Cells from rat bone marrow exhibit the proliferation-differentiation sequence of osteoblasts, form mineralized extracellular matrix in vitro and release alkaline phosphatase into the medium. Membrane-bound alkaline phosphatase was obtained by method that is easy to reproduce, simpler and fast when compared with the method used to obtain the enzyme from rat osseous plate. The membrane-bound alkaline phosphatase from cultures of rat bone marrow cells has a MWr of about 120 kDa and specific PNPP activity of 1200 U/tng. The ecto-enzyme is anchored to the plasma membrane by the GPI anchor and can be released by PIPLC (selective treatment) or polidocanol (0.2 mg/mL protein and 1% (w/v) detergent). The apparent optimum pH for PNPP hydrolysis by the enzyme was pH 10. This fraction hydrolyzes ATP (240 U/mg), ADP (350 U/ mg), glucose 1-phosphate (1100 U/mg), glucose 6-phosphate (340 Wing), fructose 6-phosphate (460 U/mg), pyrophosphate (330 U/mg) and (3glycerophosphate (600 U/mg). Cooperative effects were observed for the hydrolysis of PPi and beta-glycerophosphate. PNPPase activity was inhibited by 0.1 mM vanadate (46%), 0.1 mM ZnCl2 (68%), 1 mM levamisole (66%), 1 mM arsenate (44%), 10 mM phosphate (21%) and 1 mM theophylline (72%). We report the biochemical characterization of membrane-bound alkaline phosphatase obtained from rat bone marrow cells cultures, using a method that is simple, rapid and easy to reproduce. Its properties are compared with those of rat osseous plate enzyme and revealed that the alkaline phosphatase obtained has some kinetics and structural behaviors with higher levels of enzymatic activity, facilitating the comprehension of the mineralization process and its function. Foram conduzidos um ensaio de metabolismo para determinar o valor nutricional do milho termicamente processado (MP) e não-processado (MNP) e outro de desempenho para comparar o uso desses alimentos em dietas pré-iniciais para frangos de corte. Não foram observadas diferenças entre a digestibilidade da matéria seca (MS), do extrato etéreo, da proteína bruta e do amido e valores de energia metabolizável aparente dos tipos de milho avaliados. A energia metabolizável aparente corrigida do MP (3.537kcal/kg de MS) foi maior (P<0,05) que a determinada para o MNP (3.411kcal/kg de MS). No ensaio de desempenho, os tratamentos foram: T1- dieta formulada com MNP; T2 - dieta formulada com MP; e T3 - dieta T1, com substituição isométrica do MNP por MP. Na primeira semana, as aves alimentadas com MP (T2) apresentaram maior consumo (P<0,05) e pior conversão (P<0,05) em relação às alimentadas com MNP (T1). A substituição isométrica do MNP por MP não influenciou no desempenho das aves. No período de 1 a 42 dias, os tratamentos não influenciaram o desempenho dos frangos.

The objective of this work was to evaluate the effect of the physical form and quantity of pre-starter diet on the broiler chicks performance. In the first trial, 684 day old males were distributed in a randomized design of three treatments with six replicates of 38 birds. The physical forms of pre-starter diets tested were: mash diet; crumb diet and pellet diet. Another was conducted with 912 day old male distributed in a randomized design of three treatments with eight replicates of 38 birds. The treatments consisted in feeding the broiler chickens with 150 g/bird; 200 g/bird and 250 g/bird. After that, all the birds were fed with the same feed program. In the first week and at 21 days old the birds fed with mash pre-starter diet had body weight gain lower than those fed crumbed and pelleted diets. The variables studied were not affected by the quantity of diets. In conclusion, the physical form of pre-starter diet was important for initial performance and the quantity of pre-starter tip to 150 g/bird did not improve the development of broiler chicks.

O objetivo deste trabalho foi avaliar o efeito da formulação de rações para frangos de corte, com a utilização de valores de energia metabolizável (EM) dos alimentos determinados por diferentes métodos. O delineamento experimental foi inteiramente casualizado, com quatro tratamentos, que consistiram na formulação de rações baseadas na: em aparente corrigida e em aparente, ambas determinadas pelo método da coleta total com pintos; em aparente corrigida, determinada pelo método da coleta total com galos; e em verdadeira corrigida, determinada pelo método da alimentação forçada com galos. Na fase inicial, foi obtido maior consumo de ração, ganho de peso e melhor conversão alimentar com a formulação baseada na em aparente corrigida (pintos). Durante a fase final, a formulação com em verdadeira corrigida (galos) promoveu maior ganho de peso e melhor conversão alimentar. No período de 1 a 49 dias, as aves alimentadas com as rações formuladas com em aparente corrigida (pintos) tiveram melhor desempenho. As características de carcaça não foram influenciadas pelos tratamentos. Até os 21 dias de idade, deve-se considerar os valores de em aparente corrigida (pintos), para a formulação das rações de frangos de corte; após essa idade, deve-se considerar os valores de em aparente corrigida (galos) ou em verdadeira corrigida (galos).

Esta pesquisa foi realizada com o objetivo de determinar o efeito da inclusão de aditivo químico e da inoculação de bactérias homo e heterofermentativas sobre a estabilidade aeróbia de silagens de capim-marandu e da ração total. Foram conduzidos três experimentos para avaliação do benzoato de sódio e de dois inoculantes, um contendo Lactobacillus plantarum + Propionibacterium e o segundo Lactobacillus buchneri. Após 60 dias de fermentação, os silos foram abertos e as silagens e a ração total (RT) contendo silagens de capim-marandu foram colocadas em caixas de isopor e transferidas para câmara climática, a 25 ± 1ºC, para determinação das variações de temperatura na ração total e na silagem, das recuperações de MS e das alterações no pH da silagem. O delineamento experimental foi o inteiramente casualizado, em esquema de parcelas subdivididas. Houve perdas de MS e elevação dos teores de pH quando as silagens foram colocadas em condições de aerobiose. A temperatura das silagens e da RT teve discreto aumento durante os seis dias de aeração. O uso de bactérias ou de benzoato de sódio não influenciou a estabilidade aeróbia das silagens.

Chronic hepatitis C virus (HCV) infection is a worldwide health problem that may evolve to cirrhosis and hepatocellular carcinoma. Incompletely understood immune system mechanisms have been associated with impaired viral clearance. The nonclassical class I human leukocyte antigen G (HLA-G) molecule may downregulate immune system cell functions exhibiting well-recognized tolerogenic properties. HCV genotype was analyzed in chronic HCV-infected patients. Because HLA-G expression may be induced by certain viruses, we evaluated the presence of HLA-G in the liver microenvironment obtained from 89 biopsies of patients harboring chronic HCV infection and stratified according to clinical and histopathological features. Overall, data indicated that HCV genotype 1 was predominant, especially subgenotype 1a, with a prevalence of 87%. HLA-G expression was observed in 45(51%) liver specimens, and it was more frequent in milder stages of chronic hepatitis (67.4%) than in moderate (27.8%; p = 0.009) and severe (36.0%; p = 0.021) stages of the disease. Altogether, these results suggest that the expression of HLA-G in the context of HCV is a complex process modulated by many factors, which may contribute to an immunologic environment favoring viral persistence. However, because the milder forms predominantly expressed HLA-G, a protective role of this molecule may not be excluded. (C) 2012 American Society for Histocompatibility and Immunogenetics. Soluble, ionically bound peroxidase (POD) and polyphenoloxidase (PPO) were extracted from the pulp of peach fruit during ripening at 20degreesC Ionically bound form was purified 6.1 -fold by DEAE-cellulose and Sephadex G-100 chromatography. The purified enzyme showed only one peak of activity on Sephadex G-100 and PAGE revealed that the enzyme was purified by the procedures adopted. The purified enzyme showed a molecular weight of 29000 Da, maximum activity at pH 5.0 and at 40degreesC the calculated apparent activation energy (Ea) for the reaction was 10.04 kcal/mol. The enzyme was heat-labile in the temperature range of 60 to 75degreesC with a fast inactivation at 75degreesC Measurement of residual activity showed a stabilizing effect of sucrose at various temperature/sugar concentrations (0, 10, 20 %, w/w), with an activation energy (Ea) for inactivation increasing with sucrose concentration from 0 to 20% (w/w). The Km and V-max values were 9.35 and 15.38 mM for O-dianisidine and H2O2, respectively. The bound enzyme was inhibited competitively by (.)ferulic, caffeic and protocatechuic acids with different values of Ki,. L-cysteine, p-coumaric and indolacetic acid and Fe++ also inhibited the enzyme but at a lower grade. N-ethylmaleimide and p-CMB were not effective to inhibit the enzyme demonstrating the non-essentiality of SH groups.

A finite-difference scheme is used to calculate bound electronic states of an electron in a hydrogen atom subject to a magnetic field. The numerical results are in good agreement with exact results, in the absence of the magnetic field, and with a two-parameters variational calculation, when the magnetic field is applied.

The Dirac equation is solved for a pseudoscalar Coulomb potential in a two-dimensional world. An infinite sequence of bounded solutions are obtained. These results are in sharp contrast with those ones obtained in 3 + 1 dimensions where no bound-state solutions are found. Next the general two-dimensional problem for pseudoscalar power-law potentials is addressed consenting us to conclude that a nonsingular potential leads to bounded solutions. The behaviour of the upper and lower components of the Dirac spinor for a confining linear potential nonconserving- as well as conserving-parity, even if the potential is unbounded from below, is discussed in some detail. The Klein - Gordon and the Dirac equations with vector and scalar potentials are investigated under a more general condition, V(v) + V(s) = constant. These intrinsically relativistic and isospectral problems are solved in the case of squared hyperbolic potential functions and bound states for either particles or antiparticles are found. The eigenvalues and eigenfuntions are discussed in some detail and the effective Compton wavelength is revealed to be an important physical quantity. It is revealed that a boson is better localized than a fermion when they have the same mass and are subjected to the same potentials.
