Antimicrobial peptides (AMPs) are a promising solution to face the antibiotic-resistant problem because they display little or no resistance effects. Dimeric analogues of select AMPs have shown pharmacotechnical advantages, making these molecules promising candidates for the development of novel antibiotic agents. Here, we evaluate the effects of dimerization on the structure and biological activity of the AMP aurein 1.2 (AU). AU and the C- and N-terminal dimers, (AU)2K and E(AU)2, respectively, were synthesized by solid-phase peptide synthesis. Circular dichroism spectra indicated that E(AU)2 has a coiled coil structure in water while (AU)2K has an α-helix structure. In contrast, AU displayed typical spectra for disordered structures. In LPC micelles, all peptides acquired a high amount of α-helix structure. Hemolytic and vesicle permeabilization assays showed that AU has a concentration dependence activity, while this effect was less pronounced for dimeric versions, suggesting that dimerization may change the mechanism of action of AU. Notably, the antimicrobial activity against bacteria and yeast decreased with dimerization. However, dimeric peptides promoted the aggregation of C. albicans. The ability to aggregate yeast cells makes dimeric versions of AU attractive candidates to inhibit the adhesion of C. albicans to biological targets and medical devices, preventing disease caused by this fungus. © 2013 Springer-Verlag Wien.

Chalcones have shown potential to several pharmacological applications including antimalarial properties. We employed multivariate data analysis to correlate the antimalarial activity with electronic structure descriptors obtained through quantum mechanical calculations. The results show high statistical significance and bring valuable insights in order to design new compounds. © 2013 Springer Science+Business Media New York.

The purpose of this study was to validate noninvasive endocrine monitoring techniques for Pampas deer and to evaluate seasonal changes in testicular steroidogenic activity and their correlation to reproductive behavior, antler cycle and group size. Thus, fecal samples, behavioral data and observations of antler status were collected at monthly intervals during 1 year from free-ranging Pampas deer stags (three radio-collared individuals and 15 random individuals) living in Emas National Park, Brazil (18 degrees S latitude). Fecal steroids were extracted using 80% methanol and steroid concentrations were quantified by a commercial enzyme immunoassay (EIA). Fecal testosterone concentrations peaked in December-January (summer), March (early autumn) and in August-September (winter-spring), with minimal values from April-July. Reproductive behavior had two peaks, the first in December-January, characterized by predominately anogenital sniffing, flehmen, urine sniffing, chasing and mounting behavior, and the second peak in July-September (behavior primarily related to gland marking). There were significant correlations between fecal testosterone and reproductive behavior (r = 0.490), and between fecal testosterone and antler phases (r = 0.239). Antler casting and regrowth occurred under low testosterone concentrations, whereas velvet shedding was associated with high concentrations of testosterone. We inferred that Pampas deer stags exhibited a seasonal cycle that modulated sexual behavior and the antler cycle, and we concluded that fecal steroid analysis was a practical and reliable non-invasive method for the evaluation of the endocrine status of free-ranging Pampas deer. Phospholipases A(2) are components of Bothrops venoms responsible for disruption of cell membrane integrity via hydrolysis of its phospholipids. A class of PLA(2)-like proteins has been described which despite PLA(2) activity on artificial substrate, due to a D49K mutation, is still highly myonecrotic. This work reports the X-ray structure determination of two Lys49-PLA(2)s from Bothrops neuwiedi pauloensis (BnSP-7 and BnSP-6) and, for the first time, the comparison of eight dimeric Lys49-PLA2s. This comparison reveals that there are not just two (open and closed) but at least six different conformations. The binding of fatty acid observed in three recent Lys49-PLA(2) structures seems to be independent of their quaternary conformation. Cys29 polarization by Lys122 is not significant for BnSP-7 and BnSP-6 or other structures not bound by fatty acids. These structures may be in an active state when nothing is bound to them and the Lys122/Cys29 interactions are weak or absent. (C) 2003 Elsevier B.V. All rights reserved.

A comparison of the thermal regime and oxygen distribution patterns of some Brazilian reservoirs was made. A strong latitudinal dependence of surface temperature, annual mean and annual range was found. Except for Tucurui Reservoir, a reservoir of the Equatorial zone, the lowest surface temperature of the year in the other compared reservoirs was observed from June to August while the highest extended from October to February. The decreasing trend of annual mean temperatures and thermal ranges was due to the increasing seasonal variability of insolation with the latitude. A positive relationship between the increase on thermal surface-bottom differences and the depth of water column was found for reservoirs of similar latitudes. A long thermal stratification (around four months) was evidenced in the lacustrine zone of reservoirs with a residence time higher than 40 days. Low fluctuation (<2%) of the annual variability of heat contents was observed for the Tucurui Reservoir, while in das Garcas Reservoir a manmade lake located in the frontier between tropical and temperate regions, the annual coefficient of variation attained 13%. Concerning the heat budgets, the value for the Tucurui Reservoir was two times higher than in das Garcas Reservoir. Both the morphometric and climatological factors affected the heat contents of the two compared reservoirs. In deep eutrophic reservoirs, a significant reduction on the oxygen concentrations in the hypolimnetic zone was frequently observed. In some oligotrophic stratified reservoirs, a decrease on oxygen with depth occurred when the temperature of the hypolimnion was higher than 20 degrees C and caused a biochemical oxygen demand. In das Garcas Reservoir, the actual oxygen deficits ranged from 0.40 to 1.52 mg.O-2.cm(-2) and appear to be linked to oxygen consumption after the senescence of Microcystis aeruginosa populations in the spring. But, other factors such as the allochthonous loads of organic matter also had an important role on the oxygen balance of das Garcas Reservoir.

This study aimed to characterize the populational structure of Clibanarius vittatus (Bosc, 1802), as well as to determine the morphometric relations between the animal's size and the variables: length of their chelar propodus and the size of the shell opening. C. vittatus is a relatively abundant pagurid on the west Atlantic coast, it occurs in the intertidal region from 38 degrees N to 28 degrees S. The animals in this study were randomly sampled every month on the Paranapu (a) over tilde Beach (46 degrees 23' S e 23 degrees 59' W), S (a) over tilde $ o Vicente (SP), Brazil. The biometrics data were adjusted to the power equation (y = ax(b)), by means of the minimum square method. A total of 427 individuals were collected. The size of the cephalothoraxic shield ranged from 2.5 to 12.7 mm, evidencing a representative sample of the population. The of most commonly genus of Gastropoda shells occupied by the hermits was Thais (97,26%). In relation to the morphometric analyses performed, it was observed a positive correlation between the animal size and the variables length of the chelar propodus and the size of the shells.
