O objetivo deste trabalho foi estudar o controle genético da formação de embriões somáticos da cultivar IAS-5 de soja. O experimento foi conduzido em casa de vegetação, cultivando-se quatro plantas por vaso, sob fotoperíodo de 14 horas e temperatura em torno de 28°C. Efetuaram-se cruzamentos entre os parentais não-embriogênicos (cultivares IAC-6, Paraná e IAC-15) e embriogênico (cultivar IAS-5) e retrocruzamentos para obtenção das gerações F1, F2,RC1P1 e RC1P2. Cotilédones imaturos, com 4-6 mm de comprimento, derivados dos parentais das gerações F1, F2, RC1P1 e RC1P2 foram cultivados em placas de Petri contendo meio N10, por um período de 90 dias, em câmara de crescimento. Os embriões somáticos derivados da indução foram contados, e os números, usados para obtenção dos parâmetros genéticos. Os resultados obtidos mostraram que o caráter capacidade de produção de embriões somáticos da cultivar IAS-5 é de natureza quantitativa e controlado por, aproximadamente, 20 genes.

Background: Sugarcane is an increasingly economically and environmentally important C4 grass, used for the production of sugar and bioethanol, a low-carbon emission fuel. Sugarcane originated from crosses of Saccharum species and is noted for its unique capacity to accumulate high amounts of sucrose in its stems. Environmental stresses limit enormously sugarcane productivity worldwide. To investigate transcriptome changes in response to environmental inputs that alter yield we used cDNA microarrays to profile expression of 1,545 genes in plants submitted to drought, phosphate starvation, herbivory and N-2-fixing endophytic bacteria. We also investigated the response to phytohormones (abscisic acid and methyl jasmonate). The arrayed elements correspond mostly to genes involved in signal transduction, hormone biosynthesis, transcription factors, novel genes and genes corresponding to unknown proteins.Results: Adopting an outliers searching method 179 genes with strikingly different expression levels were identified as differentially expressed in at least one of the treatments analysed. Self Organizing Maps were used to cluster the expression profiles of 695 genes that showed a highly correlated expression pattern among replicates. The expression data for 22 genes was evaluated for 36 experimental data points by quantitative RT-PCR indicating a validation rate of 80.5% using three biological experimental replicates. The SUCAST Database was created that provides public access to the data described in this work, linked to tissue expression profiling and the SUCAST gene category and sequence analysis. The SUCAST database also includes a categorization of the sugarcane kinome based on a phylogenetic grouping that included 182 undefined kinases.Conclusion: An extensive study on the sugarcane transcriptome was performed. Sugarcane genes responsive to phytohormones and to challenges sugarcane commonly deals with in the field were identified. Additionally, the protein kinases were annotated based on a phylogenetic approach. The experimental design and statistical analysis applied proved robust to unravel genes associated with a diverse array of conditions attributing novel functions to previously unknown or undefined genes. The data consolidated in the SUCAST database resource can guide further studies and be useful for the development of improved sugarcane varieties.

Dados de rebanhos bovinos comerciais foram analisados com o objetivo de estimar as interações dos efeitos genéticos com o ambiente que podem influenciar a avaliação de características de crescimento em rebanhos de animais puros e cruzados. O conjunto de dados analisado foi obtido a partir de animais das raças Hereford, Nelore e seus cruzamentos. As características em estudo foram os pesos à desmama e ao sobreano dos animais. As análises estatísticas foram realizadas pelo método dos quadrados mínimos e o modelo proposto incluiu os efeitos de região, grupo de contemporâneos dentro de região, mês de nascimento e sexo do bezerro, os efeitos lineares e quadráticos para a idade do bezerro e idade da vaca ao parto, ambas analisadas dentro de sexo, e os efeitos de grupo genético e da interação grupo genético × região. de modo geral, o desempenho de todos os grupos genéticos foi influenciado pelo efeito de região. Além disso, observou-se tendência de que o aumento da proporção de genes zebuínos promoveu diminuição na diferença de desempenho entre as regiões. Todos os genótipos foram beneficiados no ambiente menos restritivo, o que indica a existência de interação genótipo-ambiente e comprova a importância de que sistemas de cruzamento sejam realizados de forma a manter a adaptação das matrizes e de seus produtos.

O objetivo do trabalho foi investigar a depressão endogâmica (DE) na mamoneira, espécie de reprodução sexuada mista. de uma população derivada da cultivar Guarani, amostraram-se 60 plantas-mãe. de cada uma foram obtidos três tipos de progênies: de autofecundação (AU), de cruzamentos obrigatórios (CR) e de polinização livre (PL). A produtividade de grãos das progênies for avaliada por meio de experimentos em blocos incompletos em dois locais. Houve forte interação de progênies x locais o que levou a obter estimativas dentro de cada local. Verificou-se ampla variação na depressão endogâmica, com médias de 6,7% e 13,4%, comparando-se as progênies AU com as PL. Verificou-se que a população tem alto potencial para selecionar linhagens promissoras. Foi baixa a freqüência de plantas-mãe gerando progênies com alta capacidade geral de combinação e baixa depressão endogâmica, simultaneamente. Seleção recorrente aumentará a ocorrência de genitoras associando essas duas propriedades, necessárias para obtenção de variedades sintéticas superiores.

The aim of this study to evaluate by means of quantitative traits, the genetic dissimilarity among sunflower cultivars in different locations, in addition to the agreement between the methods in order to extract lines for future crosses. There were eight sunflower hybrids grown in two locations in northwestern Rio Grande do Sul, mainly with soil type Oxisol. Multivariate methods were used to determine the genetic diversity, using the Mahalanobis distance. Although the different yield between locations and cultivars, methods of grouping agreed among them selves. To obtain segregating populations, regardless of location, the cultivate 'Olisun 5', demonstrated greater potential for hybridization, with major contributions through number of achenes per chapter and height of the insertion section.

In this paper, by using the Poincare compactification in R(3) we make a global analysis of the Lorenz system, including the complete description of its dynamic behavior on the sphere at infinity. Combining analytical and numerical techniques we show that for the parameter value b = 0 the system presents an infinite set of singularly degenerate heteroclinic cycles, which consist of invariant sets formed by a line of equilibria together with heteroclinic orbits connecting two of the equilibria. The dynamical consequences related to the existence of such cycles are discussed. In particular a possibly new mechanism behind the creation of Lorenz-like chaotic attractors, consisting of the change in the stability index of the saddle at the origin as the parameter b crosses the null value, is proposed. Based on the knowledge of this mechanism we have numerically found chaotic attractors for the Lorenz system in the case of small b > 0, so nearby the singularly degenerate heteroclinic cycles.

A imunidade na glândula mamária pode ser classificada, assim como em outros sistemas, em inata ou inespecífica e adaptativa ou específica. A imunidade inata é a defesa predominante durante os estágios iniciais da infecção. As respostas inespecíficas estão presentes no local da infecção ou são ativadas rapidamente por numerosos estímulos e não aumentam pela exposição repetida ao mesmo agente etiológico. O primeiro obstáculo enfrentado por um patógeno para adentrar o úbere é composto pela barreira formada pelo esfíncter do teto e pelo tampão de queratina formado pelo epitélio queratinizado. Uma vez que o microrganismo tenha atravessado o canal do teto e alcançado a cisterna mamária, passam a atuar diversos fatores solúveis e celulares. Dentre os fatores solúveis, estão presentes: lactoperoxidase, sistema complemento, citocinas, lactoferrina, lisozima e NAGase. As defesas celulares inespecíficas na glândula mamária são representadas pelos neutrófilos, pelos macrófagos e pelas células natural killer. Na medida em que esses mecanismos funcionam adequadamente, a maioria dos patógenos será rapidamente eliminada antes que o sistema imune específico seja ativado, sem resultar em alterações na quantidade ou qualidade do leite produzido. Uma melhor compreensão sobre os mecanismos de defesa da glândula mamária e suas alterações durante os períodos críticos da infecção é imprescindível para o desenvolvimento de métodos mais eficazes de profilaxia e controle da mastite, a principal doença dos ruminantes leiteiros. O presente estudo revisou os principais aspectos responsáveis pelo desenvolvimento da imunidade inata na glândula mamária bovina.

Growth hormone (GH), insulin-like growth factors 1 and 2 (IGF1 and IGF2) and their associated binding proteins and transmembrane receptors (GHR, IGF1R and IGF2R) play an important role in the physiology of mammalian growth. The objectives of the present study were to estimate the allele and genotype frequencies of microsatellite markers located in the 5'-regulatory region of the IGF1 and GHR genes in beef cattle belonging to different genetic groups and to determine effects of these markers on growth and carcass traits in these animals under an intensive production system. For this purpose, genotyping was performed on 384 bulls including 79 Nellore, 30 Canchim (5/8 Charolais + 3/8 Zebu) and 275 crossbred animals originating from crosses of Simmental (1/2 Simmental, n = 30) and Angus (1/2 Angus, n = 245) sires with Nellore females. The effects of substituting L allele for S allele of GHR microsatellite across Nellore, Canchim and 1/2 Angus were significant for weight gain and body weight (P < 0.05). The IGF1 microsatellite allele substitutions of 229 for 225 within Nellore group and of 225 for 229 within 1/2 Angus were not significant for any of the traits.

Molecular biology techniques are of help in genetic improvement since they permit the identification, mapping and analysis of polymorphisms of genes encoding proteins that act on metabolic pathways involved in economically interesting traits. The somatotrophic axis, which essentially consists of growth hormone releasing hormone (GHRH), growth hormone (GH), insulin-like growth factors I and II (IGF-I and IGF-II), and their associated binding proteins and receptors (GHRHR, GHR, IGF-IR and IGF-IIR), plays a key role in the metabolism and physiology of mammalian growth. The objectives of the present study were to estimate the allele and genotype frequencies of the IGF-I/SnaBI, IGF-IR/TaqI and GHRH/HaeIII gene polymorphisms in different genetic groups of beef cattle and to determine associations between these polymorphisms and growth and carcass traits. For this purpose, genotyping was performed on 79 Nellore animals, 30 Canchim (5/8 Charolais+3/8 Zebu) animals and 275 crossbred cattle originating from the crosses of Simmental (n=30) and Angus (n=245) sires with Nellore females. In the association studies, traits of interest were analyzed using the GLM procedure of SAS and least square means of the genotypes were compared by the Tukey test. Associations of IGF-I/SnaBI genotypes with body weight and subcutaneous backfat were significant (p < 0.05), and nearly significant for longissimus dorsi area (p=0.06), with the 1313 genotype being favorable compared to the AB genotype. No significant associations were observed between this polymorphism and weight gain or carcass yield (P > 0.05). The IGF-IR/TaqI and GHRH/HaeIII polymorphisms showed no association with production traits. (c) 2004 Elsevier B.V All rights reserved.

The knowledge of the buccal nerve anatomy is of fundamental importance not only for the anesthesia but also for a safe intervention in the retromolar area. The aim of this work was to study its trajectory, in the area where it is related to the anterior margin of the ramus of the mandible, therefore providing important data for a safe intervention in the region. In this study we used 10 hemi-heads from male and female adults, from different ethnic groups. They were fixed in formol, and belong to the Anatomy Laboratory at the Faculty of Dentistry in Araraquara UNESP. These hemi-heads were dissected by lateral access, preserving the buccal nerve in its trajectory related to the anterior margin of the ramus of the mandible until its penetration in the buccinator muscle. Next, we desinserted the masseter muscle so that all the ramus of the mandible were exposed. Then, the following measurements were carried out: from the base of the mandible until the buccal nerve and from the base of the mandible until the apices of the mandibular coronoid process. These measurements were accomplished with a Mitutoyo CD-6'' CS digital paquimeter. The following average values were obtained: 32.26 mm (to the left side) and 32.04 mm (to the right side), from the base of the mandible until the buccal nerve and 59.09 mm (to the left side) and 58.95 mm (to the right side) from the base of the mandible until the apices of the coronoid process. We have concluded that normally, the buccal nerve crosses the anterior margin of the ramus of the mandible in an area which is above the superior half of the ramus of the mandible and also that the interventions in the retromolar region do not offer great risks of injury in the buccal nerve.

Eighty-one lines of cauliflower (Brassica oleracea var. botrytis) from 12 populations used to produce commercial hybrids in Brazil were screened for polymorphism in the acid phosphatase system, in order to evaluate the usefulness of this marker for the determination of the parental contamination level in hybrid seeds. Little polymorphism was detected in the examined lines, but the system appeared to be very useful for hybrid identification, since the only condition required was polymorphism between the two parental lines. If the analyzed lines were used for hybrid production, 8.4% and 12.3% of the possible crosses would result in hybrids which can be positively identified using the APS-1 and B1 loci, respectively. If only one plant of each homozygous type (SS or FF) was analyzed in each population, 41% and 50% of the possible crosses would result in hybrids which can be positively identified using the APS-1 and B1 loci, respectively.
