Piaractus mesopotamicus were fed diets supplemented with three vitamin C and/or E levels (zero, 250, and 500 mg vitamin/kg dry ration) and challenged with Aeromonas hydrophila. The fish were fed during the first 60 days with diets without vitamins C and E, in an attempt to reduce vitamin sources. After this period, test diets were offered during 60 days. At the end of the experiment, all fish were intraperitoneally injected with 6 x 10(6) Colony forming units (cfu) of A. hydrophila per fish. It was concluded that for pacu, vitamin C and E are essential for protection of erythrocytes. Vitamin C induces an increase in the number of circulating thrombocytes in a dose-response relation. However, just like vitamin deficiency should be avoided, excess vitamins can also cause damage to fish as observed in the hematocrit and hemoglobin values. Based on the hematological responses obtained, the recommended vitamin C and vitamin E levels for P mesopotamicus juveniles are 500 and 250 mg/kg feed, respectively. (C) 2007 Elsevier B.V. The present study assessed the kinetics of cell accumulation at the site of inflammation induced by thioglycolate, Escherichia coli lipopolysaccharide (LPS) and heat-inactivated Aeromonas hydrophila, in the pacu, Piaractus mesopotamicus (Characidae), swim bladder. A quantitative, as well as qualitative, assessment was done of all the cells present in the exudate at 6, 24, and 48 h (n = 8) after inoculation of inflammatory agents. The results show that the thioglycolate was the irritant to induce higher total inflammatory cell accumulation when compared to the control group, 6 h after insult (P < 0.05). Inoculation of heat-inactivated Aeromonas hydrophila induced progressive accumulation of total inflammatory cells, with cell number peaking after 24 h and being significantly higher than observed in the other groups (P < 0.05). Injection of LPS also induced greater cell accumulation when compared to the control group (P < 0.05), although in lower numbers than those induced by the other two irritants. All irritants injected induced significantly greater accumulation of lymphocytes and thrombocytes when compared to the control group (P < 0.05).

This study evaluated the effects of dietary supplementation with 0.3% Saccharomyces cerevisiae yeast cell wall and of vaccination against Streptococcus agalactiae on the cellular component of acute inflammation induced in the coelomic cavity of Nile tilapia Oreochromis niloticus and on survival of the fish after challenge. A total of 84 tilapia of mean (+/- SD) weight 125.0 +/- 1.5 g were distributed among twelve 310 l fiberglass tanks according to a 2 x 2 x 3 factorial design in the following manner: with and without supplementation; 2 stimulations (oily solution without S. agalactiae vaccine and vaccination); 15 d later all fish were intracoelomically challenged with 10(8) CFU ml(-1) of a homologous strain of S. agalactiae, and evaluated after 6, 24 and 48 h, with 7 replicates. The fish received the non-supplemented or supplemented diet for a total of 77 d. The vaccination was performed on the 60th day, intracoelomically, as a single injection of 0.5 ml of the vaccine containing 10(8) CFU ml(-1). Fifteen days later, all the fish were challenged with S. agalactiae by means of an intracoelomic inoculation of 10(8) CFU ml(-1). No mortality was observed among the supplemented fish. The fish that were fed the non-supplemented diet and immunized with the bacterium presented a mortality rate of 28.5%. Among the non-supplemented and non-immunized fish, the mortality rate was 38.09%. Supplementation, in both vaccinated and non-vaccinated fish, induced larger accumulations of thrombocytes, lymphocytes and macrophages at the inflammatory focus. The results suggest that supplementation with 0.3% yeast cell wall, in both vaccinated and non-vaccinated fish, improved the inflammatory response of the fish and protected against the challenge. Vaccination increased the defense response, but the effect was stronger when associated with supplementation with S. cerevisiae.

This work aimed to study the reproduction and description of technique for digital sampling images during rats gait and determination of the sciatic functional index (SFI), through a glass walkway to obtain shoots with a digital camera. After controlled injury by strangulation of sciatic nerve, with 3mm of length, during 30 seconds, using hemostatic forceps, a group of 32 rats was performed 24 hours before the lesion which served as control and 24 hours, 7, 14 and 21 days after injury. The tests consisted in the filming and shooting each animal in order to observe the view from below (by a mirror to 45 degrees) and subsequently analyzed using the IMAGE-J program. Measures were taken from the lengths of the legs (right and left), and the distance between the ankle. In the analysis of IFC, values close to zero (0) suggest that the function of the sciatic nerve is still preserved and values coming from "less one hundred" (-100) indicate total loss of function. It was verified in this study that 24 hours before surgery the average SFI was -7.07 +/- 7.88 and 24 hours after injury these values rose to an average of -77.95 +/- 13.81, being about 10 times larger, where 78% of the animals showed 60 to 100% of functional loss in motor activity, demonstrated by the gradual recovery over the days analyzed, confirming the accuracy and effectiveness of the proposed methodology. These results suggest that studies can be conducted to simplify and reduce costs using the technique for digital images of footprints during rats gait in the laboratory.

This work evaluated piaucu juveniles (Leporinus macrocephalus) efficiency on the control of Dolops carvalhoi in pacu (Piaractus mesopotamicus) under artificial infestation. 30 fish were equally distributed in six aquarium and acclimated for seven days. After this period the pacus were unnaturaly infested with 20 D. carvalhoi per fish to four days. on the fifth day, for aquaria were added with five uninfected piaucus each and the fish were checked eight times to evaluted the total number of parasites in fish body and aquarium wall. After 24 hours in the aquarium with piaucus the total number of Dolops was significant lower than only pacus suggesting that piaucu preyed the parasite. The parasite number was reduced 15 minutes after the addition of piaucus and after 24 hours, no parasites was found in pacu body and aquarium walls, suggesting that piaucu fingerling may be used to branchiurans infestations in other species of fish.

The objective of this work was to evaluate the skin, liver and kidney histology of young pacus supplemented in diet with trivalent chromium and to evaluate the residue of chromium in water effluent. Randomised delineation by using factorial scheme 4x2, with four levels of chromium (0, 6, 12, 18 mg/L), two stocking densities (4 and 20 kg/m(3)), and four repetitions were used. Pacus with 100 g were used, for 90 days. An increase of the number of mucus cells and club cells in the epidermis of the pacus supplemented with 18 chromium mg/kg was observed in any stocking density. The same effect was observed in the liver, and in the highest density (20 kg/m(3)) the alterations already occurred at the level of 6 mg/kg with the inclusion of chromium, initially with congestion of sinusoids and getting worse at the level of 18 mg/kg with necrosis. The kidney didn't present visible morphologic alterations when supplemented and submitted to the stocking densities. The concentration of chromium in the effluent of pacus was 3.27 mu g/L in the higher level of supplementation (18 mg/L). In conclusion the level of 18 supplement mg/kg was not shown beneficial to the growth of pacus and the stocking density, due to the possible stress, accelerated the alteration histology process of tissues.

This work describes intestine histopathology of Pseudoplatystoma fasciatum infected by endohelminths. Nineteen specimens were caught from Aquidauna River, Mato Grosso do Sul State, in October 2005 and submitted to necropsy, when intestine of each specimen was fixed with AFA solution during 12 h. Soon after, samples were immersed in buffered formalin and 24 h after, transferred to alcohol 70%. Study of histology sections was made to 5; m and stained with haematoxylin and eosin. It showed structures displaying helminths encapsulated by connective tissue in intestinal muscle with fibroplasy in several stages of capsule development. Were observed focus of necrosis in intestinal mucous and sub-mucous and helminthes cut obliquely in intestinal lumen. In addition, there was discreet mucous desquamation, associated to basal membrane thickness.

The inflammatory response and hernatological parameters among Nile tilapia (Oreochromis niloticus) supplemented with Saccharomyces cerevisiae were evaluated six and 24 h after inoculation with inactivated Aeromonas hydrophila into the swim bladder. Six groups were formed (n = 10 each): G1 was treated with non-supplemented feed+injection with 0.65% saline solution; G2 with non-supplemented feed+ inoculation with A. hydrophila: G3 with feed containing 2% yeast+ injection with saline; G4 with feed containing 2% yeast + inoculation with A. hydrophila: G5 with feed containing 0.3% cell wall + injection with saline: and G6 with feed containing 0.3% cell wall + inoculation with A. hydrophila. In the groups inoculated with bacteria, the responses were more intense (P<0.05) than in those injected with saline. The groups receiving supplement that were inoculated with A. hydrophila accumulated a greater total number of cells at the lesion site (P<0.05) than did the non-supplemented groups, after six and 24 h. The groups receiving cell wall presented greater total accumulation of cells (P<0.005) that did those receiving yeast. The differential count showed that there were significantly greater number of thrombocytes (P< 0.05) and lower number of neutrophils, macrophages and lymphocytes (P<0.05) in the groups that received supplement, after 6 and 24 h, in relation to the non-supplemented groups. The values in the erythrocyte count, hemoglobin concentration and blood measurement indices did not differ statistically. The variation in circulating thrombocyte and leukocyte counts suggests that the inflammatory stimulus caused recruitment from reserve compartments to the blood. The groups that received yeast or yeast cell wall supplements presented increased nonspecific acute inflammatory response, thus suggesting that this has a beneficial effect on the immunological defense system. The cellular and molecular characteristics of a cell line (BME26) derived from embryos of the cattle tick Rhipicephalus (Boophilus) microplus were studied. The cells contained glycogen inclusions, numerous mitochondria, and vesicles with heterogeneous electron densities dispersed throughout the cytoplasm. Vesicles contained lipids and sequestered palladium meso-porphyrin (Pd-mP) and rhodamine-hemoglobin, suggesting their involvement in the autophagic and endocytic pathways. The cells phagocytosed yeast and expressed genes encoding the antimicrobial peptides (microplusin and defensin). A cDNA library was made and 898 unique mRNA sequences were obtained. Among them, 556 sequences were not significantly similar to any sequence found in public databases. Annotation using Gene Ontology revealed transcripts related to several different functional classes. We identified transcripts involved in immune response such as ferritin, serine proteases, protease inhibitors,. antimicrobial peptides, heat shock protein, glutathione S-transferase, peroxidase, and NADPH oxidase. BME26 cells transfected with a plasmid carrying a red fluorescent protein reporter gene (DsRed2) transiently expressed DsRed2 for up to 5 weeks. We conclude that BME26 can be used to experimentally analyze diverse biological processes that occur in R. (B.) microplus such as the innate immune response to tick-borne pathogens. O experimento teve como objetivo avaliar os sistemas de alimentação convencional ou de livre escolha, utilizando 6 dietas a base de milho em grão ou moído e concentrado protéico sobre o desempenho de frangos de corte do l° ao 45° dia de idade. O delineamento experimental foi inteiramente casualizado em esquema fatorial 6x2 (6 tratamentos e 2 sexos), com 3 repetições de 30 aves. Os tratamentos foram: T1-Milho moído + Concentrado do 1° ao 45° dia, fornecidos misturados; T2-Milho moído + Concentrado do 1° ao 45° dia, fornecidos separados em 2 comedouros; T3- Milho moído + Concentrado, fornecidos misturados, do l° ao 14° dia e do 15° ao 45° dia, milho em grão + concentrado, fornecidos misturados; T4 Milho moído + Concentrado, fornecidos misturados, do 1° ao 14° dia e do 15° ao 45° dia, milho em grão + concentrado, fornecidos separados; T5 - Milho moído + Concentrado, fornecidos misturados do 1° ao 21° dia e do 22° ao 45° dia, milho em grão + concentrado, fornecidos misturados; T6- Milho moído + Concentrado, fornecidos separados, do l ° ao 21° dia e do 22° ao 45° dia, milho em grão + concentrado, fornecidos misturados. Pêlos resultados e condições que realizou-se o presente trabalho conclui-se que: o milho em grão pode ser fornecido a frangos de corte misturado ao concentrado a partir do 15° dia de idade; o milho moído não proporcionou bons resultados de desempenho no programa de alimentação de livre escolha: as aves alimentadas com milho em grão apresentaram maior peso de moela, bem como acumularam mais gordura na carcaça; autilização de milho em grão fornecido junto ao concentrado, embora tenha proporcionado desempenho ligeiramente inferior à ração convencional, pode ser considerado satisfarório, sendo uma alternativa para pequenos produtores avícolas.

O objetivo deste trabalho foi avaliar os efeitos da restrição alimentar qualitativa precoce sobre o desempenho, as características do trato gastrintestinal e a carcaça de frangos de corte. Um total de 975 pintos de corte de um dia da linhagem Ross, 15 parcelas com 30 machos cada e 15 parcelas com 35 fêmeas, foi distribuído em delineamento experimental inteiramente casualizado, em esquema fatorial 5 x 2 (forma de restrição x sexo), com três repetições por tratamento. As formas de restrição estudadas foram: 1 - testemunha, sem diluição; 2 -diluição com 15% de calcário; 3 - diluição com 30% de calcário; 4 - diluição com 15% de casca de arroz; e 5 - diluição com 30% de casca de arroz. Os frangos foram alimentados ad libitum com ração padrão, no período de 1 a 7 dias e de 15 a 45 dias de idade. A restrição alimentar por diluição das rações, conforme cada tratamento, foi aplicada do 8º ao 14º dia de idade. Para o período de restrição alimentar, foram formuladas rações com níveis normais de minerais e vitaminas, reduzindo-se os outros nutrientes, conforme a diluição. Os frangos submetidos à restrição alimentar qualitativa precoce, pela diluição com 15% de calcário e, 15 ou 30% de casca de arroz, apresentaram ganho compensatório, com bom desempenho final. Entretanto, a restrição com 30% de calcário foi muito severa, prejudicando o desempenho. Houve alteração nas características do trato gastrintestinal com a restrição alimentar, porém houve recuperação até o momento do abate. Os frangos submetidos à restrição alimentar qualitativa precoce apresentaram menor deposição de gordura e maior deposição de proteína na carcaça em comparação ao controle.

Objetivou-se avaliar as equações para predição das exigências protéicas de matrizes pesadas em crescimento, desenvolvidas em estudos anteriores na Faculdade de Ciências Agrárias e Veterinárias da UNESP, Jaboticabal, tendo como padrão de comparação as recomendações estabelecidas pelo manual de criação da linhagem. Foram utilizadas 288 matrizes pesadas da linhagem Hubbard Hy-Yield, com cinco semanas de idade, em delineamento inteiramente casualizado, com dois tratamentos e seis repetições, sendo 24 aves por unidade experimental até 14 semanas e, de 15 a 20 semanas de idade, 20 aves por unidade experimental. Um tratamento foi representado pelas recomendações protéicas para a linhagem (testemunha) e o outro pelas seguintes equações de predição das exigências de proteína: de 5 a 8 semanas PB (g/ave/dia) = 2,02.P0,75+ 0,350.G, 9 a 14 semanas - PB = 2,02.P0,75 + 0,406.G e de 15 a 20 semanas PB = 2,02.P0,75 + 0,463.G, em que P é o peso corporal (kg) e G, o ganho de peso diário (g). Os resultados indicaram que as equações de predição para proteína, apesar de não comprometerem o peso ou a uniformidade corporal em relação ao tratamento testemunha, determinaram menor porcentagem de peito e maior de gordura abdominal e maior teor de gordura na carcaça, sendo atribuído à menor ingestão de proteína proporcionada pelas equações de predição e, conseqüentemente, deficiência em relação a alguns aminoácidos que não foram suplementados.

O objetivo deste experimento foi avaliar o desempenho de bovinos em confinamento alimentados com rações suplementadas com concentrados protéicos à base de farelo de algodão (AL), uréia (UR) ou amiréia (AM), tendo como volumoso silagem de milho. O confinamento teve duração de 80 dias. As rações foram compostas mantendo a proporção volumoso:concentrado no nível de 63:37, com aproximadamente 13% PB. Não se constataram diferenças quanto ao ganho de peso diário (GDP) durante o período experimental total entre os tratamentos AL, UR ou AM (1,14; 1,17; e 1,23 kg/anim. dia), ou na ingestão de MS em % PV (2,63; 2,60; e 2,60% PV), em g/kg0,75 dia (136,19; 134,74; e 134,81) ou kg/anim. dia (9,42; 9,35; e 9,40), respectivamente. Os valores de ingestão de PB foram de 1,28; 1,24; e 1,34 kg de PB/anim. dia e de 18,54; 17,86; e 19,24 g/kg0,75 dia para os tratamentos AL, UR ou AM, não diferindo entre si. Valores obtidos para conversão alimentar da matéria seca foram de 8,41; 8,01; e 7,67 kg MSI/kg GPD e para conversão protéica, 1,15; 1,06; e 1,09 kg PBI/kg GPD, respectivamente, para os tratamentos AL, UR e AM, que foram semelhantes.
