Objetivou-se avaliar a ingestão de matéria seca, o ganho de peso diário, a conversão alimentar, o peso vivo ao abate e o período de confinamento em cordeiros Santa Inês puros e ½Dorset ½Santa Inês, alimentados com dietas isoenergéticas (76,59% de NDT) e isoprotéicas (17,48% de PB) contendo diferentes fontes de óleo vegetal (óleos de soja, canola e linhaça) e uma dieta controle (sem inclusão de óleo vegetal). A relação volumoso:concentrado foi de 30:70 e utilizou-se feno de aveia como volumoso. Realizou-se também um ensaio de digestibilidade, utilizando quatro cordeiros não-castrados, distribuídos em delineamento quadrado latino, avaliando-se ingestão, excreção fecal e digestibilidade total dos nutrientes das rações. A ingestão de matéria seca, expressa em porcentagem do peso vivo, foi menor nos cordeiros que receberam dieta contendo óleo de canola que naqueles que receberam dieta controle. Porém, todas as rações proporcionaram ganhos de peso e conversão alimentar satisfatórios. Os valores de digestibilidade total da matéria seca (76,02%) e matéria orgânica (76,82%) da dieta controle foram superiores aos da dieta contendo óleo de linhaça (72,11% e 72,97%, respectivamente), embora não tenham diferido das dietas contendo óleos de soja (72,94 e 73,71%) e canola (73,45 e 74,25%). A digestibilidade do extrato etéreo foi menor na dieta controle (84,02%), enquanto as demais dietas apresentaram valor médio de 91,98%. Os óleos vegetais reduziram a digestibilidade da matéria seca e da matéria orgânica, não afetando a ingestão e digestão dos demais nutrientes.

Sugarcane workers in Brazil are exposed to various genotoxic compounds, including polycyclic aromatic hydrocarbons (PAHs), derived from an incomplete combustion process of burnt sugarcane fields. The effects of the occupational exposure to sugarcane fields burning were measured in urine samples of sugarcane workers from the northwest of the State of São Paulo when exposed (harvesting) and when non-exposed (non-harvesting). The urinary levels of 1-hydroxypyrene (1-OHP) and the influence of the genetic polymorphisms CYP1A1, GSTM1, GSTT1 and GSTP1 were evaluated. Our results showed that the 1-OHP levels were significantly higher (P < 0.0000) in the exposed sugarcane workers (0.318 mu mol mol(-1) creatinine) than in the non-exposed workers (0.035 mu mol mol(-1) creatinine). In an unvaried analysis, no influence regarding the polymorphisms was observed. However, multivariate regression analysis showed that the CYP1A1*4 polymorphism in the exposed group, and age and the GSTP1 polymorphism in the non-exposed group significantly influenced urinary 1-OHP excretion levels (P < 0.10). The utilization of energetic and protein feeds, highly digestible as well, becomes more and more necessary due to the quality of formulated rations, fish performance and relationship with the environment. Apparent digestibility coefficient (ADC) of dry matter, protein, energy, phosphorus, and amino acids of corn starch, corn, wheat, rice, soybean, and cottonseed meal, corn gluten and fish meal were determined for Nile tilapia. ADC was determined using a reference diet based on albumin, gelatin and corn starch, was used inert indicator chromium III oxide (Cr(2)O(3)). Each test diet composed by 70% of reference diet and 30% of the test diet. Feces were collected using mofied Guelph system. ADC values for protein and average ADC of amino acids were as follows: corn 89.76 and 96.43%, rice meal 95.88 and 92.26%, wheat meal 93.54 and 84.41%, fish meal 82.59 and 86.36%, corn gluten 89.82 and 87.98%, soybean meal 94.13 and 91.93%, cotton meal 87.10 and 77.47%, respectively. According to the results of this work, ADC of protein is not a reliable indicator of ADC values of amino acids, even more so for wheat meal, corn, and cotton meal. Among protein feeds, soybean meal was found to have the highest ADC for protein and amino acids, while corn was the energetic feed with the highest ADC (86.15%) for energy.

Introduction: Helicobacter pylori infection is an established risk factor for gastric cancer development, but the exact underlying mechanism still remains obscure. The inactivation of tumor suppressor genes such as p53 and p27(KIP1) is a hypothesized mechanism, although there is no consensus regarding the influence of H. pylori cagA(+) in the development of these genetic alterations. Goals: To verify the relationship among H. pylori infection, p53 mutations and p27(Kip1) Protein (p27) expression in gastric adenocarcinomas (GA) seventy-four tissues were assayed by PCR for H. pylori and cagA presence. Mutational analysis of p53 gene was performed by single-strand conformation polymorphism (SSCP). Seventy tissues were analyzed by an immunohistochemical method for p27 expression. Results: From the samples examined, 95% (70/74) were H. pylori positive, 63% cagA(+). Altered p53 electrophoretic mobility was found in 72% of cases and significantly more frequent in the presence of cagA. Considerable reduction in p27 expression (19%) was found with a tendency for association between cagA(+) and p27(-), although the results were not statistically significant. Concomitant alterations of both suppressor genes were detected in 60% of cases. In the cases cagA(+), 66.7% of them had these concomitant alterations. Conclusions: The data suggest that H. pylori cagA(+) contributes to p53 alteration and indicate that concomitant gene inactivation, with reduced p27 expression, may be a mechanism in which H. pylori can promote the development and progression of gastric cancer. This study was carried out to determine the best digestible protein/digestible lysine ratio that should be present in feed fed to Nile tilapias. Two hundred and sixteen tilapias (11.0 +/- 0.43g) were distributed in 36 fish tanks (205 L) at a density of 6 fish/tank. Twelve feeds were formulated with three different digestible protein (DP) levels 22.0; 26.0 and 30.0% (based on digestible amino acids) and four different lysine percentages of 4.5; 6.0; 7.5 or 9.0% in relation to digestible protein. The fish were fed ad libtum during a 60-day period. There was significant effect of the digestible protein and digestible lysine ratio on weight gain, because the increase in lysine levels in feeds with 26 and 30% DP promoted linear increase in this parameter. The best values for feed conversion were obtained at the levels 26 and 30% DP that increased the daily consumption of digestible protein. The lysine level caused a linear reduction in feed conversion and linear increase in the protein efficiency rate. The results suggested that the 26% DP level might be used in ration to feed Nile tilapia juveniles; however, these should contain digestible amino acids and the digestible lysine/digestible protein ratio should be 6.0%. However, for levels higher than 26% DP, a maximum digestible lysine level of 7.5% DP improved weight gain for the species.

This study was carried out to determine the best digestible energy and digestible protein ratio in feeds for Nile tilapia (Oreochromis niloticus) juveniles 30.0 +/- 4.21 g) based on digestible amino acids and the ideal protein concept). Twelve rations were formulated with protein levels 22.0; 26.0; 30.0 and 34.0% of digestible protein and levels 3,000, 3,300 and 3,600 kcal/kg digestible energy. The digestible energy/digestible protein ratio was between 8.94 and 15.19 kcal/g. Three hundred and twenty four tilapias were randomly distributed in thirty six 250 L circular tanks at a density of 9 fish/tank, a total of 12 treatments with three replications. After 60 days, there was no significant difference in weight gain, daily weight gain and feed conversion ratio among the studied treatments. A linear increase was observed in fillet yield with increasing digestible protein. With respect to feed cost/kg weight gain, the treatment with 30.0% DP and 3,000 kcal/kg DE presented low cost and better cost effectiveness index. Therefore, it was concluded that digestible energy did not influence the productive performance parameters and that effective feeds can be formulated with DP levels lower than 34% when feeding juvenile tilapias. The ration should be formulated based on the concept of ideal protein.

The TL, optical absorption (OA) and EPR properties of natural Brazilian alexandrite and chrysoberyl have been investigated. The TL measurements for natural alexandrite show five peaks between 100 and 450°C, with their emission spectrum having 370 and/or 570 nm components. The intensity of the 320°C TL peak was found to be enhanced with pre-annealing treatment, more prominently above 600°C. The OA and EPR measurements showed that this kind of heat treatment induces the Fe2→ Fe3+ conversion in the natural sample. Chrysoberyl samples exhibited the TL peaks at the same temperatures as alexandrite samples, but the glow curves were more than 200 times less intense than alexandrite ones.

We present results of thermally stimulated depolarization current (TSDC) measurements in synthetic and natural alexandrite, which show TSDC bands related to the presence of electric dipoles in both types of samples. Synthetic material shows a wide TSDC band with a peak at 179 K, which can be fitted by two distinct relaxing dipole distributions. For natural alexandrite the TSDC band has a maximum around 195 K and can be fitted by three different distributions. Both samples present one of the calculated curves with a peak about 179 K, with activation energy of 0.57 eV and constant relaxation time of 1 × 10-14 sec. Photo-induced TSDC shows that TSDC bands can also be generated by simultaneous application of light and an electric field at 77 K.

The salivary glands of Culex quinquefasciatus female mosquitoes are paired organs composed of two lateral lobes with proximal and distal secretory portions, and a medial lobe. All portions comprise a simple epithelium that surrounds a salivary duct. In the apical portion of the medial lobe, non-secretory cells strongly resemble cells involved in ion and water transport. The general architecture of the secretory portions is similar between lobes. The appearance of the secretory material and the morphological aspect of the apical cell membrane are the most distinctive features among the three secretory portions. Cells in the lateral proximal lobe display thin membrane projections extending into a translucent and finely filamentous secretory product. At the lateral distal portion, the apical cell membrane forms an intricate meshwork that encloses a dark secretory product. Medial lobe secretory cells also contain secretory cavities surrounded by intracytoplasmic vesicles, all containing a very dark and uniform product. Scattered cells holding numerous vacuoles, some of them containing a small and electron-dense granule eccentrically located and resembling those of the diffuse endocrine system, are frequently observed in the periphery of all secretory portions. Immunofluorescence assays revealed that the distal portion of the lateral lobes contains apyrase, an enzyme putatively responsible for platelet aggregation inhibition, diffusely distributed in the cell cytoplasm. © 2003 Elsevier Ltd. The development of fast, inexpensive, and reliable tests to identify nontuberculous mycobacteria (NTM) is needed. Studies have indicated that the conventional identification procedures, including biochemical assays, are imprecise. This study evaluated a proposed alternative identification method in which 83 NTM isolates, previously identified by conventional biochemical testing and in-house M. avium IS1245-PCR amplification, were submitted to the following tests: thin-layer chromatography (TLC) of mycolic acids and PCR-restriction enzyme analysis of hsp65 (PRA). High-performance liquid chromatography (HPLC) analysis of mycolic acids and Southern blot analysis for M. avium IS1245 were performed on the strains that evidenced discrepancies on either of the above tests. Sixty-eight out of 83 (82%) isolates were concordantly identified by the presence of IS1245 and PRA and by TLC mycolic acid analysis. Discrepant results were found between the phenotypic and molecular tests in 12/83 (14.4%) isolates. Most of these strains were isolated from non-sterile body sites and were most probably colonizing in the host tissue. While TLC patterns suggested the presence of polymycobacterial infection in 3/83 (3.6%) cultures, this was the case in only one HPLC-tested culture and in none of those tested by PRA. The results of this study indicated that, as a phenotypic identification procedure, TLC mycolic acid determination could be considered a relatively simple and cost-effective method for routine screening of NTM isolates in mycobacteriology laboratory practice with a potential for use in developing countries. Further positive evidence was that this method demonstrated general agreement on MAC and M. simiae identification, including in the mixed cultures that predominated in the isolates of the disseminated infections in the AIDS patients under study. In view of the fact that the same treatment regimen is recommended for infections caused by these two species, TLC mycolic acid analysis may be a useful identification tool wherever molecular methods are unaffordable.

Endophytic fungi were isolated from leaves, flowers and fruit of healthy apple trees (Malus domestica, BORKH.) growing in southern Brazilian orchards under three different cultivation systems (conventional, integrated and organic), during two vegetative cycles. The greatest total number of endophytic isolates was obtained from the orchards under organic cultivation when compared to integrated and conventional cultivation systems. Filamentous fungi from the genera Colletotrichum, Xylaria and Botryosphaeria were the most frequent ones and the most representative yeast genera were Sporobolomyces, Rhodotorula, Debaryomyces and Cryptococcus. It is suggested that some isolates may be used as indicators of the different management systems. © 2005 Wiley-VCH Verlag GmbH & Co. KGaA.

Over the past few years there have significantly been increased the articles that approach the constructions in wood or with structure in wood in the specialized Brazilian magazines. This increase brings up indications that the incorporated values to these habitations are modifying, however it is not so simple to conclude that these issues can be associated to the development of incorporated technologies to the constructive system. The work presents, firstly, a survey performed in these publications that had the objective to verify which the constructive systems in wood is being more executed, under which cultural and technician standards. From that survey it was performed a study of the habitations constructed in mixing system whose structures are timbers and walls in masonry. The aesthetic and cultural questions involved are argued considering mainly that these habitations belong to a social class whose purchasing power increased.

The majority of the Brazilian schools of Civil Engineering have taught the subject works with wood in the traditional only by disciplines of Metallic Structures and Wood at a topic in the content of the program on disciplines at the Materials of Civil Construction. To extend of the wood studies is being made possible that all the pupils have a wider understanding of this material and its adapted use is the goal of the proposal, that by the means of the education modality e-learning, disclosing the first one: Module I - Introduction to the Study of the Wood. This methodology tries to use the theoretical beddings in projects that are related to the pupil daily tasks and its practical professional. The article presents the theoretical bases of the conception proposal and its structure, and at the conclusions, the problems found during its application are displayed. Basically, the methodology considers that the professor and the pupil must share the learning, and adopt it as main in the interdisciplinary.

The present study aimed at evaluating clinical and laboratory aspects during experimental envenomation by Crotalus durissus terrificus in dogs treated with antiophidic serum. Twenty-one dogs were divided into three groups of seven animals each. Group I received 1mg/kg venom (sc); Group II received 1mg/kg venom (sc), 50mg antiophidic serum (iv), and fluid therapy including 0.9% NaCl solution (iv); and Group III received 1mg/kg venom (sc), 50mg antiophidic serum (iv), and fluid therapy including 0.9% NaCl solution containing sodium bicarbonate diluted to the dose of 4mEq/kg. The clinical signs of ataxia, sedation, flaccid paralysis, mydriasis, eyeball paralysis, mandible ptosis, sialorrhea, vomiting and diarrhea observed in the dogs were very similar to those observed in humans. The decrease in hemoglobin, hematocrit, erythrocyte, platelet and fibrinogen levels, prolongation of clotting time, prothrombin time (PT) and activated partial thromboplastin time (APTT), as well as hypocellularity in the bone marrow characterized anemia, thrombocytopenia and blood incoagulability, as well as hypofibrinogenemia and decreased bone-marrow activity. Important bleeding was not observed. Increased numbers of leukocytes and neutrophils and decreased numbers of lymphocytes and eosinophils characterized an acute inflammatory response and stress caused by generalized pain. The employed antiophidic serum was effective and all animals survived.

The present work shows laboratory aspects, electrocardiogram and histopathology results during experimental envenomation by Crotalus durissus terrificus in dogs treated with antiophidic serum. Twenty-one dogs were divided into three groups of seven animals each. Group I received 1mg/kg venom (sc); Group II received 1mg/kg venom (sc), 50mg antiophidic serum (iv) and fluid therapy including 0.9% NaCl solution (iv); and Group III received 1mg/kg venom (sc), 50mg antiophidic serum (iv) and fluid therapy including 0.9% NaCl solution containing sodium bicarbonate diluted to the dose of 4mEq/kg. Urinalysis showed brown urine, proteinuria, occult blood and myoglobinuria. Respiratory acidosis and hypotension were also observed. At the venom inoculation site, there was discreet edema, popliteal lymph node response, musculature presenting whitish areas and necrotic myositis with myoregenerative activity. There was not evidence of electrocardiographical and biochemical alterations.
