypodermal cells are not responsible for the secretion of phytomelanin, as previously described in the literature; in contrast, this function is assigned to the adjacent fibres, which have an organisation typical of cells that secrete carbohydrates. © 2012 Elsevier Ltd.

Richards' gland is known for the majority of Epiponini wasps, and despite few experimental evidences, the taxonomic distribution in swarm-founder species and the function of this gland remain rather unclear. This work presents a morphological description of Richards' gland in Protonectarina sylveirae. The gland is formed by a cluster of class 3 cells underneath the anterior margin of the fifth metasomal sternite, and a reservoir formed by the intersegmental membrane between the fourth and fifth metasomal sternites where the secretion can be stored. The secretory cells contain a branched end apparatus that carries the secretory products towards the duct cell. Externally, the cuticle of the sternite, where the duct cells penetrate, is characterized by modifications as scales with very numerous pores. The presence of Richards' gland according to the model proposed by Samacá et al. 2013 in Protonectarina corroborates the single origin of this gland in Epiponini. The occurrence of a Golgi apparatus and smooth endoplasmic reticulum suggests pheromone production.
