mRNA stability is modulated by elements in the mRNA transcript and their cognate RNA binding proteins. Poly(U) binding protein 1 (Pub1) is a cytoplasmic Saccharomyces cerevisiae mRNA binding protein that stabilizes transcripts containing AU-rich elements (AREs) or stabilizer elements (STEs). In a yeast two-hybrid screen, we identified nuclear poly(A) binding protein 2 (Nab2) as being a Pub1-interacting protein. Nab2 is an essential nucleocytoplasmic shuttling mRNA binding protein that regulates poly(A) tail length and mRNA export. The interaction between Pub1 and Nab2 was confirmed by copurification and in vitro binding assays. The interaction is mediated by the Nab2 zinc finger domain. Analysis of the functional link between these proteins reveals that Nab2, like Pub1, can modulate the stability of specific mRNA transcripts. The half-life of the RPS16B transcript, an ARE-like sequence-containing Pub1 target, is decreased in both nab2-1 and nab2-67 mutants. In contrast, GCN4, an STE-containing Pub1 target, is not affected. Similar results were obtained for other ARE- and STE-containing Pub1 target transcripts. Further analysis reveals that the ARE-like sequence is necessary for Nab2-mediated transcript stabilization. These results suggest that Nab2 functions together with Pub1 to modulate mRNA stability and strengthen a model where nuclear events are coupled to the control of mRNA turnover in the cytoplasm.

The chemopreventive potential of an Agaricus blazei (Ab) Murrill mushroom meal was investigated in a medium-term rat liver carcinogenesis assay. Male Wistar rats initiated for hepatocarcinogenesis with diethylnitrosamine (DEN, 200 mg/kg i.p.) were fed during a 6-week period with the dry powdered mushroom strains Ab 29 or 26, each one with opened (OB) or closed basidiocarp (CB), mixed at 10% level in a basal diet. All experimental animals and controls were subjected to partial hepatectomy at week 3 and killed at week 8. Chemopreventive activity of the mushroom meal was observed for the Ab 29 (OB and CB) and Ab 26 (CB) strains in terms of the number of putative preneoplastic altered foci of hepatocytes which express either the enzyme glutathione S-transferase, placental form (GST-P+) or the transforming growth factor-alpha, and for the Ab 29 (OB) and Ab 26 (CB) strains on the size of GST-P-divided by foci. This was associated with inhibition of foci cell proliferation in the animals fed the Ab 29 (013) and Ab 26 (CB) strains. The results suggest that the protective influence of the Ab meal against the DEN potential for rat liver carcinogenicity depends on both the strain and period of mushroom harvest. Ants frequently harvest seeds from faeces of frugivorous vertebrates. By transporting these seeds to nests, ants may influence seed dispersal success of plants. Seed removal by ants from vertebrate faeces is influenced by the seed species involved. Faeces from different vertebrates differ in chemical composition and physical characteristics. It remains unclear, however, whether the faeces itself affects the ant-seed interaction. In this study experimental seed-containing faecal portions were prepared using defecations of birds, marsupials, and monkeys and seeds of two aroids (Philodendron corcovadense and P. ppendiculatum, Araceae) and one liana species (Schlegelia parviflora, Bignoniaceae). Faecal portions were arranged along a transect established in the understory of a lowland rainforest in southeast Brazil. For P. appendiculatum the probability of detection and the proportion of seeds removed were identical between marsupial and monkey faeces. For P. corcovadense and S. parviflora, the probability of detection was affected by seed species and, apparently, also by the interaction between seed species and type of faeces (P = 0.097), but not by the type of faeces itself (bird or monkey). Both factors (i.e., seed species and type of faeces) affected the proportion of seeds removed (faeces type was marginally significant; P = 0.08), whereas the interaction between them was not significant. The results indicate that seed species affects seed removal by ants, while the type of faeces probably interacts with seed traits to influence faeces detection.

Os solos agrícolas podem atuar como dreno ou fonte de C atmosférico, dependendo do sistema de manejo adotado. Este estudo foi desenvolvido em experimento de longa duração (22 anos), durante o período de 30 dias do outono, com o objetivo de avaliar o impacto de sistemas de preparo de solo (preparo convencional-PC e plantio direto-PD) nas emissões de C-CO2 de um Latossolo Vermelho distrófico, em Cruz Alta, RS. As emissões de C-CO2 do solo foram avaliadas com câmaras dinâmica (Flux Chamber 6400-09, Licor) e estática (com captação em solução alcalina), imediatamente após a colheita da soja. A temperatura e a umidade do solo foram registradas, concomitantemente com as emissões de C-CO2, por meio de sensor de temperatura e TDR manual, respectivamente, integrantes do Licor-6400. Estimou-se que, em 30 dias, uma quantidade equivalente a menos de 30 % do C aportado pelos resíduos de soja foi emitida na forma de C-CO2. As emissões de C-CO2 no solo em PD foram similares às emissões do solo em PC, independentemente do tipo de câmara utilizada. Diferenças entre sistemas de preparo quanto à emissão de C-CO2, avaliadas com a câmara dinâmica, foram verificadas somente a curto prazo (leituras diárias), com o PD apresentando maiores emissões do que o PC no início do período experimental e menores no final. A câmara dinâmica foi mais eficiente do que a estática em captar as alterações das emissões de C-CO2 em função da variação da temperatura e a porosidade preenchida por água (PPA) no solo em PD, as quais explicaram 83 e 62 % das emissões de C-CO2, respectivamente. O fator Q10, que avalia a sensibilidade da emissão de C-CO2 à temperatura do solo, foi estimado em 3,93, indicando alta sensibilidade da atividade microbiana à temperatura do solo durante o outono. As emissões de C-CO2 registradas no solo em PD com a câmara estática foram correlacionadas às da câmara dinâmica, porém com valores subestimados em relação àquela notadamente nos maiores valores de fluxo. em condições de baixa temperatura e PPA, o preparo de solo induziu limitado incremento de emissão de C-CO2.

Ceratocystis fimbriata foi encontrado em tubérculos de inhame (Colocasia esculenta), apresentando lesões escuras, pouco profundas, contendo estruturas de reprodução do fungo, cuja coloração variava do cinza ao negro. As amostras foram coletadas em supermercados, quitandas e varejões nos Estados de São Paulo, Rio de Janeiro, Bahia, Rondônia e Distrito Federal que, na maioria dos casos, comercializavam inhame produzido no Estado de São Paulo. Os sintomas de podridão negra indicam se tratar de uma doença de pós-colheita. Seqüências de rDNA indicam que os isolados de Colocasia sp. pertencem ao clado da America Latina do complexo C. fimbriata, embora esses isolados sejam mais agressivos em pseudo-pecíolos de C. esculenta do que os isolados de Ficus carica e Mangifera indica.

Objetivou-se avaliar o efeito do tratamento químico com ureia (3 ou 5% na MS) e amônia anidra (3% na MS) no feno de resíduo pós-colheita de sementes de Brachiaria brizantha, cv. Marandu, contendo diferentes teores de umidade (15, 25 ou 30%). O delineamento experimental foi em blocos ao acaso com oito tratamentos e quatro repetições (camadas de fardos dentro das pilhas). O feno tratado com 3% de amônia anidra com 15% de umidade ocasionou redução de 84,3 para 79,1% nos teores de fibra em detergente neutro (FDN) e elevação de 37,3 para 55,5% na digestibilidade in vitro da matéria seca (MS) em relação ao grupo controle. A variação na umidade não alterou de maneira significativa a ação da amônia, cujos valores médios foram 77,6% de FDN e 57,3% de digestibilidade in vitro da MS. O feno com 5% de ureia reduziu os teores de FDN de 84,3 para 79,6% em relação ao feno não-tratado, o que tornou necessário o aumento da umidade para 30% para maior efeito sobre a digestibilidade da MS, que aumentou 12 unidades percentuais.

The content of ascorbic acid was assayed in acerolas harvested in three phases of maturation: green-yellow fruits (I); light red (II) and wine-coloured (III). Phase I and Phase II fruit were packed in aluminium sheets and stoppered flasks and stored in freezer (-10o.C) and in refrigerator (8o.C). Samples of 8 fruits from each experimental condiction were analysed for ascorbic acid determination by 2-chlorophenol indophenol discolouration method. The averages of 1.393,5 mg./100g. for Phase I sample, 1024,9 for Phase II and 756,5 for Phase III fruits, showed a statistically significative linear decreasing of the ascorbic acid content related with the maturation extent Phase I samples stored in freezing showed statitically significative decreasing of that vitamin at 408 hours of storage in both: aluminium sheet and stoppered flask package; in chilling temperature there was significative reduction of ascorbic acid content after 240 and 312 hours, respectively, for fruits packed in aluminium sheet and stopped flasks. Phase Il samples showed significative lost at 72 hours of storage when maintained in freezing temperature either, in aluminium sheet or in stoppered flasks: When stored in chilling temperature showed progressive lost of ascorbic acid in all measuring periods in every package. After 144 hours suffered deterioration suggested by colour changes.

The present work was conducted at FCAV-UNESP, Campus de Jaboticabal, in late harvest crop time of 1991. One hundred interpopulational hybrids obtained from a top-cross between Dent Composite and Flint Composite populations were evaluated. The analyzed characteristics were: Spodoptera frugiperda damages, lodging, damaged ears percentage, and productivity. Estimates of heritability, variance between progenies and genetical gain for each character of available population were determined. Estimates of the progenies variance, like those from heritability for the different analysed characters, indicate that there is an adequate genetical variance for the utilization of that material on subsequent genetic breeding programs, though allowing genetical gains, on the following selection cycles for the characteristics: grain weight, lodging and damages caused by armyworm larvae, whereas for the other characteristics the obtained results were not satisfactory.

Colletotrichum spp. cause anthracnose in various fruits post-harvest and are a particularly important problem in tropical and subtropical fruits. The disease in fruits of avocado, guava, papaya, mango and passion fruit has been reported to be caused by C. gloeosporioides, and in banana by C. musae. In subtropical and temperate crops such apple, grape, peach and kiwi, the disease is caused by C. acutatum. The variation in pathogenic, morphological, cultural and molecular characteristics of Brazilian isolates of Colletotrichum acutatum Simmonds and isolates from post-harvest decays of avocado, banana, guava, papaya, mango and passion fruit was evaluated. The fruits were inoculated with mycelium of C. acutatum, Colletotrichum spp. and C. musae on a disc of potato dextrose agar. The morphological, cultural and molecular characteristics studied were conidia morphology, colony growth at different temperatures, colony coloration and PCR with primers CaInt2 and ITS4 for C. acutatum and CgInt and ITS4 for C. gloeosporioides. C. acutatum was pathogenic to avocado, guava, papaya, mango and passion fruit, but it was not pathogenic to banana. The morphological, cultural and molecular studies indicated that the avocado, papaya, mango and passion fruit isolates were C. gloeosporioides. The natural guava isolate was identified as C. acutatum, which had not been found previously to produce anthracnose symptoms on guava in Brazil.
A model for optimal chemical control of leaf area damaged by fungi population - Parameter dependence

We present a model to study a fungi population submitted to chemical control, incorporating the fungicide application directly into the model. From that, we obtain an optimal control strategy that minimizes both the fungicide application (cost) and leaf area damaged by fungi population during the interval between the moment when the disease is detected (t = 0) and the time of harvest (t = t(f)). Initially, the parameters of the model are considered constant. Later, we consider the apparent infection rate depending on the time (and the temperature) and do some simulations to illustrate and to compare with the constant case.
