Phospholipases A(2) are components of Bothrops venoms responsible for disruption of cell membrane integrity via hydrolysis of its phospholipids. A class of PLA(2)-like proteins has been described which despite PLA(2) activity on artificial substrate, due to a D49K mutation, is still highly myonecrotic. This work reports the X-ray structure determination of two Lys49-PLA(2)s from Bothrops neuwiedi pauloensis (BnSP-7 and BnSP-6) and, for the first time, the comparison of eight dimeric Lys49-PLA2s. This comparison reveals that there are not just two (open and closed) but at least six different conformations. The binding of fatty acid observed in three recent Lys49-PLA(2) structures seems to be independent of their quaternary conformation. Cys29 polarization by Lys122 is not significant for BnSP-7 and BnSP-6 or other structures not bound by fatty acids. These structures may be in an active state when nothing is bound to them and the Lys122/Cys29 interactions are weak or absent. (C) 2003 Elsevier B.V. All rights reserved.

The ultrastructure of ovarian sperm storage of Helicolenus dactylopterus dactylopterus is described, before and after the spawning period. The spermatozoa remain inside cryptal structures that are situated in the interlamellar gaps and are connected to the ovarian lumen by a duct. This complex forms a highly specialised structure. During the long storage period, crypts are richly vascularised. Their surrounding simple epithelia have intercellular junctions that may serve to protect the spermatozoa from the female immune system. At the moment during which insemination of mature oocytes occurs, the sperm may be expelled from cryptal structures by means of a spasmodic contraction. During the post spawning period, residual spermatozoa that remain in the crypts are eliminated by cryptal phagocytes. At the end of the process the crypts contain only an amorphous material.

The total number and distribution per antennal flagellomere of sensilla placodea (olfactory disks), sensilla coeloconica, sensilla ampullacea and sensilla campaniformia were determined in workers of Nannotrigona testaceicornis Lepeletier a stingless bee species quite common in Brazil. The distribution of the sensilla was uniform, with the largest number occurring in flagellomere 10 and gradually decreasing in the direction of the basal flagellomeres in a way similar to that observed in Scaptotrigona postica Latreille.Nannotrigona testaceicornis had a larger number of sensilla ampullacea and a smaller number of sensilla coeloconica and sensilla campaniformia than Scaptotrigona postica. Although Nan notrigona testaceicornis does not communicate through the formation of pheromone trails, this species presents a larger quantity of sensilla placodea (relative to the length of the flagellum) than Scaptotrigona postica.

Human purine nucleoside phosphorylase (PNP) is a ubiquitous enzyme which plays a key role in the purine salvage pathway, and PNP deficiency in humans leads to an impairment of T-cell function, usually with no apparent effect on B-cell function. PNP is highly specific for 6-oxopurine nucleosides and exhibits negligible activity for 6-aminopurine nucleosides. The catalytic efficiency for inosine is 350,000-fold greater than for adenosine. Adenine nucleosides and nucleotides are deaminated by adenosine deaminase and AMP deaminase to their corresponding inosine derivatives which, in turn, may be further degraded. Here we report the crystal structures of human PNP in complex with inosine and 2',3'-dideoxymosine, refined to 2.8 Angstrom resolution using synchrotron radiation. The present structures provide explanation for ligand binding, refine the purine-binding site, and can be used for future inhibitor design. (C) 2003 Elsevier B.V. All rights reserved.

The electrophile Ca2+ is an essential multifunctional co-factor in the phospholipase A(2) mediated hydrolysis of phospholipids. Crystal structures of an acidic phospholipase A(2) from the venom of Bothrops jararacussu have been determined both in the Ca2+ free and bound states at 0.97 and 1.60 angstrom resolutions, respectively. In the Ca2+ bound state, the Ca2+ ion is penta-coordinated by a distorted pyramidal cage of oxygen and nitrogen atoms that is significantly different to that observed in structures of other Group I/II phospholipases A(2). In the absence of Ca2+, a water molecule occupies the position of the Ca2+ ion and the side chain of Asp49 and the calcium-binding loop adopts a different conformation. (c) 2005 Elsevier SAS. Protein C activation initiated by the thrombin-thrombomodulin complex forms the major physiological anticoagulant pathway. Agkistrodon contortrix contortrix protein C activator, a glycosylated single-chain serine proteinase, activates protein C without relying on thrombomodulin. The crystal structures of native and inhibited Agkistrodon contortrix contortrix protein C activator determined at 1.65 and 1.54 angstrom resolutions, respectively, indicate the pivotal roles played by the positively charged belt and the strategic positioning of the three carbohydrate moieties surrounding the catalytic site in protein C recognition, binding, and activation. Structural changes in the benzamidine-inhibited enzyme suggest a probable function in allosteric regulation for the anion-binding site located in the C-terminal extension, which is fully conserved in snake venom serine proteinases, that preferentially binds Cl1- instead of SO42-.

The protein content of many snake venoms often includes one or more phospholipases A(2) (PLA(2)). In recent years a growing number of venoms from snakes of Agkistrodon, Bothrops and Trimeresurus species have been shown to contain a catalytically inactive PLA(2)-homologue in which the highly conserved aspartic acid at position 49 (Asp49) is substituted by lysine (Lys49). Although demonstrating little or no catalytic activity, these Lys49-PLA(2)s disrupt membranes by a Ca2+-independent mechanism of action. In addition, this family of PLA(2)s demonstrates myotoxic and cytolytic pharmacological activities, however the structural bases underlying these functional properties are poorly understood. Through the application of X-ray crystallography in combination with biophysical and bioinformatics techniques, we are studying structure/function relationships of Lys49-PLA(2)s. We here present results of a systematic X-ray crystallographic and amino acid sequence analysis study of Lys49-PLA(2)s and propose a model to explain the Ca2+ independent membrane damaging activity. Three dimensional exactly solvable quantum potentials for which an extra term of form 1/r(2) has been added are shown to maintain their functional form which allows the construction of the Hamiltonian hierarchy and the determination of the spectra of eigenvalues and eigenfunctions within the Supersymmetric Quantum Mechanics formalism. For the specific cases of the harmonic oscillator and the Coulomb potentials, known as pseudo-harmonic oscillator and pseudo-Coulomb potentials, it is shown here that the inclusion of the new term corresponds to rescaling the angular momentum and it is responsible for maintaining their exact solvability.

We point out that a common feature of integrable hierarchies presenting soliton solutions is the existence of some special ''vacuum solutions'' such that the Lax operators evaluated on them, lie in some abelian subalgebra of the associated Kac-Moody algebra. The soliton solutions are constructed out of those ''vacuum solitons'' by the dressing transformation procedure.

Some properties of the Clifford algebras Cl-3,Cl-0, Cl-1,Cl-3, Cl-4,Cl-1 similar or equal to C circle times Cl-1,Cl-3 and Cl-2,Cl-4 are presented, and three isomorphisms between the Dirac-Clifford algebra C circle times Cl-1,Cl-3 and Cl-4,Cl-1 are exhibited, in order to construct conformal maps and twistors, using the paravector model of spacetime. The isomorphism between the twistor space inner product isometry group SU( 2,2) and the group $pin(+)(2,4) is also investigated, in the light of a suitable isomorphism between C circle times Cl-1,Cl-3 and Cl-4,Cl-1. After reviewing the conformal spacetime structure, conformal maps are described in Minkowski spacetime as the twisted adjoint representation of $ pin(+)(2,4), acting on paravectors. Twistors are then presented via the paravector model of Clifford algebras and related to conformal maps in the Clifford algebra over the Lorentzian R-4,(1) spacetime.We construct twistors in Minkowski spacetime as algebraic spinors associated with the Dirac-Clifford algebra C circle times Cl-1,Cl-3 using one lower spacetime dimension than standard Clifford algebra formulations, since for this purpose, the Clifford algebra over R-4,R-1 is also used to describe conformal maps, instead of R-2,(4). Our formalism sheds some new light on the use of the paravector model and generalizations.
