Reintroduction can be enhanced by data from long-term post-release monitoring, which allows for modeling opportunities such as population viability analysis (PVA). PVA-relevant data were gathered via long-term monitoring of reintroduced red-billed curassows at the Guapiacu Ecological Reserve (REGUA), located in Rio de Janeiro, Brazil, over 25 months. In the present article, we (1) assess the robustness of the reintroduction plan, (2) evaluate the viability of the current reintroduced population, and (3) examine mitigation options to increase the viability of this population. VORTEX indicates that the initial plan, fully implemented, was likely to establish a viable population at REGUA. The current population is unviable; the best mitigation strategies are to eliminate hunting altogether, or at least reduce it by half, and to supplement ten immature pairs in 2015. A positive long-term outcome at REGUA is still possible; we encourage the Brazilian government and private stakeholders to consider population supplementation, both to achieve success at REGUA and to improve the evidence base for future reintroductions. According to the Convention on International Trade in Endangered Species, 36 wild feline species are threatened by extinction or severely endangered, and to save them is the target of several conservation programs. This study aimed to assess the viability of the freeze-drying technique for domestic cat sperm cells, with the ultimate goal of transferring this technology to the wild feline species. The domestic cat is an excellent experimental model for wild felids. It is in this scenario that the freeze-drying process (low-temperature vacuum dehydration) of sperm cells shows its value in preserving male cats' germplasm. Results from membrane and DNA integrity analysis are promising and validates the use of frozen-dried sperm samples in intracytoplasmic sperm injections (ICSIs). Further studies are still necessary to evaluate the ICSI embryo production using domestic cat frozen-dried sperm and the possibility of using such technology with wild felines.

Maintaining the viability of probiotic microorganisms from production to consumption has long been a technological challenge for the food industry. The objectives of this study were to evaluate the in vitro interaction between Lactobacillus acidophilus La-5 and Williopsis saturnus var. suaveolens and the effect of this yeast on acidification kinetics, viability of Lactobacillus acidophilus and post-acidification in fermented milk during refrigerated storage at 5 °C. The in vitro study showed a positive interaction between the acid cell free-supernatant (CFS) of probiotic bacteria La-5 and the yeast. The addition of W. saturnus var. suaveolens increased the fermentation time due to consumption of the organic acids produced by L. acidophilus. During the refrigerated storage of the samples, the presence of the yeast increased the viability of L. acidophilus and reduced post-acidification. However, the mechanism of such interaction of bacteria and yeast is not fully understood.

Mouse embryo production by superstimulation is a multifactorial process. To minimize the number of sacrificed animals and to maximize the results of the superstimulatory treatment, it should be possible to predict the risk of do not get embryos from such a treated animal. This work aimed to evaluate if the variables - hormone concentration and the timing of its administration, the copulatory plug presence and individual male used to mating – could be predictive factors on the mouse embryo production. Females were distributed in four groups (cross-over design) related to scheduled superstimulation treatment (1300h or 1700h) and eCG/hCG administered concentration (5 or 10IU). After the hCG treatment, females were put to mate. On the next morning it was verified the presence of a copulatory plug (D0.5). Embryo recovery was performed from D2.5 to D4.5 by flushing the oviducts and uterine horns. Total structures recovered (TSR) and the viable embryos (VE) were classified by its morphology. Viability rate (VR) was calculated with VE in relation to TSR (x100). Group comparison was analyzed with 5% of significance. There were no significant differences among groups, even when only main effects were analyzed (hormone concentration and timing of its administration). There was significant difference in VR from animals with or without plug and from the worst and best males used. It was concluded that neither the hormone concentration nor timing of its administration - or their association – was significant as predictive factors for the embryo production. However, the plug presence was related to higher VR.

Contents Fibroblast growth factor (FGF10) acts at the cumulus oocyte complex, increasing the expression of cumulus cell expansion-related genes and oocyte competency genes. We tested the hypothesis that addition of FGF10 to the maturation medium improves oocyte maturation, decreases the percentage of apoptotic oocytes and increases development to the blastocyst stage while increasing the relative abundance of developmentally important genes (COX2, CDX2 and PLAC8). In all experiments, oocytes were matured for 22h in TCM-199 supplemented with 0, 2.5, 10 or 50ng/ml FGF10. In Experiment 1, after maturation, oocytes were stained with Hoechst to evaluate meiosis progression (metaphase I, intermediary phases and extrusion of the first polar body) and submitted to the TUNEL assay to evaluate apoptosis. In Experiment 2, oocytes were fertilized and cultured to the blastocyst stage. Blastocysts were frozen for analysis of COX2, CDX2 and PLAC8 relative abundance. In Experiment 1, 2.5ng/ml FGF10 increased (p<0.05) the percentage of oocytes with extrusion of the first polar body (35%) compared to 0, 10 and 50ng/ml FGF10 (21, 14 and 12%, respectively) and FGF10 decreased the percentage of oocytes that were TUNEL positive in all doses studied. In Experiment 2, there was no difference in the percentage of oocytes becoming blastocysts between treatments and control. Real-time RT-PCR showed a tendency of 50ng/ml FGF10 to increase the relative abundance of COX2 and PLAC8 and of 10ng/ml FGF10 to increase CDX2. In conclusion, the addition of FGF10 to the oocyte maturation medium improves oocyte maturation in vitro, decreases the percentage of apoptotic oocytes and tends to increase the relative abundance of developmentally important genes.

The aim of the present study was to evaluate the effect of bovine somatotropin (bST; 500 mg) administration on lactating buffalo donors submitted to two different ovum pick-up (OPU) and in vitro embryo production schemes with a 7 or 14 d intersession OPU interval. A total of 16 lactating buffalo cows were randomly assigned into one of four experimental groups according to the bST treatment (bST or No-bST) and the OPU intersession interval (7 or 14 d) in a 2 x 2 factorial design (16 weeks of OPU sessions). The females submitted to OPU every 14d had a larger (P < 0.001) number of ovarian follicles suitable for puncture (15.6 +/- 0.7 vs. 12.8 +/- 0.4) and an increased (P = 0.004) number of cumulus-oocyte complexes (COCs) recovered (10.0 +/- 0.5 vs. 8.5 +/- 0.3) compared to the 7 d interval group. However, a 7 or 14 d interval between OPU sessions had no effect (P = 0.34) on the number of blastocysts produced per OPU (1.0 +/- 0.1 vs. 13 +/- 0.2, respectively). In addition, bST treatment increased (P < 0.001) the number of ovarian follicles suitable for puncture (15.3 +/- 0.5 vs. 12.1 +/- 0.4) but reduced the percentage (18.9% vs. 10.9%; P = 0.009) and the number (1.4 +/- 0.2 vs. 0.8 +/- 0.1; P = 0.003) of blastocysts produced per OPU session compared with the non-bST-treated buffaloes. In conclusion, the 14d interval between OPU sessions and bST treatment efficiently increased the number of ovarian follicles suitable for puncture. However, the OPU session interval had no effect on embryo production, and bST treatment reduced the in vitro blastocyst outcomes in lactating buffalo donors.

In conservation agriculture, mainly under direct seeding, maintaining the vegetation ground cover is essential, since this serves as a reservoir of nutrients which are slowly released to plants by microorganisms. Some authors have sought to study increases in the amount of straw in the soil, in addition to slowing down the process of decomposition, with hormesis being one of the techniques used. This technique states that all chemical substances are both poisonous and nonpoisonous, with only the dosage determining whether they are lethal or not. This study aimed to evaluate the dry weight and agronomic characteristics of a crop of black oat subjected to hormesis. The experimental design was of randomised blocks, with 12 treatments and 4 replications, giving a total of 48 experimental lots. The treatments were: Haloxyfop-R Methyl Ester at dosages of 0.625, 1.25 and 2.50 g ha(-1); Glyphosate at dosages of 12.50, 25.00 and 50.00 g ha-1; 2,4-D dimethylamine salt at dosages of 100.00, 200.00 and 300.00 g ha(-1); Alterbane at a dosage of 500.00 g ha(-1); Salicylic acid at a dose of 100 g ha(-1); and a control. It was concluded that for the subdosages under test, the herbicides 2,4-D at medium dosage and Verdict at low dosage were shown to be the best treatments for conserving straw as ground cover under direct seeding.
