During the period from October/92 to September/94 experiments were carried out at the Seed Laboratory, FCAV/UNESP, Jaboticabal, SP, Brazil, using soybean seeds of different genotypes in order to evaluate the effect of genotype on the electrical conductivity (bulk conductivity) of soaked seeds. Seed moisture content (105+/-3 degrees C, 24 h), standard germination (four 50-seed samples, paper towel, 30 degrees C), and vigor-accelerated aging (42 degrees C, 48 h) were first determined. Undamaged soybean seeds were soaked in deionized water (four 50-seed samples, 75 ml, 25 degrees C, 24 h) and electrical conductivity (mu mhos.cm(-1).g(-1)) was measured. Significant differences in conductivity were observed among genotypes having the same pattern of germination and vigor. The results have showed that electrical conductivity can be significantly influenced by genotype.

The present work aimed to define the field maturity of Avena strigosa Schreb seeds cv. Comum. Three experiments were carried out in field conditions during 1986, 1987 and 1989 in Botucatu, SP, Brazil. The sowings were made in May. The characteristics evaluated were: panicles and spikelet aspects, the weight of the dry matter of 100 seeds with and without glumes, the seed moisture, the percentage of germinated seeds (sum of normal + abnormal seedlings), the percentage of germination and vigor (first count of germination test). Based on the dry matter accumulation and the gemination percentage of the seeds, it was concluded that the field maturity of the seeds was reached around 28 and 3 5 days after 100% of panicles emergence of the field. At this stage, the field presented a few percentage of particles (+/- 20%) not yet all yellowish, with spikelets at semi-hard stage and the seeds with 20 to 30% of moisture.

The objective of this study was to evaluate the effect of mechanical damage and physiological events in harvesting and processing of soybean cv. Mosoy RR 8000. The samples were taken during harvest manual, mechanical harvesting and during processing (receipt, pre-cleaning, cleaning, spiral separator, classification and gravity table). The physiological and physical quality was analized through the purity, germination, vigor (first germination count, seedling dry matter, accelerated aging, electrical conductivity, tetrazolium, mechanical damages and seedling field emergence) tests. The statistical design used was a entirely randomized with nine treatments (9 sampling points) with 4 replications, being the means compared by the Tuckey test at 5% probability. In the purity and seedling field emergence were observed highly significative difference between the sampling process, also this differences were obtained the first germination count, seedling dry weight matter, accelerated aging and electrical conductivity which showed smaller results for the mechanical harvesting when compared with the manual harvesting. The germination was obtained differences at 5% for the manual harvesting in relation to the mechanical harvesting were obtained smaller results, being the main cause of reducing the soybean seed quality, when compared with the manual harvesting.

The aim of this paper was to evaluate the effects of the combinations of substrata on germination and development of seedlings of lettuce cultivars. The experiment was carried out in a nursery, in Mato Grosso do Sul State University. There wend compared the following combinations of substrata: washed sand + earthworm humus (2: 1); manure bovine + earthworm humus (2: 1); Plantmax(R) + earthworm humus (2: 1); washed sand + Plantmax(R) (2: 1) and manure bovine + Plantmax(R) (2: 1), and three lettuce cultivars: Crespa sem cabeca, Americana Julia and Baba de Verao. The experimental design was entirely at random, in a 5x3 factorial outline, with four replications and 16 seeds perplot. There were considered for evaluation 10 central seedlings per plot. The characteristics evaluated were: germination percentage (%), index of germination speed (IVE) and dry mass of the whole seedling (g). For all the analyzed variables, the cultivars Baba de Verao and Crespa sem cabeca presented the best results. Manure + humus was the best substratum to obtain seedling with larger development (vigor), althout it did not increase IVE and germination percentage.

The onion (Allium cepa L.) belongs to the family Alliaceae, and the second vegetables in economic importance. It horticultural culture of high relevance, in central-south and in the far south, is highlighting the states of São Paulo, Rio Grande do Sul, Santa Catarina and Parana. The bulbificacion only begins when the combination of the determinants of bulbificacion (photoperiod and temperature) of each cultivar is reached. It Can be grown in tropical regions with the use of vernalization of the bulbs. The objective of this study is to identify the efficiency in the production of seed, onion depending on the cut or not the bulb before the period of vernalization. The analysis of the variables consisted: humidity; number of seeds; mass of seeds for umbel; umbels number; number of seeds for umbel; total number of flowers and of fecundated flowers; number of seeds for capsule; total mass of seeds; emergency percentage; vigor percentage; index of emergency speed and mass of 100 seeds humidity; number of seeds; mass of seeds for umbel; umbels number; number of seeds for umbel; total number of flowers and of fecundated flowers; number of seeds for capsule; total mass of seeds; emergency percentage; vigor percentage; index of emergency speed and mass of 100 seeds, using the cultivars Baia Periforme and Texas Grano. The experimental design was randomized blocks with four replicates, there was 80 plants for replicate, each parcel was conduced with 20 plants distributed in 4 lines of 5 plants with spacing of 20cm between plants and lines which was avaliated the 6 central plants. The seeds production not variated between cultivars Texas Grano e Baia Periforme. The cultivar Texas Grano showed seeds with more physiological quality. The cut or not cut of the bulbs not changed the physiological quality of the seeds. But utilization of whole bulbs resulted more yield of seeds with more flowers number and fertilized flower in Baia Periforme, more value of the total mass of seeds, number of seeds by plant and number and mass of seed by umbel in two cultivars.

This work was carried out at the Fazenda Experimental Lageado, at Botucatu, SP, to evaluate the physiological quality of cucumber (Cucumis sativus L.), seeds harvested at different fruit maturity stages and post-harvest resting periods. Fruits of cultivar Pérola were harvested at 6 ages (20, 25, 30, 35, 40 and 45 days after anthesis) and they were stored at room temperature for 0, 5, 10 or 15 days before seed extraction. After this, seeds were fermented for 2 days, washed and dried. External visual characteristics of fruits and seeds were observed at harvest. Seed quality was evaluated as for moisture, dry matter content, dimensions, germination, and vigour. Results showed that seed germination and vigour increased in fruits aging from 20 to 40 days. Post-harvest periods improved seed quality of 20-day old fruits not reaching the seed quality levels observed in 40 or 45-day old fruits, without post-harvest resting period. Seeds from fruits at 25, 30 and 35 days of age, with 10 or 15 days of post-harvest resting period reached the same quality levels of seeds from fruits with 40-45 days old.

This work was carried out in order to evaluate if there was a relationship between genotypes and the physiological soybean (Glycine max L.) seed quality. It was conducted during three years using seven cultivars each year. The seeds were harvested at: 1) yellow radicle or expanded pod stage, 2) yellow pod or physiological maturity (R7), 3) harvest maturity (R8), and 4) R8 + 21 days delay. Seed moisture content, standard germination, and vigor tests were performed. The germination and vigor evaluated by accelerated aging and electrical conductivity did not show physiological seed quality differences among genotypes as harvested at physiological maturity. Then, the evaluation of seed germination and vigor, when the environment is not a considered factor, is not an efficient method to show differences among soybean genotypes in terms of seed quality.

In order to identify the desiccation tolerance of E. edulis seeds, four fruitlots were collected from the Campinas Agronomic Institute Palmae collection, located at Ubatuba, state of Sao Paulo, Brazil, and taken in moisture-proof containers to the College of Agriculture in Botucatu, state of Sao Paulo. There, the fruits were shelled and the seeds were dried, either under silica gel conditions or in a drying chamber. The effects of dehydration were evaluated by measurements of the germination, embryo protrusion, germinative button, plumule, speed of emergence, seedling length, seed dry matter and water content. Differences among seedlots and between harvest season for most of the traits were observed. Euterpe edulis seedlots seem to tolerate water content reductions until 39% without significant reduction in germination and vigor. Below this value, most of the evaluated traits were negatively affected. Total loss of germination was observed whenever seed water content was reduced to values below 21%.

The aim of the present work was to evaluate plasma membrane integrity, motility, vigor and morphology of fresh and frozen goat spermatozoa with or without seminal plasma. Semen samples were diluted in Tris solution, before and after thawing, with a combination of carboxifluorescein diacetate and propidium iodide. The results showed differences (P < 0.01) for motility and minor defects in the presence or absence of seminal plasma, for both fresh and frozen samples. Periods of collection had a significant effect on motility, probably due to changes in the photoperiod. Plasma membrane integrity was significantly reduced by the freezing process, whether seminal plasma was present or absent. In conclusion, removal of seminal plasma decreased motility and vigor rates in frozen samples. The photoperiod probably decreased the testosterone level, contributing negatively to the high percentage of sperm abnormalities, mainly damaged membranes. The use of fluorescent probes allowed a better estimation of the percentage of functional cells, instead of only estimating the percentage of motile cells or morphology defects. © 2001 Elsevier Science B.V. In Brazil, garden and cut rose bushes are propagated through cuttings or grafting. The rootstocks used in most of the traditional areas of cultivation are not identified, being known just by the vulgar name. There is not any information in the literature on the morphologic characteristics, nor about the productivity of different cultivars on the rootstocks available in Brazil. The objective of this work was to characterize and determine productivity of nine rootstocks (Rosa multiflora 'Paulista'; R. multiflora 'Japones'; R. multiflora 'Iowa'; R. multiflora 'Kopmans'; R. indica 'Mayor'; R. indica x multiflora; R. sp. 'Natual Brier'; R. manetti and R. canina 'Inermis') grafted with 'Versilia' and 'Tineke' scions. Morphologic studies were undertaken to characterize branches of adult plants to determine shape and dimensions of the leaf; number of prickles/15 cm of stem; vigor of the stems; incidence of Diplocarpon rosae (blackspot) and coloration of the leaf. The productivity (number of commercial flowers/plant/month) was evaluated for 6 months (October, 2000 to March, 2001), in the city of Andradas, Minas Gerais State, located at 1251 meters above sea level. Since there was significant difference in just two months for both rootstocks, 'Tineke' and 'Versilia', no rootstocks could be considered inadequate. Nonetheless, in general, the rootstocks that showed a better performance with 'Tineke' were, R. multiflora 'Kopmans' and R. manetti and with 'Versilia', R. multiflora 'Kopmans' and Rosa sp. 'Natual Brier'.

The nitrogen is the nutrient required in greatest quantities by the bean crop and the response to its application depends on the applied N rate as well as the time of its application. The objective of this work was to evaluate yield components, grain productivity and physiological quality of bean seeds, affected by different nitrogen (urea) rates top-dressed at three crop growth stages. The experiment was conducted in no-till system. The treatments were constituted by 0, 40, 80, 120, 160, 200 and 240 kg ha-1 N top-dressed at V4-5, R5 and R6 growth stages, corresponding to 21, 32 and 38 days after plant emergence, respectively. Seed physiological quality was evaluated by germination and vigor tests. Nitrogen applied at different crop growth stages did not interfere on the yield components, but interfered on bean productivity. Maximum grain productivity was obtained with top-dressed 164 kg ha-1 N, independently of growth stage application. Seed physiological quality was not affected by the treatments, fitting in the category of seeds for commercialization.
