OBJETIVO: avaliar a influência do alcoolismo crônico experimental na morfologia e biomecânica da artéria aorta de ratos. MÉTODOS:ratos Wistar foram divididos em dois grupos de 22 animais. Grupo alcoolista: os ratos receberam água com concentração crescente de etanol e ração ad libitum durante 6 meses. Grupo controle: os animais receberam água e ração ad libitum pelo mesmo período. Os animais foram sacrificados com overdose de pentobarbital e as aortas abdominais foram excisadas para realização de histologia (hematoxilina-eosina, tricrômio de Masson, Calleja, Picrosirius red), histomorfometria e avaliação biomecânica (carga máxima, alongamento na carga máxima, limite de proporcionalidade, alongamento no limite de proporcionalidade e coeficiente de rigidez). RESULTADOS: A histologia e a histomorfometria não mostraram diferenças significantes na morfologia da aorta em ambos os grupos. A avaliação biomecânica mostrou aumento do alongamento no limite de proporcionalidade no grupo alcoolista (p<0.05). CONCLUSÃO: o alcoolismo crônico experimental não provocou alterações morfológicas na parede da aorta, mas causou aumento da sua elasticidade, sem modificar as outras propriedades mecânicas avaliadas.

O objetivo deste trabalho foi verificar se a dose de ranitidina 2mg kg-1, aplicada via intravenosa, causa êmese ou hipotensão em cães saudáveis. Foram selecionados 10 cães da raça Retriever do Labrador, 10 da raça Beagle e 10 cães sem raça definida, sendo cinco animais de cada sexo. Os animais foram submetidos ao exame clínico e à avaliação da pressão sanguínea antes da aplicação do fármaco e também 10 minutos e quatro horas após a administração deste. Após a aplicação, observou-se que 13,3% dos animais apresentaram-se normais; 6,7% dos cães apresentaram apatia; 50% dos animais apresentaram salivação e 30% apresentaram apatia, salivação, mímica de vômito ou êmese. Não houve diminuição significativa da pressão arterial após a administração do fármaco. Conclui-se que o uso de ranitidina na dose terapêutica, aplicada via intravenosa, pode provocar apatia, salivação, mímica de vômito e êmese.

Dois experimentos foram realizados para avaliar a digestibilidade aparente da EB, MS e PB em alimentos energéticos e protéicos utilizados para cães adultos. Foram utilizados quatro cães adultos (13,1 ± 2,0 kg), dois machos e duas fêmeas, sem raça definida, na avaliação de cada alimento. Os animais receberam a mesma quantidade de ração por unidade de peso metabólico. No experimento 1, foi determinado o valor nutritivo dos alimentos energéticos e, no experimento 2, os coeficientes de digestibilidade dos alimentos protéicos. Os coeficientes de digestibilidade da EB do milho extrusado (ME), do milho gelatinizado (MG), da gordura de coco e do óleo de soja, em dois níveis de inclusão (OS1 e OS2), e da gordura suína foram, respectivamente, 85,1; 84,4; 92,5; 92,1; 96,2 e 98,6%. Os coeficientes de digestibilidade da MS e PB do ME e MG foram, respectivamente, 84,2 e 65,3 e 84,5 e 65,0%. Na soja integral extrusada e nas farinhas de carne, de carne extrusada, de vísceras, de vísceras extrusada, de peixe extrusada e de pena extrusada, foram obtidos, respectivamente, os seguintes coeficientes de digestibilidade: 80,0; 73,3; 80,7; 87,6; 91,2; 91,1 e 79,8% da EB; 80,0; 68,4; 87,8; 86,7; 88,1; 85,2 e 76,0% da MS; e 83,7; 74,7; 82,3; 88,0; 88,9; 91,9 e 82,3% da PB.

The fragmentation pattern of a homologous series of piperidine alkaloids isolated from S. spectabilis was investigated using electrospray ionization tandem mass spectrometry (ESI-MS/MS). The ESI-MS and ESI-MS/MS analyses of EtOH extracts and fractions from flowers and fruits of S. spectabilis allowed to elucidate the structures of four new compounds. The identification of these co-metabolites, based on the fragmentation patterns of previously isolated compounds, and further confirmed by accurate mass spectrometry defines this technique as a powerful tool to determine the metabolomic profile of species which has pharmacological importance. ©2005 Sociedade Brasileira de Química.

Objective: To evaluate the results of the kangaroo mother method in Brazil. Methods: A prospective cohort study comparing 16 units that have or do not have the second phase of the kangaroo mother method: eight were national centers of excellence for the kangaroo mother method (study group) and eight were part of the Brazilian Neonatal Research Network (control group). A total of 985 newborn infants with birth weights of 500 to 1,749 g were enrolled. Multivariate analyses employedmultiple linear regression and Poisson regression with robust adjustment. Results: The adjusted analysis (controlled for birth weight, gestational age, Score for Neonatal Acute Physiology Perinatal Extension II, Neonatal Therapeutic Intervention Scoring System, and maternal age and educational level) demonstrated that mean length of hospital stay (p = 0.14) and intercurrent clinical conditions in the intermediate or kangaroo unit were equal for both groups. Weight (p = 0.012), length (p = 0.039) and head circumference (p = 0.006) at 36 weeks' corrected gestational age were all lower at the kangaroo units. The kangaroo units exhibited superior performance in relation to exclusive breastfeeding at discharge (69.2 vs. 23.8%, p=0.022). Conclusions: The evidence suggests that the humanization strategy adopted by the Brazilian Ministry of Health is a safe alternative to conventional treatment and a good strategy for promoting breastfeeding. By remote sensing, geodatabase digital processing, information and expeditions to Capivara's sub-basin, was possible to identify the changes in the landscape mainly the expansion of eucalyptus, sugar-cane, and orange trees, being the last two, mainly on the Periferic Depression of Basaltic Cuesta. A significant portion of the soil on this geological area is formed from sandstones, providing high permeability to them, making them important places to groundwater recharge areas as sensible to contamination by pesticides. Throughout last decade was observed that the native vegetation fragments stabilization, keeping a reason of 26.5% on the land used between 2000 and 2010. The pasture decrease being substituted by eucalyptus, sugar-cane and orange trees call attention for the changes in the agribusiness model demanded by the current economic and social necessity. Pasture decrease followed by erosions decrease on sub-basin is evidence that these two aspects are strongly related.

In the Nilo Coelho irrigation scheme, Brazil, the natural vegetation has been replaced by irrigated agriculture, bringing importance for the quantification of the effects on the energy exchanges between the mixed vegetated surfaces and the lower atmosphere. Landsat satellite images and agro-meteorological stations from 1992 to 2011 were used together, for modelling these exchanges. Surface albedo (α0), NDVI and surface temperature (T0) were the basic remote sensing retrieving parameters necessary to calculate the latent heat flux (λE) and the surface resistance to evapotranspiration (rs) on a large scale. The daily net radiation (Rn) was obtained from α0, air temperature (Ta) and short-wave transmissivity (τsw) throughout the slob equation, allowing the quantification of the daily sensible heat flux (H) by residual in the energy balance equation. With a threshold value for rs, it was possible to separate the energy fluxes from crops and natural vegetation. The averaged fractions of Rn partitioned as H and λE, were in average 39 and 67%, respectively. It was observed an increase of the energy used for the evapotranspiration process inside irrigated areas from 51% in 1992 to 80% in 2011, with the ratio λE/Rn presenting an increase of 3 % per year. The tools and models applied in the current research, can subsidize the monitoring of the coupled climate and land use changes effects in irrigation perimeters, being valuable when aiming the sustainability of the irrigated agriculture in the future, avoiding conflicts among different water users. © 2012 SPIE.

The municipality of Petrolina, located in the semi-arid region of Brazil, is highlighted as an important agricultural growing region, however the irrigated areas have cleared natural vegetation inducing a loss of biodiversity. To analyze the contrast between these two ecosystems the large scale values of biomass production (BIO), evapotranspiration (ET) and water productivity (WP) were quantified. Monteithś equation was applied for estimating the absorbed photosynthetically active radiation (APAR), while the new SAFER (Simple Algorithm For Evapotranspiration Retrieving) algorithm was used to retrieve ET. The water productivity (WP) was analysed by the ratio of BIO by ET at monthly time scale with four bands of MODIS satellite images together with agrometeorological data for the year of 2011. The period with the highest water productivity values were from March to April in the rainy period for both irrigated and not irrigated conditions. However the largest ET rates were in November for irrigated crops and April for natural vegetation. More uniformity of the vegetation and water variables occurs in natural vegetation, evidenced by the lower values of standard deviation when comparing to irrigated crops, due to the different crop stages, cultural and irrigation managements. The models applied with MODIS satellite images on a large scale are considered to be suitable for water productivity assessments and for quantifying the effects of increasing irrigated areas over natural vegetation on regional water consumption in situations of quick changing land use pattern. © 2012 SPIE.

Background: Smoking is the most relevant environmental factor that affects the development of aortic aneurysm. Smokers have elevated levels of elastase activity in the arterial wall, which leads to weakening of the aorta. The aim of this study was to verify whether cigarette smoke exposure itself is capable of altering the aortic wall. Methods: Forty-eight Wistar rats were divided into 2-, 4-, and 6-month experimental periods and into 2 groups: smokers (submitted to smoke exposure at a rate of 40 cigarettes/day) and nonsmokers. At the end of the experimental periods, the aortas were removed and cross-sectioned to obtain histologic specimens for light microscopic and morphometric analyses. The remaining longitudinal segments were stretched to rupture and mechanical parameters were determined. Results: A degenerative process (i.e., a reduction in elastic fibers, the loss of lamellar arrangement, and a reduction of smooth muscle cells) was observed, and this effect was proportional in intensity to the period of tobacco exposure. We observed a progressive reduction in the yield point of the thoracic aorta over time (P < 0.05). There was a decrease in stiffness (P < 0.05) and in failure load (P < 0.05) at 6 months in the abdominal aorta of rats in the smoking group. Conclusions: Chronic exposure to tobacco smoke can affect the mechanical properties of the aorta and can also provoke substantial structural changes of the arterial wall. © 2013 Elsevier Inc. BackgroundEvidence is emerging that highlights the far-reaching consequences of a high-fat diet (HFD) on kidney morphology and function disorders.MethodsThe present study was performed on 3-, 5-, 7- and 9-week-old HFD female rats compared with the appropriate gender and age-matched animals. We evaluated the kidney expression of angiotensin type II receptor and fibrotic and epithelial-to-mesenchymal transition (EMT) markers, by immunoblotting and immunohistochemical and histological techniques, in parallel with kidney function.ResultsIn the current study, the time-course HFD-treated group showed, by immunoblotting and immunohistochemical analysis, an early time-course increase in the expression of transforming growth factor β-1 (TGFβ-1) in the entire kidney of HFD-treated rats, compared with that observed in the control group. Simultaneously, the study shows a transient increase in the expression of ZEB2 in the HFD whole kidney accompanied by a fall in the E-cadherin expression and increased collagen and fibronectin deposition. A pronounced decrease in fractional urinary sodium excretion was also demonstrated in the long-term HFD-treated rats. The decreased FENa + was accompanied by a fall in FEPNa + and FEPP Na +, which occurred in association with significantly decreased CCr and, certainly on the sodium-filtered load. The reduction in the glomerular filtration rate (GFR) occurred in parallel to proteinuria and glomerular desmin overexpression.ConclusionsThe results of the current study suggest that podocyte injury in parallel with observed proteinuria and evidence of EMT transformation are associated with long-term loss of kidney function and renal sodium and water retention. © 2013 The Author. SOCS3 is an inducible endogenous negative regulator of JAK/STAT pathway, which is relevant in inflammatory conditions. We used a model of LPS-induced periodontal disease in rats to correlate SOCS3 expression with the inflammatory status. In vitro we used a murine macrophage cell line to assess the physical interaction between SOCS3 and STAT3 by coimmunoprecipitation. 30 ug of LPS from Escherichia coli were injected in the gingival tissues on the palatal aspect of first molars of the animals 3x/week for up to 4 weeks. Control animals were injected with the vehicle (PBS). The rats were sacrificed at 7, 15, and 30 days. Inflammation and gene expression were assessed by stereometric analysis, immunohistochemistry, RT-qPCR, and western blot. LPS injections increased inflammation, paralleled by an upregulation of SOCS3, of the proinflammatory cytokines IL-1β, IL-6, and TNF-and increased phosphorylation of STAT3 and p38 MAPK. SOCS3 expression accompanied the severity of inflammation and the expression of proinflammatory cytokines, as well as the activation status of STAT3 and p38 MAPK. LPS stimulation in a macrophage cell line in vitro induced transient STAT3 activation, which was inversely correlated with a dynamic physical interaction with SOCS3, suggesting that this may be a mechanism for SOCS3 regulatory function. © 2013 João Antônio Chaves de Souza et al.
