Three D-glucans were isolated from the mycelium of the fungus Botryosphaeria rhodina MAMB-05 by sequential extraction with hot-water and hot aqueous KOH (2% w/v) followed by ethanol precipitation. Following their purification by gel permeation chrornatography on Sepharose CL-4B, the structural characteristics of the D-glucans were determined by FT-IR and C-13 NMR spectroscopy and, after methylation, by GC-MS. The hot-water extract produced a fraction designated Q(1A) that was a beta-(1 -> 6)-D-glucan with the following structure:[GRAPHICS]The alkaline extract, when subjected to repeated freeze-thawing, yielded two fractions: KIP (insoluble) that comprised a beta-(1 -> 3)-D-glucan with beta-D-glucose branches at C-6 with the structure:[GRAPHICS]and K1SA (soluble) consisting of a backbone chain of alpha-(1 -> 4)-linked D-glucopyranosyl residues substituted at O-6 with alpha-D-glucopyranosyl residues:[GRAPHICS](c) 2008 Elsevier Ltd. The antibody response against the antigen sheep red blood cells (SRBC) was investigated in mice pre-treated with formalin-killed Paracoccidioides brasiliensis or with cell wall fractions of the fungus. Pre-treatment with P. brasiliensis, as well as with the F1 fraction and beta-glucan significantly increased the anti-SRBC antibody response in the experimental groups as compared to the control group that received only SRBC. This immunomodulatory effect varied with the different doses employed and with pre-treatment time. We conclude that the cell wall fractions of P. brasiliensis might play an important role in the hypergammaglobulinemia associated with Paracoccidioidomycosis. © 1993 Kluwer Academic Publishers.

The putative eukaryotic translation initiation factor 5A (eIF5A) is a highly conserved protein among archaea and eukaryotes that has recently been implicated in the elongation step of translation. eIF5A undergoes an essential and conserved posttranslational modification at a specific lysine to generate the residue hypusine. The enzymes deoxyhypusine synthase (Dys1) and deoxyhypusine hydroxylase (Lia1) catalyze this two-step modification process. Although several Saccharomyces cerevisiae eIF5A mutants have importantly contributed to the study of eIF5A function, no conditional mutant of Dys1 has been described so far. In this study, we generated and characterized the dys1-1 mutant, which showed a strong depletion of mutated Dys1 protein, resulting in more than 2-fold decrease in hypusine levels relative to the wild type. The dys1-1 mutant demonstrated a defect in total protein synthesis, a defect in polysome profile indicative of a translation elongation defect and a reduced association of eIF5A with polysomes. The growth phenotype of dys1-1 mutant is severe, growing only in the presence of 1 M sorbitol, an osmotic stabilizer. Although this phenotype is characteristic of Pkc1 cell wall integrity mutants, the sorbitol requirement from dys1-1 is not associated with cell lysis. We observed that the dys1-1 genetically interacts with the sole yeast protein kinase C (Pkc1) and Asc1, a component of the 40S ribosomal subunit. The dys1-1 mutant was synthetically lethal in combination with asc1Δ and overexpression of TIF51A (eIF5A) or DYS1 is toxic for an asc1Δ strain. Moreover, eIF5A is more associated with translating ribosomes in the absence of Asc1 in the cell. Finally, analysis of the sensitivity to cell wall-perturbing compounds revealed a more similar behavior of the dys1-1 and asc1Δ mutants in comparison with the pkc1Δ mutant. These data suggest a correlated role for eIF5A and Asc1 in coordinating the translational control of a subset of mRNAs associated with cell integrity. © 2013 Galvão et al.

Sporotrichosis is a subcutaneous mycosis that is caused by the dimorphic fungus Sporothrix schenckii. This disease generally occurs within the skin and subcutaneous tissues, causing lesions that can spread through adjacent lymphatic vessels and sometimes leading to systemic diseases in immunocompromised patients. Macrophages are crucial for proper immune responses against a variety of pathogens. Furthermore, macrophages can play different roles in response to different microorganisms and forms of activation, and they can be divided into classic or alternatively activated populations, as also known as M1 and M2 macrophages. M1 cells can lead to tissue injury and contribute to pathogenesis, whereas M2 cells promote angiogenesis, tissue remodeling, and repair. The aim of this study was to investigate the roles of M1 and M2 macrophages in a sporotrichosis model. Toward this end, we performed phenotyping of peritoneal exudate cells and evaluated the concomitant production of several immunomediators, including IL-12, IL-10, TGF-β, nitric oxide, and arginase-I activity, which were stimulated ex vivo with cell wall peptide-polysaccharide. Our results showed the predominance of the M2 macrophage population, indicated by peaks of arginase-I activity as well as IL-10 and TGF-β production during the 6th and 8th weeks after infection. These results were consistent with cellular phenotyping that revealed increases in CD206-positive cells over this period. This is the first report of the participation of M2 macrophages in sporotrichosis infections. © 2013 Springer Science+Business Media Dordrecht.

A water-soluble polysaccharide was extracted with alkali from the cell wall of Verticillium lecanii (also called Lecanicillium lecanii). After freezing and thawing, the water-soluble fraction was purified by gel filtration chromatography on Sepharose CL-6B and eluted as one peak by HPSEC/RID. Monosaccharide analysis showed galactose and glucose (1.1:1), with traces of mannose (<1%). The structural characteristics were determined by spectroscopic analysis, FT-IR and 1D and 2D 1H and 13C NMR, and methylation results. On the basis of the data obtained, the following structure of the polysaccharide (E3SIV fraction) was established: where n ≈ 22 and m ≈ 22. © 2013 Elsevier Ltd. All rights reserved.

Xylella fastidiosa 9a5c (XF-9a5c) and Xanthomonas axonopodis pv. citri (XAC) are bacteria that infect citrus plants. Sequencing of the genomes of these strains is complete and comparative analyses are now under way with the genomes of other bacteria of the same genera. In this review, we present an overview of this comparative genomic work. We also present a detailed genomic comparison between XF-9a5a and XAC. Based on this analysis, genes and operons were identified that might be relevant for adaptation to citrus. XAC has two copies of a type II secretion system, a large number of cell wall-degrading enzymes and sugar transporters, a complete energy metabolism, a whole set of avirulence genes associated with a type III secretion system, and a complete flagellar and chemotatic system. By contrast, XF-9a5c possesses more genes involved with type IV pili biosynthesis than does XAC, contains genes encoding for production of colicins, and has 4 copies of Type I restriction/modification system while XAC has only one.

No presente trabalho foram determinadas a composição química e a digestibilidade de diversas frações de Brachiaria brizantha e Brachiaria humidicola, após 70 dias de crescimento. O delineamento experimental foi inteiramente casualizado com cinco repetições para cada espécie. As plantas coletadas foram subdivididas nas frações apical, mediana e basal para as folhas e mediana e basilar para caules, de acordo com sua localização. Foram determinadas as concentrações de fibra em detergente neutro (FDN), proteína bruta (PB), lignina, ácido p_cumárico, ácido ferúlico e açúcares neutros (glicose, xilose e arabinose) e a digestibilidade in situ após 48 horas de período de incubação ruminal. As diferentes frações das espécies estudadas apresentam distinta composição química, cujos efeitos são observados na digestibilidade. A B. brizantha apresentou maiores concentrações de FDN no caule e PB nas folhas. Isto resultou em coeficientes de digestibilidade maiores em relação à B. humidicola. A diferença de digestibilidade entre caule e folhas e nas frações mais velhas pode estar relacionada ao tipo de condensação da lignina presente nos tecidos. Evidências na concentração e na proporção dos ácidos p_cumárico e ferúlico sugerem esta relação. A concentração de ácidos fenólicos esteve relacionada com a digestibilidade da matéria seca e a lignina com a digestibilidade da FDN. A análise dos ácidos fenólicos pode se constituir em importante ferramenta para avaliar o grau de condensação da lignina na parede celular dos diferentes tecidos das plantas forrageiras. A concentração de açúcares neutros não apresentou um padrão definido na composição dos diferentes tecidos. A arabinose foi o único açúcar que apresentou relações com a digestibilidade da matéria seca e com a concentração de ácidos fenólicos.

The almost complete absence or misdistribution of water as a natural resource frequently constitute a limiting factor for plant growth and development in the semi-arid northeastern Brazil. In this context, the use of appropriate irrigation techniques is an essential and indispensable factor for proper functioning of the primary and secondary metabolisms in plants. This study aimed to assess the metabolism of the Syrah grapevine in the semi-arid northeastern Brazil, by using three irrigation strategies (controlled deficit irrigation [CDI], deficit irrigation [DI], and full irrigation [FI]). The research was conducted at Embrapa Semiarid (Embrapa-Brazilian Agricultural Research Corporation), Experimental Field of Bebedouro, municipality of Petrolina, state of Pernambuco, Brazil. The statistical design was randomized blocks, with a 3 x 6 factorial scheme. Six samplings were performed throughout the experiment cycle at 54, 60, 73, 87, 101, and 115 days after pruning (DAP). We analyzed the total soluble sugars, reducing sugars, total soluble protein, and invertase activity. Analysis of variance and the F test were performed for all analyzed variables. The means were compared using the Tukey test at 5% significance. At the end of the experiment cycle, DI was found to increase the average acid invertase activity in the plant cell wall. Further, by the end of the experiment cycle, the total soluble sugars and reducing sugars increased in all plants in the three irrigation strategies. Thus, we conclude that it is possible to reduce water consumption in vineyards of the semi-arid northeastern Brazil, without significantly affecting the protein and sugar metabolisms in the plants.
