Comparative mapping data on evolutionary conserved coding sequences and synteny maps between human and cattle are insufficient to define the extent and distribution of conserved segments between these two species, because the order of loci is often rearranged. A 5000-rad cattle whole-genome radiation hybrid (WG-RH) panel was constructed to provide high-resolution comparative maps and also to integrate linkage maps of microsatellites with evolutionary conserved genes and transcripts in a single ordered map. We used the WG-RH panel to construct radiation hybrid maps of bovine Chromosomes (Chrs) 15 and 29 (BTA15 and BTA29), integrating microsatellites from published linkage maps with selected genes. The comprehensive map of BTA15 consists of 24 markers. 13 of which were placed in the framework map. Eleven molecular markers compose the comprehensive map of BTA29. seven of which were placed in the framework map. We identified the homologous regions between bovine Chr 15 (BTA15) and human Chrs 5 and 11 (HSA5 and HSA11), as well as between BTA29 and HSA11, the present study demonstrates that WG-RH mapping is an efficient method for integrating multiple genetic maps into one map and for incorporating monomorphic Type I loci into ordered maps for comparison between species.

Heros Augusto Santos Lobo & Edvaldo Cesar Moretti: Tourism in Caves and the Conservation of the Speleological Heritage: The case of Serra da Bodoquena (Mato Grosso do Sul State, Brazil)The Serra da Bodoquena is the region in the state of Mato Grosso do Sul, Brazil in which the Municipality of Bonito is located. This municipality is the primary calling card for tourism in the state and is one of the most developed areas of ecotourism and speleotourism in the country The tourism there is entitled ecotourism, and is designed to be sustainable. The present case study focuses on the ecologically Sustainable aspects of the spleotourism practiced there, especially the proposals for tourist carrying capacity adopted. The results and discussion provide Suggestions for the adoption of a different formulation of carrying capacity focusing on both operational and quantitative aspects. Ecologically Sustainable speleotourism in the Serra da Bodoquena should be possible as long as new proposals limiting visitation are adopted which conform to technical environmental management procedures and consider the interests of local stakeholders.

1. This study aimed to link basic ethnobiological research on local ecological knowledge (LEK) to the conservation of Brazilian streams, based on two case studies: original data on LEK of fishermen about freshwater fish in the Negro River, Amazon, and previously published data about LEK of farmers on the ecological relationship between forest and streams in the Macabuzinho catchment, Atlantic Forest.2. Information was obtained from fishermen through interviews using standard questionnaires containing open-ended questions. Informants for interview were selected either following some defined criteria or applying the 'snowball' method.3. Fishermen's LEK about the diets and habitats of 14 fish species in the Negro River provided new biological information on plant species that are eaten by fish, in addition to confirming some ecological patterns from the biological literature, such as dependence of fish on forests as food sources.4. In the Atlantic Forest, a comparison between farmers' LEK and a rapid stream assessment in the farmers' properties indicated that farmers tended to overestimate the ecological integrity of their streams. Farmers recognized at least 11 forest attributes that correspond to the scientific concept of ecosystem services. Such information may be useful to promote or enhance dialogue among farmers, scientists and managers.5. These results may contribute to the devising of ecosystem management measures in the Negro River, aimed to conserve both rivers and their associated floodplain forests, involving local fishermen. In the Atlantic Forest, we proposed some initiatives, such as to allow direct economic use of their forests to conciliate conflicting perceptions of farmers about ecological benefits versus economic losses from reforestation. Despite their cultural, environmental and geographical differences, the two study cases are complementary and cost-effective and promising approaches to including LEK in the design of ecological research. Ant communities are sensitive to fragmentation responding through: extinction of specialist species compared with large habitats, invasion of generalist species, changes in community composition and richness of species. Because of these characteristics, this review aims at examining the main risks that ants have been facing to maintain their communities, as well as to report current conservation practices in Brazil and in the world.

Biodiversity is organised into complex ecological networks of interacting species in local ecosystems, but our knowledge about the effects of habitat fragmentation on such systems remains limited. We consider the effects of this key driver of both local and global change on both mutualistic and antagonistic systems at different levels of biological organisation and spatiotemporal scales.There is a complex interplay of patterns and processes related to the variation and influence of spatial, temporal and biotic drivers in ecological networks. Species traits (e.g. body size, dispersal ability) play an important role in determining how networks respond to fragment size and isolation, edge shape and permeability, and the quality of the surrounding landscape matrix. Furthermore, the perception of spatial scale (e.g. environmental grain) and temporal effects (time lags, extinction debts) can differ markedly among species, network modules and trophic levels, highlighting the need to develop a more integrated perspective that considers not just nodes, but the structural role and strength of species interactions (e.g. as hubs, spatial couplers and determinants of connectance, nestedness and modularity) in response to habitat fragmentation.Many challenges remain for improving our understanding: the likely importance of specialisation, functional redundancy and trait matching has been largely overlooked. The potentially critical effects of apex consumers, abundant species and supergeneralists on network changes and evolutionary dynamics also need to be addressed in future research. Ultimately, spatial and ecological networks need to be combined to explore the effects of dispersal, colonisation, extinction and habitat fragmentation on network structure and coevolutionary dynamics. Finally, we need to embed network approaches more explicitly within applied ecology in general, because they offer great potential for improving on the current species-based or habitat-centric approaches to our management and conservation of biodiversity in the face of environmental change.

The jacutinga Pipile jacutinga was formerly one of the most abundant game bird cracids in the Atlantic forest of Brazil. Nowadays this species is vulnerable to extinction due to hunting and habitat loss. The ecology of the jacutinga was studied at Parque Estadual Intervales, Sao Paulo, Brazil from October 1993 to December 1995 and in adjacent areas. Jacutingas were observed to feed mainly on the sugar-rich fruit of 41 species. We recorded a low index of abundance for the jacutinga (0.018) or c.1.7 birds/km2 at Intervales, one of the best protected areas within their range. Surveys carried out in the Atlantic forest of Sao Paulo found jacutinga populations in 14 localities. Probably < 1500 birds survive in the best protected areas. The species' stronghold in southeastern Brazil is in the mountains of Serra de Paranapiacaba, an area protected by several parks suffering from hunting and palm heart harvesting and threatened by a hydroelectric project.

It was aimed to extend the postharvest conservation of 'Tommy Atkins' mango fruits harvested in break maturity stage. Fruits were submitted at the following treatments: hot water treatment (55°C for 5 minutes) and benomyl 1,000 mg.L-1; irradiation with 0,8 or 1,0 kGy; irradiation associated at carnaúba wax; and control. The fruits were stored at 10°C and 85 - 90%RH during 21 days, and then removed to ambient temperature (25,7±0,7°C and 87,1±2,2%RH). Through the storage time, the evolution of fresh weight, color, rottenness, total soluble solids (TSS), total titratable acidity (TTA), and TSS/TTA ratio were measured. 'Tommy Atkins' mango fruits can have shelf life notably increased, when they were submitted to hot water treatment (55°C for 5 minutes) or γ radiation (0,8 and 1,0 kGy), associated with carnaúba wax application, before cold storage. These treatments increased the fruit resistance at refrigerated storage, and improved shelflife after transferring to ambient temperature.

Natural environments have been worldwide affected by the growing impact of anthropogenic actions that promote the reduction or the extinction of several vertebrate species. Aquatic ecosystems represent one of the most affected environments and many fish species and/or populations have been increasingly fragmented distributed due to habitat degradation, predatory fishing, introduction of exotic species, river sedimentation, deforestation, pollution, reduction of food resource, and construction of hydroelectric dams. Actually, more than 150 Brazilian fish species, including freshwater, estuary and coastal species, can be considered threatened. Information on the diversity, conservation biology and population analysis on threatened species or populations, with several DNA markers, can be extremely useful for the success of fish species-recovery and maintenance programs. Although DNA analysis in Neotropical fish species are just beginning, they tend to increase with the widespread attention to the use of molecular approaches to minimize problems related to the risk of extinction. The accumulation of information on biology and pattern of genetic variation of fish species, associated with ecological and demographic data, and also education and respect to the nature, constitutes a crucial task to develop efficient conservation strategies in order to preserve the genetic diversity in aquatic environments.

An integrated and interdisciplinary research programme with native medicinal plants from tropical forests has been performed in order to obtain new forest products for sustainable use in regional markets vis-à-vis ecosystem conservation. For the success of this programme ethnopharmacological studies are very important with respect to (i) identification of useful plants including medicinal and aromatic species; (ii) recuperation and preservation of traditional knowledge about native plants; and (iii) identification of potential plants with economic value. The plants are selected with a view to evaluate efficacy and safety (pharmacological and toxicological studies), and phytochemical profile and quality control (phytochemical and chromatographic characterization). These studies are very important to add value to plant products and also to mitigate unscrupulous exploitation of medicinal plants by local communities, since multiple use of plants represents an excellent strategy for sustaining the tropical ecosystem through ex situ and in situ conservation. Thus, conservation of tropical resources is possible in conjunction with improvements in the quality of life of the traditional communities and production of new products with therapeutic, cosmetic and 'cosmeceutic' value. © NIAB 2005.

Spatial patterns in assemblage structures are generated by ecological processes that occur on multiple scales. Identifying these processes is important for the prediction of impact, for restoration and for conservation of biodiversity. This study used a hierarchical sampling design to quantify variations in assemblage structures of Brazilian estuarine fish across 2 spatial scales and to reveal the ecological processes underlying the patterns observed. Eight areas separated by 0.7 to 25 km (local scale) were sampled in 5 estuaries separated by 970 to 6000 km (regional scale) along the coast, encompassing both tropical and subtropical regions. The assemblage structure varied significantly in terms of relative biomass and presence/absence of species on both scales, but the regional variation was greater than the local variation for either dataset. However, the 5 estuaries sampled segregated into 2 major groups largely congruent with the Brazilian and Argentinian biogeographic provinces. Three environmental variables (mean temperature of the coldest month, mangrove area and mean annual precipitation) and distance between estuaries explained 44.8 and 16.3%, respectively, of the regional-scale variability in the species relative biomass. At the local scale, the importance of environmental predictors for the spatial structure of the assemblages differed between estuarine systems. Overall, these results support the idea that on a regional scale, the composition of fish assemblages is simultaneously determined by environmental filters and species dispersal capacity, while on a local scale, the effect of environmental factors should vary depending on estuary-specific physical and hydrological characteristics © 2013 Inter-Research.

Dendrobium is a large genus in the family Orchidaceae that exhibits vast diversity in floral characteristics, which is of considerable importance to orchid breeders, biotechnologists and collectors. Native species have high value as a result of their medicinal properties, while their hybrids are important as ornamental commodities, either as cut flowers or potted plants and are thus veritable industrial crops. Thus, preservation of Dendrobium germplasm is valuable for species conservation, breeding programs and the floriculture industry. Cryopreservation represents the only safe, efficient and cost-effective long-term storage option to facilitate the conservation of genetic resources of plant species. This review highlights 16 years of literature related to the preservation of Dendrobium germplasm and comprises the most comprehensive assessment of thorough studies performed to date, which shows reliable and reproducible results. Air-drying, encapsulation-dehydration, encapsulation-vitrification, vitrification and droplet-vitrification are the current cryopreservation methodologies that have been used to cryopreserve Dendrobium germplasm. Mature seeds, pollen, protoplasts, shoot primordia, protocorms and somatic embryos or protocorm-like bodies (PLBs) have been cryopreserved with different levels of success. Encapsulation-vitrification and encapsulation-dehydration are the most used protocol, while PLBs represent the main explant explored.

O Pontal do Paranapanema, extremo oeste do Estado de São Paulo, é marcado pela recente devastação florestal, e ações visando à recuperação de áreas onde outrora havia cobertura florestal tornam-se indispensáveis para a conservação da biodiversidade na região, principalmente quando se trata de áreas de Reserva Legal ou Preservação Permanente. Diante do exposto, esta pesquisa teve como objetivo analisar a dinâmica inicial da composição florística em uma área de Reserva Legal restaurada. O trabalho foi realizado no assentamento Nova Esperança, Município de Euclides da Cunha Paulista, em uma área de 26,75 ha. O levantamento das espécies foi realizado pelo método de parcelas (16 parcelas de 20 x 30 m) em duas fases distintas do plantio, e as análises foram feitas utilizando-se o software Fitopac. Os resultados evidenciaram diminuição da biodiversidade pela alta mortalidade de espécies, levando à conclusão de que em projetos de restauração as intervenções são necessárias para a manutenção do plantio mesmo em sua fase inicial.
