Mesenchymal stem cells (MSCs) are adult multipotent cells with fibroblastoid morphology and adherent to plastic. Furthermore, they can be obtained from different sources. Besides bone marrow, these cells are taken from umbilical cord blood, umbilical vein, saphenous vein, peripheral blood, arteries, liver and fetal pancreas, placenta, dental pulp and adipose tissue. MSCs derived from adipose tissue are important because of the abundant number of cells that can be obtained from this tissue, easy access and little discomfort to the patient. This study compared two techniques for obtaining MSCs from adipose tissue: mechanical dissociation (MD) and enzymatic digestion (ED). We also analyzed the inter-species cross-reactions using commercial monoclonal antibodies directed against surface antigens of stem cells from different species: mouse, horse, rabbit, monkey and human. We found that MD technique is favorable in relation to ED within 15 days of culture, and ED is more efficient in the first days of culture. The data also showed that MD causes less damage to cellular DNA. About inter-species cross-reactions, the monoclonal antibody A69 directed against stem cells from rabbits, which can be used in veterinary medicine, particularly in research involving horses

Experimental models composed by human and animal cell lines are simplified and informative, allowing them to be widely used for biomedical research. Most laboratories that use in vitro cultivated cells maintain a variation of cell lines stored and cultivated. Therefore, misidentification and cross-contamination events can happen during cell lines handling. This problem can generate a repertoire of dubious results and papers, which may prejudice biomedical research. Recently it was created the International Cell Line Authentication Committee (ICLAC), which aims to spread knowledge about cross-contamination and misidentification of in vitro cell lines. Despite of the efforts spent trying to aware scientific community about the importance of the correct identification of cells, the number of papers based on misidentified cell lines it´s still worrying, compromising the reliability of out coming results and conclusions regarding them. The present study aims to analyze and discuss the main advantages and limitations of eukaryote in vitro cell lines use, characterizing the cell lines authentication problems. Therefore, compilation and critical analyses of literature data was realized, aiming to improve the understanding about this subject. Based on information about 445 cell lines with issues published by ICLAC it´s clear that contamination in human cell lines represented 89,2 % of mentioned problems. HeLa cell line was the responsible for most contamination, especially in 92 normal tissue cell lines, representing 44,6% of the contamination. These results reinforce the importance of periodic maintenance of cell lines cultures by labs and implementation of authentication methods as polymorphic STRs, besides obtaining cell lines from reliable sources and cell banks

A monoclonal antibody (mAb) is an important tool in medical biotechnology and the production of biopharmaceuticals, especially for disease diagnosis and treatment of infections, because the antibodies have a significant advantage over chemical agents used in conventional therapies . The last thirty years the technology of production of monoclonal antibodies developed mainly the technique of obtaining in vitro, but also of their production is laborious, the cost is high. A major element of the high cost of production is the fact that the long-term culture consumes a large amount of imported inputs with high added value. A major contribution of this work is to promote cell growth more quickly and efficiently. Currently, a great race to discover new technologies and techniques to synthesize new antibodies and significantly increase the production of murine mAbs. New technologies such as laser and LED are innovations and widespread in modern life, so much so that its use has proliferated worldwide, primarily in the medical field. Recent studies show a series of results from the influence of the LED light in biological tissues such as: increasing the rate of cell proliferation, increased production rate of fibroblasts, increasing the rate of synthesis of RNA and DNA synthesis of ATP, etc. To assess the contribution of the LED in the culture of Myeloma NS1murino compared to the standard procedure. - NS1 cells were provided and followed the criteria of culture medium of the Laboratory of Cellular Engineering Center of Botucatu (POPs). The same amount of cells was grown in bottles of 25 cm2 polystyrene Tissue Culture Treated, specifically marked and kept in special medium RPMI 1640 Gibco BRL  supplemented with fetal bovine serum 10%, essential amino acids and non-essential, glucose, insulin and antibiotics. We present a succinct review of the canonical formalism of classical mechanics, followed by a brief review of the main representations of quantum mechanics. We emphasize the formal similarities between the corresponding equations. We notice that these similarities contributed to the formulation of quantum mechanics. Of course, the driving force behind the search of any new physics is based on experimental evidence

Nowadays, bamboo is being studied because of their strength properties according with their specific mass and speed of growth, which makes it an important alternative as a new resource that will help reduce pressure on forests and helpping them favoring the minimization of uncontrolled deforestation in many regions of Brazil. This study aimed, in general, to analyze physical and mechanical properties of the material with the divulgation of its potential for industrial application. To do so, in this research were determined in relation to the physical properties, moisture content, dimensional stability and the apparent densities and the mechanics and basic, just a tension parallel to grain, in order to observe the interference of various kinds of treatments (chemical, thermal and natural) on the strength and modulus of elasticity in this request. The species used was the Guadua angustifolia, a species native of Brazil. All tests were performed at Universidade Estadual Paulista - Campus Experimental Itapeva in the laboratory of Materials Properties. The methodology used for testing of moisture, density and tension parallel to grain were based on NBR 7190/1997 for the wood, and dimensional stability tests were based on much the same as in COPANT 462/1972 (South American) . The preservative treatments conducted followed the recommendation of each manufacturer. The values obtained in tests of physical properties were satisfactory especially with respect to density and dimensional stability analyzed by the coefficient of anisotropy, showed that, compared to wood, excellent quality for the shrinkage test, obtaining a coefficient of 1.2. With respect to parallel tensile tests to fibre results showed, in most cases, that test specimens with the presence of us have lower values of resistance and modulus of elasticity when compared with those without us. Prostate cancer is the second leading cause of cancer death in the country. Due to this factor, the interest of showing what relationship exists between vasectomy surgery and the incidence of prostate carcinoma has started. Some epidemiological studies showed an increased risk of prostate cancer in vasectomized men (Emard et al., 2001). On the other hand other authors have argued that there is no correlation (Patel et al., 2005) and there are those who said that vasectomy is linked to reduced risk of prostate cancer (Ross, 1983). Faced with an analysis of published works, such discussion remains today. The vasectomy, or deferentectomia, is a contraceptive male method, which is the section of the vas deferens of man by preventing the sperm from being expelled along with the seminal fluid during ejaculation, and is one of the most simple, economical, uncomplicated post- operative. This study aims to evaluate the process of cell proliferation and to evaluate immuno-histochemically the expression of specific markers of PCNA (Proliferating Cell Nuclear Antigen) and Ki-67, in the prostate of the gerbil after chemical induction by intraperitoneal injection of the carcinogen N-methyl -N-nitrosourea (MNU), because the regulation of the functional balance between cell proliferation and apoptosis is associated with hyperplasia and prostatic carcinoma. Airplane Motor Cradles have a complex geometry, since they require different conbinations between different tubes and TIG welded in several angles. In T-25 aircraft and Universal T-27 Tucano (EMBRAER / FAB), besides having to bear the engine balance, these components maintain fixed the nose landing gear in another extremity. They are considered critical to flight safety, and for this reason, the aviation standards are extremely rigid in their production, imposing a zero index” of defects on the final weld metal quality. These structures may be containing an historical of welding repairs, whose effects on their structural integrity are not computed. In this work we analyzed the standardised AISI 4130 steel and the raw steel of tubes to the Airplane Motor Cradles. First of all, microscopy and microanalysis of the base steel, then we analyzed the effects of the TIG weld. The development of technology for structural composites has as one of its ends form a set of materials that combine high values of mechanical strength and stiffness and low density. Today, companies like Embraer and PETROBRAS and research institutions like NASA, working with these materials with recognized advantages in terms of weight gain, increased performance and low corrosion. We have developed a systematic study to determine the bond strength between composite carbon fiber / epoxy and fiberglass / epoxy laminate both bonded to a carbon steel which are widely used in the petrochemical industry and repair. For morphological evaluation and bonding between materials of different natures, ultrasound analysis, optical microscopy and stereoscopy were performed. To simulate actual conditions, the composites were subjected to conditioning by using heat shock temperatures from -50 to 80 ° C for 1000 cycles for composite carbon fiber / epoxy composites and 2000 cycles for fiberglass / epoxy . The use of composites studied here proved to be efficient to perform repairs in metallic pipes with application petrochemical, as when exposed to sudden changes of temperature (-50 ° to 80 ° C) cycling at 1000 to 2000 times, its mechanical properties (shear and tensile) practically do not change

In 2002, the Ministry of Education (MEC) published the Curriculum Guidelines for Engineering courses in the country. The Guidelines contain subjects like the profile of graduates, the course structure, curriculum, internships and skills and abilities, all geared to guide the training of engineers in Brazil. The objective of this study is to evaluate together with teachers of the Production Department of this University and students of the fifth year of Production Engineering is how student learning of Production Engineering for the development and importance of skills within the course and skills for engineers who are present in the Curriculum Guidelines for engineering courses developed by MEC

The use of pesticides in Brazil has intensified over the years. And since 2009 it was ranked as the largest market for pesticides worldwide. The consequential diffuse contamination of the environment is therefore due to the increasing number of chemicals of different classes, origins and modes of action. Little is known about the action of pesticides on human health in situations of co-exposure. Possible toxic effects are not restricted to agricultural and industrial workers, but also the general population that may be exposed continuously to its residues in food and water. Although these pesticides are mostly present in the environment at low doses, it must be considered that possible cumulative or synergistic effects may occur when there are concurrent or sustained exposure for two or more of these agents, which can lead to late manifestation of subclinical damages, sometimes irreversible. Thus, the specific objective of this study was to assess the effect of carcinogenesis promotion of a mixture of pesticides at low doses and analyze the phenomena of cell proliferation and apoptosis in rat liver. A total of 50 male Lewis rats was separated into 5 groups for 8 weeks in a medium term hepatocarcinogenesis model. The three different classes of pesticides (dieldrin, dicofol, endosulfan, dichlorvos and permethrin), whose residues were detected by ANVISA during the period from 2001 to 2005 in tomatoes cultures, were added to the feed of rats initiated to hepatocarcinogenesis with diethylnitrosamine (DEN- 200mg/kg ip). We used two different mixtures, one with no toxic effects at doses (MEX1) referring to the NOEL (no-observed-effect level) and another at doses LOEL / LEL / LOAEL (Lowest-observed-effect level / Lowest-effect level / Lowest -observed-adverse-effect level), to the installation of adverse effects (MEX2), derived from chronic studies. The need to reduce environmental damage and add value to waste causes more and more new alternatives appear to unite these two points. One of the main ways to achieve this in timber industries and the use of waste for making panels. This work was aimed at studying the influence of particle size and density in Eucalyptus mechanical compressive strength of cement composite wood. For this study was performed production and physico-mechanical characterization of specimens, using portland cement, water and waste eucalyptus. The methodology consists of a statistical study of the results obtained by calculating the density and axial compression tests and a subsequent comparison of these results with other studies. The results showed that there are significant differences in density and compressive strength when using different particle sizes the particles of eucalyptus. In general, the smaller the particle size, the lower the compression strength and the greater the density when the samples are produced with the same trait

The study of physical and mechanical properties of wood is essential for its structural use and it is of great importance to the construction industry. Thus, this study aimed to determine the physical and mechanical properties of the wood Amaru - a hybrid of Eucalyptus, developed by Plantar Projects and Forest Products Ltda. In order to determine the properties of Amaru, round samples were used, which were provided to the Laboratory of Wood and Wooden Structures of the School of Engineering of São Carlos, University of São Paulo - LaMEM / EESC / USP. For the characterization of the physical properties, the apparent specific gravity and moisture content of the samples were determined. To the mechanical characterization, the following properties were evaluated: strength and stiffness in compression, strength and stiffness in bending, shear and tension. The procedures of the tests performed in this study were done according to the recommendations of the Brazilian Wood Standard ABNT NBR 7190:1997. The specimen used were confectioned in actual dimensions, according to as those used in the construction system proposed by Plantar. The results obtained from the tests performed showed that the mechanical properties approached the values proposed by the Wood Standard NBR 7190. The visual grading was important to provide a primary idea about the failure modes to be obtained from the tests performed. The bending test showed the modulus of elasticity (MOE) and Modulus of Rupture (MOR), which resulted in 15822 MPa and 101,7 MPa, respectively. The compression test resulted in values Ec0,m and fc0, 15698 MPa and 50,7 MPa. The tensile strength (ft0) of this hybrid was calculated and its value obtained was 60,8 MPa. The shear strength (fv0) was 8,2 MPa. The results obtained from the tests are the basis for engineers and architects to design structures using wood species Amaru

Malformations and possible damages to the urogenital system can be originated in the embryonic period. Moreover, fire guns, knives and accidents, where there is the disruption of the urethra, also cause these lesions. The objective was to analyze the contribution of tissue engineering in the construction of neo-urethra, developed by bioengineering. We performed an urothelial ex vivo expansion of cells in 3D scaffolds (platelet gel matrix and acellular porcine aorta) to assess the contribution of this technique in the construction of a neo-urethra. Mechanical dissociation was made of the inner wall of 10 North Folk rabbit’s bladder, weighing 2.5 to 3.0 kg. After dissociation the cell content was centrifuged and obtained a pellet of urothelial cells. The pellet was ressuspended in culture medium DMEM F12 and cells were maintained in culture for 15 days. Immunohistochemical analysis characterized the urothelial culture. The cells were then implanted in the scaffold - platelet gel. In a second experiment using aortic porcine acellular matrix were implanted urothelial cells alone and urothelial cells on platelet gel, on the inner wall of the scaffold - aorta, with space for setting bordered by a urethral probe. The complex probe - cells - aorta and probe - cells in platelet gel - aorta, were sealed with suture material and culture were maintained in a humidified 37ºC incubator with 5% CO2 in air for 12 days to subsequent histological analysis of urothelium cell adhesion to the scaffolds. By observation under an optical microscope, we could see the growth of cells in the scaffold platelet gel, from a monolayer in to a three-dimensional structure. In the acellular porcine aortic matrix containing the platelet gel, we could observe a few quantity of urothelial cells adhered. However with the acellular porcine aortic matrix in which was implanted only the urothelial cells, we have obtained adhesion to the wall

This work has the propose to discuss the National Examination Performance of students for the course in Production Engineering , from 2005 up to 2011, subsidies are shown how this came about assessment and how the course has been evaluated in recent years . Compares higher education from public and private universities in Brazil and is also discussed how important a good performance in the exam. Qualitative and quantitative surveys were conducted at different times that the work that enables a critical analysis of the examination , the institution and also the students themselves , so the list is the main motivators for the exam and also what should be done to obtaining good results in academic performance. Motivators for the exam has highlighted its importance is paramount and the consent of all involved from the beginning of the course to ensure that the examination is compulsory for all graduates of undergraduate manufacturing engineering in Brazil
