The aim of this study was to adapt the methodology of electrical conductivity (EC) test and determine the physiological quality of seeds of B. virgilioides. Seeds were separated into three batches according to tegument color (1-yellow, 2-orange, and 3-red/black). The EC test analyzed three quantities of seeds (25, 50 and 100), three volumes of water (25, 50 and 100 mL), and six times of seed imbibition (12, 24, 36, 48, 60 and 72), at 25 °C with 6 replications. Germination test was employed to compare the results. The EC test allowed the differentiation of batches in three levels of vigor - the same was observed in germination test: batch 1 presented the best seed vigor, followed by batches 2 and 3. The use of 25, 50 or 100 seeds imbibed in 50 mL of water for 24 hours is recommended.

The tangerine 'Sunki' are highly relevant for the Brazilian citrus industry by presenting important traits for genetic improvement and to be considered a great portaenxento. In this context, the objective of the present work to study the characterization of the fruits of citrus rootstock 'Sunki', depending on the position on the plan and also the germination of their seeds. The physical characteristics evaluated were fruit weight, longitudinal and equatorial diameters of the fruit longitudinal diameter / equatorial diameter, number of buds per fruit, weight of buds and bark and number of viable seeds per fruit. As for the chemical characterization was evaluated titratable, acidity, soluble solid, "Ratio", pH and vitamin C. For the evaluation of these variables, the fruits were obtained from the northeast, southwest, northwest and southeast quadrants. Regarding the evaluation of seed germination, different doses of gibberellic acid and Stimulate were used. The seeds extracted from ripe fruit, washed, dried in the shade and stored in BOD. Immediately after, they were treated for 24 hours with plant growth regulators. After soaking, the seeds were placed in a germinator in germitest paper. After initiation of the germination process, the evaluations were performed daily until the thirty- fourth day. Fruits of 'Sunki'mandarin with uneven as the fruit position within the canopy, the use of gibberellic acid and growth promoter is promising for commercial use on seed germination of this rootstock.

Two experiments were conducted in order to study the effects of seed number, temperature and time of soaking sesame seeds on the results of the electrical conductivity test for physiological quality of seeds. In the experiment I evaluated the electrical conductivity varying the number of seeds (25, 50 and 100) and temperature (20, 25 and 30° C) in experiment II is different soaking periods (2, 4, 6, 8, 12, 16, 20 and 24 h). The tests were performed with three cultivars (cv. Trebol, cv. Cnpa G4 e cv. Comum). The treatments in factorial 3x3x3 and 3x8 in experiments I and II, respectively. The experimental design was completely randomized, with four replications. Means were compared by Tukey test at 5% probability. The characterization of cultivars held through the germination and vigor tests (physical, physiological and biochemical), which rated the Trebol cultivar seeds with better physiological then the Cnpa G4 and Comum. The electrical conductivity test was conducted with four sub-samples of 25 pure seeds soaked in 75 mL of deionized water at 25° C, to separate the lots from 2 hours of soaking, being feasible to evaluate the quality sesame seeds.

During the period from October/92 to September/94 experiments were carried out at the Seed Laboratory, FCAV/UNESP, Jaboticabal, SP, Brazil, using soybean seeds of different genotypes in order to evaluate the effect of genotype on the electrical conductivity (bulk conductivity) of soaked seeds. Seed moisture content (105 ± 3°C, 24 h), standard germination (four 50-seed samples, paper towel, 30°C), and vigor-accelerated aging (42°C, 48 h) were first determined. Undamaged soybean seeds were soaked in deionized water (four 50-seed samples, 75 ml, 25°C, 24 h) and electrical conductivity (μmhos.cm+1.g+1) was measured. Significant differences in conductivity were observed among genotypes having the same pattern of germination and vigor. The results have showed that electrical conductivity can be significantly influenced by genotype.

The objective of this work was to develop a methodology for seed processing and X-ray analysis and to study imbibition pattern in seeds of candeia (Eremanthus incanus). Seeds were gathered in 2001 and 2002 in Morro do Pilar and Lavras respectively, processed and stored in a cold chamber (5ºC/60%RH) until the start of experiments. In order to identify unfilled seeds, a protocol was developed for X-ray use combining radiation energy (Kv) and exposure time (seconds). For elimination of unfilled seeds, an experiment was conducted using a South Dakota seed blower whereby different opening settings and ventilation times were combined. Original seed lots and categorized lots following radiograph viewing were tested by germination tests, with seeds being cleansed in sodium hypochlorite and scattered over blotting paper in 'gerboxes' and then taken to germinators with alternating temperatures of 20º-30ºC and 10 hours of light, for up to 15 days. Imbibition curves were determined under the conditions 30ºC, 20º-30ºC/10 hours of light and 30º-20ºC/10 hours of light. The X-ray protocol that best allowed viewing of internal seed structures was 30Kv for 45 seconds. Seed separation using the blower setting at a 3.0 opening for 30 seconds raised the number of embryonic seeds to values exceeding 99%, also raising the germination rates as a consequence. The imbibition pattern is three-phased and the germination process is completed in 72 hours. Under alternate temperatures, primary root growth is favored.
