Com o intuito de se estudar a presença de suínos portadores renais de leptospiras foram colhidas 131 amostras sanguíneas e os respectivos rins de animais durante o abate em abatedouro da região de Botucatu-SP. Pela prova de Soroaglutinação Microscópica obteve-se 48 amostras sorológicas positivas para um ou mais sorovar de Leptospira spp., com uma taxa de ocorrência de anticorpos anti-leptospira de 36,64%, e maior importância para o sorovar icterohaemorrhagiae. Para a pesquisa do agente nos rins, das 88 amostras renais submetidas a cultura em meio de EMJH e analisadas pela prova de PCR, foi isolado e detectado o agente em uma única amostra renal, pertencente a um animal soropositivo. Embora não tenha sido possível a comparação estatística, em termos de sensibilidade e especificidade das duas provas de detecção do agente a partir de amostras renais, a PCR mostrou-se mais rápida e prática na pesquisa de portadores renais. Pelo isolamento obtido, ressalta-se a importância desses animais como possíveis transmissores da doença para trabalhadores de abatedouro e inspetores de carne.

Três búfalos e três bovinos zebuínos adultos com cânulas ruminais foram alimentados à vontade com dieta de cana-de-açúcar fresca e picada, suplementada com 3 kg de concentrado/animal, durante 13 semanas. Após duas semanas de adaptação, amostras do conteúdo ruminal foram coletadas semanalmente, em dois tempos de amostragem: antes da alimentação e 40 minutos após para determinação da concentração e composição da fauna ruminal. Na quinta semana do experimento, os conteúdos ruminais foram misturados e reinoculados entre animais de mesma espécie e, na última semana, estimadas as degradabilidades in situ da MS, PB e FDN da cana-de-açúcar e MS e PB do concentrado. A concentração média de protozoários foi maior em bovinos, de 4,85 x 10(5)/mL, que em búfalos, de 3,82 x 10(5)/mL. As composições genéricas para Entodinium e subfamília Diplodiniinae foram 79,2 e 6,2% em bovinos e 32,0 e 54,9% em búfalos, respectivamente. Não houve interação significativa entre espécies animais e tempo de amostragem. Houve aumento da concentração média dos holotricos após 40 minutos da alimentação. As degradabilidades efetivas dos nutrientes estudados foram semelhantes entre as duas espécies animais, observando-se diferenças na cinética da degradação. Concluiu-se que os búfalos apresentaram menor concentração de ciliados no rúmen, maior composição de Diplodiniinae e menor de Entodinium que os bovinos; os holotricos exibiram capacidade de migração e seqüestro no rúmen em ambas as espécies; e as diferenças na fauna ruminal não influenciaram a degradabilidade efetiva dos nutrientes da cana-de-açúcar e do concentrado.

Quatro bubalinos e quatro bovinos, fistulados no rúmen, foram distribuídos aleatoriamente a dois quadrados latinos (4 x 4) para avaliar os efeitos de rações com quatro níveis de FDN (54, 60, 66 e 72%) e de dois tamanhos de poros em sacos de náilon (53 e 100 mm) sobre a população de protozoários ciliados no conteúdo ruminal e dentro dos sacos incubados em diversos tempos. Não houve diferença nos diversos gêneros estudados e no total de protozoários nas amostras dos sacos de náilon entre 53 e 100 mm. As concentrações de Entodinium e de total de protozoários do conteúdo ruminal elevaram-se até o nível de 66% de FDN, com queda acentuada em 72% nos bubalinos, enquanto nos bovinos houve redução com o aumento de FDN na ração. Houve diferenças nas concentrações dos protozoários da subfamília Diplodiniinae, Epidinium e Dasytricha entre bubalinos e bovinos, dentro das amostras incubadas em sacos com poros de 53 mm. A concentração de Diplodiniinae aumentou com 72% de FDN na dieta em ambas as espécies animais. Os bubalinos apresentaram valores médios de pH e taxa de passagem do líquido ruminal mais elevados e volumes ruminais menores que os bovinos.

The water buffalo (Bubalus bubalis) is an important natural host for Neospora caninum. Serologic responses to N. caninum were studied in experimentally and naturally infected water buffaloes in Brazil. Antibodies were assayed by the indirect fluorescent antibody test (IFAT) using a cut off value of 1:25. Six buffaloes were each inoculated subcutaneously with 5 x 106 live culture-derived tachyzoites of the cattle Illinois strain of N. caninum, and two calves were kept as uninoculated controls. Post-inoculation (p.i.) blood samples were collected weekly for 8 weeks and then monthly until 1 year p.i. All inoculated buffaloes developed IFAT titers of 1:100 or more between 7 and 11 days p.i. and the titers remained elevated until 7 weeks p.i. Antibody titers peaked to 1:1600 in 1, 1:800 in 3 and 1:400 in 2, usually by 3 weeks p.i. Antibody titers declined to 1:25 or 1:50 in all the six buffaloes by 12 months p.i. IFAT titers to N. caninum remained at an undetectable level (< 1:25) in both control uninoculated buffaloes. To follow the dynamics of N. caninum antibodies, sera from 29 buffaloes and their calves were collected for 1 year and assayed for N. caninum antibodies; 23 of 29 calves were seropositive (IFAT of 1:100 or more) at 1-2 day of age. of these 23 calves, 17 remained seropositive during the study, while six became seronegative at four (two calves), six (one calf) seven (two calves) and eight (one calf) months of age. These findings suggest a high rate of neonatal transmission of N. caninum in buffaloes. Published by Elsevier B.V.

Quatro búfalos com cânulas ruminais foram distribuídos aleatoriamente em quadrado latino 4 x 4, compreendendo quatro rações com feno de capim-coastcross (Cynodon dactylon) (50% MS) e níveis crescentes de polpa cítrica peletizada em substituição ao milho em grão moído no concentrado (0, 33, 66 e 100% MS). Foram avaliadas a degradabilidade dos nutrientes e a população de protozoários no rúmen. Houve efeito quadrático da fração solúvel dos nutrientes entre os tratamentos, com valores maiores nos dois extremos. A taxa de degradação da PB do feno e da MS da polpa cítrica apresentou-se linear com aumento da polpa cítrica na ração. As rações contendo polpa cítrica promoveram menores concentrações de Entodinium e número total de ciliados/mL que a dieta só com milho. Houve predominância dos Entodinium nas dietas contendo somente milho ou somente polpa cítrica, enquanto em mistura predominaram os ciliados da subfamília Diplodiniinae. Observou-se efeito de interação da polpa cítrica com o milho, promovendo modificações no metabolismo ruminal, especialmente na solubilidade dos nutrientes, na curva do pH e na composição da fauna.

Toxocara vitulorum, a parasite of the small intestine of cattle and water buffaloes, is mainly acquired by calves via the colostrum/milk from infected cows. To understand the development of immune responses in calves, antibody levels to a soluble extract antigen (Ex) from T. vitulorum infective larvae were measured by an indirect ELISA with sera of 15 buffalo calves, which were sampled every 15 days for the first 180 days after birth and 9 buffalo cows during the perinatal period. From all serum samples examined during the first 180 days, antibody level was lowest and highest in calves at 1 day of age before and after suckling colostrum, respectively, suggesting that the origin of antibodies was the colostrum. Immediately after birth, antibody levels in suckled calves remained at high levels until day 15, began to decrease to lower levels between 15 and 30 days and remained relatively stable until 120 days. By comparing the immune responses of these animals with their parasitological status it was considered possible to determine if passively acquired or actively produced antibodies provided protection against the infection. High numbers of T. vitulorum eggs in the feces between 30 and 60 days indicated that passively acquired antibodies did not provide protection against the infection, at least during these first days, and the maximum fecal egg counts during 30-45 days were coincident with decreased antibody levels. Between 60 and 120 days, when serum antibodies were detected at reduced, but stable levels, adult nematodes were expelled from the intestines and no more T. vitulorum eggs were found, suggesting development of acquired resistance. However, the potential and functional protective role of the antibodies against T. vitulorum infection and the process of self-cure requires further investigation. (C) 2001 Elsevier B.V. B.V. Genetic parameters for the relation between the traits of milk yield (MY), age at first calving (AFC) and interval between first and second calving (IBFSC) were estimated in milk buffaloes of the Murrah breed. In the study, data of 1578 buffaloes at first lactation, with calvings from 1974 to 2006 were analyzed. The MTDFREML system was used in the analyses with models for the MY, IBFSC traits which included the fixed effects of herd-year-season of calving, linear and quadratic terms of calving age as covariate and the random animal effects and error. The model for AFC consisted of the herd-year-season fixed effects of calving and the random effects of animal and error. Heritability estimates MY, AFC and IBFSC traits were 0.20, 0.07 and 0.14, respectively. Genetic and phenotypic correlations between the traits were: MY and AFC = -0.12 and -0.15, MY and IBFSC = 0.07 and 0.30, AFC and IBFSC = 0.35 and 0.37, respectively. Genetic correlation between MY and AFC traits showed desirable negative association, suggesting that the daughters of the bulls with high breeding value for MY could be physiological maturity to a precocious age. Genetic correlation between MY and IBFSC showed that the selection of the animals that increased milk yield is also those that tend to intervals of bigger calving.

The objectives this paper were to estimate genetic parameters and genetic and phenotypic trends of birth weight (BWT) and weights adjusted to 205 (WT205), 365 (WT365) and 550 (P550) days of age of beef buffaloes born from 1985 to 2003 in Brazil. For BWT and WT205 the model included direct and maternal genetic and maternal environment as random effects and contemporary and genetic groups as fixed effects. For WT365 and WT550 the same model was used except without direct maternal and maternal environmental effects. The genetic and phenotypic trends were estimated by regression of means of dependent variables on birth year of animals Regressions were obtained by using two methodologies: 1) linear regression; and 2) non-parametric splined regression. The direct heritability estimates were 0.09, 0.45, 0.46 and 0.58 for BWT, WT205, WT365 and WT550, respectively. The direct genetic trends from linear regression were 0.01, 0.23, 0.58 and 1.40 kg per year for PN, WT205, VVT365 and WT550, respectively (P<0.001 for all). Phenotypic trends were strongly positive while genetic trends were consistently positive but small. Genetic parameters indicate potential for increased rate of genetic change with full implementation of genetic improvement programs.

In view of the importance of venereal transmission of bovine leptospirosis, the objective of the present study was to apply the polymerase chain reaction (PCR) to 26 serovars of Leptospira interrogans, L. borgpetersenii, L. santarosai, L. noguchii and L. biflexa, to determine the detection threshold in semen samples and to evaluate the possibility of differentiation among serovars using 19 restriction endonucleases. The results showed that all serovars were amplified and the detection threshold in semen samples of a bull was 100 bacteria/ml. Using endonucleases we could classify the 26 serovars into eight groups. The present results show that PCR is a method of great potential for the detection of Leptospira spp, at bovine artificial insemination centers. (C) 2000 Elsevier B.V. B.V. All rights reserved.

Genetic and environmental effects on parturition interval (PI) and duration of lactation (DL) were evaluated in 107 Jafarabadi, 98 Mediterranean, 1027 Murrah and 624 crossbred buffalo females (n=1856), based on data from the Buffalo Genetic Improvement Program-PROMEBUL from 1980 to 2003. The statistical model included effects of herd, parturition year and month, calf's sex, parturition order and genetic group, composing 11, 34, 12, 2, 12 and 4 classes, respectively. A significant effect over PI was observed (P<0.01) in all classes, excepting sex. Mean parturition intervals per genetic group presented significant differences through SNK test (P<0.05), with mean values of 451.29; 429.47; 406.97 and 389.78 days in Mediterranean, crossbred, Murrah and Jafarabadi, respectively. Mean DL values were 276.68; 270.33; 258.03 and 235.59 days for Mediterranean, Murrah, crossbred and Jafarabadi groups, respectively. No significant differences in DL were observed in relation to genetic groups. However, herd and parturition order, year and month significantly influenced DL (P<0.01). The herd was the main source of variation over DL, followed by parturition year and month. A regression based on parturition month in relation to PI and DL showed that females giving birth in the last months of the year presented higher PI and DL.

This research aimed to report the occurrence of subclinical mastitis in a buffalo from a study carried out with 548 milk samples of 137 Murrah and Mediterranean buffalos from seven milk properties, located in the cities of Jau, Botucatu and Sorocaba, State of São Paulo. The animals of the study were submitted to a clinical examination of the mammary gland by the inspection and to the diagnosis of clinical and subclinical mastitis by California Mastitis Test (CMT), being the milk samples later directed to the laboratory for microbiological studies and also to the test of Whiteside Modified (WSM). The isolated agents were identified by the morphological characteristics of its colonies and through microcultive staining with blue cotton. Two (02) pure fungi samples were isolated representing 2.86% of the total isolated microorganism, corresponding to two mammary rooms in one animal, and were classified as pertaining to the Aspergillus fumigatus specie. The animal in question showed reaction of ++ to the CMT in both affected rooms and negative reaction to the WSM. In this way it is concluded that the Aspergillus fumigatus participates in a discrete way as a determinant agent of bubaline subclinical mastitis, however it is important because the affected animals can act as potential reservoirs and may be able to generate the infection in human beings.

Buffalo meat production has been arising interest breeder and emerges as alternative to consumer, more and more demanding of the quality products. Thus, this research was conducted to study the chemical composition and tenderness of Longissimus dorsi muscle from 10 non-castrated Murrah buffaloes slaughtered at different weights. The research was carried in feedlot of School of Veterinary Medicine and Animal Science of the São Paulo State University, Botucatu, São Paulo, Brazil. Animals were divided in two groups, received diet ad libitum and slaughtered when reached 450 and 500 kg of live weight. The experiment design was completely randomized, with five repetitions to each treatment. Meat sample from Longissimus dorsi muscle, taken between 12(th) and 13(th) ribs, were carried analysis of moisture, crude protein, fat, ash, Longissimus muscle area (LMA), fat thickness (FT), marbling, calorie and tenderness. It did not have significant difference between the groups. Average values at of 76.0; 20.7; 2.1 and 1.2 of moisture, crude protein, fat and ash respectively, were obtained. Calorie, tenderness, LMA, FT and marbling were obtained at average values of 132 kca1/100g; 3.94 kgf; 34.2 cm(2); 5.9 mm and 2 points, respectively. Values obtained for tenderness are similar in the literature and has been proving that buffalo meat is tender (< 5kgf). Positive correlation was observed between the protein percentage and the shear force of the meat. The buffalo meat is excellent alternative source of red protein of high biological value to feeding of Brazilian consumers.

Newborn Holstein-Friesian and Nelore cattle and Murrah buffalo blood samples were collected for hemograma and serum constituents testing. Results indicated that: 1. buffalo red blood cell (RBC) counts were lower than RBC counts for cattle during the first 30 days after birth; 2. buffalo leukocytes, neutrophil and lymphocyte counts increased in older animals; 3. The highest fibrinogen levels occurred in older calves; 4. buffalo glucose levels were lower than Nelore cattle and decreased in the older calves; 5. buffalo iron, urea, and aspartate aminotransferase levels were higher than in cattle with the highest concentration at the time of birth; 6. cattle bilirubin and alanine aminotranferase levels were higher than buffalo with the highest value at the time of birth; 7. alkaline phosphatase and gammaglutamyltransferase activities were lower in the older calves; 8. buffalo magnesium levels were higher than cattle with lower concentrations at the time of birth..
