This work was carried out at the Fazenda Experimental Lageado, at Botucatu, SP, to evaluate the physiological quality of cucumber (Cucumis sativus L.), seeds harvested at different fruit maturity stages and post-harvest resting periods. Fruits of cultivar Pérola were harvested at 6 ages (20, 25, 30, 35, 40 and 45 days after anthesis) and they were stored at room temperature for 0, 5, 10 or 15 days before seed extraction. After this, seeds were fermented for 2 days, washed and dried. External visual characteristics of fruits and seeds were observed at harvest. Seed quality was evaluated as for moisture, dry matter content, dimensions, germination, and vigour. Results showed that seed germination and vigour increased in fruits aging from 20 to 40 days. Post-harvest periods improved seed quality of 20-day old fruits not reaching the seed quality levels observed in 40 or 45-day old fruits, without post-harvest resting period. Seeds from fruits at 25, 30 and 35 days of age, with 10 or 15 days of post-harvest resting period reached the same quality levels of seeds from fruits with 40-45 days old.

The final levels of ethanol (levels of ethanol produced plus that added initially to the media) reached by the thermotolerant yeasts were highest (16.5-20.3%, v/v) at 8% initial ethanol. The thermotolerant yeasts were found to have the following characteristics: constant levels of ethanol formation (10.5-12.3%, v/v), fog additions of external ethanol within the range 2-8% (v/v) of initial ethanol; constant values of product coefficients when initial ethanol was in the range of 2-6%, which increased or decreased, depending on the strain, when initial ethanol exceeded 6%; growth activity was inhibited at different levels of addition of external ethanol when final biomass and specific rate of growth were compared; significant differences among the yeast strains in the amount of external ethanol capable of reducing biomass formation by one half. In addition, the viability of the strains (early stationary phase) varied with the amount of external ethanol, the lowest viabilities occurring at concentrations of initial ethanol ranging from 4 to 7% and the highest in the range of 7 to 8% (v/v). The relative levels of trehalose (with/without 7% ethanol added initially) in the yeast strains (the stationary phase) ranged from 1.03 to 1.75, suggesting that the effect of produced ethanol on trehalose accumulation was stronger than that of external ethanol. The levels of final ethanol shown by the yeast strains were also correlated with the cellular levels of glycerol-3-phosphate dehydrogenase (increase in enzyme levels with decrease in final ethanol) for cells harvested at the stationary phase.

The objective of this experiment was to analyze the rumen fermentation of silages made from corn harvested at milk stage (MS), milk early dough stage (MEDS), medium dough stage (MDS) and semi-hard dough stage (SHDS). Rumen fluid was collected from sheep by esophageal tube at 0, 1, 3 and 6 hours after feeding. There were no differences among silages for ammonia nitrogen (NH3-N) and methylene blue reduction time (MBRT). Only the MS and SHDS silages differed in rumen pH (6.82 and 6.53, respectively). Differences in total rumen VFA and acetic acid concentrations (mmoles/L) were observed among stages, but not between MS (36.40 and 22.13) and MEDS (42.49 and 25.73), nor between MDS (64.52 and 40.34 respectively) and SHDS (64.09 and 43.61, respectively). The periods of 1 and 3 hours after feeding showed the smallest pH values (6.47 and 6.63), the highest NH3-N concentrations (9.75 and 10.56 mg/dL) and the highest concentrations of total VFA, and acetic and propionic acids (60.33, 37.05 and 16.73; 59.40, 35.28 and 16.84 mmoles/L, respectively). On the whole, the MDS and SHDS silages showed the best rumen fermentation patterns based on pH and total and individual VFA values.

This work was carried out in order to evaluate if there was a relationship between genotypes and the physiological soybean (Glycine max L.) seed quality. It was conducted during three years using seven cultivars each year. The seeds were harvested at: 1) yellow radicle or expanded pod stage, 2) yellow pod or physiological maturity (R7), 3) harvest maturity (R8), and 4) R8 + 21 days delay. Seed moisture content, standard germination, and vigor tests were performed. The germination and vigor evaluated by accelerated aging and electrical conductivity did not show physiological seed quality differences among genotypes as harvested at physiological maturity. Then, the evaluation of seed germination and vigor, when the environment is not a considered factor, is not an efficient method to show differences among soybean genotypes in terms of seed quality.

Fruits of okra (Abelmoschus esculentus (L.) Moench.) cv. Amarelinho were harvested at three times: 35, 45 and 55 days after anthesis. Half part of the fruits of each harvest time was shelled and the seeds were dried in natural environment of laboratory or in dry chamber. The other half was dried unshelled in the same two conditions. Water contents of seeds were evaluated at harvest time and before the germination test, that was carried out when the seeds were in hygroscopic equilibrium with the two environments. The physiological quality of seeds was affected by the interaction effects of fruit age, drying method and drying condition. The highest values for percentage of germination were obtained from seeds taken from fruits 55 days old (up to 92%) and the hard seeds percentage was not affected by drying method and drying condition. The seeds from fruits of 35 and 45 days old had the germination percentage increased when dried inside the fruit in natural environment.

The objective of this research was to evaluate the residue from cassava, known as manipueira, as an alternative for tomato (Lycopersicon esculentum) fertilization. This experiment was carried out in UNESP with the cultivar Rio Grande. The experimental design was a completely randomized blocks in a 3 × 3 factorial arrangement with five blocks. Treatments constituted the following: 1) witness; 2) 54 m3/ha of manipueira; 3) 108 m3/ha of manipueira; 4) half recommended mineral fertilization; 5) half recommended mineral fertilization + 54 m3/ha of manipueira; 6) half recommended mineral fertilization + 108 m3/ha of manipueira; 7) recommended mineral fertilization; 8) recommended mineral fertilization + 54 m3/ha of manipueira and 9) recommended mineral fertilization + 108 m3/ha of manipueira. Tomato yield, number of fruits per plant, fruit diameter and lenght, total soluble solids (TSS), total titratable acidity (TTA) and the ratio TSS/TTA were evaluated. The results obtained indicated that manipueira contributed significantly to yield and quality of pos-harvested tomato fruits.

A greenhouse experiment was conducted in a factorial design of randomized complete blocks, with three replications, to study plantage response of soybean cultivars on efficiency: update (EAB); conversion (EC) and utilization, as well as utilization index (IU) by soybean cultivars. Four soybean cultivars (IAC 17, IAC 18, IAC 11 e FT 2) were grown in nutrient solutions with either adequate or deficient K levels (0.5 and 1.82 mmol. L-1) and harvested 20, 40 and 60 days after plant emergence (DAPE). The plants were separated in top, median and lower leaves, stems and roots, dried, ground and analyzed for K contents. Regardless of cultivar, in deficient plants K was translocated from the other organs to median leaves, in order to maintain its K contents. This behavior was particularly evident in 'IAC 11' and 'IAC 18'. Well nourished plants showed higher EAB than deficient plants. 'IAC 17' and 'IAC 18' showed higher EAB at either adequate or deficient K. levels. 'FT 2' showed highest EUK in deficient solutions at 40 DAPE (immediately before flowering), and the lowest EUK at 60 DAPE. The utilization index was helpful in determining cultivar plasticity to adapt to K deficiency. The highest plasticity was observed for 'FT 2' at 40 DAPE and for 'IAC 18' at 60 DAPE. K-deficient plants were more efficient in EC, yielding more dry matter for K uptake unit.

Objective: The present study was performed to investigate the influence of different routes of perfusion on the distribution of the preservation solutions in the lung parenchyma and upper airways. Methods: Pigs were divided into four groups: control (n = 6), pulmonary artery (PA) (n = 6), simultaneous PA + bronchial artery (BA) (n = 8), and retrograde delivery (n = 6). After preparation and cannulation, cardioplegia solution and Euro- Collins solution (ECS) for lung preservation were given simultaneously. After removal of the heart, the double lung bloc was harvested. Following parameters were assessed: total and regional perfusion (dye-labeled microspheres), tissue water content, PA, aorta, left atrial and left ventricular pressures, cardiac output and lung temperature. Results: Our data show that flow of the ECS in lung parenchyma did not reach control values (9.4 ± 1.0 ml/min per g lung wet weight) regardless of the route of delivery (PA 6.3 ± 1.5, PA + BA 4.8 ± 0.9, retrograde 2.7 ± 0.9 ml/min per g lung wet weight). However, flow in the proximal and distal trachea were significantly increased by PA + BA delivery (0.970 ± 0.4, respectively, 0.380 ± 0.2 ml/min per g) in comparison with PA (0.023 ± 0.007, respectively, 0.024 ± 0.070 ml/min per g), retrograde (0.009 ± 0.003, respectively, 0.021 ± 0.006 ml/min per g) and control experiments (0.125 ± 0.0018, respectively, 0.105 ± 0.012 ml/g per min). Similarly the highest flow rates in the right main bronchus were achieved by PA + BA delivery (1.04 ± 0.4 ml/min per g) in comparison with 0.11 ± 0.03 in control, 0.033 ± 0.008 in PA, and 0.019 ± 0.005 ml/min per g in retrograde group. Flows in the left main bronchus were 0.09 ± 0.02 ml/min per g in control, 0.045 ± 0.012 ml/min per g in PA, and 0.027 ± 0.006 ml/min per g in retrograde group. The flow rates were significantly (P = 0.001) increased by PA + BA delivery of the storage solution (0.97 ± 0.3 ml/min per g). Conclusions: Our data show that the distribution of ECS for lung preservation is significantly improved in airway tissues (trachea and bronchi) if a simultaneous PA + BA delivery is used.

Studies were conducted to show the effect of different temperatures in the drying process on the amount and quality of essential oils of Cymbopogon citratus (DC) Stapf. Leaves were harvested in the experimental field of the Agronomical Sciences College, UNESP, Botucatu, SP, Brazil in September, 1996. Blades of the leaves were cut in small parts (about 1-1,5 cm length), dried for several days at 30°, 50°, 70° and 90°C, until establishment of the weights. In the following process a hydrodistillation, during 2.5 hours, by Clevenger apparatus, was subsidized to extract the essential oils. A higher amount of oil could clearly be collected with the lower drying temperatures, except at 30°C, affected by fungus growing. Aspergillus sp., Penicillium sp., Rhyzopus sp., Cladosporium sp., Trichoderma sp. and Alternaria sp. were observed in the leaves. The analysis of the oil by GC-MS showed the variation of citral concentration of the treatments (86,1 to 95,2%). The results proved it is worthwhile to spend more time and effort in the production process using longer times of careful drying.

An experiment was carried out at Sâo Manuel Experimental Farm, FCA-UNESP, Botucatu-SP, in 1994 and 1995, aiming to determine the biomass production and essential oil yield of mentrasto (Ageratum conyzoides L. - Asteraceae) in different stages of development. The statistical design used was a complete randomized block with 8 replications and 3 treatments (pre-flowering, flowering and post-flowering). The spacing was 70 cm between rows and 50 cm between plants, with 70 plants/parcel. The seedlings were transplanted 40 days after sowing. After harvesting, the fragments (roots, stems, leaves, flowers) and whole plants were washed, separated and weighted. The extractions of essential oil were done by hydrodistillation, using Clevenger apparatus, for two hours. There were statistic differences (Tukey 5%) in fresh weight of all vegetal parts w'hen harvested in different development stages. About essential oil yield, statistic differences were not verified among the treatments.

This experiment was developed in order to evaluation the efficiency of pheromone to control the pink bollworm and the total time of its release in cotton field. The experiment was installed in field conditions, in Chapadao do Sul/SP/Brazil, from January to April, 1998. The treatments consisted of 2 areas, being one of 30ha, where it was applied the pheromone and another of 10ha that was chosen as control area and did not receive pheromone. In the treated area, the laboratory synthesized sex pheromone (PB-Rope) was used thought of dispensers that allowed the slow and gradual release of the active substance. A total of 250 dispenser per hectare were evenly hand distributed in the area. The dispensers were wrapped around the plants. Both areas (treated area and untreated area) were monitored by delta trap. For evaluation of the boll damage, the treatment area was divided into 4 sub-areas. Twenty five green bolls were collected at random from each sub-area at 48 and 65 days after pheromone treatment. Bolls were cracked open by hand, and number of the bolls with symptoms of pink bollworm attacks was recorded. For evaluation of the productivity four areas were demarcated in each treatment, where all fibers and seeds harvested were weighted. Release rate of pheromone from dispenser was evaluated through of the weigh of the dispensers. Were marked and weighed in analytic scale, 20 dispensers contend the pheromone, being placed 10 dispensers under the cotton plants in treated area and other 10 dispensers in an open area. To every 15 days the dispensers were retired and weighed in analytic scale and soon after put back in the field in the same places. The results showed that only one application of mating disrupt pheromone, used in a dosage of 250 dispenser/ha, reached 80% of control for pink bollworm. the release period of pheromone from dispenser, after the application, was 120 days.

An antigen-competitive enzyme-linked immunosorbent assay (Ag-C-ELISA) was developed for the detection of infectious bronchitis virus (IBV) antigens, M41 strain, in tissues from experimentally infected chickens, or in allantoic fluid harvested from inoculated embryonated eggs. The detection limit of IBV in the Ag-C-ELISA was 104.1 median embryo infective doses (EID50)/well. Tracheal and lung samples from chickens vaccinated with 102.5 EID50 of live attenuated infectious bronchitis (H120) vaccine were negative in the direct detection Ag-C-ELISA. The results indicate that the Ag-C-ELISA has the potential to detect IBV, either directly in tissue samples or when combined with the passage of material in embryonated eggs, thereby constituting an alternative method for the diagnosis of IBV.

Seed ageing is natural phenomenon that occurs in all seeds, including those stored in dry and low temperature rooms. Phaseolus vulgaris L. cv. 'Carioca' seeds harvested in five different years were stored in at 15°C. Seed samples were germinated and evaluated according to The Brazilian Rules for Testing Seeds. The newest seed sample was submitted to artificial ageing. Small (∼1g) samples of all materials (naturally and artificially aged) were ground and proteins extracted. Equal quantities of protein were loaded onto the gels and electrophoresis carried out. Although seeds submitted to 24, 48, 72 and 96 hours of artificial ageing did not show any statistical difference (5%) in relation to unaged seeds in physiological parameters related to emergence, some statistically different parameters related to dry matter and length of shoots and roots occurred. A 24 h of artificial ageing at 41°C improved these factors. Protein patterns were changed after 72 h of artificial ageing and naturally aged seeds showed alterations after two years storage at 15°C. Aged seeds, naturally and artificially, had decreasing germination, vigour, and changes in the characteristic banding pattern of proteins. Physiological parameters and electrophoresis examination showed that for naturally aged seeds physiological parameters were more sensitive while artificially aged seeds, (41°C/100% RH), had electrophoretic profiles that were more efficient for seed lot discrimination.

Studies were conducted to show the effect of different temperatures in the drying process on the amount and quality of essential oils of peppermint (Mentha piperita L.) The leaves were harvested in the Demeter Farmer, Botucatu, SP, Brazil in december, 1997. The leaves were dried at 40°C, 60°C and 80°C, until establishment of the weights. The essential oil was extracted by destilation in Clevenger apparatus and analysed by GC-MS. Higher drying temperature sharply decreased the essential oil content (% v/w) from 1.0% (40°C) to 0.14% (60°C) and 0.12% (80°C). Higher drying temperatures also affected the composition, decreasing the contents of 1,8 cineol and citronelal until 80°C, and increasing the contents of menthol and neomenthol until 60°C.
