Objetivos Neste estudo foi investigado o efeito do consumo habitual de suco de laranja no perfil dos lípides e lipoproteínas em homens e mulheres normolipidêmicos. Métodos Todos os voluntários (n=29) consumiram 750mL/dia de suco de laranja durante 60 dias. Variáveis bioquímicas como perfil lipídico, apolipoproteínas, glicose, paraoxonase1 e o tamanho de HDL foram medidas antes e após o período de suplementação com suco de laranja. Também foram realizadas medidas antropométricas e inquéritos dietéticos. Resultados O consumo crônico de suco de laranja reduziu significativamente o colesterol total nos homens (11%, p<0,05) e nas mulheres (10%, p<0,05) e o LDL-C nos homens e mulheres (15%, p<0,05). O HDL-C e a apoA-I também diminuíram, refletindo a redução do colesterol total. Os triacilgliceróis, apo B, PON1, tamanho da HDL, IMC, gordura corporal e circunferência abdominal não foram modificados com o tratamento com suco de laranja. Conclusão Neste estudo, mostrou-se que o suco de laranja apresenta propriedade redutora sobre o colesterol, e foi sugerido que a associação dos flavonóides cítricos com a vitamina C previne o estresse oxidativo e o desenvolvimento da aterosclerose.

OBJETIVO: Investigar as alterações dos marcadores inflamatórios em obesos com glicemia de jejum alterada. MÉTODOS: Foram avaliados 125 indivíduos adultos tendo sido pesquisados: Índice de Massa Corporal, circunferência da cintura, glicemia de jejum, proteína C reativa ultra-sensível, ácido úrico e homocisteína. Os grupos apresentando glicemia de jejum normal (grupo 1) e glicemia de jejum alterada (grupo 2) foram comparados entre si e em associação aos subgrupos de obesos, sobrepesos e eutróficos. RESULTADOS: O grupo 2 apresentou maiores valores de circunferência da cintura (p<0,05), principalmente quando associados ao excesso de peso. Adicionalmente, o grupo 2 mostrou valores de proteína C reativa e ácido úrico superiores, sendo os dos obesos maiores que os com sobrepeso e os eutróficos, enquanto a homocisteína foi semelhante entre obesos, com sobrepeso e eutróficos. O Índice de Massa Corporal correlacionou-se positivamente com ambos ácido úrico (r=0,39, p<0,01) e proteína C reativa (r=0,37; p<0,01). A circunferência da cintura apresentou correlação apenas com o ácido úrico (r=0,53, p<0,01). Porém, a correlação entre Índice de Massa Corporal e proteína C reativa foi significante no grupo 2 (r=0,66, p<0,01), mas não no grupo 1 (r=0,25, p>0,05). O mesmo resultado foi encontrado em relação à circunferência da cintura e à proteína C reativa, que se correlacionaram significantemente somente na presença de glicemia de jejum alterada (r=0,40, p<0,05). Na regressão logística, a circunferência da cintura foi a variável explicativa (11%, p<0,01) da alteração da glicemia de jejum. CONCLUSÃO: Os estados pró-oxidativo e pró-inflamatório estiveram significantemente associados à glicemia de jejum alterada na presença de adiposidade corporal.

A regulação da homeostasia intra e extra-celular da glicose está diretamente relacionada ao controle preciso da expressão dos genes que codificam as diferentes isoformas de proteínas transportadoras de glicose, as quais se expressam de maneira tecido-específica, em conseqüência do padrão de ativação dos fatores transcricionais reguladores de cada gene, em cada tipo celular. A síndrome metabólica (SM) abrange uma grande variedade de alterações fisiopatológicas, todas de repercussões sistêmicas, acometendo os mais distintos territórios do organismo, nos quais alterações nos transportadores de glicose presentes são observadas em maior ou menor grau. A presente revisão abordará as alterações na expressão de transportadores de glicose claramente demonstradas na literatura, cujas repercussões nos fluxos territoriais de glicose auxiliam na compreensão de mecanismos fisiopatológicos da SM, assim como dos tratamentos propostos para esta entidade.

Fourteen samples of particulate matter and semi-volatile organic compounds were collected during 6 months in the city of Campo Grande, South Mato Grosso State, Brazil. Particle-bound polycyclic aromatic hydrocarbons (PAHs) were collected on Fluoropore PTFE filters and gas-phase PAHs were collected into sorbent tubes with XAD-2 resin. Both types of samples were extracted with a dichloromethane/methanol mixture (4:1 v/v), then the extracts were subjected to gas chromatography-mass spectrometry (GC-MS) analysis. PAHs, oxidized PAH (oxy-PAHs), phenols and methoxyphenols were identified by use of GC retention indices and MS files. The average value obtained for the sum of 15 PAHs was 21.05 ng m(-3) (range: 8.94-62.5 ng m(-3)). The presence of specific tracers and calculations of characteristic ratios (e.g. [Phe]/[Phe] + [Ant]) were used to identify the sources of the emissions of PAHs in the atmospheric samples. Levoglucosan (the anhydride of beta-glucose), retene (1-methyl-7-isopropylphenanthrene) and methoxyphenols (derivatives of syringol and guaiacol) and tracers for wood burning were identified. This study demonstrates that biomass burning from the rural zone is the main source of PAHs and emissions of other substances in the investigated site of Campo Grande. (c) 2004 Elsevier Ltd. In a cross-sectional study, we assessed beta-cell function and insulin sensitivity index (ISI) with hyperglycemic clamps (10 mmol/l) in 24 subjects with impaired fasting glycemia (IFG, fasting plasma glucose [FPG] between 6.1 and 7.0 mmol/l), 15 type 2 diabetic subjects (FPG >7.0 mmol/l), and 280 subjects with normal fasting glycemia (NFG, FPG <6.1 mmol/l). First-phase insulin release (0-10 min) was lower in IFG (geometric mean 541 pmol/l (.) 10 min; 95% confidence interval [CI] 416-702 pmol/l (.) 10 min) and in type 2 diabetes (geometric mean 376 pmol/l (.) 10 min; 95% CI 247-572 pmol/l (.) 10 min) than NFG (geometric mean 814 pmol/l (.) 10 min; 95% CI 759-873 pmol/l (.) 10 min) (P < 0.001). Second-phase insulin secretion (140-180 min) was also lower in IFG (geometric mean 251 pmol/l; 95% CI 198-318 pmol/l; P = 0.026) and type 2 diabetes (geometric mean 157 pmol/l; 95% CI 105-235 pmol/l; P < 0.001) than NFG (geometric mean 295 pmol/l; 95% CI 276-315 pmol/l): IFG and type 2 diabetic subjects had a lower ISI (0.15 +/- 0.02 and 0.16 +/- 0.02 mumol/kg fat-free mass [FFM]/min/ pmol/l, respectively) than NFG (0.24 +/- 0.01 mumol/kg FFM/min/pmol/l, P < 0.05). We found a stepwise decline in first-phase (and second-phase) secretion in NFG subjects with progressive decline in oral glucose tolerance (P < 0.05). IFG subjects with impaired glucose tolerance (IGT) had lower first-phase secretion than NFG subjects with IGT (P < 0.02), with comparable second-phase secretion and ISI. NFG and IFG subjects with a diabetic glucose tolerance (2-h glucose >11.1 mmol/l) had a lower ISI than their respective IGT counterparts (P < 0.05). We conclude that the early stages of glucose intolerance are associated with disturbances in beta-cell function, while insulin resistance is seen more markedly in later stages.

Fungi producing γ-linolenic acid (GLA) were isolated from soil of the Ecological Station of Juréia-Itatins, SP. This essential fatty acid has aroused great interest due to its increasing by applications in pharmaceutical industry. The GLA production by zygomycetous fungi is an alternative way of comparing seed extraction. Thirty-two zygomycetous strains of Mucorales were isolated, most of them belonging to Mucor genus. The GLA production was evaluated after 4 days of incubation at 25°C on a rotary shaker at 150 rpm in medium containing 2% glucose, and 1% yeast extract, following new medium addition (20%) and incubation for an additional period of 3 days at 12°C, without agitation. The GLA production varied according to the microorganism and the strain.

The biosynthetic origins of the isoprene units of 4-nerolidylcatechol (1), the major constituent of Potomorphe umbellata, have been studied through feeding experiments with [14C]- and [13C]-glucose, and with precursors of the mevalonic acid and triose/pyruvate pathways, namely, [2- 14C]-mevalonolactone and [U-14C]-glyceraldehyde-3- phosphate, respectively. The pattern of incorporation of label from [1- 13C]-glucose into 1 was determined by quantitative 13C NMR spectroscopy. The labelling pattern revealed that the additive was specifically incorporated, and that the isoprene units of the sesquiterpenoid moiety of 4-nerolidylcatechol were derived from both the mevalonic acid and the triose/pyruvate pathways. The results indicate that both plastidic and cytoplasmic pathways are able to provide isopentenyl diphosphate units for the biosynthesis of 1. ©2005 Sociedade Brasileira de Química.

Tithonia diversifolia, also known as Mexican arnica, has been used in traditional medicine to treat inflammatory refractory with absence of citotoxicity. The possible health risks associated with the consumption of ingestion of the infusion (tea) plant makes it is necessary to identify the potential pharmacological activity or toxicity to prove certain plants that are acclimated in Brazil. Considering the limited number of pharmacological studies regarding the Tithonia diversifolia, the aim of this study was evaluate the effects of this infusion in platelet aggregation. Venous blood was collected with informed consent from healthy volunteers who denied taking any medication in the previous 14 days. Whole blood was transferred into polypropylene tubes containing one-tenth of final volume of acid citrate dextrose (ACD-C; citric acid 3%, trisodium citrate 4%, glucose 2%; 1:9 v/v) and centrifuged at 200g for 15 min. Platelet rich plasma was added of wash buffer solution (NaCl 140mM, KCl 5mM, sodium citrate 12mM, glucose 10mM and saccharose 12mM; pH 6; 5:7 v/v) and centrifuged at 800g for 12 min at 20°C. Platelet pellet was gently resuspended in Krebs-Ringer solution and counts were performed on a Neubauer chamber. Aggregation assay was carried out with 400 μL of platelet suspension (1.2x10 8 platelets/mL) in a cuvette at 37°C with constant stirring. Platelet suspension was incubated for 3 min with aqueous extract infusion (0.6-20μg/mL) prior to addition of thrombin (100 mU/mL). Percentage of platelet aggregation was recorded with an aggregometer (Chrono-log Lumi-Aggregometer model 560-Ca, USA). Our results show an inhibition of thrombin induced platelet aggregation in the presence of 0.6-20 ug/mL Tithonia diversifolia infusion leaves. The Tithonia diversifolia infusion leaves inhibits thrombin induced washed platelet aggregation.
