A trial was carried out to evaluate efficacy of different herbicides and spray volumes when sprayed to ryegrass for chemical management and control of remaining seeds. The treatments were: three formulations of glyphosate at 1080 g ha(-1) (Original Round up, Round up Transorb, WG Round Up), paraquat + diuron at 300+500 and 150+250 g ha(-1) and glufosinate ammonium at 400 and 600 g ha(-1). All treatments were used in spray volumes of 200 and 400 l ha(-1). The effectiveness of the desiccation of ryegrass plants was assessed at 1, 3, 7, 14 and 21 days after application. At the last evaluation samples were collected for dry mass production analysis, and also panicles with seeds for germination test. Glyphosate in different formulations and volumes and glufosinate ammonium at 600 g ha(-1) when sprayed with into 400 l ha(-1) provided the best controls regarding ryegrass chemical management. Treatments with gliphosate (Roundup Original) and paraquat + diuron (300 +150 g ha(-1)) in volumes of 400 and 200 L ha(-1), respectively, were the ones that showed the lowest values of dry matter, differing from the control. All the ryegrass seeds were killed by treatments with herbicides (paraquat + diuron) at 500 + 250 g ha(-1) and glufosinate-ammonium at 600 g ha(-1) when used 200 l ha(-1).

The gene encoding glycogen synthase in Neurospora crassa (gsn) is transcriptionally down-regulated when mycelium is exposed to a heat shock from 30 to 45 degrees C. The gsn promoter has one stress response element (STRE) motif that is specifically bound by heat shock activated nuclear proteins. In this work, we used biochemical approaches together with mass spectrometric analysis to identify the proteins that bind to the STRE motif and could participate in the gsn transcription regulation during heat shock. Crude nuclear extract of heat-shocked mycelium was prepared and fractionated by affinity chromatography. The fractions exhibiting DNA-binding activity were identified by electrophoretic mobility shift assay (EMSA) using as probe a DNA fragment containing the STRE motif DNA-protein binding activity was confirmed by Southwestern analysis. The molecular mass (MM) of proteins was estimated by fractionating the crude nuclear extract by SDS-PAGE followed by EMSA analysis of the proteins corresponding to different MM intervals. Binding activity was detected at the 30-50 MM kDa interval. Fractionation of the crude nuclear proteins by IEF followed by EMSA analysis led to the identification of two active fractions belonging to the pIs intervals 3.54-4.08 and 6.77-7.31. The proteins comprising the MM and pI intervals previously identified were excised from a 2-DE gel, and subjected to mass spectrometric analysis (MALDI-TOF/TOF) after tryptic digestion. The proteins were identified by search against the MIPS and MIT N. crassa databases and five promising candidates were identified. Their structural characteristics and putative roles in the gsn transcription regulation are discussed.

The present work describes a more efficient methodology for the chlorination of water containing disperse dyes, where the chlorinated byproducts identified by mass spectra are compared. this investigation, we tested the degradation of Cl Disperse Blue 291 dye, 2-[(2-Bromo-4,6-dinitrophenyl)azo]-5-(diethylamino)-4-methoxyacetanilide) a commercial azo dye with mutagenic properties. The present work evaluates the photoelectrocatalytic efficiency of removing the Cl Disperse Blue 291 dye from a wastewater of the textile industry. We employed NaCl as a supporting electrolyte. It should be noted that photoelectrocatalytic techniques are non-conventional method of generating chlorine radicals. The by-products formed in this process were analyzed using spectrophotometry, liquid chromatography, dissolved organic carbon, mass spectral analysis and mutagenicity assays. The process efficiency was compared with the conventional chlorination process adopted during sewage and effluents treatment processes. This conventional chlorination process is less efficient in removing color, total organic carbon than the photoelectrochemistry technique. Furthermore, we shall demonstrate that the mutagenicity of the generated by-products obtained using photoelectrocatalysis is completely different from that obtained by the conventional oxidation of chloride ions in the drinking wafer treatment process. As imagens obtidas pelos raios X vêm sendo utilizadas na análise da qualidade de sementes desde os anos 50 e, atualmente, vêm se destacando por ser um método rápido, de boa precisão e não destrutivo. Deste modo, o presente trabalho foi realizado com o objetivo de avaliar as características morfológicas dos embriões de sementes de Tecoma stans L. Juss. ex Kunth pertencentes a diferentes classes de massa através da análise de imagens obtidas pelo uso dos raios X, correlacionando-as com a germinação e morfologia das plântulas. Foram utilizadas sete classes de massa com trinta sementes em cada classe, que foram submetidas aos raios X para a obtenção das imagens dos embriões. Os embriões, de cada uma das classes, foram separados de acordo com suas características morfológicas em 4 categorias (embriões sem defeito, embriões com pequenos defeitos, embriões deformados e sementes sem embrião). As sementes das diferentes classes de massa foram colocadas para germinar (emissão da raiz primária) e após dez dias foi observado o número de plântulas normais em cada uma das categorias e classes de massa. A maioria das sementes das classes 1 e 2, mais leves, apresenta-se com embriões deformados ou sem embrião, enquanto que as de maior massa (classes 3 a 7) apresentam embriões sem defeitos, em sua maioria ou totalidade. Quase a totalidade das sementes com embriões sem defeito germina, porém, nem todas dão origem a plântulas normais.

Five minute-averaged values of sky clearness, direct and diffuse indices, were used to model the frequency distributions of these variables in terms of optical air mass. From more than four years of solar radiation observations it was found that variations in the frequency distributions of the three indices of optical air mass for Botucatu, Brazil, are similar to those in other places, as published in the literature. The proposed models were obtained by linear combination of normalized Beta probability functions, using the observed distributions derived from three years of data. The versatility of these functions allows modelling of all three irradiance indexes to similar levels of accuracy. A comparison with the observed distributions obtained from one year of observations indicate that the models are able to reproduce the observed frequency distributions of all three indices at the 95% confidence level.

A new quinonemethide triterpene named as salacin, has been isolated from the root bark of Salacia campestris in addition to the known pristimerin, maytenin, 20 alpha-hydroxymaytenin, and netzahualcoyene. Salacin was identified on the basis of NMR-spectral and mass spectrometric analysis. The free-radical scavenging activities of the quinonemethide triterpenes salacin (1), pristimerin (2), maytenin (3), 20a-hydroxymaytenin (4), and netzahualcoyene (5) towards DPPH have been evaluated and showed absorbance variation (AA) of 19, 20, 39, 28, 55, and 10%, respectively, having rutin (74% at 50 pm) and BHT (7% at 50 mu M) as standard compounds.

A lectin-like protein from the seeds of Acacia farnesiana was isolated from the albumin fraction, characterized, and sequenced by tandem mass spectrometry. The albumin fraction was extracted with 0.5 M NaCl, and the lectin-like protein of A. farnesiana (AFAL) was purified by ion-exchange chromatography (Mono-Q) followed by chromatofocusing. AFAL agglutinated rabbit erythrocytes and did not agglutinate human ABO erythrocytes either native or treated with proteolytic enzymes. In sodium dodecyl sulfate gel electrophoresis under reducing and nonreducing conditions, AFAL separated into two bands with a subunit molecular mass of 35 and 50 kDa. The homogeneity of purified protein was confirmed by chromatofocusing with a pI=4.0+/-0.5. Molecular exclusion chromatography confirmed time-dependent oligomerization in AFAL, in accordance with mass spectrometry analysis, which confers an alteration in AFAL affinity for chitin. The protein sequence was obtained by a liquid chromatography quadrupole time-of-flight experiment and showed that AFAL has 68% and 63% sequence similarity with lectins of Phaseolus vulgaris and Dolichos biflorus, respectively.

Accurate long-term monitoring of total ozone is one of the most important requirements for identifying possible natural or anthropogenic changes in the composition of the stratosphere. For this purpose, the NDACC (Network for the Detection of Atmospheric Composition Change) UV-visible Working Group has made recommendations for improving and homogenizing the retrieval of total ozone columns from twilight zenith-sky visible spectrometers. These instruments, deployed all over the world in about 35 stations, allow measuring total ozone twice daily with limited sensitivity to stratospheric temperature and cloud cover. The NDACC recommendations address both the DOAS spectral parameters and the calculation of air mass factors (AMF) needed for the conversion of O-3 slant column densities into vertical column amounts. The most important improvement is the use of O-3 AMF look-up tables calculated using the TOMS V8 (TV8) O-3 profile climatology, that allows accounting for the dependence of the O-3 AMF on the seasonal and latitudinal variations of the O-3 vertical distribution. To investigate their impact on the retrieved ozone columns, the recommendations have been applied to measurements from the NDACC/SAOZ (Systeme d'Analyse par Observation Zenithale) network. The revised SAOZ ozone data from eight stations deployed at all latitudes have been compared to TOMS, GOMEGDP4, SCIAMACHY-TOSOMI, SCIAMACHY-OL3, OMI-TOMS, and OMI-DOAS satellite overpass observations, as well as to those of collocated Dobson and Brewer instruments at Observatoire de Haute Provence (44 degrees N, 5.5 degrees E) and Sodankyla (67 degrees N, 27 degrees E), respectively. A significantly better agreement is obtained between SAOZ and correlative reference ground-based measurements after applying the new O-3 AMFs. However, systematic seasonal differences between SAOZ and satellite instruments remain. These are shown to mainly originate from (i) a possible problem in the satellite retrieval algorithms in dealing with the temperature dependence of the ozone cross-sections in the UV and the solar zenith angle (SZA) dependence, (ii) zonal modulations and seasonal variations of tropospheric ozone columns not accounted for in the TV8 profile climatology, and (iii) uncertainty on the stratospheric ozone profiles at high latitude in the winter in the TV8 climatology. For those measurements mostly sensitive to stratospheric temperature like TOMS, OMI-TOMS, Dobson and Brewer, or to SZA like SCIAMACHY-TOSOMI, the application of temperature and SZA corrections results in the almost complete removal of the seasonal difference with SAOZ, improving significantly the consistency between all ground-based and satellite total ozone observations.

We discuss the relationship between exact solvability of the Schroedinger equation, due to a spatially dependent mass, and the ordering ambiguity. Some examples show that, even in this case, one can find exact solutions. Furthermore, it is demonstrated that operators with linear dependence on the momentum are nonambiguous.
