O objetivo do presente estudo foi comparar a produção de IFN-γ, IL-12 e IL-4 entre camundongos jovens (5, 12 e 19 dias de idade) e adultos (30 dias de idade). As avaliações foram feitas por estimulação, in vitro, de células esplênicas com Concanavalina A (ConA) , Staphylococcus aureus (S. aureus) e lipopolissacarídeo (LPS). Diferentes concentrações de cada estímulo foram testadas e os sobrenadantes das culturas foram coletados após 48 horas de incubação e as concentrações de IFN-γ, IL-12 e IL-4 determinadas por ELISA. Células de camundongos jovens e adultos produziram níveis igualmente elevados de IFN-γ após estímulo com ConA. Somente animais adultos produziram IFN-γ em resposta ao estímulo com S. aureus. Em culturas estimuladas com LPS, a produção desta citocina foi baixa e similar nos animais jovens e significativamente elevada nos animais adultos. Somente células de animais adultos estimuladas com S. aureus foram capazes de produzir IL-12. O único estímulo capaz de induzir níveis detectáveis de IL-4 foi ConA, sendo que estes níveis foram mais elevados nos animais com 12 e 19 dias de idade em comparação com animais neonatos e adultos. A diminuição das doses ótimas dos estímulos não mudou o perfil de produção de cada citocina nos animais jovens. Estes resultados permitem concluir que a idade afeta a produção de citocinas: ocorre maior produção de IL-4 em camundongos jovens e maior produção de IL-12 e IFN-γ em animais adultos. Estas informações são importantes devido ao papel destas citocinas na polarização das respostas imunes nos sentidos Th1 e Th2. Autoimmune bullous dermatoses are diseases in which blisters and vesicles are the primary and fundamental types of skin lesion. Their classification is based on the location of the blister: intraepidermal and subepidermal. Patients produce autoantibodies against self-specific structures of the skin detectable by immunofluorescence techniques, immunoblotting and ELISA. Recent advances in molecular and cellular biology have brought to knowledge these self-antigens, against which patients are sensitized, and which are found in epidermis or in the dermo-epidermal junction. These are low incidence, but high morbidity diseases that may be fatal. The aim of this article is to review and describe the progress of four autoimmune vesiculobullous disorders: endemic pemphigus foliaceous (wild fire), pemphigus vulgaris, bullous pemphigoid and dermatitis herpetiformis. ©2009 by Anais Brasileiros de Dermatologia.

Background: Protein-calorie malnutrition (PCM) is the most common type of malnutrition. PCM leads to immunodeficiency and consequent increased susceptibility to infectious agents. In addition, responses to prophylactic vaccines depend on nutritional status. This study aims to evaluate the ability of undernourished mice to mount an immune response to a genetic vaccine (pVAXhsp65) against tuberculosis, containing the gene coding for the heat shock protein 65 from mycobacteria. Methods: Young adult female BALB/c mice were fed ad libitum or with 80% of the amount of food consumed by a normal diet group. We initially characterized a mice model of dietary restriction by determining body and spleen weights, hematological parameters and histopathological changes in lymphoid organs. The ability of splenic cells to produce IFN-gamma and IL-4 upon in vitro stimulation with LPS or S. aureus and the serum titer of specific IgG1 and IgG2a anti-hsp65 antibodies after intramuscular immunization with pVAXhsp65 was then tested. Results: Dietary restriction significantly decreased body and spleen weights and also the total lymphocyte count in blood. This restriction also determined a striking atrophy in lymphoid organs as spleen, thymus and lymphoid tissue associated with the small intestine. Specific antibodies were not detected in mice submitted to dietary restriction whereas the well nourished animals produced significant levels of both, IgG1 and IgG2a anti-hsp65. Conclusion: 20% restriction in food intake deeply compromised humoral immunity induced by a genetic vaccine, alerting, therefore, for the relevance of the nutritional condition in vaccination programs based on these kinds of constructs. © 2009 Ishikawa et al; licensee BioMed Central Ltd.

The incentive for consumption and production in large quantity in modern society generates enormous amounts of urban solid residues in the form of municipal solid waste (MSW). With the intention of reducing these residues of the municipal waste tips and to generate energy, briquettes with mixtures of MSW and residues of Eucalyptus grandis were produced. The briquettes were manufactured with 0, 5, 10, 15, 20 and 25% of MSW in the mixture with wood waste and 12% of moisture content. The analyzed parameters used to choose the best treatments were combustion analysis versus ash content, mechanical strength and energy content. The briquettes up to 10% of MSW showed low resistance, and above 15% showed large increase in ash content. Therefore, the treatment that fulfilled the requirements for combustion versus ash content and mechanical resistance was of at least 15% of MSW, since the source of the ash is unidentified. Considering the net energy content, the best treatment was 25% of MSW, with 17,175 kJ kg-1. Nevertheless, it is strongly advised that further studies related to gas emissions are necessary.

Background: Our group previously demonstrated that a DNA plasmid encoding the mycobacterial 65-kDa heat shock protein (DNA-HSP65) displayed prophylactic and therapeutic effect in a mice model for tuberculosis. This protection was attributed to induction of a strong cellular immunity against HSP65. As specific immunity to HSP60 family has been detected in arthritis, multiple sclerosis and diabetes, the vaccination procedure with DNA-HSP65 could induce a cross-reactive immune response that could trigger or worsen these autoimmune diseases. Methods: In this investigation was evaluated the effect of a previous vaccination with DNA-HSP65 on diabetes development induced by Streptozotocin (STZ). C57BL/6 mice received three vaccine doses or the corresponding empty vector and were then injected with multiple low doses of STZ. Results: DNA-HSP65 vaccination protected mice from STZ induced insulitis and this was associated with higher production of IL-10 in spleen and also in the islets. This protective effect was also concomitant with the appearance of a regulatory cell population in the spleen and a decreased infiltration of the islets by T CD8+ lymphocytes. The vector (DNAv) also determined immunomodulation but its protective effect against insulitis was very discrete. Conclusion: The data presented in this study encourages a further investigation in the regulatory potential of the DNA-HSP65 construct. Our findings have important implications for the development of new immune therapy strategies to combat autoimmune diseases. © 2009 Santos et al; licensee BioMed Central Ltd.

The purpose of the present study was to evaluate, using a biomechanical test, the force needed to remove implants with surface modification by laser (Nd:YAG) in comparison with implants with machined surfaces. Twenty-four rabbits received one implant with each surface treatment in the tibia, machined surface (MS) and laser-modified surface (LMS). After 4, 8 and 12 weeks of healing, the removal torque was measured by a torque gauge. The surfaces studied were analyzed according to their topography, chemical composition and roughness. The average removal torque in each period was 23.28, 24.0 and 33.85 Ncm for MS, and 33.0, 39.87 and 54.57 Ncm for LMS, respectively. The difference between the surfaces in all periods of evaluation was statistically significant (p < 0.05). Surface characterization showed that a deep and regular topography was provided by the laser conditioning, with a great quantity of oxygen ions when compared to the MS. The surface micro-topography analysis showed a statistical difference (p < 0.01) between the roughness of the LMS (R a = 1.38 ± 0.23 μm) when compared to that of the MS (R a = 0.33 ± 0.06 μm). Based on these results, it was possible to conclude that the LMS implants' physical-chemical properties increased bone-implant interaction when compared to the MS implants. © 2009 Sociedade Brasileira de Pesquisa Odontológica.

O trabalho teve objetivo estudar a variabilidade temporal da temperatura do ar, precipitação pluviométrica e umidade relativa do ar na cidade de Botucatu-SP, Brasil, utilizando técnicas geoestatísticas. Os dados de precipitação pluviométrica, temperatura do ar e umidade relativa do ar utilizados no presente estudo são provenientes da Estação Meteorológica da Fazenda Lageado, da Faculdade de Ciências Agronômicas-UNESP. As observações foram realizadas no período de 1988 a 2007, referem-se ao total mensal de precipitação pluvial expressa em altura de lâmina d'água (mm), médias mensais de temperatura em ºC e umidade relativa em %. Os dados foram avaliados por meio da estatística clássica e geoestatística. As variáveis climáticas tiveram sua dependência verificada por variogramas, apresentando dependência temporal maior que 76%. A série temporal de umidade relativa do ar foi a que apresentou maior alcance (8,67 meses) e, conseqüentemente, maior estabilidade climática. O conhecimento da distribuição temporal das variáveis climáticas é importante para o estudo e realização do zoneamento agroclimático, bem como para o dimensionamento do sistema de irrigação das culturas.

Background: Several anti-inflammatory drugs have been used to reduce pain and discomfort after periodontal surgeries. This study evaluates the efficacy of using etoricoxib and dexamethasone for pain prevention after open-flap debridement surgery. Methods: For this prospective, double-masked, crossover, placebo-controlled, randomized clinical trial, open-flap debridement surgeries were performed on 15 patients (eight males and seven females, age range 20 to 56 years: mean age ± SD: 40 ± 9.7 years) who presented with chronic periodontitis after nonsurgical periodontal therapy at three quadrants. Each patient underwent three surgical procedures at intervals of 30 days and received one of the following premedication protocols 1-hour before surgery: group 1 = placebo, group 2 = 8 mg dexamethasone, and group 3 = 120 mg etoricoxib. Rescue medication (750 mg acetaminophen) was given to each patient who was instructed to take it when necessary. Pain intensity and discomfort were evaluated by a 101-point numeric rate scale and a four-point verbal rate scale, respectively, hourly for the first 8 hours after surgery and three times a day on the following 3 days. Results: The results demonstrate that groups 2 and 3 present reduced postoperative pain-intensity levels compared to group 1. There were statistically significant differences at the 4, 5, 6, 7, and 8 hour-periods after surgery (Friedman test; P<0.05). Furthermore, rescue-medication intake was significantly lower for groups 2 and 3 than for group 1 (analysis of variance; P<0.02). Conclusion: The adoption of a preemptive medication protocol using etoricoxib or dexamethasone may be considered effective for pain and discomfort prevention after open-flap debridement surgeries.

The purpose of this work is to evaluate the capacity of full polarimetric L band data to discriminate macrophyte species in Amazon wetland. Fieldwork was carried out almost simultaneously to the acquisition of the full polarimetric PALSAR data. Coherent and incoherent attributes were extracted from the image, and macrophyte morphological variables were measured on the ground. The image attributes and the macrophyte variables were compared in order to evaluate their application for discriminating macrophytes species. The findings suggest that polarimetric information could be adopted to discriminate plant species based on morphology, and that estimation of plant biomass and productivity could be improved by using the polarimetric information. © 2010 IEEE.

Background: Although there is some information in the literature discussing differences of the estrous cycle of Bos taurus and Bos indicus cattle, most of the data derive from studies performed in temperate climate countries, under environmental and nutritional conditions very different than those found in tropical countries. Moreover, the physiological basis for understanding the differences between Bos taurus and Bos indicus estrous cycles are still unknown. This review explores the physiological and metabolic bases for understanding the key differences between the Bos taurus and Bos indicus estrous cycle. Moreover, it presents recent results of studies that have directly compared reproductive variables between Zebu and European cattle. Review: The knowledge of reproductive physiology, especially the differences between Bos taurus and Bos indicus, is important for the development and application of different techniques of reproductive management in cattle. In this regard, overall, Bos indicus have a greater number of small ovarian follicles and ovulatory follicles are smaller as compared to Bos taurus. Consequently, Zebu cattle also have smaller corpus luteum (CL). Nevertheless, circulating concentrations of steroid and metabolic hormones are not necessarily higher in European cattle. In fact, some studies have shown that despite ovulating smaller follicles and having smaller CL, Bos indicus cows or heifers have higher circulating concentrations of estradiol, progesterone, insulin and IGF-I compared to Bos taurus females. In addition, there are also substantial differences between Bos indicus and Bos taurus cattle in relation to follicle size at the time of selection of the dominant follicle. Conclusion: Data from very recent studies performed in Brazil have corroborated results from previous reports that have observed substantial differences in the estrous cycle variables of Bos indicus versus Bos taurus cattle. Those differences are probably related to distinct metabolism and metabolic hormone concentrations between Zebu and European cattle. This increased knowledge will allow for the establishment of more adequate reproductive management protocols in both breeds of cattle.

By remote sensing, geodatabase digital processing, information and expeditions to Capivara's sub-basin, was possible to identify the changes in the landscape mainly the expansion of eucalyptus, sugar-cane, and orange trees, being the last two, mainly on the Periferic Depression of Basaltic Cuesta. A significant portion of the soil on this geological area is formed from sandstones, providing high permeability to them, making them important places to groundwater recharge areas as sensible to contamination by pesticides. Throughout last decade was observed that the native vegetation fragments stabilization, keeping a reason of 26.5% on the land used between 2000 and 2010. The pasture decrease being substituted by eucalyptus, sugar-cane and orange trees call attention for the changes in the agribusiness model demanded by the current economic and social necessity. Pasture decrease followed by erosions decrease on sub-basin is evidence that these two aspects are strongly related.

Studying the physical environment of a watershed is the basic condition for a successful planning of the riparian forest preservation, and for water production and conservation. The aims of the present study were to analyze and quantify the spatial and temporal evolution (1984 and 2010) using Landsat-5 satellite images of Cintra Stream sub-watershed, Botucatu, São Paulo State, Brazil, processed by the software IDRISI Andes, as well as to analyze the water quality through the parameters pH, EC, DO and BOD5 at 4 different sites in the years 1999, 2008 and 2009. Considering the 1076.48ha area of the sub-watershed, the pasture class of 1984 was reduced by 25.55% in 2010, resulting in an increase in the remaining classes. The most important class was native forest and reforestation since it had an increase of 5.08%, which indicates recovery of the riparian forest. Degraded areas were identified close to the inferior limit of the sub-watershed (P3 and P4), as well as local contamination (P1 and P2) with worsening of the water quality in the remaining sites in the periods 2008 and 2009. Recovery and management of the ecological succession of degraded areas and water quality monitoring at 1 and 2 sites will be necessary to reestablish the natural condition of the area studied.

The experiment was conducted to evaluate the effect of supplementing glutamine and nucleotides on growth performance and development intestinal morphology in broiler chicks. In the trial, 600 male broiler chicks distributed in randomized blocks in a 3x2 factorial arrangement (consisting of a uniform basal diet supplemented with: 0.0, 0.5 or 1.0% glutamine, and 0.0 or 0.04% nucleotides), for a total of 6 treatments with 25 birds each. Means of performance (weight, weight gain, feed intake, feed conversion and mortality) were obtained 7, 21 and 42 days of age. At the end of the experiment 4 broilers per treatment were used to determine organs weight and development intestinal morphology. Glutamine supplementation (1%) improved the body weight, feed intake and feed conversion in the first week. Glutamine and nucleotides supplementation did not affect performance in broiler chicks in the period one to 21 and one to 42 days of age. In conclusion, it was demonstrated that feeding 1.0% glutamine improved growth performance of broiler birds at 21 days of age.

This study evaluated using of Multicriteria Evaluation in a GIS, specifically by Weighted Linear Combination Method for generation of map of priority areas for forest restoration in the initial part of River Pardo Basin, SP, in order to water resources conservation. Aiming to define criteria and restrictions it was used Participatory Techniques, and the following factors had been selected: proximity of the hydrographic network, proximity of forest cover, slope and erodibility of soil. To calculate the weight to each factor it was used the decision-making process, known as Analytic Hierarchy Analysis, this method consists of a paired comparison of factors to determine the relative importance of each. According to Weighted Linear Combination, the very high priority areas have a more limited spatial distribution, with an apparent concentration around the water bodies, outlining a buffer to the river system. The proximity factor of the hydrographic network, and enables the connection forestry, contributed, along with the factor of proximity to forest cover, so there would be the definition of most of the areas with the highest priority in the basin, which concentrate the largest areas of forest and native riparian areas along the hydrographic.

Development within the cleidoic egg of birds and reptiles presents the embryo with the problem of accumulation of wastes from nitrogen metabolism. Ammonia derived from protein catabolism is converted into the less toxic product urea or relatively insoluble uric acid. The pattern of nitrogen excretion of the green iguana, Iguana iguana, was determined during embryonic development using samples from allantoic fluid and from the whole homogenized egg, and in hatchlings and adults using samples of blood plasma. Urea was the major excretory product over the course of embryonic development. It was found in higher concentrations in the allantoic sac, suggesting that there is a mechanism present on the allantoic membrane enabling the concentration of urea. The newly hatched iguana still produced urea while adults produced uric acid. The time course of this shift in the type of nitrogen waste was not determined but the change is likely to be related to the water relations associated with the terrestrial habit of the adult. The green iguana produces parchment-shelled eggs that double in mass during incubation due to water absorption; the eggs also accumulate 0.02. mM of urea, representing 82% of the total measured nitrogenous residues that accumulate inside the allantois. The increase in egg mass and urea concentration became significant after 55. days of incubation then were unchanged until hatching. © 2012 Elsevier Inc.
