To evaluate the biochemical profile and protein concentration of whey from milk samples of healthy Murrah primiparous and pluriparous buffaloes, 30 female buffaloes were analyzed during a complete lactation. The animals were divided into three groups: G1 = 10 primiparous buffaloes, G2 = 10 pluriparous buffaloes with 2-3 lactations and G3 = 10 pluriparous buffaloes with > 3 lactations. The lactation period was divided into: early stage (I: 1-3 months of lactation), intermediate stage (T: 4-6 months of lactation) and final stage (F: 7-9 months of lactation). Before milk sampling, physical examination of the mammary gland, strip cup test and California Mastitis Test (CMT) were performed. After mammary quarters asepsis, 20mL of milk were collected monthly from each mammary quarter, during a complete lactation, in sterilized plastic bottles without preservative, in order to perform microbiological isolation, biochemical profile and protein electrophoresis in sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE), and 30mL of milk from each mammary quarter were collect, in sterilized plastic bottles containing preservative bronopol to perform the somatic cell count (SCC). A total of 1,042 milk samples were collected from the experimental groups during lactation, of which 923 samples showed negative reaction to CMT and negative microbiological isolation and were selected to biochemical profile analysis and protein electrophoresis in SDS-PAGE. There were influence of parity order and stage of lactation in biochemical profile and protein concentration of healthy Murrah buffaloes'whey. Primiparous buffaloes (G1) showed higher gamma-glutamyltransferase (GGT: 2,346 U/L), alkaline phosphatase (ALP: 181 U/L), phosphorus (P; 56.6mg/dL), potassium (K; 32.0mg/dL) and alpha-lactalbumin (458mg/dL). Buffaloes with 2-3 lactations (G2) showed higher SCC (70,700 cells/mL) and higher concentrations of total protein (1.55g/dL), albumin (100mg/dL), magnesium (Mg; 8.80mg/dL), chlorides (Cl; 176mg/dL), iron (Fe; 10.7 mu g/dL), sodium (Na; 178mMol/L) and lactoferrin (59.5mg/dL). Bufalloes with > 3 lactations (G3) showed higher concentrations of total calcium (Ca; 41.8mg/dL), ionized calcium (iCa; 2.92mMol/L), immunoglobulin A (IgA; 1.32mg/dL), serum albumin (99.1mg/dL), immunoglobulin G (IgG; 49.7mg/dL) and beta-lactoglobulin (1,068mg/dL). During lactation it was observed increase in SCC, GGT, ALP, total protein, albumin, P, Mg, Cl, Na, lactoferrin, serum albumin, IgG and alpha-lactalbumin, as well as decrease in concentrations of Ca, Fe, iCa, K, IgA and beta-lactoglobulin in buffaloes'whey. The results may be used as reference for buffaloes and to support diagnosis and prognosis of diseases common to lactation periods.

A total of 3.035 lactations of Holstein cows from four farms in the Southeast, to check the influence of data structure of milk yield on the genetic parameters. Four dataset with different structures were tested, weekly controls (CW) with 122.842 controls, monthly controls (CM) 30.883, bimonthly controls (CB) with 15,837 and quarterly controls (CQ) with 12,702. The random regression model was used and was considered as random additive genetic and permanent environment effects, fixed effects of the contemporary groups (herd-year-month of test-day) and age of cow (linear and quadratic effects). Heritability estimates showed similar trends among the data files analyzed, with the greatest similarity between dataset CS, CM and CB. The dataset submitted all the CB estimates of genetic parameters analyzed with the same trend and similar magnitude to the CS and CM dataset, allowing the claim that there was no influence of the data structure on estimates of covariance components for the dataset CS, CM and CB. Thus, milk recording could be accomplished in a CB structure.

Persistent Organic Pollutants (POPs) are chemical substances extraordinarily toxic and persistent in the environment. Due to these properties, POPs are targets of the Stockholm Convention which aims to provide means to eliminate production and use by encouraging the reduction and, where possible, eliminating the release of these contaminants to the environment. One of the articles of this _Convention aims to assess the basal levels of such substances in the environment to monitor both the reduction of release to the environment, and to assess human exposure, using analyzes of milk and air for example. Although efforts to control POPs production and release to the environment have been made, data about their levels in Brazil are still scarce, mainly in the atmosphere or indoor environment. Considering the permanence of people inside its house, this study presents a method for POP extraction from XAD-2 sorbent employed in indoor air sampling, with identification and quantification by GC-ECD. GC-ECD linear range of the studied analytes, congeners of polychlorinated biphenyls (PCB) (28, 52, 101, 118, 138, 153 and 180), DDTs and their metabolites (DDDs, DDEs) and endosullan (isomers c - and 13 - and metabolite endosulfan sulfate) ranged between 0.5 and 16.0 ng mL-I with r2 > 0.967. The method developing consist of selecting the extraction solvent system by agitation (90 rpm, 1 hour) with n-hexan,e (2 mL) or n-hexane:acetone (1:1, v/v) (2 mL), concentration the extract with a gentle N2 stream, and evaluation of breakthrough with passage or air through the fortified XAD-2 cartridge before extraction, employing a low volume air suction pump (Pump Model 224 universal-PCXR8; 5-5000 mL min-1, SKC Inc., Eighty Four, PA, USA), Calibrator Dry-Cal DC-Lite (BIOS, Butler, NJ, USA)). This paper reports the results of an extension project carried out at Unesp/Veterinary Medicine course in Araçatuba city with milk farmers of the region. The aim of the project was to supply information to farmers concerning to good quality milk production and, at the same time, to follow up the evolution of milk quality parameters, according to the current Brazilian legislation. Every 45 days, approximately, lectures were presented to milk farmers in Araçatuba city region, approaching chemical and microbiological composition of milk, prevention and detection of mastitis, hygiene proceedings in milking and conservation of milk, cleaning and sanitization operations of facilities and equipments and prevention of adulteration. During the intervals between lectures, milk samples were collected from collective milk cooling tanks and analyzed for microbiological, hygienic and physicochemical tests. The main inadequacies in milk quality were high total bacteria and somatic cells counts, low solids contents and water addition. These problems did not proceed to betterment during the project time. So, it was concluded that the time for instruction of farmers was not enough for a progress in the quality of the milk produced in the region, pointing out the need in continuing this kind of work.

Brazil is one of the largest milk producers in the world, with an estimated total production of 37 billion liters in 2014. Of this total, 33% is not inspected, being destined for the informal market. Objective: The aim of this study was to collect and analyze data on raw milk consumption in the city of Araraquara (SP, Brazil). Methods: Regular milk consumers were provided with exploratory questionnaires in locations chosen for convenience. The reproducibility of each question was tested by Kappa statistics. Associations of interest were 2 detected by the chi-squared (χ ) or Fisher's exact test. Statistical significance was established when p0.05. Results: Most of the volunteers (97.6%) reported consuming milk daily, the type of milk chosen most, mainly for ease of purchase, being boxed UHT. Regarding raw milk, 15.3% of volunteers said they consume it, the greatest consumption being observed among subjects with complete secondary or higher education. The majority of the participants reported knowing of the possibility of diseases being transmitted by unprocessed milk. Most respondents pointed to "flavor" as the main reason for consumption. Only 15.3% reported that someone in their house had become ill from drinking milk, and only 4.1% attributed this to the ingestion of raw milk. Conclusion: It was found that 15.3% of consumers drink raw milk in the city of Araraquara, which is more than the expected proportion in a medium sized city located in the state of São Paulo.

Milk is considered a nutritionally complete food but it is also an excellent substrate for microbial growth. Therefore, it may pose a risk to consumers’ health. The aim of this study was to collect data on the occurrence of gastroenteritis outbreaks caused by the intake of milk and dairy products in the state of São Paulo, from 2000 to 2010. The data were collected from the records on foodborne illness outbreaks kept by the Division of Waterborne and Foodborne Diseases of the Center for Epidemiological Surveillance of the Department of Health of the State of São Paulo. A total of 239 outbreaks were notifi ed. They involved 2.418 cases related to the consumption of dairy products during the study period. The etiological agents were identifi ed in 79 outbreaks (33.0%). They were mostly bacteria (84.8%), especially Staphylococcus aureus. Most outbreaks occurred at home (59.4%) and during the months of January (14.2%) and July (12.5%). Most (24.1%) of the affected individuals aged 20 to 49 years. The results shows the need of better manufacturing and handling practices, as well as increasing the awareness of the population and relevant agencies about the importance of notifying cases and implementing preventive measures to ensure food safety.

Brucellosis is a zoonosis caused by bacteria of the genus Brucella. Man infection occurs through contact with reproductive secretions as placenta and its lochia, semen and penile secretion of infected animals or by consuming unpasteurized milk and dairy products. With the objective of investigating the presence of bacteria in milk, 30 samples of raw milk sold illegally in the region of Botucatu, São Paulo, Brazil, as well as 50 samples of milk delivered to a dairy industry previously to its pasteurization were evaluated by the polymerase chain reaction (PCR) technique. Of the 80 samples analyzed, 10 samples (12.5%) were positive and 70 (87.5%) were negative. Among the positive samples, 5 (16.6%) were from illegal traders and other 5 (10%) were obtained from the dairy industry. Brucella spp. positivity shows that the pathogen is representatively present in Botucatu, São Paulo, Brazil, and the risk associated to public health due to the commercialization of illegal products without pasteurization is real.

Mastitis is an acute or a chronic inflammation of the udder, represented for physical, chemical and bacteriological milk alterations. It is notable that ovine milk represents high economic importance, and it can be used in the production of cheeses and other derivates, contributing for the farmer’s recipe enlargement. However, the mastitis in animals can determine the reduction of milk quantity and quality in infected animals. This may determine lambs’ weight loss and increase animal’s mortality. By the other hand, considering Public Health issues, mastitis can cause milk nutrients losses and promote the microorganisms’ multiplication that may lead to diarrheas and to other zoonosis in humans. This study aimed to evaluate 159 sheep mastitis status in 20 small farms located in Bauru (SP) region. Of the 309 colected samples, in 97 (31.4%) were isolated microorganisms of the genus Staphylococcus sp., which them 27.5% being coagulase negative Staphylococcus and 3.9% coagulase positive Staphylococcus. Of 177 isolates, 97 (54.8%) were represented by the genus Staphylococcus sp., which them 57 (58.8%) from pure culture and 40 (41.2%) in association. Most agents isolated were sensitive to antibiotics used in the test. Thus, we could verify the isolation of infectious microorganisms such as coagulase positive and negative Staphylococcus and, as well in dairy cattle, preventive measures to obtain hygienic milk are extremely important in order to get a final product with quality and safe in food and microbiological point of view.

Milk that is adequate for consumption must be of hygienic quality, nutritional value, and should maintain its organoleptic properties. Isolation of fecal and/or total coliforms from bovine milk is considered an indicator of hygiene and good management practices, and can be used as a quality indicator. This study aimed to isolate, identify, and assess the resistance profile of coliforms isolated from collective bulk tanks and individual milk tanks. A total of 89 milk samples were collected from collective bulk tanks and, from these, 21 Klebsiella spp., one E. coli, and 29 Enterobacter spp. were isolated, whereas 102 milk samples from individual tanks showed isolation of one Klebsiella spp. and seven Enterobacter spp. In collective bulk tanks, at least 47% of Klebsiella spp. and Enterobacter spp. were resistant to cephalexin and 30% to ampicillin. From these, at least 24% showed multidrug resistance. Among the microorganisms isolated from the individual tanks, 85% or more were resistant to ampicillin. The ESBL phenotype and the blaTEM gene were detected in strains of Klebsiella spp. isolated from both tanks. It was concluded that contamination of milk with resistant total coliforms, and especially the storage of raw milk from several small producers in the collective bulk tank increase the risk of contamination.
