Despite recent advances in understanding the biological basis ofprostate cancer (PCa), the management of this disease remains a challenge. Chemoprotective agents have been usedto protect against or eradicateprostatemalignancies. Here, we investigated the protective effect of -tocopherol on N-methyl-N-nitrosourea (MNU)-induced epithelial dysplasia in the rat ventral prostate (VP). Thirty-two male Wistar rats were divided into four groups (n=8): control (CT): healthy control animals fed a standard diet; control+-tocopherol (CT+T): healthy control animals without intervention fed a -tocopherol-enriched diet (20mg/kg); N-methyl-N-nitrosourea (MNU): rats that received a single dose of MNU (30mg/kg) plus testosterone propionate (100mg/kg) and were fed a standard diet; and MNU+-tocopherol (MNU+T): rats that received the same treatment of MNU plus testosterone and were fed with a -tocopherol-enriched diet (20mg/kg). After 4months, the VPs were excised to evaluate morphology, cell proliferation and apoptosis, as well as cyclooxygenase-2 (Cox-2), glutathione-S-transferase-pi (GST-pi) and androgen receptor (AR) protein expression, and matrix metalloproteinase-9 (MMP-9) activity. An increase in the incidence of epithelial dysplasias, such as stratified epithelial hyperplasia and squamous metaplasia, in the MNU group was accompanied by augmented cell proliferation, GST-pi and Cox-2 immunoexpression and pro-MMP-9 activity. Stromal thickening and inflammatory foci were also observed. The administration of a -tocopherol-enriched diet significantly attenuated the adverse effects of MNU in the VP. The incidence of epithelial dysplasia decreased, along with the cell proliferation index, GST-pi and Cox-2 immunoexpression. The gelatinolytic activity of pro-MMP-9 returned to the levels observed for the CT group. These results suggest that -tocopherol acts as a protective agent against MNU-induced prostatic disorders in the rat ventral prostate.

The focus of this study was to evaluate the effect of lysine on the productive (weight and length gain, apparent feed conversion, specific growth rate and condition factor) and reproductive performance of Rhamdia voulezi males confined in net cages during the first reproductive cycle. The following parameters were assessed: seminal parameters (motility rate and duration, survival, sperm concentration, morphology, pH and osmolarity), hormonal parameters (cortisol and testosterone), testicular tissue (histomorphology), organosomatic indices (gonadosomatic, hepatosomatic and viscerosomatic indices) and composition of essential amino acids, crude protein and moisture of whole carcass. Four hundred fish were used, distributed in a random experimental design with four treatments and four replications in 16 net cages with 25 fish each. The treatments consisted of four different diets prepared so as to contain the following levels of lysine: 1.20, 1.40, 1.60 and 1.80%, with 30% crude protein and 3500 kcal kg(-1) digestible energy for 185 days (Jul./12-Jan./13). Eighteen males were selected per treatment, and they all released semen after slight abdominal pressure. The males were weighed, measured, submitted to hypophysation (2.5 mg kg(-1) carp pituitary extract), and then had their semen and blood collected. The fish were sacrificed by cervical dislocation, dissected, and the testes, liver, fat and guts were removed and weighed. The effects (p < 0.05) for the means of final weight, weight gain, apparent feed conversion and condition factor were observed for the analysis of productive performance. With regard to the reproductive parameters, only the seminal volume was affected (p < 0.05). Thus, the levels of testosterone showed quadratic effect (p < 0.05). The anatomy and the histomorphology of the testes were similar between the treatments during the spermiation period. With regard to the organosomatic indices, there was no influence (p > 0.05) between the treatments. The amino acids in the carcass were not affected (p > 0.05). The increment of lysine in the diet provided linear increase for weight gain and seminal volume and linear decrease for feed conversion in R. voulezi broodstocks confined in net cages. Adequate testosterone levels are necessary for the development, growth and maintenance of the male reproductive system. Testosterone deficiency is common in men with diabetes in whom it may contribute to impaired performance, with consequent reduction of the activity of the androgen regulated organs, such as the prostate. However, little attention has been given to the plasma dihydrotestosterone (DHT) level, the most potent androgen, nor to the expression of the androgen receptor (AR), insulin-like growth factor type I (IGF-1) and receptor (IGF-1R) in target tissues. Here, we investigated the effect of type I diabetes mellitus on DHT plasma levels and on prostate AR, IGF-1 and IGF-1R expression during rat pubertal growth. Diabetes was induced in prepubertal male rats through administration of streptozotocin (STZ; 40 mg/kg). Diabetic, diabetic treated with insulin, and age-matched control animals were killed by overdoses of pentobarbital. The ventral prostatic lobe (VP) was dissected, weighed and processed for immunohistochemistry for AR, IGF-1 e IGF-1R; plasma T and DHT levels were also determined. Hyperglycemia at puberty reduced VP weight gain to about 50% and plasma T level to about 80% of the control levels. In contrast there were no changes in plasma DHT levels. Insulin replacement restored the VP weight gain, but not the plasma T levels, which remained 90% below the ones of controls. Immunohistochemistry showed that AR, IGF-1 and IGF-1R expression in the prostate epithelial cells did not change with hyperglycemia or insulin replacement. Thus, the AR expression in the prostate epithelial cells appears to be regulated by DHT, and to a minor extent it also controls glandular growth

It is known that exposure to substances in the environment can contribute to various reproductive disorders, especially if such exposure occurs during critical periods of development such as the intra-uterine and postnatal. The female reproductive system may be the target of androgens, both as a result of exposure to environmental chemicals, or by pathological conditions (polycystic ovary syndrome or congenital adrenal hyperplasia).Usually, little attention is given off in relation to the study of androgenic effects in the female reproductive axis. This study aims to evaluate the effects of exposure to androgens on the development, structure and reproductive function in rats whose mothers were exposed to testosterone propionate from gestational day 12 (DG12) after weaning - postnatal day 21 (DPN21) . For this purpose, pregnancy rats were divided into four groups: a control group that received corn oil (vehicle) and three groups receiving testosterone propionate in doses of 0.05 mg / kg / day, 0.1 mg / kg / day and 0.2 mg / kg / day, all under the same experimental conditions. The possible effects of exposure were assessed using reproductive parameters, such as a measure of anogenital distance, count areolas / nipples, age at vaginal opening and first estrus (puberty indicative installation), weight and histological evaluation of the reproductive organs ( uterus and ovaries), weight of the kidneys, liver and pituitary hormone levels, regularity of the estrous cycle, sexual behavior and fertility. Such analysis is important in understanding the effects of androgen exposure on the female genital system, especially on the reproductive potential, and processes that may involve morphofunctional changes. In these experimental conditions, it is concluded that treatment with PT caused reduction in body weight and initial masculinization in females without cubs, however, commit further sexual development
