Inflammatory cells surround breast carcinomas and may act promoting tumor development or stimulating anti-tumor immunity. N-acetylglucosaminidase (NAG) has been employed to detect macrophage accumulation/activation. Myeloperoxidase (MPO) is considered a marker for neutrophils activity/accumulation. Vascular Endothelial Growth Factor (VEGF) is as strong pro-angiogenic cytokine. The aim of this study was to measure the systemic inflammatory response by measuring serum levels of NAG, MPO and VEGF in women diagnosed with breast cancer and associate this response to the peritumoral inflammatory infiltrate and to prognostic factors. Serum samples obtained from women with no evidence of disease (n = 31) and with breast cancer (n = 68) were analyzed for the activities of NAG, MPO and VEGF by enzymatic assay. Serum levels of NAG and VEGF were higher in healthy volunteers (P < 0.0001) and serum levels of MPO were higher in patients with breast cancer (P = 0.002). Serum levels of NAG were positively correlated to serum levels of MPO and VEGF (P < 0.0001 and P = 0.0012, respectively) and MPO and VEGF serum levels had also a positive correlation (P = 0.0018). The inflammatory infiltrate was not associated to serum levels of the inflammatory markers, and higher levels of MPO were associated to lymphovascular invasion negativity (P = 0.0175). Human salivary gland tumors originated from intercalated ducts present a broad range of histologic and cytologic patterns, mainly due to the presence of myoepithelial cells. The aim of this study is to verify the differentiation grade of neoplastic cells and a possible relation between myoepithelial cell differentiation and the presence of luminal secretory contents. The expression of vimentin and cytokeratin (CK) intermediate filaments, actin myofilament and epithelial membrane antigen (EMA) was investigated by double labeling immunocytochemical technique, in thirty salivary gland neoplasms: 5 pleomorphic adenomas, 5 myoepitheliomas, 3 basal cell adenomas, 7 adenoid cystic carcinomas (ACC) and 10 polimorphous low grade adenocarcinomas (PLGA). Tumors with intercalated duct differentiation (pleomorphic adenomas, basal cell adenomas and ACC) express CKs 7, 8, 18 and 19 in the luminal cells and coexpress eventually CK14 with these CKs. Some luminal cells stained with anti-EMA antibody, mainly where a secretory content in the lumen was observed. Outer ductal cells and other myoepithelial-like cells express vimentin, sometimes coexpressing actin and/or CK14 with vimentin. Plasmacytoid cells in myoepitheliomas and pleomorphic adenomas express vimentin and rarely CKs 7, 8, 18 and 19, sometimes coexpressing these CKs with CK14 but they are negative for the remaining antigens. Tumors without intercalated duct differentiation (solid basal cell adenoma and PLGA) express vimentin and CKs 7, 8, 14 and 18, sometimes coexpressing CKs 8 and 18 with CK14. In conclusion, in tumors with intercalated duct differentiation, myoepithelial cells express vimentin and sometimes coexpress actin and/or CK14 with vimentin, never coexpressing other CKs with vimentin. CK14 and actin are independently expressed by myoepithelial cells, so their expression is probably induced by different stimulus. Despite the remarkable improvements in breast cancer (BC) characterization, accurate prediction of BC clinical behavior is often still difficult to achieve. Some studies have investigated the association between the molecular subtype, namely the basal-like BC and the pattern of relapse, however only few investigated the association between relapse pattern and immunohistochemical defined triple-negative breast cancers (TNBCs). The aim of this study was to evaluate the pattern of relapse in patients with TNBC, namely the primary distant relapse site:One-hundred twenty nine (129) invasive breast carcinomas with follow-up information were classified according to the molecular subtype using immunohistochemistry for ER, PgR and Her2. The association between TNBC and distant relapse primary site was analyzed by logistic regression. Using multivariate logistic regression analysis patients with TNBC displayed only 0.09(95% CI: 0.00-0.74; p = 0.02) the odds of, the non-TNBC patients of developing bone primary relapse. Regarding visceral and lymph-node relapse, no differences between in this cohort were found.Though classically regarded as aggressive tumors, TNBCs rarely development primary relapse in bone when compared to non-TNBC, a clinical relevant fact when investigating a metastasis of an occult or non-sampled primary BC. A decreased radioiodine uptake is frequently detected in differentiated thyroid carcinomas (DTC) and is associated with high recurrence rate and reduced survival. We investigated the correlation between NIS mRNA expression levels in the primary tumor and patient outcome using a quantitative real-time RT-PCR method. NIS expression was decreased in 17 DTC (21.04 +/- 39.66 pg Eq) compared to four autoimune thyroid disease (180.51 +/- 92.63 pg Eq) and 14 normal tissues (75.71 +/- 66.98 pg Eq) (p < 0.0001). The 17 thyroid differentiated carcinoma patients were submitted to surgery complemented by radioiodine ablation and had at least 24 months of follow-up, under levothyroxine continued suppressive therapy. According to their outcome, we could characterize a group of papillary carcinoma patients with aggressive carcinomas, whose NIS mRNA levels were markedly lower than a group with non-aggressive carcinomas (0.62 +/- 0.79 versus 54.87 +/- 53.79: p < 0.005). We suggest that the quantification of NIS mRNA relative levels in the primary tumor may predict poor outcome. (C) 2003 Elsevier Ireland Ltd. All rights reserved.

A metilação de ilhas CpG em regiões regulatórias de vários genes tem sido descrita como um processo importante no silenciamento de genes supressores de tumor e diretamente envolvida no processo de carcinogênese de uma série de tumores. O estudo desses genes afetados pela metilação em tumores visa identificar possíveis marcadores moleculares para o diagnóstico, prognóstico e tratamento de tumores, além de possibilitar a descoberta de fatores importantes no processo biológico do câncer. Os genes MX1 e ADAM23 foram identificados como diferencialmente metilados em linhagens celulares provenientes de carcinoma de cabeça e pescoço. Neste sentido, ao estudar o perfil diferencial de metilação de genes em carcinomas de células escamosas de cabeça e pescoço, o gene MX1 foi identificado como diferencialmente expresso. Dessa forma, para elucidar a função destes genes no controle da carcinogênese, o presente estudo buscou identificar ligantes celulares das proteínas MX1 e ADAM23 por meio de rastreamentos de duplo-híbrido, usando bibliotecas de cDNA de cérebro fetal humano. Os rastreamentos com a proteína ADAM23 não geraram resultados positivos por isso não são aqui discutidos. Foi realizado rastreamento com a proteína MX1, que resultou em aproximadamente 1,0x106 transformantes, dos quais 74 foram confirmados pelo ensaio de duplo-híbrido e codificam para 9 ligantes já conhecidos e 21novos ligantes prováveis de MX1. Entre esses novos ligantes prováveis estão proteínas que já haviam sido descritas como ligantes de MX1, incluindo a própria proteína MX1, o que valida os resultados obtidos com este rastreamento. Além disso, grande parte dos ligantes identificados são fatores envolvidos no processo de SUMOilação de proteínas, na formação de corpúsculos nucleares denominados PML-NB e uma série de proteínas relacionadas ao controle da transcrição e apoptose... O presente trabalho de conclusão de curso visou à otimização e preparação da síntese de Lapdesf-IUNI e avaliação do potencial de inibição de urease de três compostos previamente sintetizados, direcionados especificamente para o tratamento de infecções por Helicobacter pylori, bactéria mundialmente reconhecida como uma das principais causadoras de infecções no mundo, responsável pelos casos de infecções gástricas, como gastrites crônicas, úlceras (gástricas e duodenais) e carcinomas. Lapdesf-IUNI é um composto derivado de metronidazol, com atividade potencialmente inibidora de urease, enzima presente neste microrganismo e essencial a sua sobrevivência no ambiente gástrico. O produto foi obtido, porém com baixo rendimento, por isso sua metodologia ainda necessita ser aperfeiçoada, para que, em seguida este composto possa ser testado. Outros três compostos, um derivado de tiouréia e dois derivados guanidínicos, com potencial atividade inibidora de urease, T-6, R-17 e R-21, sintetizados pela equipe do Prof. Eduardo Perez, foram avaliados com relação a sua atividade inibidora frente à enzima urease e IC50, apresentando bons resultados, que permitem a continuidade dos estudos, comparando inclusive com fármacos utilizados nos protocolos de tratamento padrões para o tratamento das infecções causadas por H. pylori

Nevoid basal cell carcinoma (NBCCS) or Gorlin-Goltz syndrome (GS) is a multidisciplinary problem, the early diagnosis of which allows secondary prophylaxis that follows an appropriate regimen to delay progression of the syndrome. The aim of this study was to present a case of delayed diagnosis of GS in a young patient who received multidisciplinary treatment 5 years after onset. The patient presented for evaluation with painless swelling of the left maxilla. Histological examination confirmed the diagnosis of a keratocyst odontogenic tumor (KOT) that was enucleated. On presentation, the patient’s symptoms and clinical signs were not related to complications of GS, and the possibility of GS was initially rejected, as he did not have a family history of the syndrome. Four years after the first surgery to remove the lesion, the patient came to our clinic with a brown, pigmented lesion. Computed tomography revealed ectopic lamellar calcification of the falx cerebri, which was the conclusive factor for the diagnosis of GS. It is important that clinicians recognize the clinical signs of GS, which mainly manifests itself as multiple basal cell carcinomas in the skin.
