IgG, IgM and IgA antibodies to GP43 (glycoprotein fraction of Paracoccidioides brasiliensis) were measured by ELISA in 63 samples from 23 patients with paracoccidioidomycosis before and twice after chemotherapy was started. Antibodies against P. brasiliensis were detected by indirect immunofluorescence (IF) (IgG, IgM and IgA isotypes), counterimmunoelectrophoresis (CIE) and complement fixation. Two control groups composed of 19 healthy individuals and 12 patients with other diseases (six with histoplasmosis, three with tuberculosis and three with other mycoses). The highest efficiency percentages were found with IgG and IgA- ELISA (100%), IgG-IF (96.2%), CIE (94.4%) and the lowest with CF (75.9%). Highest positive and negative predictive values (100%) were observed for IgG and IgA ELISA. IgG and IgM-ELISA antibodies are more often found in patients with acute than chronic disease (P = 0.01). Four to six months after treatment follow-up showed decreased levels of IgG and IgM-ELISA for acute cases and decreased titres of CIE for chronic cases in relation to pretreatment levels. This study suggests that IgG-ELISA anti-GP43 represents a good marker to monitor clinical response to therapy.

The problem of existence and uniqueness of polynomial solutions of the Lamé differential equation A(x)y″ + 2B(x)y′ + C(x)y = 0, where A(x),B(x) and C(x) are polynomials of degree p + 1,p and p - 1, is under discussion. We concentrate on the case when A(x) has only real zeros aj and, in contrast to a classical result of Heine and Stieltjes which concerns the case of positive coefficients rj in the partial fraction decomposition B(x)/A(x) = ∑j p=0 rj/(x - aj), we allow the presence of both positive and negative coefficients rj. The corresponding electrostatic interpretation of the zeros of the solution y(x) as points of equilibrium in an electrostatic field generated by charges rj at aj is given. As an application we prove that the zeros of the Gegenbauer-Laurent polynomials are the points of unique equilibrium in a field generated by two positive and two negative charges. © 2000 American Mathematical Society.

We compute the analytical solutions of the generalized relativistic harmonic oscillator in 1+1 dimensions, including a linear pseudoscalar potential and quadratic scalar and vector potentials which have equal or opposite signs These are the conditions in which pseudospin or spin symmetries can be realized We consider positive and negative quadratic potentials and present their bound-state solutions for fermions and an-tifermions. We relate the spin-type and pseudospin-type spectra through charge conjugation and γ5 chiral transformations. Finally, we establish a relation of the solutions found with single-particle states of nuclei described by relativistic mean-field theories with tensor interactions and discuss the conditions in which one may have both nucleon and antin-ucleon bound states.

Several antiseptic substances have been used as adjuncts to routine mechanical procedures of oral hygiene, based on their antimicrobial effects. The objective of this study was to assess in vitro the antimicrobial efficiency of 2 mouthwash containing Triclosan/Gantrez and sodium bicarbonate in comparison to both positive and negative controls. Standard strain samples of Escherichia coli, Pseudomonas aeruginosa, Actinomyces viscosus and Bacillus subtilis were used. Samples of Streptococcus mutans and Gram-negative bacilli were collected from 20 volunteers (10 with a clinically healthy periodontium and 10 presenting biofilm-associated gingivitis). Evaluation of the antimicrobial activity was performed by determining the minimal inhibitory concentration (MIC). The results indicated that the test solution inhibited the growth of both Gram-negative and Gram-positive microorganisms from the volunteers' saliva as well as that of the standard strains at the MIC dilution of 1:20, whereas the MIC dilution of 0.12% chlorhexidine against the same bacteria was 1:80. Thus, even though the tested mouthrinse solution presented an in-vitro antimicrobial activity superior to that of a placebo, it was inferior to that of chlorhexidine.

The pyrH-encoded uridine 5′-monophosphate kinase (UMPK) is involved in both de novo and salvage synthesis of DNA and RNA precursors. Here we describe Mycobacterium tuberculosis UMPK (MtUMPK) cloning and expression in Escherichia coli. N-terminal amino acid sequencing and electrospray ionization mass spectrometry analyses confirmed the identity of homogeneous MtUMPK. MtUMPK catalyzed the phosphorylation of UMP to UDP, using ATP-Mg 2+ as phosphate donor. Size exclusion chromatography showed that the protein is a homotetramer. Kinetic studies revealed that MtUMPK exhibits cooperative kinetics towards ATP and undergoes allosteric regulation. GTP and UTP are, respectively, positive and negative effectors, maintaining the balance of purine versus pyrimidine synthesis. Initial velocity studies and substrate(s) binding measured by isothermal titration calorimetry suggested that catalysis proceeds by a sequential ordered mechanism, in which ATP binds first followed by UMP binding, and release of products is random. As MtUMPK does not resemble its eukaryotic counterparts, specific inhibitors could be designed to be tested as antitubercular agents. © 2010 Elsevier Inc. All rights reserved.

Naringenin and quercetin are considered antioxidant compounds with promising activity against oxidative damage in human cells. However, no reports have described their effects on reactive oxygen species (ROS) production by phagocytes during microbicidal activity. Thus, the present study evaluated the effects of naringenin and quercetin on ROS production, specifically hypochlorous acid (HOCl), and their involvement in the microbicidal activity of neutrophils. Naringenin and quercetin inhibited HOCl production through different systems, but this inhibition was more pronounced for quercetin, even in the cell-free systems. With regard to the microbicidal activity of neutrophils, both naringenin and quercetin completely inhibited the killing of Staphylococcus aureus. Altogether, these data indicate that the decrease in the oxidant activity of neutrophils induced by these compounds directly impaired the microbicidal activity of neutrophils. Naringenin and quercetin exerted their effects by controlling the effector mechanisms of ROS production, with both positive and negative effects of these antioxidant agents in oxidative stress conditions and on ROS in the microbicidal activity of phagocytes. The present results challenge the traditional view of antioxidants as improvers of pathological conditions. © 2013 Francielli de Cássia Yukari Nishimura et al.

Infection by Cryptosporidium serpentis is one of the most important diseases in reptiles and is characterized by chronic clinical or subclinical infection and the presence of hypertrophic gastritis, food regurgitation, progressive weight loss, mortality, and intermittent or continuous shedding of oocysts in the feces. The objectives of this study were to standardize an indirect enzyme-linked immunosorbent assay (ELISA) to detect antibodies against C. serpentis and to evaluate the clinical, parasitological, and humoral immune response in snakes naturally infected with C. serpentis. Twenty-one snakes naturally infected with C. serpentis and housed at the Butantan Institute, São Paulo, Brazil, underwent clinical and parasitological analyses for C. serpentis infection through daily records of clinical signs and a monthly survey of fecal shedding of oocysts using the Kinyoun's acid-fast staining. The serological evaluation was performed monthly by indirect ELISA using crude total antigen from oocysts of C. serpentis to detect anti-C. serpentis antibodies. Clinical symptoms consisted of food regurgitation, inappetence, and progressive weight loss. The parasitological analysis revealed intermittent fecal shedding of a variable number of oocysts in all snakes, with positivity in 85.32% (157/184) of the samples. The indirect ELISA was positive in 68.25% (86/126) of the samples. A humoral immune response was observed in most animals; however, fluctuating antibodies levels, leading to alternating positive and negative results, were observed in most snakes. © 2013 Elsevier B.V. Blood is one of the most important and powerful elements in magical and religious symbology. As it is an essential substance for animal survival, it does not sound strange that through the ages blood has been given many significant symbolic values (both positive and negative). It can provide life, protection and prosperity in the same proportion that it can cause calamities, destruction and even death. In Opus agriculturae, a farming treatise written by Palladius (V AD), more specifically in Book I, Chapter 35, the author presents prescriptions to protect farms against scourges and climatic phenomena. He mentions some procedures in which blood is a fundamental component in the success of the prescriptions. The aim of the present article is to identify any magic power associated with these practices, their specific symbolic value, especially when related to menstrual blood and to other feminine elements with some magical or religious value. An evaluation is also made of the extent of these symbologies in the context of other practices and situations linked not only to agricultural magic, but to religion and to the ritual practices of other communities, whether in ancient times or not.

This study aimed to investigate the antitumor and cytotoxicity activities of Kielmeyera coriacea and Pyrostegia venusta extracts. Therefore, the hydroalcoholic extracts of P. venusta flowers and K. coriacea leaves were prepared. The extracts were evaporated and the dry extracts were diluted at concentrations of 1.0, 0.1, 0.01 and 0.001 mg/ml for carrying out the bioassays. Artemia salina eggs were incubated in saline solution at 28°C for 24 h. The larvae were treated with different extracts concentrations and the mortality was evaluated after 24 and 48 h. Five discs of potato were placed in Petri dishes and 50 μl of inoculum of Agrobacterium tumefaciens were added to it at 28°C for 24 h incubation. So, 50 μl of the extracts in different concentrations were added. Positive and negative controls were made. The P. venusta and K. coriacea extracts did not show statistically significant acute toxicity. K. coriacea extract showed (mean% of tumor ± standard deviation) 15.30 ± 3.24, 6.34 ± 3.82, 7.57 ± 2.92 and 5.77 ± 2.85 and P. venusta showed 25.82 ± 5.15, 38.40 ± 8.28, 15.75 ± 4.44 and 13.38 ± 7.92, with their concentrations for the antitumor bioassay, and the positive control showed 25.80 ± 6.14. According to the obtained results it was established that the K. coriacea and P. venusta extracts showed antitumor activity but did not show significant cytotoxic activity in A. salina test.

The prodrug hydroximethylnitrofurazone (NFOH) presents antichagasic activity with greatly reduced toxicity compared to its drug matrix nitrofurazone (NF). Besides these new characteristics, the prodrug was more active against the parasite T. cruzi amastigotes. These advantages make the prodrug a possible therapeutic alternative for the treatment of both acute and the chronic phase of Chagas disease. However, the knowledge of pharmacokinetic profile is crucial to evaluate the feasibility of a new drug. In this study, our objective was to evaluate the in vivo formation of NF from the NFOH single administration and to evaluate its pharmacokinetic profile and compared it to NF administration. A bioanalytical method to determine the NF and NFOH by LCMS/MS was developed and validated to perform these investigations. Male albino rabbits (n=15) received NF intravenously and orally in doses of 6.35 and 63.5 mg / kg respectively, and NFOH, 80.5 mg / kg orally. The serial blood samples were processed and analyzed by mass spectrometry. The system operated in positive and negative modes for the analites determination, under elution of the mobile phase 50:50 water: methanol. The administration of NFOH allowed the calculation of pharmacokinetic parameters for the prodrug, and the NF obtained from NFOH administration. Using the pharmacokinetic profile obtained from the NF i.v. administration, the oral bioavailability of NF from the administered prodrug was obtained (60.1%) and, as a key parameter in a prodrug administration, should be considered in future studies. The i.v. and oral administrations of NF differ in the constant of elimination (0.04 vs 0.002) and elimination half-life (17.32 min vs 276.09 min) due to the low solubility of the drug that hinders the formation of molecular dispersions in the digestory tract. Still, there was observed no statistical differences were observed between the pharmacokinetic parameters of orally administered NF and NF obtained from NFOH. The calculated area under the curve (AUC 0-∞) showed that the exposure to the parental drug was fairly the same (844.79 vs 566.44) for NF and NF obtained from the prodrug administration. The tendency to higher NF's mean residence time (MRT) as observed in the prodrug administration (956.1 min vs 496.3 min) guarantees longer time for the action of the drug and it allows the expansion of the administration intervals. These findings, added with the beneficial characteristics of the prodrug encourage new efficacy tests towards the clinical use of NFOH.
